The inhibitory effect on asthma by CD8α-CD11c+ dendritic cells from Schistosoma japonicum infected mice

Abstract: Abstract: Objective To study the inhibitory effect on allergic asthma by adoptive transfer of    CD8α-CD11c+ dendritic cells (DC) from Schistosoma japonicum-infected mice. Methods CD8α-CD11c+DC from S.japonicum-infected mice and naive were isolated and purified using microbeads respectively,. Twenty BALB/c mice were divided into four groups randomly.5×105 CD8α-CD11c+DCs from S.japonicum-infected mice were injected into the tail vein of each mouse in Group A,while 5×105 CD8α-CD11c+ DCs of na?ve were injected into the tail vein of each mouse in Group B. One hour after injection,mice in Group A、B and C were sensitized and challenged with ovalbumin to induce asthma.Four weeks later,all the mice were killed,and the left lungs were cut for histopathologica1 analysis.Results Compared with mice in Group B and C,pulmonary inflammation of mice in Group A diminished markedly.According to Underwood’s standard,average total scores of Group A,B,C and D were 7.67±2.34,l0.86±1.35,11.75±2.22and 4.5±1.73 respectively ( P< 0.05). Conclusion CD8α-CD11c+ DC from S.japonicum infected mice can significantly inhibit allergic asthma.
