莲心碱促进膀胱癌 5637 细胞凋亡增强Caspase-7 表达及活化

Liensinine promotes apoptosis of bladder cancer 5637 cells and enhances Caspase-7 expression and activation

摘要: 目的 研究莲心碱(Liensinine)对膀胱癌细胞株 5637 凋亡的影响, 初步探讨其作用机制。方法 使用CCK-8 法及克隆形成实验检测莲心碱对 5637 细胞增殖的抑制作用; 通过流式细胞术 AnnexinV-FITC/PI 双染法测定莲心碱对 5637 细胞凋亡的影响; 采用 Western blot 技术分析细胞凋亡相关基因 Caspase-7 表达状况。结果 CCK-8法及克隆形成实验结果均表明莲心碱对膀胱癌细胞 5637 增殖有明显的抑制作用, 并呈剂量-效应关系。流式细胞术结果提示莲心碱可使膀胱癌 5637 细胞发生早期凋亡; Western blot 检测结果显示莲心碱增强 5637 细胞 Caspase-7表达并促进其活化。结论 莲心碱可抑制 5637 细胞增殖, 使其发生早期凋亡, 其作用机制可能与促进 Caspase-7 表达及活化有关。

Abstract: Objective To study the effects of Liensinine on apoptosis of 5637 cells, and its mechanism thereof. Methods CCK-8 method and the colony formation test were used to detect cell viabilities, and then inhibition rates were calculated. Flow cytometry was used to detect the effects of Liensinine on apoptosis of 5637 cells. Western blot assay was used to detect Caspase-7 protein expression. Results CCK-8 assay and colony formation test indicated that Liensinine inhibited the cell proliferation significantly. Results of flow cytometry indicated that Liensinine induced early apoptosis of 5637 cells. Western blot assay showed that Liensinine improved the expression of Caspase-7 and enhanced the activation of Caspase-7 in 5637 cells. Conclusion Liensinine could inhibit the proliferation of 5637 cells, induce early apoptosis, which may be related with the enhanced expression of Caspase-7 and its activation.
