Sudy on the Relationship between PI3K/Akt Signal Transduction Pathway and DDP Resistance of Ovarian Carcinoma

Abstract: Abstract Objective:Study the relationship between cisplatin resistance of ovarian carcinoma and PI3K/Akt singnal transduction pathway. Methods:Western blot was used to detect the expression level of PI3K and Akt in SKOV3 and SKOV3/DDP cells.The effection of LY294002 and DDP to expression of PI3K and Akt in Ovarian carcinoma cells were detected by Western Blot. MTT assay was used to analyze the synergism of LY294002 to DDP and the effection of LY294002 to control ovarian tumor cells growth .Results:The correspondence density of PI3K and Akt in SKOV3 cells were lower than that in SKOV3/DDP cells.LY294002 could reduce the expression of PI3K and Akt in SKOV3 and SKOV3/DDP cells. After interferenced by LY294002 40μmol/l, the sensitivity of SKOV3 cells to DDP increased 67.36% and the sensitivity of SKOV3/DDP cells to DDP increased 76.56%. Conclusion:The abnomal PI3K Akt/signaling pathways related to Cisplatin resistance of Ovarian carcinoma. Inhibiting PI3K Akt/signaling pathways could reverse ovarian cisplatin drug-resistant phenotype.
