自噬相关蛋白在肺鳞癌和腺癌组织中的表达及相关性研究

Expression and Correlated Analysis of autophagy-related proteins in Human squamous cell carcinoma and adenocarcinoma of lung

摘要: 目的:探讨自噬相关蛋白Beclin1、Atg1、mTOR在不同分化程度肺鳞癌(SCC)和腺癌中的表达情况及其相关性。方法:采用免疫组织化学方法,检测82例肺癌组织及17例癌旁正常肺组织Beclin1、Atg1、mTOR的表达情况。结果:癌旁组织中Beclin1、Atg1阳性率高,而mTOR的阳性率较低;SCC和肺腺癌组织中,随分化程度的降低,Beclin1、Atg1呈递减趋势,而mTOR的阳性表达呈递增趋势。相关性分析显示,不同分化程度SCC中Beclin1与Atg1表达呈正相关(R=0.609,P<0.01),Beclin1、Atg1分别与mTOR呈负相关(R=-0.617,-0.444,P<0.01);不同分化程度肺腺癌中,Beclin1与Atg1表达呈正相关(R=0.458, P<0.01),Beclin1、Atg1分别与mTOR呈负相关(R=-0.497,-0.541,P<0.01)。结论: mTOR随SCC和肺腺癌分化程度的降低,表达逐渐增高,而Beclin1、Atg1表达逐渐降低,提示SCC和肺腺癌的恶变程度与自噬有关。

Abstract: Objective:To explore the expression of Beclin1、autophagy-related protein 1 (Atg1)、target of rapamycin (mTOR) in different squamous cell carcinoma(SCC) and adenocarcinoma of lung and relationship among the three factors. Methods: The expression of Beclin1、Atg1、mTOR in different human SCC and adenocarcinoma of lung were detected by immunohistochemical techniques .There were 17 cases of normal tissue,82 cases of lung cancer. Results: The expression of Beclin1 and Atg1 showed a tendency to increase from high differentiated SCC and adenocarcinoma of lung to low differentiated ones. But mTOR showed a tendency to decrease. Compared with the expression of Beclin1,the expression of Atg1 in SCC and adenocarcinoma of lung presented a positive correlation. And Compared with the one of mTOR,Beclin1 and Atg1 show a negative correlation. Conclusion:Autophagy is related with development and SCC and adenocarcinoma of lung.
