Abstract: Abstract Objective:To investigate the mechanism of angiotensin II type\(AT1)receptor blocker, telmisartan in OLETF rats with insulin resistance.Methods:Forty-seven male OLETF rats were fed with high-fat diet for 22 weeks to establish the insulin resistance model with hyperlipidemia.Forty-seven OLETF rats with insulin resistance models were randomly assigned into five groups, the model group,in which the rats were fed with high-fat diet;the telmisartan group,in which the rats were fed with high-fat diet and telmisartan;the metformin group in which the rats were fed with high-fat diet and metformin;the pioglitazone group,in which the rats were fed with high-fat diet and pioglitazone ;the low pioglitazone group,in which the rats were fed with high-fat diet and low pioglitazone .After 26 weeks of drug intervention, the plasma levels of fasting blood glucose(FBG),fasting insulin(FINS),free fatty acids(FFA),lipids, omentin, retinol binding protein 4 and visfatin were detected in five groups. Results:After 26 weeks of telmisartan treatment, the insulin resistance and lipid metabolism were improved in OLETF rats. The serum omentin was significantly higher in the telmisartan group than that of the model group(F=6.888,P<0.001). The serum retinol binding protein 4 and visfatin were significantly lower in the telmisartan group than that of the model group(F=7.236,P<0.001 and F=4.165,P=0.003).The serum lipid level and insulin resistance were significantly improved. Conclusion:Telmisartan can improve blood glucose and insulin resistance by inhibiting the expression of retinol binding protein 4 and visfatin ;enhancing the expression of omentin and regulating serum lipid leve1.
