参麦注射液对肠缺血再灌注肺脂质过氧化损伤的影响

Effects of ShenMai Injection on Lipid Peroxidation in the Lung Induced by Intestinal Ischemia/Reperfusion(I/R)

摘要: 目的:探讨参麦注射液对肠缺血/再灌注肺脂质过氧化损伤的防护作用。方法:SD大鼠24只,随机均分为对照组、模型组、治疗组。对照组只分离出肠系膜上动脉;模型组、治疗组复制大鼠肠缺血/再灌注(I/R)损伤模型;治疗组于缺血前、再灌前、再灌后30min尾静脉推注参麦注射液0.9mL/100g体质量。观察各组外周血白细胞粘附聚集(LAA)、肺组织湿/干比(W/D)、肺系数以及血浆和肺组织超氧化物歧化酶(SOD)、丙二醛(MDA)、髓过氧化物酶(MPO)的变化及参麦注射液对上述指标的影响;并进行肺组织病理学检测。结果:(1)模型组LAA、肺组织W/D、肺系数分别为(26.39±1.35)%、(3.23±0.33)、(1.065±0.163)%,均高于对照组(P <0.01),光镜下肺组织病理损害亦明显重于对照组;治疗组上述指标均明显下降,分别为(17.39±2.07))%、(2.44±0.17)、(0.751±0.064)%,与模型组比较差异显著(P <0.01);(2)模型组血浆和肺组织SOD活性[(177.37±10.84)U/mL、(22.47±2.44)U/mgpro]低于对照组(P <0.01);MPO活性[(0.327±0.040)U/mL、(2.961±0.314)U/g湿片]及MDA含量[(4.61±1.18) nmol/mL、(6.26±1.44)nmol/mgpro]高于对照组(P <0.01),治疗组上述各检测指标均较模型组明显改善,分别为(226.82±15.53)U/mL、(37.64±2.93)U/mgpro、(0.230±0.031)U/mL、(1.948±0.249)U/g湿片、(2.02±0.47) nmol/mL、(3.15±0.65)nmol/mgpro (P<0.01)。结论:参麦注射液对肠I/R后肺损伤具有保护作用,其机制可能与抑制中性粒细胞的聚集,对抗脂质过氧化损伤有关。

Abstract: Abstract Objective: To explore the effects of shenmai injection(SM) on lipid peroxidiation in the lung induced by intestinal ischemia/reperfusion(I/R). Methods: 24 SD rats were randomly divided into three groups:sham group,I/R group,treatment group. In sham group ,superior mesenteric artery was separated only;Models of intestinal I/R injury in I/R group and treatment group were estabished. In treatment group, SM(0.9mL/100g) was injected through tail vein before ischemia, before reperfusion and after reperfusion 30min respectively. The state of leukocyte adhesiveness/aggregation(LAA) in peripheral blood, lung wet/dry weight ratio,lung coefficient, Superoxide dismutase(SOD), malondialdehyde(MDA) , myeloperoxidase (MPO) in serum and lung tissue were measured, as well as the effects of SM were observed. The changes of lung histology were examined by light microscope. Results: (1)In I/R group ,LAA in peripheral blood, lung wet/dry weight ratio,lung coefficient were significantly increased than that in sham group(P<0.01); the damage to lung tissue was increased in severity with light microscope. The above-mentioned indexes of treatment group were obviously lower in comparison with I/R group(P<0.01).(2)The activity of SOD in serum and lung tissue were significantly lower in I/R group than that in sham group (P<0.01); The content of MDA , the activity of MPO in serum and lung tissue were significantly increased in I/R group than that in sham group (P<0.01).The above-mentioned indexes of treatment group were obviously ameliorated in comparison with I/R group(P<0.01). Conclusion: Shenmai injection can protect the lung injury induced by intestinal ischemia/reperfusion(I/R) ,which may be mediated by inhibiting lipid peroxidation and aggregation of neutrophils.
