γH2AX对肺癌放射敏感性的预测效果*

摘要:

【摘要】目的 探讨人肺癌细胞受照后γH2AX表达量变化与肺癌细胞放射敏感性之间的关系。方法 选择肺腺癌A549和小细胞肺癌SBC-3细胞株,采用细胞克隆形成实验检测经不同剂量(0、2、4、6、8、10Gy)照射后A549 和SBC-3细胞的克隆形成率,并绘制细胞生存曲线;应用Western blot检测经2Gy放射剂量照射后不同时间点(0 min、30min、1h、2h、6h、12h、24h、36h、48h)的细胞中γH2AX蛋白表达量。分析γH2AX蛋白表达量变化与肿瘤细胞放射敏感性的相关性。结果 小细胞肺癌SBC-3细胞的放射敏感性明显高于肺腺癌A549细胞。两种细胞受照后1h和6h,γH2AX的表达量均与肿瘤细胞的平均致死剂量(D0)相关。结论 γH2AX可以作为肺癌细胞放射敏感性的一个预测指标。

Abstract:

[Abstract] Objective To observe the relationship between expression changes ofγH2AX and the radiosensitivity of lung cancer cells in vitro.Methods The radiosensitivity of lung cancer cell lines A549and SBC-3was measured by clone forming assay. The DSBs damage of lung cancer cell lines A549and SBC-3was determined by Western blot assay.Re? sults The clone forming rates of lung cancer cell lines A549and SBC-3were gradually decreased with the increased radia? tion dose. γH2AX expression was related to the cell radiosensitivity1hour and6hours after radiated. Conclusion The phosphorylated histoneγH2AX is a powerful tool to monitor DNA DSBs and to predict the radiosensitivity in lung cancer ra? diotherapy.
