摘要: Prestin蛋白的体外稳定高表达模型的建立 于 利 (辽宁医学院基础学院生理教研室,辽宁 锦州 121001) 摘要 目的 建立体外稳定高表达prestin蛋白的多克隆CHO细胞株。方法 采用转染prestin基因并在CHO细胞进行表达,高浓度抗生素筛选,多克隆细胞株传代培养,并进行prestin蛋白表达的检测。 结果 成功获得稳定高表达prestin蛋白的多克隆CHO细胞株。结论 通过简便、快捷、易操作的转染筛选的方法,成功建立多克隆的稳定高表达prestin蛋白的体系,为进一步研究其功能奠定基础。

Abstract: Construction of high level of stably expression of Prestin protein in vitro YU Li Department of Physiology, Fundamental School, Liao Ning Medical University, Jin Zhou Liao Ning 121001 Abstract Objective To construct stable expressions of prestin in CHO cell line in vitro. Methods CHO cells were transfected by prestin plasmids, which were selected by high dose antibiotic. Multi-colony cells which selected by antibiotic were subcultured and measured for expressing of prestin protein. Result It is successful that stably high expression of prestin protein in CHO cell line was constructed. Conclusion Multi-colony highly stably expression of prestin protein in cell line was constructed in which was transfected and selected by a simple, short-term, easy to operate method. The research is also lay a base for next studies on prestin protein.
