    Abstract ( 1390 )   PDF (3030KB) ( 2016 )
ABSTRACT Objective: Study the mechanism of action of hypoxia inducible factor-1α(HIF-1α), signal transducers and activators of transcription 3(STAT3) in lymphatic metastasis of colorectal carcinoma. Methods: The levels of HIF-1α, STAT3 and VEGF-C mRNA transcription and protein expression of adenocarcinoma and surgical margin tissues gotten from 60 cases with CRC were determined by real-time quantitative PCR using fluorescence and immunohistochemistry, as well as lymph vessel density was counted using anti-podoplanin monoclonal antibody. And correlations of above indexes and lymphatic metastasis of colorectal carcinoma were analyzed. Results: The levels of HIF-1α, STAT3 and VEGF-C of tumor tissues were significant higher than that of surgical margin in either mRNA transcription or protein expression(P<0.05). The levels of HIF-1α, STAT3 and VEGF-C of tumor tissues were more significant relative not only in either mRNA transcription or protein expression but also in lymphatic metastasis group(P<0.05). Conclusion: The expression of lymphatic vessels system in colorectal carcinoma is higher than that in surgical marginal tissue. HIF-1α,STAT3 and VEGF-C are the key regulators in lymphatic vessel formation of colorectal carcinoma, and are closely related to lymphatic metastasis.

After the MSCs were infected with Cbfa1 through non-viral vector FuGene HD, the expressions of mRNA for osteoblast gene markers such as Cbfa1, alkaline phosphatase, osteocalcin, osteopontin and type I collagen were analyzed with real-time reverse transcriptase polymerase chain reaction (real-time RT-PCR ) at 7d and 14d. Alkaline phosphatase ( ALP ) activity and expression of osteocalcin were measured at 1, 2, and 3 weeks after transfection. Results: The real-time RT-PCR indicated that all the osteoblast gene markers were up-regulated in the Cbfa1 treatment group. The morphous of hBMSCs modified with Cbfa1 were similar to osteoblasts, and expressed ALP and osteocalcin. Conclusion: FuGene HD-Cbfa1 can directly promote the osteogenic differentiation of hBMSCs.

    2011, 39 (8):  748-750 .
    Abstract ( 1229 )   PDF (2131KB) ( 2262 )
Abstract Objective: To investigate the clinical characteristics and post-operation outcome in infertile women following tubal pregnancy. Methods: We retrospectively analyzed the data of consecutive in-hospital female infertility cases which the last pregnancy was tubal pregnancy and rule out male infertility from Dec 2008 to Aug 2009. Results: A total of 602 female infertility cases was admitted to Pingjin Hospital, Medical College of Chinese People’s Armed Policed Forces during this period. In these patients, infertility following tubal pregnancy accounted for 15.1% (91/602). All of them were managed with a combined interventional procedure (laparoscopy and hysteroscopy) in our department. Under laparoscopy the gross pathological changes of fallopian tubes involved normal appearance (20.7%), dropsy and atresia (23.3%), adhesions (59.3%), partial resection (10.7%), and other diseases of pelvic cavity (14.3%). After laparoscopic operation, infertility case with tubal blockage accounted for 63.3% under hysteroscopic tubal catheterization. The patency rate of blocked fallopian tubes after hysteroscopy directed guidewire tubal catheterization was 66.3%. Conclusion: previous tubal pregnancy which cause abnormal oviduct morphology and tubal blockage is the important reason of female infertility. Laparoscopy and hysteroscopy can advantage infertility women due to previous tubal pregnancy to resume reproductive.

Percutaneous transluminal angioplasty for the treatment ofchronic arterial occlusive disease below the knee
    2011, 39 (8):  750-751 .
    Abstract ( 1353 )   PDF (1430KB) ( 2261 )
Abstract Objective: To evaluate the clinical effects of Percutaneous transluminal angioplasty(PTA) in the treatment of infrapopliteal arterial occlusive disease. Methods: The data of 39 patients(43limbs) who underwent infrapopliteal PTA from Jun 2008 to July 2009 in our hospital were reviewed. Results: Procedures were performed on 40 of 43 limbs(92%) successfully. During a mean follow up time of (8 range,3~18) months, ankle-brachial index (ABI) was increased from 0.41±0.12 before PTA to 0.82±0.05 after PTA(P<0.01).There was no death in hospital The limb salvage rate and complication rate was 91.4% and 13%(6/43),respectively.Conclusion: Infrapopliteal PTA can be performed with favorable clinical results in patients with chronic occlusive disease and reduce the amputation rate.PTA should be considered as an effective and safe therapy for lower extremity ischemia caused by infrapopliteal occlusive disease.

Morphological features of prostate intraepithelial neoplasia and its significance
    2011, 39 (8):  768-739 .
    Abstract ( 1553 )   PDF (2223KB) ( 2238 )
【Abstract】 Objective To investigate the morpaology of prostate intraepithelial neoplasia(PIN) and its significance. Methods 103 specimen of prostate biopsy, including 26 cases with prostate carcinoma and 77 cases of benign prostate hyperplasia (BPH), were collected. Morphological features were reviewed. The expression of 34BE12, P63 and p504s were assayed in all patients by immunohistochemistry. Results High grade intraepithelial neoplasia(HGPIN) was found in 23 of 26 case with prostate carcinoma. There was no low grade intraepithelial neoplasia (LGPIN) to be observed in the patients above. Of 77 cases with BPH, intraepithelial neoplasia were found in 24 cases,(including 18 cases of LGPIN and 6 cases of HGPIN ). The histological types of HGPIN were mainly cluster and micropapilla structures (accounting for 92%). Immunohistochemistry: Among 29 cases with HGPIN , the discontinuous expression of 34βE12 was observed in basal cell around gland. In those of prostate carcinoma with HGPIN, P504s expression was from poor to moderate positive in the lesions of HGPIN near carcinoma(+-++), whereas was from moderate to strong expression in those away from carcinoma(++-+++); In the patients who were BPH with HGPIN, the positive expression was only observed in the two of 6 cases. The positive ratio of P504s was 82% in the case with HGPIN. Conclusion HGPIN is only precancerous lesion of prostate carcinoma. It must be differentiated from atypical hyperplasia of small alveolar gland and intraductocarcinoma of prostate; P504s is specific to HGPIN and carcinoma of prostate. The expression of it is probably associated with the progression of these tumors.; The correct diagnosis of HGPIN must depend on morphologic features and immunohistochemistry stain.
