钴铬合金与钛合金修复材料的细胞相容性研究

摘要: 摘要: 目的 研究 2 种牙科金属材料钴铬合金、 钛合金浸提液对人牙龈成纤维(HGF)细胞的细胞相容性。方法 以钴铬合金、 钛合金的浸提液培养 HGF 细胞, 以含 10%胎牛血清的 DMEM 培养基作为空白对照。采用 MTT 法在细胞水平评价 2 种牙科金属材料的细胞毒性, 使用试剂盒检测细胞中还原型谷胱甘肽 (rGSH) 的含量, 采用 ELISA 法检测 2 种牙科金属材料浸提液对 HGF 细胞肿瘤坏死因子 (TNF) -α分泌的影响, 采用实时 PCR(real-time PCR) 在基因水平检测 2 种牙科金属材料浸提液对 HGF 细胞凋亡相关基因 caspase-3 mRNA 表达的影响。结果 钴铬合金组抑制了 HGF 细胞增殖, 与空白对照组差异有统计学意义, 钛合金组促进了 HGF 细胞增殖。与空白对照相比, 钴铬合金组抑制了 HGF 细胞 rGSH 的产生 (P < 0.05), 促进了 HGF 细胞 TNF-α的分泌 (P < 0.05), 而钛合金对 HGF 细胞 rGSH 产生、 TNF-α分泌没有明显影响; 钴铬合金组 caspase-3 的基因表达高于空白对照组(P < 0.05), 而钛合金对 caspase-3 基因表达无明显影响。结论 钴铬合金对 HGF 细胞具有一定的细胞毒性, 而钛合金细胞相容性良好。

Abstract: Abstract: Objective To investigate the cytocompatibility of the liching liquids of Co-Cr alloy and Ti alloy on human gingival fibroblasts (HGF). Methods The HGF were treated in vitro with leaching liquids of Co-Cr alloy and Ti alloy, re⁃ spectively. The DMEM cell medium containing 10% fetal bovine serum was served as a negative control. The viability of HGF treated by two dental alloys were evaluated by means of MTT, and the contents of intracellular reduced glutathione (rGSH) were assayed by kits. The tumor necrosis factor-α (TNF-α) contents were determined in the culture supernatant by ELISA in two groups. The effects of these alloys on the expression of caspase-3 were examined by real time-PCR method. Results Compared with the control group, HGF treated with Co-Cr alloy leaching liquids showed a lower viability (P < 0.05), while Ti alloy leaching liquid promoted the proliferation of HGF. In Co-Cr alloy group, the rGSH content was signifi⁃ cantly decreased (P < 0.05), while TNF-α content was significantly increased (P < 0.05) compared with control group. There were no significant differences in rGSH and TNF-α contents between the Ti alloy group and control group (P > 0.05). The ex⁃ pression of caspase-3 was significantly higher in Co-Cr alloy group than that of control group (P < 0.05), while there was no significant difference in the expression of caspase-3 between Ti alloy group and control group. Conclusion Results suggest that Co-Cr alloy possesses cytotoxicity, while there is better cell compatibility for Ti alloy.
