Effects of high glucose on angiopioetin-like protein 3 expression in vitro and in vivo

Abstract: Abstract:Objective To observe the effects of high glucose on angiopioetin-like protein 3(ANGPTL3) expression in vivo and in vitro .To explore the relationship and of ANGPTL3 with the diabetes mellitus (DM). Methods Take human serum and L02 as research subjects including 10 cases healthy human serum as control group(male : famale=5:5)and 10 cases of hyperglycemia serum as experimental group (M:F=5:5) from clinic. L02 cells were cultured separately in mediums containing 5.6,7.1,11.2 and 20mmol/L glucose,among which 5.6 mmol/L glucose group was taken as contro1;The relative level of ANGPTL3 protein in serum was measured by ELISA .The relative level of ANGPTL3 mRNA expression was detected by reverse transcription-polymerase chain reaction(RT-PCR). The relative level of ANGPTL3 protein in L02 was measured by Western Blot. Results The concentration of ANGPTL3 protein in hyperglycemia group was obviously higher than that of ctrol group(P<0.05).High concentration of glucose in medium could stimulate the expresion of ANGPTL3 mRNA and protein in cultured L02 cells . The differences were significant (P<0.05).Conclusion high glucose can upregulate ANGPTL3 expression both in vtro and in vivo. The expression of ANGPTL3 might by related to DM.
