    Construction and ldentification of Cell Division Cycle 2 Promoter Reporter Gene Vector

    Abstract Objective: To construct the luciferase reporter gene vector of cell division cycle 2 gene promoter and determine its transcriptional activity. Methods: Primers were designed based on human Cdc2 promoter sequence from UCSC software. Then Cdc2 promoter from human genome DNA was replicated. After pGL3-Basic Vector and Cdc2 promoter were digested with restriction enzymes ScaⅠand XhoⅠseparately, Cdc2 promoter was inserted into pGL3-Basic vector. The recombinant plasmid named pGL3-Cdc2-promoter was transiently co-transfected into U2OS cells with control vector pRL-SV40, then the activity of dual luciferase was detected. Results: pGL3-Cdc2- promoter was constructed successfully. Restriction analysis and sequencing proved the same as the design . Promoter activity of pGL3-Cdc2-promoter in U2OS was 32 times than that in negative control group. Conclusion: pGL3-Cdc2-promoter can be transcribed and activated in U2OS cells. This research provided an important basis for antitumor drug screening and evaluation.

    To Investigate the Genetic Polymorphisms of STR Loci D18S53, D18S58 and D18S488 in Tianjin Han Fetus

    Abstract Objective: To investigate the genetic polymorphisms of 3 short tandem repeat (STR) loci D18S53, D18S58 and D18S488 on chromosome 18 in fetus of Tianjin Han population. The aim of the study was to discuss the evaluation criterion of quantitative fluorescence polymerase chain reaction (QF-PCR) and to provide basic data for the use of 3 STR loci in gene diagnosis and prenatal gene diagnosis of Edward syndrome (ES). Methods: Samples of villus and 374 amniotic fluid from gravida of Tianjin Han population were collected. QF-PCR and ABI PRISM 377 sequence were used. The frequencies of the genotypes were tested with Hardy-Weinberg equilibrium. Genetic analysis was performed to conclude some data of population genetics such as the frequency of the alleles, the heterozygosity of observation(Ho), the polymorphism information content(PIC), the probability of discrimination power(DP), and the probability of exclusion(PE). Results: 15, 13 and 15 alleles of D18S53, D18S58 and D18S488 were observed respectively. The frequencies of the genotypes were in good agreement with Hardy-Weinberg equilibrium. The Ho of 3 STR loci were 0.797, 0.847 and 0.792. The PIC were 0.81, 0.75 and 0.73. The DP were 0.944, 0.901 and 0.881. The PE were 0.593, 0.689 and 0.585. Conclusion: It was the favorable genetic markers of chromosome 18 that D18S53, D18S58 and D18S488 STR loci were, so they can be used in prenatal gene diagnosis of ES.

    Construction of Recombinant Plasmid of Human RIP3 Gene and Its Expression effects on the death of Breast Cancer MCF7 Cells

    Abstract: Objective: Construct the recombinant RIP3 overexpressed plasmids and transfect them in breast cancer MCF7 cells. Identify the expression and localization of fusion protein, as well as its effect on the death way of MCF7 cells. Methods: The expression of RIP3 mRNA in four breast cancer cell lines and one mammary epithelial cell was detected by reverse transcription polymerase chain reaction (RT-PCR). The RIP3 coding sequence was amplified by polymerase chain reaction and subcloned into mCherry vector to construct recombinant plasmids. The plasmids were transfected into MCF7 cells by lentivirus after DNA sequencing, then screened by basticidin (4 mg/L) for 1 week. The efficiencies of RIP3 expression were validated by Western blotting. The death way of mCherry-RIP3-MCF7 cells was observed under the treatment of TNF-αand zVAD. Results: The lowest expression of RIP3 mRNA was seen in MCF7 cells. The sequencing results validated the well recombinant plasmids. The expression of mCherry-RIP3 fusion protein with a molecular weight of 85 ku was detected by Western blot. mCherry-RIP3 expression enhance the sensitivity of MCF7 cells to TNF-α and z-VAD induced cell death. Conclusion:The recombinant RIP3 overexpressed plasmids were successfully constructed, and the stable MCF7 cells with ectopic RIP3 transfection were obtained. The mCherry-RIP3 fusion protein was expressed in the cytoplasm and was conformed to mediate TNF-α induced necroptosis.

    Construction of siRNA expression vector targeting TLR4 gene of Vascular Smooth muscle cells and evaluation the inhibitory efficacy

    Objective: to construct a vector expressing a short hairpin small interfering RNA which targets the Toll like receptor-4 gene and to construct a TLR4 knock-downed SMC cell line with this TLR4 siRNA expressing vector. Methods: three double-strands shRNA targeting the TLR4 gene were designed, synthesized and clone into the pSilence 2.1-U6 neo vector. Positive clones were verified with double enzyme digestion and sequencing. Then the recombinants were transfected to vascular smooth muscle cells by the cationic lipid method respectively. SMCs expressing plasmid stably were screened by G418 and collected. TLR4 mRNA and protein expression was detected by RT-PCR and Weston blot. Results: the pSilence2.1-siTLR4 expression vectors were successfully constructed and a TLR4 stable knock-downed SMC cell line was establishing. RT-PCR and Weston blot analyze confirmed that the expression of TLR4 was significantly down-regulated in these infected SMC cell lines and pSilence2.1-siTLR4-1was the most efficacious recombinant vector. Conclusion: recombinant vectors carrying a shRNA targeting the TLR4 gene were successfully constructed, and a SMC cell line stably expressing TLR4 shRNA was established with this recombinant vector.

    The Significance Of Detection Of Serum HE4 In The Diagnosis Of Lung Cancer

    Objective: To discuss the value of serum HE4 in the diagnosis of lung cancer.To analysis the level of HE4 in different pathological types and TNM staging lung cancer patients. Methods: Forty-seven patients with lung cancer and thirty-one healthy subjects were selected to join this study.According to pathological type and TNM stage grouping, non-parametric test was used to compare the differences of the each group serum HE4.In pathological diagnosis as the gold standard, draw ROC curve of serum HE4 and serum CEA for diagnosis of lung cancer. Results: There were significant differences in serum HE4 assay values between lung cancer group and healthy group, and lung cancer group were higher than healthy group.There were no significant differences in serum HE4 assay values among different pathological types and different TNM staging of lung cancer patients.The AUC of serum HE4 was closed to 1,and greater than the AUC of serum CEA. Conclusion: There were no significant differences in the level of serum HE4 among different pathological types and different TNM staging of patients with lung cancer. Serum HE4 is a specific and sensitive index in lung cancer,even better than serum CEA. The detection of serum HE4 is useful for the diagnosis of lung cancer.

    Evaluation of the combination use of ultrasonographic elastography and color Doppler on breast neoplasm diagnosis and TNM staging

    Objective To investigate the effect of the combined use of ultrasonographic (US) elastography and color Doppler US on breast neoplasm diagnosis and TNM staging. Methods A cohort of 58 biopsy-proved cases, who underwent US elastographic and Doppler US imaging, were included in the current study. The diagnostic abilities of the different elastographic indices (changing rate of the diameter and area, elastographic coefficient, Doppler blood flow score) were compared by using the ROC curves. Results Comparing to other indices, the area changing rate had the highest diagnostic ability with the AUC=0.914. Moreover, the combination use of the elastography and color Doppler US could increase the AUC to 0.981. All the elastographic indices of the advanced-stage (stage Ⅱ~Ⅳ) neoplasm were high than the early-stage (stage 0~Ⅰ) ones. The elastographic and Doppler scores both increased with the progress of the TNM stages. Conclusion The elastographic area changing rate could be used to accurately diagnose the breast neoplasm. Moreover, the combined use of US elastography and color Doppler US could increase the accuracy for the diagnosis, and may also provide useful information for the TNM staging.

    The Analysis of HPV Genotypes and Related High Risk Factors in 459 Women Conducted Health Examination

    Objective To investigate the HPV infection, genotypes and related high risk factors among 459 women conducted health examination. Methods 459 cervical cytology specimens were detected with nest PCR and pyrosequencing,questionnaires were collected simultaneously and the risk factors of HPV infection were analyzed . Results 82 of 465 cases (17.9%) were infected with HPV. 7 kinds of HR-HPV were found. HPV 16 were the most common (9.8%), HPV58 (7.0%), HPV18 (5.2%) orderly. The rate of single, duplex, multiplex infection was 9.6%, 4.8%, 1.5% respectively. HPV infection was closely correlated with drinking,low economy condition,having multiple sexual partners,sexually active and cervical erosion (P<0.05). Conclusion HPV DNA genotyping can provide important reference for HPV screening at early time and the application of HPV vaccines; it also contributes to provide significant evidence for the prevention, diagnosis, and treatment of cervical cancer at early stage for high risk population.

    The relationship between hTERT gene amplification with High-risk HPV infection in cervical lesions

    【Abstract】 Objective: To investigate the relationship between the amplification of human telomerase reverse transcriptase (hTERT) gene and High-risk human papillomavirus(HR-HPV) infections in cervical intraepithelial neoplasia(CIN)and cervix carcinoma. Methods: The cytological specimens of cervix were selected from Sep 2012 to Apr 2013 in department of gynaecology second hospital of tianjin medical university。The amplification of hTERT gene of cervical epithelial cells in human normal cervical epithelium of 34 cases, CIN I of 31 cases, CIN II of 33 cases, CIN III of 34 cases and cervical carcinoma of 20 cases were detected by fluorescence in situ hybridization( FISH).All of them received HPV DNA detection. Results: With aggravated cervical lesions, the incidence of HR-HPV infection were 20.59%、54.84%、72.73%、85.29%、90.00%,respectively and there was significant difference between them (?2=41.448,P<0.05).The incidence of HR-HPV infection between normal cervical epithelium and the CIN II or the CIN III or the cervix carcinoma was significant difference(P<0.005),while there was no significant difference between the normal group and the CIN I(P>0.005). With the increase in grades of cervical lesions, the positive rate of hTERT gene were 0.00%、3.22%、18.18%、41.18%、80.00%,respectively and there was significant difference between them (?2=57.998,P<0.05).The positive rate of hTERT gene between the cervix carcinoma and normal group or the CIN I or the CIN II was significant difference(P<0.005), however there was no significant difference between the cervix carcinoma and the CIN III(P>0.005).The positive rate of hTERT gene was 33.68% with HR-HPV infection, however 8.77% without HR-HPV infection, there was significant difference between them (P<0.05).There was positive correlation between the amplification of hTERT gene and HR-HPV infection (P<0.05,r= 0.2381).Conclusion: (1)the incidence of HR-HPV infection increase as significantly as aggravating the lesions.(2)The positive rate of hTERT gene increased with the cervical lesion grade which makes it a screening test marker for CIN and may help to determine the progressive potential of individual lesions.(3)The amplification of hTERT gene may be an early stage event which related to the activation of telomerase resulted from HR-HPV infection.The detection of HPV-DNA and hTERT gene can be used as ideal tumor indexes for early cervical lesions screening.     Expression and prognosis significance of ING4 and CDX2 in colorectal cancer

    Abstract Objective:To investigate expression and prognostic significance of ING4 and CDX2 in colorectal cancer.Methods:The expressions of ING4 and CDX2 proteins were detected by immunohistochemistry in colorectal cancer tissues from 99 cases of colorectal cancer and normal colorectal tissues from 30 corresponding para-cancerous normal tissues. The data for clinic outcomes were collected. The correlations between the expressions of ING4、CDX2 and clinicopathological parameters were also analyzed. Results: The positive expression rates of ING4 and CDX2 were 67.7% and72.7% in the colorectal cancer tissues,the positive rate were significantly lower in colorectal cancer than that innormal tissue group (p<0.05).Both of ING4 and CDX2 expressions in colorectal cancer tissues were correlated with histological grade,infiltration depth,lymph node metastasis and TNM staging(p<0.05). The 5-year survival rate in the ING4 negative group(35.5%) was lower compared with the ING4 positive group (77.9%) ; The 5-year survival rate in the CDX2 negative group(48.1%) was lower compared with the CDX2 positive group (70.8%) . ING4 was positively correlated with CDX2. Conclusion: The expressions of ING4 and CDX2 are strongly associated with the carcinogenesis, development and prognosis of the cancer,which indicates that ING4 and CDX2 might be taken as prognostic markers for the colorectal cancer.

    Study of the relationship between Serum Omentin-1 and Serum high sensitive C-reactive protein in Coronary heart disease

    Objective To evaluate the correlation of Omentin-1 , the high sensitivity C-reactive protein(hsCRP) and coronary heart disease (CHD) by test the serum level of them. Methods Sixtyeight patients with CHD were divided into the unstable angina pectoris (UAP) group (n =36) and the stable angina pectoris (SAP) group (n=32), 30 normal subjects without CHD diagnosed by coronary arteriography(CAG) served as the control group. The serum level of Omentin-1 is measured by enzyme linked immune sorbent assay (ELISA) and the serum level of hsCRP is measured by turbidmetric immunoassay. Results The serum level of Omentin-1 were lower in UAP and SAP group than control group (P<0.05). The serum level of hsCRP were higher in UAP and SAP group than control group (P<0.05).The mean level of hsCRP in UAP group was differently significantly, compared with that in SAP (P>0.05) .While the Omentin-1 is the opposite. The mean level of Omentin-1 in single, double and three vessel lesion group were lower than that in control group (F=22.02,P<0.05) .The mean level of hsCRP in single, double and three vessel lesion group were higher than that in control group (F=19.78,P<0.05), but the serum levels of Omentin-1or hsCRP among single, double and three were not significant different (P>0.05). The serum level of Omentin-1and hsCRP were significantly negative correlation in Spearman correlation coefficients (r=-0.543,P<0.01). Conclusions Omentin-1 and hsCRP may correlate with CHD. However, may not reflect the severity of artery stenosis.

    The change of estradiol and TNF alpha levels in the serum of the endometriosis patients with pelvic pain

    Abstract Objective: To investigate the levels of estradiol and TNF alpha in the serum of the endometriosis patients with pelvic pain and their correlation. Methods: The levels of estradiol and TNF alpha in the serum of 113 patients with Ems pelvic pain (mild pain group of 37 cases, moderate pain group 41 cases; 35 cases of severe pain group) and 30 women without Ems (control group) were measured with ELISA. Results: No significant difference was found in the levels of estradiol and TNF alpha in the patienrs with mild pain and the normal group(P > 0.05);But in patients with moderate and severe pain,the leves of estradiol and TNF alpha were significantly higher than that of mild pain, and severe pain is higher than that of moderate pain, differences were statistically significant (P < 0.05); In mild pain, estradiol has no correlation with TNF alpha level (P > 0.05), while estradiol in moderate and severe pain is a positive correlation with the expression level of TNF alpha (all P <0.05).Conclusion: The leves of estradiol and TNF alpha in patients with moderate and severe pelvic pain of Ems were significantly higher than normal and mild pain, and the expression was positively correlated, suggests that relatively high concentrations of estradiol can induced macrophage to release TNF alpha , thus play a role in chronic pelvic pain of Ems .

    Evaluation of the effectiveness of mild hypothermia and IABP in postcardiac surgical patients with severe heart failure.

    【Abstract】: Objective To evaluate the effectiveness of mild hypothermia and IABP in postcardiac surgical patients with severe heart failure.Methods 12 postcardiac surgical patients with severe heart failure despite of the use of IABP with massive doses of catecholamine underwent mild hypothermia and IABP. Hemodynamic parameters were measured such as CI,SvO2,lac and urine volume. Results After mild hypothermia and IABP,the CI of patients improved obviously(1.6±0.2 to 2.4±0.5L/min.m2 ), Mixed venous blood oxygen saturation increased(SvO2:52±5 to 65±7%),Urine output increased obviously(1.2±0.9 to 2.9±1.9ml/kg.h),Cr decreased obviously(282±25 to 95±10 mmol/L)(P<0.05). Conclusion Combination of mild hypothermia and IABP is a effective and simple procedure for the postcardiac surgical patients with severe heart failure to improve their cardiofunction.

    The effect of amino acid configuration on the biodistribution of peptide nanofiber

    [Abstract] Objective To compare the biodistribution difference of peptide nanofibers which were self-assembled by peptide composed of L- or D-amino acids, respectively. And provide guidance for the in vivo applications of peptide nanofibers. Methods The Nap-GFFYGRGD (L-peptide) and Nap-GDFDFDYGRGD (D-peptide, F and Y were D-configuration) were synthesized with solid phase peptide synthesis (SPPS). The structure of the two peptides was identified by nuclear magnetic resonance spectroscopy (1H NMR) and high-resolution mass spectrometry (HR-MS). The two peptides could self-assemble into nanofibers during the cooling process after being boiled and the morphology of the nanofibers was observed with transmission electron microscope (TEM). The peptides were radiolabled with iodine-125 and self-assembled into nanofibers, then administrated into BALB/c mice via tail vein. The blood was collected and then the mice were sacrificed at 1, 3, 6 and 12 hours. The main organs (heart, liver, spleen, lung, kidney, stomach, large intestine, small intestine, muscle and brain) were isolated and weighed. The radioactivity of organs was detected with a gamma counter. Results The two peptides could self-assemble into nanofibers with diameter of 10-20 nanometers. The diameter and morphology of the two naofibers had no statistically significant difference. Biodistribution of the two nanofibers had statistically significant difference. The concentration of L-fiber in blood at one hour after injection was 5.9±0.30 %ID/g and maintained at a stable level during six hours after injection. While the conterpart of D-fiber was 8.17±0.32 %ID/g and then cleared rapidly from the blood. The L-fiber mainly accumulated in stomach while the D-fiber mainly accumulated in liver. Conculsion The configuration of amino acids (D/L) could affect the biodistribution of peptide nanofibers dramatically, this results may provide guidance for the medical applications of peptide nanofibers.

    Hepatic Oxidative Stress and Inflammation in Sleep Intermittent Hypoxia and Emphysema Rat Model

    Objectives To develop a sleep intermittent hypoxia (SIH) and emphysema rat model and to explore whether SHE results in more severe hepatic oxidative stress and inflammation than emphysema alone or sleep intermittent hypoxia alone .Methods Sixty male Wistar rats were assigned to 4 groups with 15 per group according to the exposure conditions. Group A (control group), sham smoke exposure(smoke exposure, exposed to smoke of 15 cigarettes twice everyday, 8weeks) and sham SIH exposure (5% O2, 8hours); Group B (Emphysema group), smoke exposure and sham SIH exposure; Group C (SIH group), sham smoke exposure and SIH exposure ; Group D (Emphysema and SIH group), smoke exposure and SIH exposure . At the end of exposure, 5 rats in each group were randomly selected for arterial blood gas analysis. In the rest 10 rats in each group, We also obtained tissue blocks of liver for pathologic scoring and measurements of liver oxidative stress (measuring hepatic oxidative stress enzymes, superoxide dismutase (SOD) activity, catalase (CAT) activity and malondialdehyde (MDA) concentration)and liver inflammation (measuring hepatic inflammatory factor interleukin -6 (IL-6) protein and tumor necrosis factor a (TNF-a) protein).Results Characteristics of emphysema were obvious in group B and D.Group B and D had higher mean linear intercept (MLI) values than group A and C (all P <0.05). group B and D had lower mean alveolar number (MAN) values than group A and C. MAN values in group D were the lowest (all P <0.05).SOD and CAT values were lower and MDA values were higher in group B, C and D than group A(all P <0.05)). SOD and CAT values were the lowest in group D ,MDA values were the highest in group D (all P <0.05); Histological scores of liver , IL-6 and TNF-α values were higher in group B, C and D than group A , group D were the highest (all P <0.05) .Conclusions Emphysema and SIH have synergistic action in causing of hepatic oxidative stress and inflammation.

    Effects of dexamethasone on bone mineral density and body composition in rats

    Objective To investigate the effects of different doses of dexamethasone (Dex) on bone mineral density (BMD) and body composition in Sprague-Dawley (SD) rats. Methods Forty female 3-month-old SD rats were divided into four groups by the doses of Dex, control group (Cont, saline), low dose group (LDG, 1mg/kg ), medium dose group (MDG, 2.5mg/kg) and high dose group (HDG, 5mg/kg). Ten rats for each group, injected intramuscularly twice a week. The BMD and body composition were measured at the beginning and 4 weeks later by DEXA densitometer. Results The body weight of Cont was increased with time, the body weight of different doses of Dex intervention groups were decreased after 1 to 4 weeks intervention when compared with the Cont. After 4 weeks intervention, the total BMD, femur BMD, total BMC, total fat mass, trunk fat mass, leg fat mass of Cont were increased while the total lean mass, trunk lean mass of Cont were decreased when compared with itself before intervention. The total BMC, femur BMD, leg lean mass, leg fat mass of LDG and MDG were lower than the cont (P<0.05). The femur BMD, leg fat mass of LDG and MDG were lower than the HDG(P<0.05). Conclusion The doses of 1mg/kg and 2.5mg/kg Dex have greater influence than the dose of 5 mg/kg Dex on the femur BMD and the leg composition of SD rats by intramuscular injections twice a week for 4 weeks, and can lead the femur BMD and leg fat mass decreased.

    The Mechanism underlying the Role of HIF-1α during Hepatocyte Apoptosis in Intermittent Hypoxia with Pulmonary Emphysema Rats

    [Abstract] Objective To investigate the effect of intermittent hypoxia (IH) with pulmonary emphysema on the expression of hypoxia inducible factor-1α(HIF-1α)Bax and Bcl-2, and reveal the mechanism underlying the role of HIF-1α in hepatocyte apoptosis. Methods Sixty rats were divided randomly into the four groups: the normal control group, rats were treated normally; IH group, rats were treated by 30s nitrogen and then 90s air, which was called one cycle, and rats were treated by 120 cycle from nine clock to seventeen clock daily;pulmonary emphysema group, rats were treated by smudging, the condition of smudging was set for half an hour, twice a day (eight clock and sixteen clock); IH with pulmonary emphysema group, rats were treated by 120 cycle which were composed of 30s nitrogen and then 90s air from nine clock to seventeen clock daily. After exposed fourteen weeks, the whole rats were killed. qRT-PCR assay was conducted to detect the expression of HIF-1α mRNA, Bax mRNA, Bcl-2 mRNA in live tissues. Correlation analysis was applied to reveal the relationship between HIF-1α and Bax, Bcl-2. Results The expression of HIF-1α, Bax, Bax/Bcl-2 in IH with pulmonary emphysema group was significantly higher than that in other groups (P<0.05), and the expression of Bcl-2 in IH with pulmonary emphysema group was significantly lower than the pulmonary emphysema group and the normal control group, but the expression of Bcl-2 in IH with pulmonary emphysema group did not show significant difference compared with the IH group. Positive correlations existed between HIF-α and Bax, Bax/Bcl-2, while negative correlations existed between HIF-α and Bcl-2. Conclusion The expression of HIF-1α mRNA, Bax mRNA, Bcl-2 mRNA was induced in hepatocyte under the condition of intermittent hypoxia with pulmonary emphysema, and the induction was greater than that under purely intermittent hypoxia and pulmonary emphysema. HIF-1α expression correlated with Bax and Bcl-2, suggesting that HIF-1α may promote the hepatocyte apoptosis through transcriptional coactivators, Bax and Bcl-2.

    The influences of pre-transfusion of donor apoptotic cells on survival of islet grafts and function of T lymphocytes

    Objective : To study the influence of apoptotic cells on the cytokines of different subsets of T cell in the peripheral blood from the recipient rats after islet transplantation in order to explore the possible way of apoptotic cells involved in the reducing of immune tolerance. Methods: apoptotic cells from the donor rats were obtained by irradiation of X-irradiation from electron linear accelerator. the diabetic rats for islet transplantation were randomly divided into four groups:each group were respectively injected by Physiological saline (A group), normal cells(B group), apoptotic donor cells(C group), and necrotic donor cells(D group). then, each group rats received islet transplantation on the seventh day after the pre-injection. The level of cytokines IFN-γ、IL-10 in peripheral blood were determined by Luminex 100 Integrated System and TGF-β1 measured by ELISA respectively at 0d, 1week, 2week after islet transplantation. Results:: The level of IFN-γ of C group has no difference before transplantation, but at 7d, 14d after transplantation it is significantly lower than other groups(P<0.050). The content of IL-10 and TGF-β1 in C group has a significant increase compared with other three groups(P<0.050)before transplantation and at 7d , 14d after transplantation, the difference disappeared after rejection. Conclusions: We have demonstrated that the pre-treatment of donor apoptotic cells can decrease the level of IFN-γ and increase the level of IL-10 and TGF-β1. All these would indicate that the transfusion of apoptotic cells might induce the immune tolerance by accommodating the balance of Th1/Th2、Th3.

    Morphological effects of ischemic acute kidney injury on neurons of hippocampus CA1 in rats

    Objective To observe neuron changes of hippocampus CA1 induced by ischemic acute kidney injury (IAKI) in rats. Methods Based on successful established IAKI animal models light and electron microscope, immunohistochemical (IHC)staining and Western blot were used for the present observations. Results Pyknotic neurons in CA1 of hippocampus were found after renal 60 min ischemia and 24 h reperfusion. The examinations of electron microscope provided facts that swollen mitochondria and dilated endoplasmic reticule in cytoplasm and shrinkage nucleus with no pyknotic chromatin were found in the pyknotic neurons. IHC staining observations revealed that the pyknotic neuron showed negative caspase-3 staining but positive PARP-1 staining. In addition, immunoblot analysis provided that only PARP-1 expressed much stronger in IAKI associated hippocampus. Conclusion The pyknotic neurons induced by IAKI might be of PARP-1 mediated cell death and of cytoplasmic death morphologically.

    The Animal Experiment Research of Dual Antiplatelet Drugs Induced Intestinal Damage

    【Abstract】 Objective To investigate the effect and its possible mechanism on the digestive tract in rats of dual antiplatelet drugs .Methods 80 SD rats were randomly allocated into four groups:these experiment groups to give clopidogrel (7.81mg/kg.d,n=20),aspirin (10.41mg/kg.d,n=20) and clopidogrel combined aspirin (hereafter referred to dual antiplatelet,n=20) respectively. Each group had the intragastric administration once daily,blank group will give normal saline(n=20),meanwhile. All rats received operation after the final intragastric administration,then observed small intestinal injury,bacterial culture of jejunal fluid,small intestinal permeability test through EB,estimated the injury degree of small intestinal mucosa by HE staining.Results 1.Each experiment group rats’small intestinal mucosa have different degree of damage ,and pathological lesions were higher than the negative control group(P<0.05),moreover,the dual antiplatelet group was the highest;2.Each experiment group rats’ small intestinal permeability increased obviously compared with the control group(P<0.05),and the dual antiplatelet group was higher than both the aspirin group and the clopidogrel group (130.533±29.631vs90.965±3.765vs66.800±4.853,P=0.000);3.The total bacteria of the dual antiplatelet group rats’ jejunal was apparently higher than the control group(61924.805±1751.159vs18154.280±1153.376,P=0.000),and the ratio of enterobacterium and enterococcus decreased obviously in the dual antiplatelet group compared with the control group(0.220±0.089vs 1.007±0.148,P=0.000).Conclusions Routine dose of dual antiplatelet drug aggravates the small intestinal injury degree of rats compared with a single drug. The increased intestinal bacteria ,dysbacteriosis, and increased small intestinal permeability may participate in the small intestinal mucosa damage process.

    [Abstract] Objective To explore the protective effect of cotreatment with antioxidants 4-hydroxy-2,2,6,6-tetramethylpiperidine 1-oxyl (tempol) and reduced glutathione (GSH) in pancreatic injury induced by IH in mice.Methods 50 male C57BL/6J mouse were equally randomly divided into control group(CON),two-week IH group(IH2w), IH+tempol+GSH group(IH+Tempol+GSH),IH+tempol group(IH+Tempol), IH+GSH group(IH+GSH).After successfully making animal model, insulin resistance level, pancreatic malondialdehyde (MDA) levels、superoxide dismutase (SOD) activity、glutathione (GSH) concentrations, and pancreaticβ-cell apoptosis were measured in all groups.Results The levels of insulin resistance were more significantly increased in IH group than those in CON group (P<0.01),but more significantly decreased in IH+Tempol+GSH group than those in IH group (P<0.01); MDA content was significantly higher (P<0.01) in the IH group than that in the CON group, but SOD activity (P<0.01) and GSH content (P< 0.01) were obviously decreased ,and MDA level was significantly lower (P<0.01) in the IH+Tempol+GSH group than that in the IH group,but SOD and GSH levels were obviously increased(P<0.01);percent of βcell apoptosis was obviously increased in IH group than that in the CON group(P<0.01), but they was significantly lower in IH+Tempol+GSH group than that in IH group(P<0.01);and there was no significantly difference in all above indexes between IH+Tempol,IH+GSH and IH groups (P>0.05). Conclusion Cotreatment with the antioxidant tempol and GSH can obviously protect IH–induced pancreatic injury in mice,however, tempol or GSH alone can not significantly inhibit this injury.

    Epidemiological status and risk factors for hypertensive disorder complicating pregnancy in Pudong New Area in Shanghai

    Objective:To comprehend the epidemiological status and risk factors in hypertensive disorder complicating in Pudong New Area in Shanghai,in order to provide a method for early intervention of hypertensive disorder complicating. Methods:A total of 4056 pregnant women in our hospital from 2009 to 2012 were retrospectively analyzded and the related factors were evaluated by multivarisate unconditional logistic analysis. Results:The incidence of hypertensive disorder complicating was 5.18%,with gestational hypertension was 2.49%,with preeclampsia slightly was 0.64%,wieh preeclampsia smi was 1.85%,with eclampsia was 0.05%,and pregnancy with chronic hypertension was 0.15%.Multivariate unconditional logistic regression analysis showed that age(OR=7.610,95%CI:2.322-24.936),body mass index(BMI)before pregnancy(OR=120.715,95%CI:61.867-235.540), the increased weight during pregnancy(OR=16.219,95%CI6.219-42.316)were risk factors for hypertensive disorder complicating. Conclutions:Age,BMI before pregnancy,the increased weight during pregnancy,primiparity,and pregnant with no antenatal examination are risk factors for hypertensive disorder complicating.While parity have no obvious correlation with hypertensive disorder complicating.

    【Abstract】Objective To evaluate the clinical value of midfrontal keyhole approach for treatment of severe intraventricular hematoma .Methods The clinical data of 21 cases of severe intraventricular hemorrhage through midfrontal keyhole approach were analyzed retrospectively. Results Evacuation of hematoma was satisfied in all cases. improved the GCS scores, intraventricular hemorrhage Graeb scores. Mian Complications after operation: diffuse brain swelling 1 case, cerebral vasospasm 3 cases, intracranial infection 1 case, pulmonary infection 7 case. According to COS, of 21 patients,5 were recovered well, 9 moderately disabled, 3 severely disabled, 1 vegetative and 3 died. Conclusions We can remove the hematoma through midfrontal keyhole approach, Restore the cerebrospinal fluid circulation , reduce the secondary brain damage, improve the prognosis of patients
