猪小肠粘膜下层作为组织工程肌腱支架材料的实验研究

摘要: 摘要 目的:探讨猪小肠粘膜下层(small intestinal submucosa,SIS)作为组织工程肌腱支架的可行性。方法:将肌腱细胞接种到经过处理的SIS卷轴上,形成细胞-支架复合物。24只罗曼鸡被随机分为两组,实验组为细胞-SIS复合物,对照组为自体移植肌腱。术后3、6、9周取材并进行大体形态、组织形态学、体式显微镜观察和生物力学测定。结果:实验组术后局部反应小,伤口一期愈合。组织学检查显示,3周时胶原纤维数量相对少,排列紊乱,第6周时细胞数量增多,形态也略显规则,第9周时纤维数量多且排列规则。生物力学测定证实9周后两组植入物抗拉力强度在统计学上无著性差别(P>0.05)。结论:以SIS与肌腱细胞构建的肌腱组织能够达到替代肌腱缺损的要求,SIS可以作为肌腱组织工程的一种理想的支架材料。

Abstract: Abstract Objective:To investigate the feasibility of the small intestinal submucosa as a scaffold for tissue engineering tendon. Methods:The cells were seeded in the SIS. The adhesion of the cell was observed by the somatotype microscope.40 Roman chickens for 12weeks old were divided into two groups randomly, cell-seeded SIS group(experimental group) and autograft group(control group). The chickens were sacrificed at 3, 6, or 9 weeks after surgery. The specimens were observated including general appearance somato- typemicroscope, histologic pattern, and mechanical test. Results:The wound was healed primarily and the reactor was little in the experimental group after surgery. Histology shows few fibers and the confused alinement of fibers at the 3rd week. The increasing amount of fibers was seen at the 6th week, the alinement of fibers become a certain sequence. At the 9th week, a most sums of fibers, spread regularly,can be seen. Mechanical strength in the two groups have not statistical different significantly at the9thweek(P>0.05).Conclusion:Cell-seeded SIS scaffold may be the suitable substitution for the defected tendons, SIS perhaps becomes a ideal scaffold in the tissue engineering tendon.
