Abstract: Objective To observe the therapeutical effect of kangnao liquid in rats with focal cerebral ischemia-reperfusion(I/R) and to investigate the expression of Pi3kmRNA and AktmRNA in rats.Methods 180 healthy male SD rats were randomly divided into 6 groups:sham operated group,model group,The high-,middle- and low-dose kangnao liquid groups and Nimodipine group.Rats in drug treatment groups were administrated with kangnao liquid in accordance with 24g.kg-1.d-1 and 12g.kg-1.d-1 and 6g.kg-1.d-1 and with Nimodipine in accordance with 1mg.kg-1.d-1.Rats in model and sham groups were treated with distilled water with the equal volume.Continuously administrated for 7 days, the animal models of middle cerebral artery occlusion (MCAO) were established by a monofilament method from right internal carotid artery.Neurological evaluation was performed 24h after reperfusion.The In situ hybridization was used to investigate the expression of Pi3kmRNA and AktmRNA in rats on 12h, 24h, 48h, 72h and 168h after ischemia for 2h.Results Compared with model group,kangnao liquid groups had significantly improved neurological responses.The expression of Pi3kmRNA and AktmRNA at different time points of brain tissue were significantly improved.Nimodipine group had also improved neurological responses and the expression of Pi3kmRNA and AktmRNA,but it was lower than high- and middle-dose kangnao liquid groups.Conclusions Kangnao liquid can treated the rats with cerebral ischemia-reprefusion injury and protect nerve cells of cerebral infarction areas.The protective effect of kangnao liquid may be associated with that kangnao liquid can incresse the expression of Pi3kmRNA and AktmRNA.
