Abstract: Abstract Objective:With the aim to further investigate the etiology of endometrial cancer, tissue chip technology was used to evaluate the expression of Insulin-like growth factor-1(IGF-1) and cyclooxygenase-2(COX-2)in endometrial cancer tissues. Furthermore, the association between their expression and the clinicopathological features of this malignancy was also examined. Methods:The tissue chip contained 150 cases endometrial cancer examples, 54 atypical and 84 normal endometrial examples were also contained.Immunohistochemistry SP method was applied to evaluate the expression of IGF-1 and COX-2 in the tissue chip. Results:(1)The expression of IGF-1 and COX-2 in endometrial cancer were significantly higher than that in the normal examples(P<0.05).(2)The expression of IGF-1 was positively associated with myometrial invasion(P<0.05).No association was found between the expression of IGF-1 and histological grade, FIGO phase and lymph node metastasis. The expression of COX-2 was found positively associated with histological grade(P<0.05).No association was found between the expression of COX-2 and myometrial invasion, FIGO phase and lymph node metastasis.(3)The effects of IGF-1 and COX-2 on the development of endometrial cancer were synergistic(P<0.05). The expression of them was positively associated. Conclusion:With the tissue chip to evaluate the expression of IGF-1 and COX-2 in endometrial cancer ,help to earlier diagnosis of endometrial carcinoma and its pre-cancerous,comprehensive evaluation its biologic behavior, and predict invasion of biology indexes, to judge the degree malignant to have the important meaning.
