Rhizoma Phragmitis ethanol extract in diabetic mice liver glycogen content and glycogen synthetase expression effect

Abstract: Abstract:objective To study Rhizoma Phragmitis ethanol extract influence on STZ induced diabetic mice liver glycogen content and glycogen synthetase. Methods Application periodic aci-Schiff reaction assay groups of laboratory mice liver glycogen content, and application reverse transcription polymerase chain reaction and Western blot assay Glycogen synthetase the expression of mice liver. Results After PAS staining the hepatic glycogen content decreased significantly after the model control group, low-dose group, high-dose group compared with model control group increased significantly (P<0.01), high-dose group was most pronounced among them; after RT-PCR, GS mRNA is lower than other three groups of model control group, low-dose group is lower than the high-dose group; after Westen blot after test model GS protein expression in the control group was lower than other three groups, low-dose group is lower than the high-dose group. Conclusions It is the dose-dependent regulation the Rhizoma Phragmitis ethanol extract influence on STZ induced diabetic mice liver glycogen, and raising the liver glycogen synthetase expression may be one of the role of the mechanism.
