dbcAMP对甲状腺鳞癌SW579细胞株增殖的影响

The Effects of dbcAMP on the Proliferation of SW579 Cell Line

摘要: 摘要 目的:探讨dbcAMP对甲状腺鳞癌SW579细胞株的增殖影响。方法:利用dbcAMP 0.5,1.0,2.0 mmol/L作用于甲状腺鳞癌SW579细胞24 h后用四甲基偶氮唑蓝比色法(MTT法)检测SW579细胞增殖的变化;ELISA法检测经dbcAMP1.0 mmol/L作用24 h后MPF的活性; WesternBlot方法检测经dbcAMP1.0 mmol/L作用24 h cdc2-Tyr15磷酸化水平。流式细胞术检测经dbcAMP1.0 mmol/L作用24 h细胞周期变化情况。结果:MTT法结果显示,在dbcAMP作用下,SW579细胞活力呈逐渐下降趋势,随dbcAMP的剂量的增加和作用时间的延长,其作用越明显,呈剂量时间依赖性。ELISA法显示,dbcAMP使MPF活性下降。对照组MPF活性为 (115.5±7.53) ng/L;实验组MPF活性为 (22.87±2.57) ng/L,两组间比较差异有统计学意义(P<0.01)。WesternBlot显示,dbcAMP上调cdc2-Tyr15磷酸化水平。流式结果显示,实验组G0/G1期细胞(62..50±1.22)%,S期细胞(10.15±0.38)%,G2/M期细胞(26.39±0.76)%;对照组G0/G1期细胞(60.83±1.61)%,S期细胞(29.19±0.97)%,G2/M期细胞(8.75±0.50)%, (G1期P>0. 05,S期P<0. 05, G2期P<0. 05)。结论:dbcAMP能够显著降低甲状腺鳞癌SW579细胞株的活力,使其处于G2/M期阻滞,细胞增殖受到明显抑制,其抑制作用可能与MPF活性下降有关。

Abstract: Abstract Objective :To study the effect of dbcAMP on the Proliferation of SW579 Cell Line.Methods: SW579 cells were treated with dbcAMP (0.5,1.0,2.0 mmol/L) at 24 h. The growth activity of cells was detected by MTT; The activity of MPF through the ELISA; the protein express of the phosphorylation level of cdc2-Tyr15 by Western Blot; the change of cell cycle will be detected by FCM . Results :MTT showed that the growth activity of SW579 cells was reduced ; ELISA showed that dbcAMP could decrease the activity of MPF. The activity of MPF was (115.5±7.53) ng/L in group without dbcAMP ; While the activity of MPF was (22.87±2.57) ng/L in group with dbcAMP . Western blot showed that the content of cdc2-Tyr15 increased. After the treatment with 1.0 mmol/L dbcAMP for 24 h, the percentage of SW579 cell in G0/G1 phase was (62.50±1.22)%, in S phase was (10.15±0.38)%, in G2/M phase was (26.39±0.76)%; otherwhile in the control group, the percentage of SW579 cell in G0/G1 phase was (60.83±1.61)%, in S phase was (29.19±0.97)%, in G2/M phase was (8.75±0.50)%. Conclusion :dbcAMP could significantly inhabit the SW579 cells proliferation and arrest cell cycle in G2/M phase. The mechanism on inhibiting the growth of thyroid cancer maybe related to the decreasing activity of MPF.
