子宫内膜异位症中子宫内膜间质细胞侵袭、转移能力的研究

Invasion and Metastasis Ability of Endometrial Stromal Cells in Endometriosis

【 摘要】 目的 检测子宫内膜异位症(EMs) 患者子宫内膜间质细胞中环氧化酶-2(COX-2)、 前列腺素 E2 (PGE2)、锌指转录因子(Snail)和 E-钙黏蛋白(E-cadherin) mRNA 表达情况, 分析 EMs 中子宫内膜间质细胞的侵袭、转移能力。 方法 收集 EMs 患者的在位及异位病灶子宫内膜组织和子宫肌瘤患者的在位内膜组织, 原代消化培养子宫内膜细胞, 取纯化的第 4 代子宫内膜间质细胞进行研究, RT-PCR 法检测细胞 COX-2、PGE2、Snail 和 E-cad? herin mRNA 的表达。 结果 EMs 和子宫肌瘤患者分泌期在位子宫内膜间质细胞 COX-2、PGE2、Snail 和 E-cadherin mRNA 表达与增殖期比较差异均无统计学意义; EMs 患者在位及异位的子宫内膜间质细胞 COX-2、 PGE2、 Snail mRNA 表达比肌瘤患者在位细胞均明显升高, 而 E-cadherin mRNA 表达降低(P< 0.05 或 P< 0.01); EMs 患者异位的子宫内膜间质细胞 COX-2、PGE2、Snail mRNA 表达比在位间质细胞升高, 而 E-cadherin mRNA 表达降低(P< 0.05 或 P< 0.01)。 结论 EMs 患者的子宫内膜间质细胞侵袭、转移能力强于非 EMs 患者, 且在 EMs 发病中异位病灶子宫内膜间质细胞的侵袭、转移能力更强。

Abstract:

[Abstract] Objective To analyze the invasion and metastasis of endometrial stromal cells in endometriosis through assessing the transcription levels of COX-2, PGE2, Snail and E-cadherin mRNA. Methods Ectopic and eutopic endometri? al tissues from patients with EMs (endometriosis) or uterine fibroids were collected and cultured. The fourth generation of pu? rified endometrial stromal cells were used for research, and the transcription levels of COX-2, PGE2, Snail and E-cadherin mRNA were analyzed by RT-PCR. Results There were no statistical differences in transcription levels of COX-2, PGE2, Snail and E-cadherin mRNA in eutopic endometrial stromal cells from patients with EMs or uterine fibroids between secra? tion stage and proliferation stage(P> 0.05). The transcription levels of COX-2, PGE2 and Snail mRNA were significantly in? creased while transcription level of E-cadherin mRNA was decreased in eutopic and ectopic endometrial stromal cells in pa? tients with EM compared with those in patients with uterine fibroid. And the difference was statistically significant(P< 0.05 or P< 0.01). The transcription levels of COX-2, PGE2 and Snail mRNA were obviously higher while transcription level of E-cadherin mRNA was lower in ectopic endometrial stromal cells than those in eutopic endometrial stromal cells from pa? tients with EMs. And the difference was statistically significant(P< 0.05 or P< 0.01). Conclusion The invasion and me? tastasis of endometrial stromal cells of patients with EMs were remarkable than those cell of patients without EMs. What’ s more, the invasion and metastasis in ectopic endometrial stromal cells were more obvious than those cells in the pathogenesis of EMs.
