The Effect of Fluvastatin on Ventricular Remodeling and VWF in Heart Failure Rats

Abstract: Abstract  Objective  To study the effect of fluvastatin on cardiac remodeling and cardiac function , and to investigate the effect of VWF. Method   CHF model was induced by soproterenol via subcutaneous injection and rats were randomly divided into Placebo group and fluvastatin group , Another normal 10 rats served as the control group. Fluvastatin group were treated with fluvastatin (20mg /kg/d), Placebo group and control group were treated with normal saline.After 6 weeks treatment,the structure and function of hearts in different groups were measured by echocardiography ;measured ventricular weight index, the serum levels of VWF and BNP were measured by ELISA; the levels of VWF mRNA in cardiac muscle were measured by RT-PCR. Results  Compared with the control group, LVEDD, LVESD, LVRW, RVRW and VWF mRNA were significant increased in the Placebo group and drug groups, while LVEF and LVFS were significant decreased;Compared with placebo group,LVEDD,LVESD,LVRW,RVRW and VWF mRNA were significant decreased in the drug groups, while LVEF and LVFS were significant increased in the drug groups;The level of VWF were positive correction with the BNP. Conclusion   Fluvastatin could improve the cardiac function and prevent the cardiac remodeling, the mechanisms may be by reducing the levels of VWF and improving endothelial function.
