Regulatory action of Ruangan Huajian granule on hepatic expression of Bcl-2 and Bax in C57 mice with experimental hepatic fibrosis

Abstract: [Abstract]    Objective   To study the effect of Ruangan Huajian granule on hepatic expression of apoptosis related of gene expression in C57 mice with experimental hepatic fibrosis. Methods    Forty C57 mice were randomly divided into four groups, i.e. normal control group, model control group, model Ruangan Huajian Granule low-dose group and high-dose group. There were 10 mice in every group. C57 mice with experimental hepatic fibrosis were induced by intraperitoneal injection of 10% CCl4. The expression of Bcl-2和Bax in liver tissue of mice was observed by immune histochemical technique. The score of masculine grade and fibrosis stage (S) in liver tissue were also analyzed. Results    Bcl-2和Bax were Mainly express in coagulative liver cells , Model control group particularly obvious. The expression of Bcl-2和Bax and the fibrosis stage in model groups were significantly higher than those in normal control group (P<0.05), the ratio of Bcl-2/Bax Significantly lower than normal control group . the expression of Bcl-2 in Ruangan Huajian Granule low-dose and high-dose group obvious enhancement than those in normal control group and model control group(P<0.05), while the expression of Bax signifecantly reduce(P<0.05), the ratio of Bcl-2/Bax Significantly higher than the normal control group and model control group(P<0.01,P<0.01). Conclusion   Ruangan Huajian granule can modulate the expression of Bcl-2 and Bax in liver tissue ,and inhibit the liver cell apoptisis , Therefor it possesses anti-fibrotic effect.
