Research on genetic polymorphisms of HLA- A31, B40, B58, DRB1*16 in the Hans of Zunyi

Abstract: Objective :To investigate the frequencies and polymorphism of hemorrhagic fever with renal syndrome (HFRS) associated HLA alleles ,HLA-A31, B40, B58 and DRB1*16 , in the Hans of Zunyi area. Methods: Polymerase chain reaction-sequence specific primers (PCR-SSP) were used to type HLA-A, B, DR genes of 200 unrelated healthy Han individuals in Zunyi area, and then the positive samples of A31, B40, B58 and DRB1*16 were selected and subtyped at high-resolution.. The allelic distribution of these alleles in Zunyi Han populations were compared with other different populations by chi-square test . Results: In HLA-A31, B58, DRB1*16 gene, only one kind of alleles was severally obtained, whereas, in HLA-B40 four kinds of alleles were obtained. Among them, B*4001, B*4002 and B*4006 were common alleles , and B*4063 was one kind of the rare alleles. Comparison of the HLA subtypes frequencies in different populations showed a close relationship of Zunyi Han population with the Han population in Southern China and the distribution of the allele subtypes in Zunyi Han population differed significantly from that of the Japanese. Conclusion: The alleles in HLA-A31、B58、DRB1*16 are relatively single, but the alleles in HLA-B40 have plenty of polymorphism. Their distributions have not only the southern Han’s characteristics but also their own territory feature.
