The Correlation study of endometrial carcinoma and insulin//Ras/MAPK signaling pathway

Abstract: Objective: To investigate the expression and correlation of Grb-2、Ras、P-ERK、CD-1,which are the main members of insulin/Ras/MAPK signal pathway in endometrial carcinoma. Methods:The expressions of Grb-2、Ras、P-ERK、CD-1 protein in tissues from 51 cases of endometrial carcinoma, 10 endometrial atypical hyperplasia, and 48 normal endometrium were detected by using immunohistochemistry Elivison method and image analysis system. The serum insulin concentration was measured by radioimmunoscintigraphy in endometrial carcinoma.Results:The positive expression rate of Grb2、Ras、P-ERK、CD-1 in normal endometrium、endometrial atypical hyperplasia and endometrial carcinoma was gradually increased. The expression rate of Grb2、Ras、P-ERK、CD-1 in endometrial carcinoma was higher than that in normal endometrium and endometrial atypical hyperplasia. In endometrial carcinoma there were positive correlation between Grb-2 and FIGO stage,either between P-ERK and FIGO stage (P<0.05). Also,there were positive correlation between Grb-2 and lymphonode metastasis(P<0.05). There were positive correlations among the expression rates of Grb-2-Ras、Grb-2-P-ERK in endometrial carcinoma. The expression of Grb-2 and P-ERK was correlate with fasting serum insulin level in endometrial carcinoma(P<0.05).Conclusion:Insulin/Ras/MAPK pathway may play an important role in the carcinogenesis and development of endometrial cancer.
