高浓度葡萄糖对人脐静脉内皮细胞中 ghrelin 表达的影响

摘要: 目的 探讨葡萄糖对人脐静脉内皮细胞(HUVECs)中 ghrelin 表达的影响。方法 体外培养 HUVECs,经 过 12 h 无糖培养后分别观察不同浓度葡萄糖和不同培养时间对 ghrelin 表达的影响。(1)细胞经 0、5、10、15、20 mmol/L 葡萄糖处理 2 h,RT-PCR 法测定 ghrelin mRNA 表达水平,酶联免疫吸附试验(ELISA)测定细胞裂解液及培 养上清中 ghrelin 蛋白水平。(2)细胞以 10 mmol/L 葡萄糖处理 0、0.5、1、2、6、12 h 后,测定 ghrelin mRNA 及其蛋白表 达情况。结果 (1)随着葡萄糖浓度的升高,ghrelin mRNA 及蛋白表达水平出现下降,但当葡萄糖浓度大于 15 mmol/L 时,继续增加葡萄糖浓度对 ghrelin mRNA 及蛋白表达无明显影响。(2)在 10 mmol/L 葡萄糖作用下,随着培养时间的 延长,ghrelin mRNA 及蛋白表达水平出现下降,当超过 6 h 后,继续增加作用时间对 ghrelin mRNA 及蛋白表达无明 显影响。结论 高浓度葡萄糖可抑制 ghrelin 的表达,可能是发生动脉粥样硬化的机制之一。

Abstract: Objective To investigate the influence of high- level of glucose on the expression of ghrelin in human umbilical vein endothelial cells (HUVECs). Methods After 12 h glucose free culturing, the effects of different concentrations of glucose and different incubation times on expressions of ghrelin were observed in HUVECs. (1) The cells were treated with 0, 5, 10, 15, 20 mmol/L glucose for 2 h, then ghrelin mRNA expression levels were detected by RT-PCR, and the protein levels of ghrelin were detected by ELISA. (2) The cells were treated with 10 mmol/L glucose for 0, 0.5, 1, 2, 6, 12 hours, ghrelin mRNA and protein levels were detected respectively. Results (1) The expression levels of ghrelin mRNA and protein decreased along with increased glucose concentrations, which showed no obvious changes when the glucose was above 15 mmol/L. (2) The expression levels of ghrelin mRNA and protein decreased with the prolonged incubation time. But more than 6 h culturing time showed no further effect on reducing the expression level of ghrelin. Conclusion High levels of glucose can inhibit the expression level of ghrelin, which may be one of the mechanisms of atherosclerosis.
