Effect of Pseudolaric acid on the Expression of VEGF in Hepatocellular Carcinoma Cells

Abstract: 【Abstract】 Objective:A study was carried to investigate the impact pseudolaric acid do on VEGF expression in human liver cancer cells BEL - 7402.Methods:Human liver cancer cell BEL -7402 were processed by 0, 5, 10, 20, 40, 80 umol?L-1 pseudolaric acid separately for 6 hours, then analysed the cell proliferation and vitality in the two ways of WST 8 and trypan blue staining , and detected the VEGF expression in human liver cancer cells BEL - 7402 using the methods of RT- PCR and Western Blotting. Results: At first, compared with the control group, there was no significant difference (P > 0.05) in liver cancer cell proliferation rate and energy that processed by different doses of pseudolaric acid. Secondly with the increasing does of pseudolaric acid , VEGF protein expression was decreased in proportion, a significant difference emerged when compared with the control group (P < 0.01) and then with the increasing does of pseudolaric acid , there were significant decrease in VEGF mRNA expression compared with the control group (P < 0.01). Conclusion: Pseudolaric acid have significant inhibiting-effect on the expression of VEGF mRNA and protein in human liver cancer cells BEL - 7402 .
