摘要: 【摘要】 目的 观察金属硫蛋白-1MT-1)在小鼠肾脏各部位的表达及细胞内定位。 方法 选取成年雄性 CD1 小鼠, 剖腹取出肾脏, 应用 Western blot法检测小鼠肾皮质和髓质中 MT-1 蛋白的表达; 应用免疫组织化学和免疫荧光技术观察MT-1 在小鼠肾小管和集合管的定位; 应用免疫荧光双标共聚焦显微扫描技术观察MT-1 在小鼠肾小管和集合管上皮细胞内的亚细胞定位。 结果 MT-1 在小鼠肾脏皮质和髓质均有表达。 在皮质迷路, 主要表达于近曲小管和远曲小管; 在髓放线和髓质, 主要表达于近直小管、远直小管和集合管。 近端小管表达最强, 远端小管次之,集合管最弱。 在近端小管位于上皮细胞游离面刷状缘及胞浆内; 在远端小管位于上皮细胞胞浆内; 在集合管位于上皮细胞基底侧。 亚细胞定位在溶酶体。 结论 MT-1 在小鼠肾脏近端小管、远端小管和集合管的细胞定位不同, 可能决定了其在肾脏各结构中发挥的作用是不同的。

Abstract: Objective   To survey the expression and cellular localization of metallothionein-1 (MT-1) in mouse kidney. Methods   To detect the protein of MT-1 in mouse renal cortex and medulla by Western blot. Immunohistochemistry,double-immunofluorescence and confocal laser scanning microscopy were applied to observe the intracellular location of MT-1 in structures of mouse kidney. Results   The expression of MT-1 was detected in cortex and medulla of mouse kidney. The expression of MT-1was mainly in proximal convoluted tubule and distal convoluted tubule of cortical labyrinth, and proximal straight tubule, distal straight tubule and collecting duct of ray medullary and medulla of kidney. The most strongest expression of MT-1 was in proximal tubule, secondary in distal tubule and last in collecting duct. The intracellular location of MT-1 was in intracytoplasm, specific in lysosome, of proximal tubule and distal tubule; whereas was in brush borde of proximal tubule and basilar part of collecting duct. Conclusions    The differdnt of MT-1 expressed in proximal tubule, distal tubule and collecting duct may determine different roles of it in kidney.
