大剂量放疗后豚鼠内耳螺旋神经节细胞Caspase3的表达

Caspase-3 Expression in spiral Ganglion Cells in Inner Ear in Guinea Pigs After Large dose 60Co Irradiation

摘要: 目的:探讨一次性大剂量放疗后内耳螺旋神经节细胞(sprial ganglion cell,SGC)的损伤。方法:白化豚鼠40只,随机分为5组,每组8只,右耳为照射耳,左耳为对照耳。豚鼠一侧耳颞部(右耳)做一次性60Co照射70Gy。于放疗后1d,4d,7d,14d,30d处死,做耳蜗中轴切片HE染色及caspase3免疫组织化学染色。结果:HE染色显示放疗后各不同时间点螺旋神经节细胞萎缩率与对照耳比较有显著性差异(P<0.01),放疗后不同时间点螺旋神经节细胞Caspase3的光密度值与对照耳比较均有显著性改变(P<0.01)。结论:放疗可引起内耳SGC的损伤,Caspase3在SGC的表达上调可能与细胞损伤有关。

Abstract: Objective To explore the damage of sprial ganglion cell inner ear after 60Co irradiation. Methods Forty guinea pigs were randomly divided into 5 groups.The right ear was radiation ear,the left ear was controll ear.The guinea pigs were sacrificed in 1、4、7、14 and 30 days after irradiation,All specimens were dehydrated,embeded in paraffin and serially cutted into 5 m slices. Sections were stained with haematoxylin and eosin and immunohistochemistry for light microscopic examination.Results It was observed that irradiation ear promoted degeneration in spiral ganglion cells at these times(p<0.01).There was a singnificant difference on radiation-induced optical density value of caspase-3 in spiral ganglion(p<0.01).Conclusions irradiation might cause spiral ganglion cell damage. Caspase-3 cascade might be involved in the course of cell damage.
