    Hypoxia and hypoxia related microRNA-210 regulate the epithelial-mesenchymal transition in gastric cancer cell MGC803

    Objective To explore the effect of anoxic microenvironment and hypoxia related microRNA- 210 (miR- 210) on the epithelial- mesenchymal transition (EMT) in gastric cancer cell line 803. Methods The mRNA relative expressions of E-cadherin and Twist1 were detected by qRT-PCR in gastric cancer cell line after being induced hypoxia by CoCl2 (100, 200 and 300 μmol/L), which was compared with normoxic group (0 mol/L). The cell proliferation of gastric cancer cell line was observed by cell proliferation assay. The relative expressions of E-cadherin and Twist1 were detected in gastric cancer cell line after transfecting miR-210 mimics. Results With the increase of the concentrations of CoCl2, the relative expression of miR-210 and the proliferation rate of gastric cancer cells showed a tendency of increase first and then decrease(P<0.05). The relative expression of E- cadherin mRNA was lower in hypoxia group than that of normoxic group. Therelative expression of Twist1 mRNA was higher in hypoxia group than that of normoxic group. The relative expressions of Ecadherin mRNA were significantly higher in 200 and 300 μmol/L CoCl2 groups than that of 100 μmol/L CoCl2 group (P<0.05). There was no significant difference in the relative expression of Twist1 mRNA between the three CoCl2 groups. The relative expression of Twist1 mRNA was higher in the transfection group than that of control group. The relative expression of E- cadherin mRNA was significantly lower in the transfection group than that of control group (P<0.05). Conclusion Hypoxia can promote the proliferation and EMT of gastric cancer cells. The up-regulation of miR-210 can also promote the procession of EMT, which may be the intermediate link in the process of EMT induced by hypoxia.

    Effects of overexpression of miR-30b on the biological function and tumor formation of human gastric cancer cells

    Objective To investigate the effect of overexpression of miR-30b on the proliferation, cell cycle, apoptosis and invasion of gastric cancer cell line SGC- 7901 and AGS, and the inhibitory effect on the tumor formation in vivo.Methods SGC-7901 and AGS cells were transfected with miR-30b mimics and miR-control, and qRT-PCR was used to detect the expression levels of miR-30b. Western blot assay was used to detect the expression of eIF5A2 protein. CCK-8 assay was used to measure the cell proliferation. Flow cytometry was used to analyze cell cycle and apoptosis. Transwell assay was used to detect cell invasion. In addition, the SGC-7901 and AGS cells transfected with miR-30b mimics and miRcontrol were injected into nude mice to observe the tumor formation and the expression of eIF5A2 protein in vivo. Results Results of qRT-PCR showed that the relative expression of miR-30b was significantly higher than that of miR-control group (P < 0.05). Western blot assay showed that the expression of eIF5A2 protein was decreased in miR- 30b mimics group.CCK-8 assay showed that cell proliferation was inhibited in miR-30b mimics group. The result of flow cytometry showed that the cell cycle decreased and the apoptosis increased in miR-30b group. Transwell assay showed that the cell invasion was significantly lower in miR-30b group than that of control group (P<0.05). Overexpression of miR-30b inhibited the formation of tumor and decreased the expression of eIF5A2 protein in vivo. Conclusion Overexpression of miR- 30b inhibits the proliferation, invasion and tumor formation of gastric cancer cells, and reduces the expression of eIF5A2 protein,which provides a potential target for gastric cancer treatment.

    Protective effect and mechanism of exenatide on HRMCs injured by fluctuating hyperglycemia

    Objective To observe the effect of exenatide on endoplasmic reticulum stress (ERS) in human renal mesangial cells (HRMCs) injured by fluctuating hyperglycemia culture, and to explore the mechanism. Methods HRMCs were randomly divided into three groups: control group (group N, cells were cultured in 5.6 mmol/L glucose for 24 h),fluctuating hyperglycemia group (group F, cells were cultured in 30 mmol/L glucose for 3 h, 5.6 mmol/L glucose for 2 h,repeated three times in one day, then 5.6 mmol/L glucose overnight), fluctuating hyperglycemia and exenatide group (group F+G, HRMCs were cultured in fluctuating hyperglycemia and 100 nmol/L exenatide). MTT assay was used to measure the viability in each group. The apoptosis rates were detected by flow cytometry in three groups. The relative expression of glucose regulated protein78 (GRP78) and CCAAT/enhancerbinding protein homologous protein (CHOP) were tested by Western blot assay. Results Compared with the group N, the cell proliferation level decreased, the cell apoptosis rate increased, and the expression levels of GRP78 and CHOP increased in F group (P < 0.05). After treatment with exenatide,the cell proliferation rate increased, cell apoptosis rate decreased (P < 0.05), and the expression levels of GRP78 and CHOP decreased in F+G group, compared with those of the group F (P < 0.05). Conclusion Exenatide can reduce the damage of fluctuating hyperglycemia on HRMCs by down-regulating the stress levels of the endoplasmic reticulum stress.

    The effects of SST for predicting the differentiation of endometrial carcinoma and on cell biological behavior

    Objective To investigate the expression levels of somatostatin (SST) gene in endometrial adenocarcinoma tissues and cell lines, and the effects of over-expression of SST gene on the proliferation and invasion of endometrial cancer cell line Ishikawa in vitro. Methods Tissue sections of normal endometrium, endometrioid adenocarcinoma and uterine papillary serous carcinoma were selected to detect the expressions of SST by immunohistochemical method. The total RNA was extracted from fresh specimens that were confirmed as endometrioid adenocarcinoma. According to FIGO staging,samples included G1 (7 cases), G2 (6 cases) and G3 (5 cases) of endometrioid adenocarcinoma. The SST expression levels were detected by real-time PCR. Three endometrial cancer cell lines, Ishikawa, HEC-1A and KLE, were selected and the expression levels of SST were detected by real-time PCR and Western blot assay. Transfection was performed with pLVXSST and pLVX. The transfection efficiency was observed by fluorescence confocal microscopy. The protein levels of SST were detected by Western blot assay. The assays of CCK- 8 and transwell were applied to examine variations in cell proliferation and invasion. Results Immunohistochemical results showed that SST expression was increased in endometrioid adenocarcinoma and uterine papillary serous carcinoma compared with that of normal endometrium. Real-time PCR showed that SST expression was significantly increased in G3 compared with that of G1 and G2 in endometrioid adenocarcinoma (P<0.05). No matter mRNA or protein, SST levels were significantly increased in endometrial cancer cell line KLE compared with those of Ishikawa and HEC- 1A, and the expression levels of SST mRNA and protein were significantly increased in HEC- 1A group than those of Ishikawa group (P<0.05). The expression of SST protein was significantly higher in the group of Ish- SST after 2 generations compared with that of Ish- ctr group. There were no significant differences in cell proliferation and invasive ability after over-expression of SST between Ishikawa cell group and control group (P>0.05). Conclusion SST is highly expressed in poorly differentiated endometrial cancer cells. The proliferation and invasion are not increased after the over-expression of SST in Ishikawa cell line of endometrial cancer.

    Effects of harmine on the epithelial mesenchymal transition of melanoma A375 cells

    Objective To explore the inhibitory effect of harmine on melanoma A375 cells and its mechanism thereof.Methods (1) Melanoma A375 cells were treated with harmine at 0, 0.5, 1, 2, 5, 10, 20, 50 and 100 mg/L for 48 h in vitro.CCK-8 method was used to detect the cell viability and confirm the experimental concentrations. (2) After the cells were treated with 0, 1, 2 mg/L harmine, the scratch and transwell assays were used to detect the cell migration and invasion ability. Western blot assay was used to detect the expression levels of epithelial mesenchymal transition (EMT) - related protein E-cadherin, N-cadherin, Snail and p53. (3) Three groups of cells were set up. The control group was transfected with empty vector only. The empty vector group was transfected with empty vector after treated with 2 mg/L harmine for 24 h. The Snail transfection group was transfected with Snail cDNA after treated with 2 mg/L harmine for 24 h. The cell migration and invasion ability were detected after the transfection. Results (1) When the concentration of harmine was above 2 mg/L, the survival rate of A375 cells was significantly lower than that of the control group with the increase of harmine concentrations(P < 0.05). Then, the concentrations of 0, 1 and 2 mg/L of harmine were used in the following experiments. (2) With the increase of the harmine concentrations, the number of cells in the scratched area and the number of trans-membrane cells in each group were significantly decreased. The migration and invasion ability of the cells were decreased gradually. The expression levels of E- cadherin and p53 were increased, while the expression levels of N- cadherin and Snail were decreased. (3) Cell transfection experiments showed that the migration and invasion ability of the cells were increased compared with those of empty vector group after transfection with Snail. Conclusion Harmine can inhibit the proliferation of A375 cells and decrease the abilities of metastasis and invasion, which may be achieved by decreasing the expression of Snail after activating the p53, thereby increasing E-cadherin and down-regulating N-cadherin to inhibit the EMT process.

    Construction of miR-196b sponge lentiviral vector and its expression in bone marrow stromal cell line ST2

    Objective To construct miR- 196b sponge lentiviral vector, and laid the foundation for studying the function of miR- 196b in bone marrow stromal cells. Methods Based on the miR- 196b mature sequence, a sequence consisting of 6 tandem repeats of the complementary sequence of miR- 196b was designed, and which was cloned into pUC19 plasmid by using reverse PCR. Then the six-repeat sequence was cut and subcloned into pLVX- shRNA2 lentiviral vector. The lentivirus was packaged using 293T cells, and titer determination was done. The pLVX-shRNA2 lentivirus was used as the control group, and the 196b-sponge-pLVX lentivirus was the experimental group. Then ST2 cells were infected with the viruses, and the infection efficiency was calculated. The protein level of forkhead box O1 (FoxO1) was detected by Western blot assay. Results The identity of the sponge sequence was verified by sequencing. The titer of the sponge virus was 1 × 108 PFU/mL, and the infection efficiency reached 80% . Compared with the control group, the expression level of FoxO1 protein was significantly increased (P < 0.05). Conclusion The miR-196b sponge lentiviral vector is successfully constructed, and which has the capability to inhibit endogenous miR-196b.

    Protective effects of theaflavins on radiation injury of thymus

    Objective To investigate the protective effect of theaflavins on thymus injury caused by total body irradiation (TBI). Methods Twenty- five C57BL/6 mice were randomly divided into 5 groups: control group, 4 Gy TBI group, 4 Gy TBI + 25 mg/kg theaflavins group, 4 Gy TBI + 50 mg/kg theaflavins group and 4 Gy TBI + 100 mg/kg theaflavins group. Thymus index and total number of thymocytes were detected at the 14th d post-irradiation to determine the optimal dose of theaflavins. According to this optimal dose, 32 C57BL/6 mice were randomly divided into 4 groups: control group,theaflavins group, 4 Gy TBI group and 4 Gy TBI + theaflavins group. Thymus histomorphology, CD4CD8 T cell subsets, and reactive oxygen species (ROS) in thymocytes were examined at the 14th d post-irradiation. Results The irradiated thymus exhibited decreased thymus index and total number of thymocytes (P<0.05), aberrant histomorphology and T cell subsets (P<0.05), and increased ROS level in thymocytes (P<0.05). Compared with 4 Gy TBI group, the thymus index and total number of thymocytes were significantly increased in 4 Gy TBI + 50 mg/kg theaflavins group (P<0.05). The total number of thymocytes was significantly higher in 4 Gy TBI + 50 mg/kg theaflavins group than that in 4 Gy TBI + 25 mg/kg theaflavins group (P<0.05). Therefore, 50 mg/kg theaflavins was chosen as the optimal dose for subsequent experiments. Moreover, the aberrant histomorphology of irradiated thymus was alleviated by theaflavins. A decline in the percentage of CD4-CD8- T cellsand an elevation of CD4+CD8- and CD4+CD8+ T cells were found in irradiated mice administered with theaflavins (P<0.05).Compared with 4 Gy TBI group, the ROS level was significantly decreased in 4 Gy TBI + theaflavins group (P<0.05).Conclusion Theaflavins exhibits a protective effect on radiation-induced thymus injury.

    The expression and the clinical guiding significance of HIF-1α and Hp in Mongolian and Han two nationalities

    Objective To detect the expressions of two protein factors hypoxia- inducible factor (HIF)- 1α and Helicobacter pylori (Hp) in gastric cancer in Mongolian and Han two nationalities respectively, and to explore the relationship of HIF-1α and Hp in the development of gastric cancer. Methods A modified Warthin-Starry (W-S) silver staining method was used to detect Hp infection in 27 Mongolian patients with gastric cancer and 30 Han patients with gastric cancer. Immunohistochemical (S-P) method and reverse transcription polymerase chain reaction (RT-PCR) method were used to detect protein and mRNA expressions of HIF- 1α in cancer tissue, paracancerous tissue and normal tissue samples. The relationship of HIF-1α protein and Hp expression was analyzed between gastric carcinoma tissue samples of Mongolian patient group and Han patient group. Results Hp infection rates were 81.5% (22/27) and 66.7% (20/30) in Mongolian and Han patients with gastric cancer. The difference in infection rate was not statistically significant between two groups of patients (χ2=1.608, P > 0.05). The positive expression levels of HIF-1α protein were 62.96% (17/27) and 70.00% (21/30) in gastric cancer tissue samples of Mongolian and Han groups. The relative expression levels of HIF-1α mRNA were 0.891 4±0.075 0 and 0.907 4±0.065 5. There was no significant difference in the HIF-1α mRNA expression between the two nationalities. No positive expressions of HIF-1α protein and mRNA were found in paracancerous tissue and normal tissue samples. There was a positive correlation in the positive expression of HIF- 1α protein and Hp infection in gastric cancer tissue samples of Mongolian and Han patient groups (rs =0.424 and 0.617, P < 0.05). Conclusion HIF-1α and Hp are pathogenic factors of gastric cancer. Hp and HIF-1α may promote the occurrence of gastric cancer together. The pathogenic mechanism of HIF-1α and Hp in gastric cancer may be the same for Mongolian and Han patients.

    Efficacy of pushen capsule on blood lipid and atherosclerosis in patients with ischemic stroke

    Objective To explore the efficacy of pushen capsule on blood lipid and atherosclerosis in patients with ischemic stroke. Methods One hundred and twenty patients with ischemic stroke were randomly divided into three groups,treatment group (pushen capsule and rosuvastatin calcium), control group- 1 (pushen capsule) and control group- 2 (rosuvastatin calcium). There were 40 patients for each group. The course of treatment was 6 months. Data of serum total cholesterol (TC), triacylglycerol (TG), high density lipoprotein cholesterol (HDL- C), low density lipoprotein cholesterol (LDL- C), homocysteine (Hcy), high sensitive C reactive protein (hs- CRP), nitric oxide (NO), endothelin - 1 (ET- 1), the number and size of carotid plaques, and changes of intima-media thickness (IMT) were detected before and after treatment in three groups. The incidences of adverse cerebrovascular events and adverse events were observed in the three groups. And the therapeutic effects for dyslipidemia were evaluated in three groups. Results The treatment indicators were obviously improved after treatment in three groups (P<0.05). There were no significant differences in treatment indicators before treatment between three groups. The levels of TG and NO increased, the level of LDL-C, the size and thickness of carotid plaques decreased after treatment in control group-2 than those of control group-1. Levels of TC, LDL-C, Hcy, hs-CRP,ET-1, IMT, and the number, size and thickness of carotid plaques were significantly lower in treatment group than those of control group- 1 (P<0.05), and the level of NO was higher in treatment group than that of control group- 1 (P<0.05).Compared with control group-2, levels of TG, LDL-C, hs-CRP and the number, size and thickness of carotid plaques were significantly lower, and levels of HDL-C and NO were significantly increased in treatment group (P<0.05). There were no significant differences in the incidences of adverse drug reactions and adverse cerebrovascular events between the three groups (P>0.05). The total effective rate was higher in treatment group than that of control group-1 (P<0.05). There wereno significant differences in total effective rates between treatment group and control group-2, and between control group-2and control group-1 (P>0.05). Conclusion Pushen capsule combined with rosuvastatin calcium can obviously improve blood lipid metabolism in patients with ischemic stroke, which shows obvious antiatherosclerosis effect and good safety.

    Objective To study the relationship between lumbar and proximal femoral bone mineral density (BMD) with gender, age and body mass index (BMI) in adolescents ranged from 12 to 19 years of age in Tianjin city, and provide preliminary reference to understand the bone health status of adolescent in the city. Methods The BMD values of lumbar spine (L1-L4), femoral neck and total hip were detected by the dual-energy X-ray absorptiometry (DXA) in 221 adolescents from Tianjin Hospital, and the Z value was used for clinical diagnosis standard. Simultaneously the height and weight of adolescents were measured, and BMI was calculated. The differences of BMD values in the lumbar, femoral neck and total hip were compared between different age, gender and BMI groups. Results The BMD values of femoral neck and total hip were significantly higher in males than those in females (P < 0.01). The BMD values of lumbar spine, femoral neck and total hip were gradually increased with age. The BMD was correspondingly increased in adolescents with higher BMI in the lumbar spine, femoral neck and total hip. Conclusion There is a close relationship between BMD with age, gender and BMI in adolescents. We should pay more attention to the nutrition health and the level of BMI in adolescents.

    Clinical observation of the effect of budesonide atomization on tracheal extubation in children

    Objective To investigate the effect of budesonide atomization on tracheal extubation in children. Methods A total of 85 patients with tracheal intubation in Tianjin Children’ s Hospital from May 2013 to September 2016 were selected in this study. Patients were randomly divided into budesonide group (n=44) and dexamethasone group (n=41).The budesonide group was given 1 mg of budesonide for inhalation 30 min before extubation, and 1 mg of budesonide inhalation immediately after extubation. Then every 8 hours for 0.5- 1.0 mg budesonide inhalation for 4 days. The dexamethasone group was given dexamethasone 0.2-0.3 mg/kg intravenously 30 min before extubation, and dexamethasone 2.5-5.0 mg inhalation immediately after extubation. Then dexamethasone 2.5-5.0 mg inhalation was given every 8 hours for 4 days. The incidence of laryngeal edema (stridor, hoarseness), the time of extinction, the expression of hypoxia, reintubation rate within 24 hours and secondary infection rate after extubation were compared between the two groups.Results There were no significant differences in the mission success rate, the incidence of laryngeal edema, the time of extinction, the incidence of hypoxia and re- intubation rate between the two groups (P > 0.05). Two patients were found secondary infection after extubation in dexamethasone group. Conclusion Both budesonide and dexamethasone show curative effects on the prevention and treatment of laryngeal edema after extubation. Budesonide atomization can replace systemic corticosteroids, thus reducing the adverse reactions of glucocorticoids. We recommend the use of budesonide in treating adverse events after extubation.

    Significance of c-Myc in prognostic evaluation of gastric mucosa associated lymphoid tissue lymphoma

    Objective To investigate the expression of c-Myc in gastric mucosa associated lymphoid tissue (MALT)lymphoma and its implications on prognosis. Methods The clinical data of 79 patients hospitalized in our hospital from 2009 to 2015 were retrospectively analyzed. In these patients, 38 patients were low- grade MALT lymphoma, 20 patients were high-grade MALT lymphoma, 21 patients were diffused large B cell lymphoma (DLBCL). Real-time PCR was used to detect the levels of c-Myc expression in gastric MALT tissues and pair-matched adjacent normal tissues. The relationship between the expression of c- Myc and prognosis of patients was evaluated combing with the clinical data. Results Compared with the normal tissues, the expression levels of c-Myc protein were 15.7% (6/38), 25% (5/20) and 28.5% (6/21)in patients with low-grade MALT lymphoma, high-grade MALT lymphoma, and DLBCL. The relative expression levels of cMyc mRNA were gradually elevated in low-grade MALT lymphoma, high-grade MALT lymphoma and DLBCL. The tumor size and depth of invasion can influence the expression level of c-Myc. Survival analysis found that the overall survival rates and relapse-free survival rates were lower in patients with c-Myc positive expression than those of patients with negative expression (P<0.05). Conclusion C-Myc plays a key role in the malignant transformation of gastric MALT lymphoma.

    The influence of different dosage of protamine neutralization heparin on perioperation of on-pump coronary artery bypass grafting

    Objective To investigate the effect of different doses of protamine neutralizing heparin on perioperation of on-pump coronary artery bypass graftting (CABG). Methods A total of 180 on-pump CABG patients hospitalized from January 2015 to November 2016 were randomly divided into three groups, the protamine group 1, protamine group 2 and protamine group 3, 60 patients in each group. Heparin (3 mg/kg) was used before extracorporeal circulation. After intracardiac operation was over, protamine was used to neutralize the heparin to adjust the activated clotting time (ACT) in protamine group 1, which was 10% higher than that of intubation. Meanwhile, protamine group 2 was neutralized to equal to the ACT before intubation, and protamine group 3 was 10% lower than that before the intubation. The differences of intraoperative and postoperative parameters were compared between the three groups. Results No death was found in the three groups during hospitalization. Comparing with protamine group 1 and protamine group 2, the time of operation, the ACT before the leaving operation room, the ACT of the first hour after returning to ICU, the amount of bleeding during operation, the time of closing and the amount of red blood for transfusion were decreased in protamine group 3 (P>0.05).The total amount of protamine for neutralizing and the ratio of protamine and heparin were significantly increased in protamine group 3 (P<0.05). The heart dysfunction after operation, perioperative myocardial infarction, pulmonary edema,pulmonary infection, renal dysfunction, poor wound healing, neurological complications, and time of in hospital stay showed no significant differences between three groups (P>0.05). Conclusion ACT below 10% of preoperation is safe, after neutralization of heparin by protamine, which can obviously reduce the bleeding, the time of sternal closure and the amount of red blood cell transfusion, showing a positive clinical significance.

    Analysis of risk factors and clinical characteristics of pulmonary embolism in patients with lung cancer

    Objective To investigate the risk factors, clinic charactertics and survival prognosis of pulmonary embolism (PE) in patients with lung cancer. Methods The clinic data of 28 lung cancer patients with PE, hospitalized in department of respiratory and critical care medicine of Tianjin Chest Hospital between June 2012 to June 2015, were retrospectively reviewed. Eleven of them were diagnosed with primary lung cancer and PE (symptomatic group), and 17 lung cancer patients were found PE accidentally (asymptomatic group). A total of 56 lung cancer patients without PE were used as control subjects (no PE group). Data of pathological types, clinical staging of lung cancer, systemic chemotherapy, white blood cell (WBC), hemoglobin (Hb), platelet (PLT), D- dimer (DD), albumin (ALB) and C- reactive protein (CRP) were analyzed by univariate analysis in the symptomatic group and asymptomatic group. Logistic regression analysis was carried out on the statistically significant indexes to observe the influencing factors. The morphology and location of the remobilization images were analyzed in lung cancer patients with PE. The median time to embolism and survival of PE patients were compared between symptomatic group and asymptomatic group. Results The proportion of adenocarcinoma,systemic chemotherapy and stage Ⅲ + Ⅳ were significantly higher in PE group than those in no PE group (P < 0.01). The ratio of white blood cells (WBC)> 11×109/L (hyperleukocytosis) and albumin (ALB) <30 g/L and D-dimer (DD)> 0.5 mg/L were significantly higher in PE group than those of no PE group (P < 0.05). There were no statistical differences in other indicators including clinical symptoms between the two groups (P > 0.05). The logistic regression analysis showed that adenocarcinoma, chemotherapy, WBC>11×109/L and DD>0.5 mg/L were the risk factors of lung cancer with PE (P < 0.05).There was higher ratio of asymptomatic PE in lung cancer patients with PE. There were no significant differences in morphology and location of the remobilization images in symptomatic group. The median time of embolization was 3.6 months(95% CI: 3.2- 4.0) in the asymptomatic group, which was significantly earlier than that in the symptomatic group (10.5months, 95% CI: 8.88-12.12, P < 0.01). The median survival time was 7.2 months (95% CI: 5.86-8.56) in the asymptomatic group, which was significantly longer than that of symptomatic group (2.8 months, 95% CI: 2.48- 3.12, P < 0.05).Conclusion Lung adenocarcinom, systemic chemotherapy, hypoproteinemia and increased D-dimer are the independent risk factors in lung cancer patients with PE. PE in lung cancer is frequently asymptomatic in the early stage. Compared to symptomatic patients, asymptomatic cases have better prognosis.

    Objective To investigate the expression and significance of syntaxin 8 (STX8) in non- small cell lung cancer (NSCLC), and analyze the relationship between STX8 expression and clinicopathological features of NSCLC.Methods Seventy samples of NSCLC and 70 samples of pericancerous tissues were collected for immunohistochemistry staining, and another 10 samples of NSCLC and pericancerous tissues were used for RNA and protein extraction. qRT-PCR was performed to detect the expression of STX8 mRNA, and Western blot assay was adopted to detect the expression of STX8 protein. The relationship between STX8 expression and clinicopathological features of NSCLC was analyzed. Results Results of qRT-PCR showed that the expression of STX8 mRNA was significantly higher in NSCLC tissues than that in the pericancerous tissues (3.962 5±0. 487 3 vs. 0.538 2±0.097 5, t=21.797, P<0.01). Immunohistochemistry results showed that the high expression rate of STX8 was significantly higher in NSCLC tissues than that in the pericancerous tissues [71.43%(50/70) vs. 38.57% (27/70), χ2=15.267, P<0.01]. Western blot results showed that the expression of STX8 protein was significantly higher in NSCLC tissues than that in the pericancerous tissues (2.496 1±0.362 5 vs. 0.340 2±0.119 1, t=17.876,P<0.01). The high expression rate of STX8 was significantly different in different histological types of NSCLC (P <0.05). The high expression rate of STX8 was higher in squamous cell carcinoma and adenocarcinoma than that in adenosquamous carcinoma, and the high expression rate of STX8 was not observed in large cell carcinoma. There were no significant differences in expressions of STX8 in NSCLC patients with different gender, ages, tumor diameters, TNM stages and with or without lymph node metastasis (P>0.05). Conclusion A high expression of STX8 in NSCLC tissues may be correlated with the occurrence and development of NSCLC.

    Objective To investigate the predicative value of midkine (MK) as a cancer biomarker for metastatic lesions in differentiated thyroid cancer (DTC) patients with positive thyroglobulin antibodies (TgAb) before the first 131I therapy. Methods MK levels were measured by enzyme- linked immunosorbent assay in 151 recruited DTC patients included in this study according to strict inclusion and exclusion criteria. There were 28 TgAb positive DTC patients with metastases and 123 DTC patients without metastases. The value of pre- 131I-ablative MK to predict metastasis was assessed by receiver operating characteristic (ROC) curves in these two groups of patients. Results MK levels were significantly higher in TgAb positive DTC patients than those in DTC patients without metastases. MK levels showed good diagnostic value, with an area under the curve of 0.856 (P<0.001), and a diagnostic accuracy of 83% at the optimal cut-off value of 550ng/L. Conclusion Results show that MK can potentially be used as a surrogate biomarker for predicting DTC metastases when thyroglobulin is not suitable due to TgAb positivity.

    Objective To explore the effect of preservation of inclined fractured permanent molars by transverse fixation with screw post, and provide a practical method for reserving the inclined fractured permanent molars. Methods Clinical data of 50 patients with inclined fractured permanent molars by transverse fixation with screw post in the Department of Stomatology of Tianjin First Central Hospital between January 2010 to December 2011 were reviewed retrospectively. After fixing the crown with steel- wire binding and root canal treatment, the molars were drilled a hole buccolingually and placed screw post. The flowable light-curing composite resin was used to seal the crack and bond screw post to prevent the relative movement. All treated molars were restored with full crown. All patients were followed up for 10-60 months. The therapeutic effects were observed. Results After 5 year-follow-up, 41 cases were cured completely, 6 cases were cured clinically, and 3 cases failed treatment. Conclusion The transverse fixation with screw post and bonding technique can preserve the inclined fractured molars to gain chewing function and avoid or reduce the extraction of affected teeth, which provides a better restoration effect.

    Expressions and significance of IFN-γ and IL-10 mRNA in peritoneal fluid mononuclear cells of patients with endometriosis

    Objective To investigate the expression levels and significance of interferon-γ (IFN-γ), interleukin (IL)-10 mRNA in peritoneal fluid mononuclear cells (PFMC) in patients with endometriosis (EM). Methods A total of 55 patients with EM diagnosed histopathologically after operation were enrolled as EM group, and 52 patients without EM were enrolled as control group. The PFMC were isolated, and the expression levels of IFN-γ and IL-10 mRNA were determined by TaqMan real-time reverse transcriptase-polymerase chain reaction (RT-PCR). Results It was found that IFN-γ mRNA expression level was significantly decreased in EM group compared with that in control group (P<0.05), while IL-10 mRNA expression level was significantly increased in EM group compared with that in control group (P<0.05). The ratio of IFN-γ/IL-10 was significantly lower in EM group than that in control group (P<0.05). The ratio of IFN-γ/IL-10 was significantlyhigher in patients with stageⅠ-Ⅱ than that in patients with stage Ⅲ-Ⅳ (P<0.05). Conclusion The imbalance of IFN-γ/IL-10 in PFMC is associated with the pathogenesis of EM.

    Gitelman’s syndrome (GS) is an autosomal recessive hereditary tubulopathy. It is caused by sodium reabsorption in the distal convoluted tubule of the renal unit, which is not yet fully cured. It is characterized by great heterogeneity with clinical manifestations. Some patients with no symptoms, were confirmed by hypokalemia with physical examination. The others can be manifested as fatigue, nocturia, polydipsia and hypotension. Less literature is reported about GS combined with osteoarthrosis. In this report, the author reported a patient of GS with osteoarthrosis characterized by hypokalemia, hypomagnesemia, metabolic alkalosis, lower blood pressure, and swelling and pain in left knee joint. Thesymptoms were improved after treatment with potassium and magnesium supplementation

    Wegener’ s granulomatosis (WG) is a kind of systemic necrotizing granulomatous vasculitis characterized by capillary and small arteriovenous involvement. The typical WG triad is lesions of the upper and lower respiratory tract and kidneys. Only changes in the respiratory tract without renal lesions are localized WG. Localized WG is very easy to bemisdiagnosed because of its nonspecific symptoms and radiological manifestations. The author found one case of localized WG, which showed pulmonary symptoms and refractory heart failure. The chest CT showed that multiple irregular nidus,blurred and density enhanced lesions in both lungs. The patient was misdiagnosed as pulmonary infection for a long time.The patient was eventually diagnosed as asantineutrophil cytoplasmic antibody (ANCA) associated vasculitis and WG after two times of examination of protease -3(PR3) ANCA were positive. This article reviewed the diagnosis and treatment of the patient, and reviewed the relevant literatures.

    The characteristic of atypical atrial flutter episodes on electrocardiogram is different from the typical atrial flutter. Due to the poor diagnosis awareness about atypical atrial flutter, which is easily misdiagnosed as other arrhythmia, for example sinus arrhythmia, atrial fibrillation, and Ⅲ°atrioventricular block. Here we reported a case of atypical atrial flutter in child. In addition to the characteristics of typical atrial flutter, such as regular and more fixed atrial rhythm, fluctuated ventricular rate, faster atrial rate than the ventricular rate, the patient was found equipotential line, slow atrial rate, and uprighted F wave on the inferior wall leads and V1 lead of electrocardiogram, which were different from typical atrial flutter.

    Objective To evaluate the clinical efficacy of polyethylene glycols (PEG) on functional constipation in children by Meta-analysis. Methods The Medline, Pubmed, Cochrane database, Embase, VIP database, China Knowledge Resource Integrated Database (CNKI) and WANFANG MED ONLINE resources were reviewed for the treatment with polyethylene glycols on functional constipation in children from the inception to December 2016 to collect all randomized controlled trials (RCTs) or comparative prospective studies. Revman 5.3 software was used to assess risk of bias of the Cochrane Collaboration’s tool. A Meta analysis of the therapeutic effect of polyethylene glycols on functional constipation in children was conducted by Stata11.0 software. The pooled standardized mean difference (SMD) and 95%confidence interval (95% CI) were calculated in order to know the clinical efficacy of polyethylene glycols on functional constipation in children. Results Seven literatures were included in our Meta analysis. There were 662 children were included in this study, and they were divided into PEG treatment group (n=318) and non-PEG treatment group (n=344). The forest plot results of Meta analysis showed that the main effect index“weekly stool frequency”was significantly higher in PEG treatment group than that of lactulose group, according to the different control measures. The main effect index“weekly stool frequency”was significantly higher in PEG treatment group than that of non- PEG treatment group, according to the mapping forest chart displayed different clinical effect observation time points. Results from Meta analysis showed that the secondary effect index“successful stool consistency improvement”was significantly higher in PEG treatment group than that of non- PEG treatment group, according to different observation time points. Conclusion Polyethylene glycols is safe and effective in the treatment of functional constipation in children.

    Atrial fibrillation (AF) is one of the most common arrhythmias in clinical practice. It often causes serious thromboembolic complications, which brings great harm to human health. The incidence and morbidity rates of AF are increasing year by year. The research on the pathogenesis of AF has been focused on the aspects of electrical remodeling and structural remodeling. Atrial electrical remodeling and structural remodeling are the main mechanism of AF for the maintenance and recurrence. The intervention of atrial remodeling is helpful to the treatment of AF. It is found that the rennin-angiotensin aldosterone system (RAAS) plays an important role in atrial remodeling. RAAS blockers can significantly reduce the onset of AF by inhibiting the activation of the renin axis, which can block the activation of RAAS and improve the atrial remodeling. This may provide a new choice for the treatment of AF. This article reviews the research progress of RAAS blocking agent in the prevention and treatment of AF.

    Research progress in the anti-cancer effect of metformin

    The incidence of malignancy tumors has increased significantly recent years in patients with type 2 diabetes mellitus (T2DM). As the first- line oral medicine of T2DM, in addition to the safe and effective hypoglycemic effect,metformin can reduce the incidence and mortality of cancer, and improve the prognosis in T2DM patients. While the exactly anti- tumor mechanisms of metformin is not yet clear, which might be the inhibition of proliferation and promotion of apoptosis of tumor cells mainly by acting on an adenosine monophosphate activated protein kinase (AMPK) and microRNAs(miRNAs). And metformin can inhibit migration and invasion of tumor cells, as well as strengthen the effects of chemotherapy drugs. This article reviews the recent research progress of clinical and epidemiological studies and anticancer mechanisms of metformin.

    Toll- like receptor 4 (TLR4) plays an important role in inflammation and immune response. MicroRNAs (miRNAs) are involved in the regulation of TLR4 signaling pathway in multiple levels and various molecules, which play an important role in inflammatory reaction. A variety of miRNAs are involved in the regulation of TLR4 signaling pathway, and the TLR4 signaling pathway can induce a variety of miRNAs. Chronic diseases such as diabetes, Alzheimer’ s disease and cardiovascular disease are closely related to inflammatory response. The regulatory role of TLR4 signaling related miRNAs has attracted much attention in inflammatory diseases. In this review, the research progress of TLR4 signaling pathway related miRNAs in the regulation of inflammatory response is summarized, which provides a new research direction for the clinical diagnosis and treatment of inflammatory response related diseases.

    In recent years, the prevalence of diabetes has increased rapidly, and which has become a serious public health problem worldwide. Diabetic nephropathy is a major cause of end-stage renal failure, which is also one of the most common chronic complication of diabetes. However, the pathogenesis of diabetes and diabetic nephropathy has not been fully elucidated up to now. IL-17 plays a key role in autoimmune disease and inflammatory disease. This article reviews the role of IL-17 in the pathogenesis of diabetes and diabetic nephropathy.
