    Wnt/β-catenin signaling in the pathogenesis of osteoarthritis

    Abstract Objectives The expression of proteins and genes of Wnt/β-catenin signaling in the articular cartilage of human OA samples from patients undergoing total knee arthroplasty.Methods Knee joint samples were dissected and embeded in paraffin. The tissue sections were stained with HE , Safranin O / Fast Green (SO/FG) and Alcian blue / orange G(AB/OG),and graded using a modified version of the Mankin scale. Immunohistochemistry staining of β-catenin and MMP-13 were performed in all samples. Total RNA extracted from articular cartilage tissue was prepared using Trizol according to the manufacture’s protocol. Real time RT-PCR was performed to test BMP-2, COL-10 and MMP-13 expression. Results 1.All samples were studied and the distribution of averaged grades allowed for stratification of human samples into three groups :control ,mild OA and moderate OA. 2.Immunohistochemistry of all samples showed a significant up-regulation of β-catenin and MMP-13 in both the mild and moderate groups compared with the normal control.3.Expression of chondrocyte marker genes such as BMP-2, COL-10 and MMP-13 were significantly increased in moderate OA group. Conclusions 1.The protein level of β-catenin and MMP-13 was increased in OA patients.2.Expression of chondrocyte maturation marker genes, such as BMP-2, COL-10, and MMP-13 were significantly increased in articular cartilage of OA patients. 3.Overexpression of β-catenin in articular chondrocytes leads to premature chondrocytes differentiation and the development of OA. These results establish a strong association between human OA and β-catenin expression.

    Inhibition of cell apoptotic volume decrease and apoptosis by blocking Cl- channels in nasopharyngeal carcinoma cells

    Abstract Objective: To investigate the roles of chloride channels in apoptotic volume decrease (AVD) and apoptosis in nasopharyngeal carcinoma cells (CNE-2Z). Methods: The changes of cell volume were monitored and analyzed by the time-lapse imaging technique. Cell apoptotic rates were measured by using nuclear staining analysis by Hoechst 33258. Results:The results showed that 5-Fu (100?mol/L) induced an early cell volume decrease just in a few minutes. The cell volume was decreased to 88.2% of the control volume in 120 min. The apoptotic rate induced by 5-Fu was 49.2% 48 h after treatments. Chloride channel blocker 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) prevented appearance of 5-Fu-induced cell shrinkage and apoptosis by 79.1% and 74.9%, respectively. Conclusion: Chloride channel blockers could inhibit 5-Fu-induced apoptotic cell volume decrease and apoptosis. The results suggest that chloride channels may contribute to 5-Fu-induced apoptosis by activation of chloride channels and inducing apoptotic volume decrease in CNE-2Z cells. The activation of chloride channels play an important role in the anti-tumour action of 5-Fu.

    Biological behavior of HepG2 hepatoma cells transfected by XPD in viro

    Objective To investigate the role of wild—type XPD in HepG2 hepatoma cells,its relationship with P52 and P21.And the apoptosis mechanism of HepG2 hepatoma cells. Methods The experiment were classified three groups: pEGFP-N2-XPD group, pEGFP-N2 group, with the same genetic background and the generations of HepG2 cells as normal group. pEGFP-N2 and pEGFP-N2-XPD were transfected into HepG2 cell line by Lipofectamine 2000. The expression of green fluorescent protein(GFP) were observed with fluorescence microscope. The expression of wild-type XPD,P52 ,P21 was detected by RT-PCR,Western blot.Cell growth was detected by MTT. Results ①HepG2-pEGFP-N2-XPD and HepG2-pEGFP-N2 express the green fluorescence protein which could be detected under the immunofluorescence microscope.②RT-PCR The relative expression of P21 mRNA, XPD mRNA in the HepG2, HepG2-pEGFP-N2-XPD and HepG2-pEGFP-N2: the relative expression of P21 mRNA, XPD mRNA in the HepG2-pEGFP-N2-XPD was significantly higher than other two controls(P<0.01). Otherwise, the relative expression of P52 mRNA was significantly lower than other two controls (P<0.01), the difference of two controls was not significant(P>0.05)③Western blot The relative expression of XPD protein,P21 protein were significantly higher than other two controls (P<0.01).Otherwise,the relative expression of P52 protein was significantly lower than other two controls(P<0.01),the difference of two controls was not significant(P>0.05).④MTT The -XPD was much more decreased than other two controls (P<0.05),the other two controls were not different(P>0.05). Conclusions The wild-type XPD could inhibit the proliferation of HepG2 cells in vitro. The wild-type XPD could decrease the expression of P52 and increase the expression of P21.

    【Abstract】 Objective Study the expression and the Significance of PRL-3 and Microvessel dencity which was marked with CD l05 in laryngeal Squamous Cell quamous Cell carcinoma and 25 normal tissues was investigated by carcinoma. Methods :Expression of PRL-3 and MVD in samples of 69 laryngeal Susingmmunohistoehemieal methods of SABC respectively. Results:The expression of PhosPhatas of regenerating liver-3( PRL-3 )and MVD has close correlation with clinic stage of laryngeal carcinoma and lymph-node metastasis in carcinoma (p<0.05). PRL-3 hasSignificant correlation with the degree of histological differentiation but Microvessel dencity has no correlation with the degree of histological differentiation; Significant correlation could be established among the expression of microvessel density and PRL-3 (P<0?05).Conclusions:PRL-3 may be involved in laryngeal squamous cell carcinoma angiogenesis and promoted laryngeal squamous cell carcinoma metastasis.MVD had a close correlation with clinic stage of laryngeal carcinoma and lymph-nodemetastasis, It may be a valuable index to evaluate the prognosis of patients with laryngeal squamous cell carcinoma,also, MVD can provide the basis for anti-vascular therapy

    The role of Interleukin-17 and CRP in patients with chronic obstructive pulmonary diseases and their correlations

    Objective By observing the serum level changes and their correlations of Interleukin-17(IL-17) and C reactive protein(CRP) in patients with chronic obstructive pulmonary disease(COPD) at acute exacerbation phase and stable phase,to investigate the role of IL-17 and CRP in pathogenesy of COPD in order to offer evidence for the judgement of illness state and treatment of COPD.Methods The serum levels of IL-17 and CRP were determined in 100 COPD patients at acute exacerbation phase and stable phase by double antibody enzyme-labeled immunosorbent assay (DA-ELISA) . Pulmonary function of all research subjects were tested and calculated for the percentage of FEV1 in prediction value (FEV1%Pre). Results The serum level of IL-17 and CRP in COPD patients at acute exacerbation phase was significantly higher than that at table phase(P<0.01). The serum levels of IL-17 were in negative correlation with FEV1%Pre in COPD patients at acute exacerbation phase, coefficient of correlation r=-0.437, P<0.05;IL-17 were in positive correlation with CRP at acute exacerbation phase, coefficient of correlation r=0.582, P<0.05;at stable phase,coefficient of correlation r=0.481, P<0.05.Conclusions IL-17 might be associated with pathogenesy of COPD,especially play an role of acute exacerbation.It can substitute CRP to monitor the activity of COPD.

    The preliminary study on the imbalance ofTh1/Th2 chemokines in patient with Graves disease

    Objective: To explore the changes of Th1/Th2 chemokines CXCL10、CCL22 and its clinical significance in the patient with Graves disease. Method: 76 outpatient with Graves disease in different stage of treatment were selected,including untreated group、remission、stable group and control group. We collected the datas including thyroid function、thyroid antibodys,as well as the level of serum CXCL10 and CCL22 detected by ELISA. Results: Serum CXCL10、CCL22 level were significantly increased in untreaded group(p<0.001),after treatment CXCL10、CCL22 levels the ratio(CXCL10/CCL22) were significantly decreased(p<0.01),and in the remission group the levels were lower than normal(p<0.01). The levels of CXCL10、CCL22 were positively correlated with FT4( r=0.538,r=0.389, respectively). Conclusion: Th1/Th2 chemokines CXCL10、CCL22 were involved in the immune regulation of Graves

    Trigeminal small-fiber dysfunction assessed with contact heat evoked potential in patients with diabetes mellitus

    【Abstract】 Objective To investigate the application value of the contact heat evoked potential (CHEP) for the trigeminal small-fiber dysfunction in patients with diabetes mellitus. Methods Heat stimuli was delivered to the skin above the eyebrow with a contact heat stimulator in 46 diabetic patients and 20 normal subjects. The peak latency of the negative component and the peak-to-peak amplitude of CHEP was recorded at the vertex (Cz). Blink reflex (BR) and direct electrical stimulation of the facial nerve were performed in the same side. The patients with diabetes mellitus were divided into the group with normal conduction (DMN group) and the group with abnormal conduction (DMA group) according to the results of the nerve conduction of the limbs. Results In DM group and the control group, the peak latency of the negative component of CHEP was (382.7±36.8) ms vs (342.2±20.8)ms (t=5.465, P=0.000), the common logarithm of the peak-to-peak amplitude of CHEP was (1.66±0.21) vs (1.84±0.24) (t=2.922, P=0.005). There were no statistical differences between two groups in the latencies of R1、R2、R2′ in BR and R/D ratio(the latency of R1/ the latency of direct electrical stimulation of the facial nerve); The peak latency of the negative component of CHEP in DMN group was longer than that of the control group. The common logarithm of the peak-to-peak amplitude of CHEP was lower in DMA group than that of DMN group. Conclusion Limb nerves may be involved in patients with diabetic neuropathy, the trigeminal nerve can also be involved coexistence of limb nerve injuries; The trigeminal small-fiber dysfunction may occur when BR or nerve conduction of the large myelinated nerve was normal, and becomes obvious when the former was abnormal. CHEP has potential clinical application value in the evaluation of the cranial nerve small-fiber function in patients with diabetes mellitus .

    Clinical Research of Prevention prophase oral cavity infection and deep fungal infection after liver transplantion

    [Abstract] Objective To observe the clinical effect of durg combination use in oral cavity on prevention of prophase oral infection and deep fungal infenction after liver transplantion. Methods To choose 162 liver transplantation patients were randomly divided into observation groupA(n=57), observation group B(n=62)and control group C(n=43),For the experiment group , alternately use 3% sodium bicarbonate gargle and 0.1% cetylpyridinium chloride gargle to cavity nursing, And observation group B use 50000 U/ml nystatin aqueous suspension combine with 0.1% cetylpyridinium chloride gargle in turn to oral nursing, control group C only with normal saline nursing oral cavity ,then the oral complication in every groups was investigated,and make plasma (1-3)-β-D-glucan test. Results The oral complication and (1-3)-β-D-glucan test level were no difference between observation groupA and observation group B(P>0.05);but showed significant differentces between observation groupA and observation group B to contrast control group C (P<0.05). Conclusions There were important meaning in use 0.1% cetylpyridinium chloride gargle combine with 3% sodium bicarbonate gargle and 50000 U/ml nystatin aqueous suspension respectively to oral nursing, on prevention of prophase oral infection and deep fungal infenction after liver transplantion.

    Detection of Serum Diamine Oxidase Activity in Chronic Urticaria and its Significance

    Objective: To detect the serum activity of diamine oxidase in chronic urticaria and analyze its correlation with urticaria severity, so as to explore its significance in the pathogenesis of chronic urticaria. Methods: Serum diamine oxidase activity were determined by spectrophotometry in 68 patients with chronic urticaria and 54 healthy control. The activity and the urticaria severity were compared by using linear correlation. Results: The serum diamine oxidase activity in 1 hour, 1.5 hour and 3 hours was lower than that of healthy control. There were significant difference between 1.5/ 3 hours and control group (P<0.01). The correlation between the activity and the urticaria severity is significant (P<0.05). Conclusion: The serum diamine oxidase activity in chronic urticaria patients decreased, which might be implicated with the chornic pathogenesis and urticaria severity.

    Abstract Objective:To investigate the law and the reason of the electrotonic modulated T wave(EMTW) after radiofrequency ablation(RFCA) in patients of preexcitation syndrome.Methods:By observing the EMTW in ECG before and immediately after RFCA in 109 cases who had preexcitation syndrome,to conclude the law of EMTW in position and amplitude.Rusults: EMTW were detected in in 78 of the 109 cases,the rate of EMTW is more lager in right free wall、right posterior septal、right posterior free wall、left anterior free wall、left free wall than other positions,and the EMTW is not only inverted.Conclution:The position and amplitude of the EMTW is relative with the positions of the pathway.

    Treatment of the varus knee osteoarthritis by total knee arthroplasty

    Objective To analysis the experiences and results of total knee arthroplasty on the varus knee osteoarthritis. Methods From January 2006 to May 2009, 800 patients (939 knees) with varus osteoarthritis were treated by TKA in our hospital, male 246 patients (296 knees), female 554 cases (643 knees); aged 45 to 85. Based on modified Krackow classification: Ⅰ type: 480 patients (512 knees); Ⅱ type 225 patients (278 knees); Ⅲ type: 93 cases (147 knees); Ⅳ type 2 cases(2 knees). The postoperative imaging and KSS score and complications were analyzed. Results The average unilateral knee surgery time was 60±7 min. 66 knee underwent bone defect filling. After 1-4 years follow-up no prosthesis loosening was found except 5 cases of deep infection. Imaging: preoperative varus angle 6°~ 30°, average 18.6°±1.2 °; weight-bearing X-ray post TKA revealed TFA 5°~7 °, average 5.8°±0.6°. KSS score: After 4 years, the clinical score and functional scores were 86.4±8.4、86.0±9.7, significant difference was found compared with presurgery (P<0.05); no significant difference was found in the three groups classified by varus angle (P>0.05). 52 cases occurred different degrees of complications in 800 patients (6.5%). Conclusion TKA is an effective treatment method for the end stage of varus-type osteoarthritis of the knee, the function restored well in the short time follow up, and modified Krackow classification had some indicating value in the soft tissue release and bone reconstruction.

    The Value of Gas Partial Pressure Evaluate in Pleural Cavity to Pneumothorax Patients with Blocked Drainage Tube

    Abstract Objective:To study the guidance value of gas in pleural cavity partial pressure evaluate twice of pneumothorax and arterial blood gas analysis in patients with blocked drainage tube for further treatment. Methods: The patients with intercostal tube and water seal bottle percutaneous drainage, but drainage tube blocking and ipsilateral lung is not reexpansion. Extracted gas in the pleural cavity twice, do gas partial pressure evaluate and arterial blood gas analysis of radial artery. In accordance with [Pp(CO2)], [Pp(O2)], [Pp(CO2)] / [Pp(O2)],[Pp(CO2)]and[p(CO2)judge different type of pneumothorax in patients with the blocked drainage tube, choose the way to treatment. Result: The group of 49 patients, 30 cases judged closed pneumothorax, 16 cases of traffic pneumothorax , 3 cases of tension pneumothorax; coincidence rate is 97.96%(48/49). Including tension pneumothorax and traffic pneumothorax are 100%,closed pneumothorax is 96.67%(29/30),97.96%(48/49)patients get a scientific treatment. Conclusion: To extract the gas of pleural cavity and evaluated twice, according to the results determine the type of patients with pneumothorax is more accurately, the exhaust in order to take a scientific and effective measures.

    The relationship between the cardiac function and the level of thyroid hormone, B-type natriuretic peptide(BNP) in the congestive heart failure patients

    Objective: To analysis the relationship between the cardiac functional grading and the level of thyroid hormone, B-type natriuretic peptide(BNP),Left ventricular ejection fraction(LVEF).Methods:The 150 patients have been divided into 4 groups,the chronic congestive heart failure(CHF) group and the control group.All the patients have been measured the levels of free triiodothyronine (FT3),total triiodothyronine(TT3),free thyroxine (FT4), total thyroxine(TT4), thyroid-stimulating hormone(TSH) and BNP in the plasma by the radioimmunoassay and the immunofluorescence method.Meanwhile to monitor the LVEF by Echocardiography.Result: The level of the FT3、TT3 and LVEF are lower in the CHF group than in the control group with significant difference,while the BNP level are higher in the CHF group with significant difference; According to the cardiac functional grading,there are significant difference in the FT3、TT3,BNP and LVEF in patients. Conclusion: The level of the thyroid hormones,LVEF and BNP can be the indicators to evaluate the extent of CHF.

    Objective:To test differences of different thickness of PermaCem-Dual on the effect of Crown’s Retention. Methods: 30 extracted human maxillary premolars were selected for orthodontics purpose and randomly divided into three groups, tooth structure was prepared according to uniform standards,the metal crowns were produced and the adhesive thickness were Group A、Group B、Group C, ten of each group. All teeth were tested for tensile strength after cyclic loaded by increasing Ladder-type load 10000 times.The data was analysised with SPSS 16.0 on single-factor analysis of variance (ANOVA) and SNK test between groups.Results: the tensile bonding strength of Group A was significantly greater than Group B and Group C,the difference was significant .(P <0.05). Conclusion: As the bond thickness increases, the tensile bond strength was decreased.

    Abstract Objective: To study the role of Ca2+/Calmodulin- dependent protein kinase II (CaMK II) in the mechanism of contract-stimulated glucose transport 4 translocation in skeletal muscle cells. Methods: C2C12GLUT4myc myotubes were divided into two groups with or without carbachol (acetylcholine receptor agonist) treatment. CaMK II inhibitor KN93 or KN62 was added to the medium prior to treatment, respectively. Then cell surface levels of GLUT4myc were measured by enzyme linked immunosorbent assay (ELISA). The phosphorylation of CaMK II was detected by immunoblotting after pre-incubation with KN93 before treatment. Results: Carbachol increased phosphorylation of CaMK II which was reduced by KN93. KN93 and KN62 inhibited the gain of cell surface GLUT4myc induced by Cch. Conclusion: CaMK II is the downstream signal of Ca2+ and mediates contraction-stimulated GLUT4myc traffic in skeletal muscle cells.

    【Abstract】Objective To explore the effects of thyroid hormone on the expression of homeobox gene Nkx2.2 in child rat and the relationship between gene expression and hormone level by supplying their hypothyroidism pregnant mother with thyroid hormone. Method 240 Wistar rats,half male and half female, were randomly divided into eight groups: normal control group,hypothyroidism group, hypothyrodism groups which were supplied with thyroid hormone in different doses in early stage(1-17d),hypothyrodism groups which were supplied with thyroid hormone in different doses in late stage(18-20d) .All the rats were fed with low-iodine food.The normal control group was given KIO3 solution and the other groups were given deionized water.After three months’raising,each group was drawn materials from the 17-day fetal rats,new-born and 20-day old offspring,and was detected the content of Nkx2.2 mRNA in afterbrain tissue by real-time fluorescence quantitative PCR techniques. Results① The expression of Nkx2.2 mRNA have significant difference between groups in 17-day fetal rats,new-born and 20-day old offsprings(F=7.080、19.007、24.652,P﹤0.01)②The time factor and dosage factor have influence on Nkx6.1 mRNA expression (F=6.992、5.939,P﹤0.01) ③Time factor and dosage factor have interaction(F=5.610,P﹤0.01)④ the expression level in low-iodine group were higher than normal group during the same period(P﹤0.05),and it increased gradually with age.⑤ The expression of Nkx2.2 mRNA in new-born and 20-day old offsprings which were supplied with thyroid hormone in high and medium dosage in late stage is close to the normal group at the same period(P﹥0.05).Conclusion The expression of Nkx2.2 in child rats is highly related to the supply dosage and supply time of thyroid hormone in hypothyrodism preganant rats.

    The effect of electroacupuncture on changes of the phosphorylation of capsaicin receptor in chronic morphine tolerance rats

    Objective:To investigate the effect of electroacupuncture(EA) on morphine tolerance of rats and on changes of the phosphorylation of transient receptor potential vanilloid subfamily member 1(TRPV1),which is a member of capsaicin receptor. Method:The 24 healthy male SD rats were randomized into 3 groups (n=8): they were administered respectively intrathecally 10 L saline for group N; 10 g morphine for group M; 10g morphine and 2HzEA on "Zu-San-Li" and "Yang-Ling-Quan" after the first administration of morphine every day for group H.Administration of the three groups were all twice daily for 7days.Mechanical withdrawal threshold (MWT) and thermal paw withdrawal latency (PWL) of rats was examined to evaluate their mechanical hyperalgesia and heat hyperalgesia. Take the L4~5 dorsal root ganglion(DRG) on the 8 day of administration ,and the expression of total TRPV1 and phosphorated TRPV1 in DRG was measured by western-blot.Result: The rats in group M were developed morphine tolerance .2Hz EA can alleviated the formation of morphine tolerance.The expression of total TRPV1 in group M were significantly higher than that in group N and group H,while there's no significant difference between group N and group H.The phosphorylation of TRPV1 in DRG of group M were increased significantly,while the expression of phosphorylated TRPV1 in group H were significantly lower compared with that of group M.Conclusion:2Hz EA has inhibitory effect on morphine tolerance and tolerance-associated thermal hyperalgesia,it may be through the inhibition of phosphorylation of TRPV1 in DRG.

    Objective To explore the damage of sprial ganglion cell inner ear after 60Co irradiation. Methods Forty guinea pigs were randomly divided into 5 groups.The right ear was radiation ear,the left ear was controll ear.The guinea pigs were sacrificed in 1、4、7、14 and 30 days after irradiation,All specimens were dehydrated,embeded in paraffin and serially cutted into 5 m slices. Sections were stained with haematoxylin and eosin and immunohistochemistry for light microscopic examination.Results It was observed that irradiation ear promoted degeneration in spiral ganglion cells at these times(p<0.01).There was a singnificant difference on radiation-induced optical density value of caspase-3 in spiral ganglion(p<0.01).Conclusions irradiation might cause spiral ganglion cell damage. Caspase-3 cascade might be involved in the course of cell damage.

    Protective Effect of Rapamycin On The Renal of Diabetic Rats

    Objective: To evaluate protective effects and possible mechanism of rapamycin (RAPA) on the renal of diabetic rat. Methods: 36 rats were randomly divided into 3 groups: control group: 12 rats; diabetic group: 12 rats; RAPA treatment group: 12 rats. Diabetic model rats were made by injecting streptozotocin into abdomen. Biochemical changes and histological changes were obsvered. The expression of proliferating cell nuclear antigen (PCNA) and Ⅳ collagen (COL-Ⅳ) were detected by westhern blot. Results: Compared with the control group, the level of Glu, Scr, BUN, and UPro were significantly increased in diabetes group. Kidney hypertrophy, glomerular hypertrophy, basement membrane thickness were significantly increased renal cortex and COL-Ⅳ、PCNA protein expression was up. After RAPA intervention, the above parameters, except blood glucose were partially inhibited. Conclusion: RAPA can reduce kidney hypertrophy in diabetic rats, inhibition of mesangial extracellular matrix accumulation, reduced the expression of COL-Ⅳ and PCNA, delay progression of diabetic nephropathy.

    The empirical study on Chronic Pancreatitis gastrointestinal motility and the intervention of trimebutine maleate

    ABSTRACT Objective: To explore the changes and mechanismof gastriointestinal motility in the rats with chronic pancreatitis and the intervention effect of trimebutine maleate. Methods: Thirty wistar rats were divided into control,model and trimebutine maleate group randomly(n=10). Rats in model group were used to induce chronic pancreatitis by injecting oleic acid into the pancreatic duct. The control and model groups, administered intragastrically with physiological saline everyday. the Trimebutine Maleate group, administered intragastrically with Trimebutine Maleate everyday. The experiment period of each group are twe weeks. Eight weeks later, bowel movement coefficient and serum cholecystokinin(CCK) were detected, contractility of gastrointestinal circular muscle stripe and the contractile response of dispersed gastrointestinal circular smooth muscle cells stimulated by Aceylcholine (Ach) were measured. RT-PCR was used to detect the expression of inducible nitric oxide synthase (iNOS), mRNA within gastrointestinal muscularis , the level of nitric oxide (NO) in cell cultures was quantified. Results: The bowel movement coefficient, gastrointestinal circular smooth muscle stripes contractility and contractile response of dispersed circular smooth muscle cells stimulated by Ach in model group were declined obviously than the control(P<0.05) while the levels of serum CCK, iNOS mRNA expression and NO production were significantly elevated(P<0.05). Conclusions: Elevated CCK secretion caused by CP up-regulated the expression of iNOS mRNA and increased the output of NO within the gastrointestinal muscularis. It might be one of the mechanisms of gastrointestinal motility dysfunction in CP that NO inhibited the contractility of gastrointestinal circular smooth muscle. The trimebutine maleate can improve the gastrointestinal motility dysfunction caused by CP efficiently, by inhibiting the release of CCK.

    The Effect of Laparoscopic Preconditioning Against Laparoscopy-induced Lung Injury in Rabbits.

    Abstract Objective: Pneumoperitoneum performed to facilitate laparoscopy is associated with splanchnic perfusion, ischemia/reperfusion (I/R) injury, and oxidative stress. The current study evaluated the effect of laparoscopic preconditioning against laparoscopy-induced lung injury. Methods: 40 rabbits were randomly assigned to following groups. The control group(N) was subjected to a sham pneumoperitoneum. The short time group (S)was subjected to 1 hour of pneumoperitoneum. The long time group (L)was subjected to 3 hours of pneumoperitoneum.The laparoscopic preconditioning group(P) was subjected to 5 min of insufflation and 5 min of desufflation followed by 3 hours of pneumoperitoneum. After 20 min of desufflation subsequent to cessation of pneumoperitoneum, blood, lung, and bronchoalveolar lavage fluid(BALF) samples were obtained from every rabbit of the groups.white blood cell(WBC) count in BALF were detennined .The myleopemxidase (MPO) activity and wet to dry weight ratio(W/D)were also measured.Results: lung permeability index(LPI) and WBC count in BALF of group (L) are higher than the other three groups(p<0.01),The lung tissue MPO activity of group (P) is much higher than group(S) and group(N) (p<0.01),but it is lower than group (L).Conclusion: Lung injury induced by more than 3 hours of pneumoperitoneum is more effective than 1 hour of pneumoperitoneum. laparoscopic preconditioning is one of the ways which can inhibit the accumulation of PMNs in lung tissue,and thus attenuate the inflammatory response induced by more than 3 hours of pneumoperit -oneum rabbit lungs.     The effects of caffeic acid phenethyl ester on the expression of VEGF and ICAM-1 in the retinae of diabetic rats

    [Abstract] Objective To observe the effccts of caffeic acid phenethyl ester on the expression of VEGF and ICAM-1 in the retinae of diabetic rats. Method Thirty diabetic SD rats Inducided by streptozotocin were divided into three groups, diabetic group (DM, n=10) ,low dose group of caffeic acid phenethyl ester(CAPE5,n=10). High dose group of caffeic acid phenethyl ester(CAPE10,n=10).Ten else rats were in normal control group.All of the rats were executed 3 months later , both side of the retinae were homogenized in each rat, and the protein level of VEGF and ICAM-1 was detected by Elisa. Results After peritoneal injection of caffeic acid phenethyl ester ,the protein level of VEGF and ICAM-1 were decreased ,in the low dose group, the protein level of VEGF and ICAM-1 were decreased by 40% and 29%,when the protein level of VEGF and ICAM-1 were decreased by 42% and 44% in high dose group. Conclusion caffeic acid phenethyl ester can reduce the expression of VEGF and ICAM-1 in retinae of diabetic rats.

    Risk factor for development of early acute renal failure in patients with open-heart surgery

    Objective:To determine the risk factor for development of early acute renal failure in patients with open-heart surgery.Methods:1660 ASAⅡ-Ⅳ patients in our hospital from October 2005 to January 2010 under open-heart surgery were divided into acute renal failure group (ARF group) and no acute renal failure group (NARF group).ARF group had 78 cases and NARF group had 1582 cases.We compared the possible risk factor between groups and then applied the logistic regression to identify the risk factor of ARF.Result:Logistic analysis indicated that serum β2-microglobin higher than normal value before operation, the cardiopulmanory bypass time and the low cardiac output syndrome after operation were closely correlated with development of early ARF after open-heart surgy.Conclusion: Serum β2-microglobin higher than normal value before operation, the cardiopulmanory bypass time and the low cardiac output syndrome after operation are the risk factors for early ARF after open-heart surgy.

    Mid-urethral sling combined with Modified pelvic floor Reconstruction in the treatment of female stress urinary incontinence

    Abstract Objective:To asses the safety and efficacy of the transobturator tape (TOT) combined with modified pelvic floor reconstruction for surgical treatment of female stress urinary incontinence. Methods:Retrospectively analyzed from January 2008 to December 2009 and 31 women with stress urinary incontinence concomitant pelvic organ prolepses underwent the TOT procedure and modified pelvic reconstruction. 31 women are aged from 43~76 years ;mean age 54.29 years .There disease course raged from 1 to 40 years ;mean 10.31 years. The follow-up time is from 6 to 30 months, mean 18.3 months. Therapeutic effects were assessed by Grouts-Blaivas and POP-Q outcome score .The results of operations, complications of operation were observed. Results:The operations were operated under epidural anesthesia . Mean operation time was 70.2 minutes .Mean blood loss was 60.8 ml. Early complications included femoral puncture site pain in 4 cases, dysuria in 2 cases and 1 cases of urinary posture change. There was no long-term complication. POP-Q and Grouts-Blaivas score to evaluate the efficacy and symptoms disappeared completely 30 cases, long-term cure rate of 96.8%, the overall rate of satisfaction 100%, without failing case. Conclusions:TOT procedure combined with modified pelvic floor reconstruction is a safe and effective approach, the short—time effect is wel1.However, the long time affect need more cases of observation.
