Effects of Sanhuangtangshenkang particles on expressions of TNF-α and IL-6 in bone tissue of T2DM rats#br#

摘要: 目的 观察三黄糖肾康颗粒对 2 型糖尿病大鼠骨组织肿瘤坏死因子 (TNF、 白细胞介素 (IL-6 表达的影响。方法 取雌性 Wistar 大鼠, 正常对照组 10 只, 制作骨质疏松模型 10 只和糖尿病模型 50 只。糖尿病模型大鼠进一步分成模型对照组, 中药预防、 高、 低剂量组和钙尔奇碳酸钙 D3(西药),每组 10 只。7 组分别给予相应干预, 喂养 20 周后取材,全自动生化仪检测空腹血糖 (FBG); 酶联免疫吸附法 (ELISA) 检测血清 TNF-αIL-6 水平; 双X 线骨密度仪 (DXA) 检测骨密度, 实时定量聚合酶链反应 (RT-PCR) 检测骨组织 TNF-αIL-6 mRNA 水平; 蛋白免疫印迹法 (Western blot) 检测骨组织 TNF-αIL-6 蛋白水平。结果 与骨质疏松模型组比较, 中药预防、 高、 低剂量组及西药组骨密度升高, 血清 TNF-αIL-6 下降, 骨组织 IL-6 mRNA 及蛋白表达下降(P < 0.05), 中药预防、 高剂量组和西药组骨组织 TNF-α mRNA 及蛋白表达下降 (P < 0.05)。与糖尿病模型对照组比较, 中药预防、 高、 低剂量组及西药组 FBG、 血清 TNF-αIL-6 均明显下降 (P < 0.05), 中药预防组、 中药高剂量组和西药组骨组织 TNF-α mRNA及蛋白表达下降 (P < 0.05), 中药预防、 低剂量和西药组骨组织 IL-6 mRNA 及蛋白表达下降 (P < 0.05)。结论 糖尿病大鼠血清和骨组织 TNF-αIL-6 表达增高, 三黄糖肾康颗粒能够改善糖尿病大鼠的炎症状况。

Abstract: Objective To observe the effects of Sanhuangtangshenkang particles on the expression levels of tumor necrosis factor (TNF)-α, interleukin (IL)-6 in bone tissue of type 2 diabetic (T2DM) rat model. Methods There were seven groups of rats in this study. Ten female Wistar rats were used as control. Ten rats were established for osteoporosis model, and fifty rats were established for diabetic model. Fifty diabetic model rats were further divided into model control group, traditional Chinese medicine (TCM) prevention group (high-dose TCM group, low-dose TCM group) and Western medicine group, with 10 rats for each group. All rats were given the corresponding intervention for 20 weeks. Automatic biochemistry analyzer was used to determine fasting blood glucose (FBG). ELISA was used to detect serum TNF-α and IL-6. Dual-energy X-ray absorptiometry (DXA) was used to assess bone density. Real-time PCR was used to appraise the mRNA of TNF-α and IL-6 in bone tissue. Western blot assay was used to measure protein expressions of TNF-α and IL-6 in bone tissue. Results Compared with osteoporosis model group, bone density was significantly increased, serum levels of TNF-α and IL-6, the mRNA and protein expression level of IL-6 were significantly decreased in bone tissue of TCM groups and Western medicine group (P < 0.05). The expression levels of TNF-α mRNA and protein were significantly decreased in bone tissue of in high-dose TCM prevention group and Western medicine group (P < 0.05). Compared with the diabetic model group, the serum levels of FBG, TNF-α and IL-6 were significantly decreased in TCM groups and Western medicine group (P < 0.05). The expression levels of TNF-α mRNA and protein were significantly decreased in high-dose TCM prevention group and Western medicine group (P < 0.05). The mRNA and protein expression levels of IL-6 were significantly decreased in lowdose TCM prevention group and Western medicine group (P < 0.05). Conclusion The expression levels of TNF-α and IL-6 are increased in serum and bone tissue of diabetic rats. Sanhuangtangshenkang particles can improve the inflammatory state to play prevention and therapeutic effect on diabetic rats.
