The effect of carboxymethyl chitosan thermosensitive hydrogel on the L929 cells and the research of its biocompatibility

Abstract: Objective: Producing the Carboxymethyl chitosan thermosensitive hydrogel,to research the proliferation of the Carboxymethyl chitosan thermosensitive hydrogel on mouse lung fibroblasts (L929),moreover, probe into its tissue compatibility through animal experiments which can provide the fundamental experiment proof for further clinical application.Methods: Carboxymethyl chitosan interworked with glycerol phosphate to a preparation of the Carboxymethyl chitosan thermosensitive。Using MTT colorimetric assay to determine the evaluation on proliferation of L929 cells at different concentrations of carboxymethyl chitosan thermosensitive hydrogel.To take the transplant carboxymethyl chitosan temperaturesensitive gel in mice’s subcutaneous tissue, and kill them separately on the third, 7th and 14th day.And make regular tissue slice, then HE dye and observe the inflammatory response on the surrounding tissue of gel, finally evaluate the tissue biocompatibility and degradability of the gel of each group.Results: After 24h,the each concentration of extract groups on L929 cell have a certain role in promoting.In 48h,72h,96h,each experimental group as compared with negative and positive control group showed a more obvious role in promoting cell proliferation,in which 0.4 times leaching liquor have the cell-multiplication function obviously (P <0.05);On the 3dth after histology injecting, the experimental group is mainly in the acute inflammation.With the time of inflammatory cells in each experimental group gradually decreased to disappearit descriptions that carboxymethyl thermosensitive hydrogels have good biocompatibility.Conclusion:The results suggest that the Carboxymethyl chitosan thermosensitive hydrogel could promote the proliferation of the L929 and have good biocompatibility., local application of the chitosan thermosensitive hydrogel for treating periodontal diseases is feasible.
