Inhibitory Effects of Periplocin from Cortex Periplocae in Human Lung Cancer Cell Line QG56
    2014, 42 (3):  197-199 .
    Abstract ( 724 )   PDF (329KB) ( 2787 )
[ABSTRACT] Objective: To investigate the inhibitory effects of Periplocin from Cortex Periplocae (CPP) in human lung cancer cell line QG56 and to discuss its mechanism. Methods: Inhibitory effect of CPP in different concentrations (1.25, 2.50, 5.00, 10.00, 20.00 ng/ml) and different time (24 h, 48 h, 72 h) on QG56 proliferation was tested by MTT method. The morphological changes of the QG56 cells treated with CPP were observed by inverted microscope. The effects of CPP on cell cycle and cell apoptosis were determined by flow cytometry (FCM). Expression of apoptosis associated genes bax on cells was assessed by RT-PCR and SP immunocytochemistry method respectively before and after treatment of CPP. Results: Marked inhibitory effect of CPP on QG56 cells was observed. The morphological changes were displayed in QG56 exposed to CPP. The results of flow cytometry showed that the proportion of tumor cells in the G0/G1 phase was increased markedly, which of S and G2/M phase cells decreased, after treatment with CPP. The apoptosis rate of QG56 cells in CPP groups increased significantly compared to the control group (P0.05). The expression of bax mRNA was increased in QG56 exposed to CPP. The result of immunocytochemistry indicated that bax protein showed stronger positive staining in cells with CPP treatment. Conclusion: CPP has significant inhibitory effect on human lung cancer cell lines QG56, which is probably related to blocking cell cycle and induction of apoptosis.

    2014, 42 (3):  200-202 .
    Abstract ( 538 )   PDF (287KB) ( 2692 )
Objective: To investigate the relationship between single nucleotide polymorphisms in mitochondrial displacement-loop and the risk of renal cell carcinoma. Methods: We selected 59 clear cell renal cell cancer patients who received nephrectomy in the Department of urinary surgery at the Fourth Hospital of Hebei University between 2002 and 2007. We also collected 68 healthy female controls. Total DNA was extracted using a Wizard Genomic DNA extraction kit (Promega, Madison, WI, USA) and stored at -20℃. PCR was performed according to the protocol of PCR Master Mix Kit (Promega) and purified prior to sequencing. Cycle sequencing was performed with the Dye Terminator Cycle Sequencing Ready Reaction Kit (Applied Biosystem, Foster City, CA, USA) and the products were then separated on the ABIPRISM Genetic Analyzer 3100 (Applied Biosystem). Polymorphisms were confirmed by repeated analyses from both strands. Results: SNPs were detected in 143 sites of the 982-bp mitochondria D-Loop region from blood samples of the ccRCC patients and the healthy controls. No statistical reference exist for distribution frequency of each SNP referring to age and sex. A statistically significant increase in SNP frequency for the 16293G, 262T alleles was observed in clear cell renal cancer patients (p<0.05), whereas the SNP frequency for the 16298C, 319A alleles was decreased in clear cell renal cell cancer patients. Conclusion: analysis of genetic polymorphisms in the D-loop may help to identify patient subgroups at a higher risk for developing RCC, thereby helping to refine therapeutic decisions for these patients.

The effect of antisense miR-30d on invasion and migration in breast cancer cells
    2014, 42 (3):  203-207 .
    Abstract ( 696 )   PDF (482KB) ( 2788 )
Objective To investigate the expression of miR-30d in breast cancer tissues and demonstrate the regulative effects of miR-30d ASO on the invasion and migration of breast cancer cells in vitro. Methods The expression of miR-30d in 108 breast cancer tissues and their adjacent tissues were detected by real-time quantitative PCR method. After transfection with miR-30dASO, the biological effects of miR-30d in breast cancer cells was measured by transwell assay and wound healing assay, and invasion-related protein expression was analyzed by western blot. Results The expression level of miR-30d was found to be overexpressed in 50.93%(55/108) of the breast cancer cases (p<0.05). miR-30d expression in breast cancer cells(transfection with miR-30d ASO)was significantly less than control group (p<0.05). Transwell and wound healing assay results showed that the invasion and migration ability decreased greatly after transfection with miR-30d ASO, furthermore,down-regulation of miR-30d resulted in obvious inactivation of MMP2 and MMP9. (p<0.05). Conclusion:miR-30d was overexpressed in human breast cancer. Reduce the expression of miR-30d can effectively inhibit the invasion and migration of breast cancer cells. miR-30d may become a new target for the regulation of invasion and migration in breast cancer.

Effects of different magnitudes of mechanical stretch on human intervertebral disc cells
    2014, 42 (3):  241-244 .
    Abstract ( 600 )   PDF (373KB) ( 2762 )
ObjectiveMechanical stress plays an important role in intervertebral disc degeneration which is closely related with low back pain. The effects of mechanical stress on disc degeneration and the mechanisms involved are poorly understood. In the present study, we investigated how different magnitudes of mechanical stretch affected human intervertebral disc cells. MethodsCells were subjected to 0%, 6%, 12%, or 18% elongation for 24 h using a Flexercell Strain Unit, and the mRNA expressions of anabolic genes (collagen-1A1, collagen-2A1, aggrecan, versican) and catabolic genes (MMP-3, MMP-13, ADAMTS-4, ADAMTS-5) were examined. ResultsThe results showed that the expression of collagen-1A1, collagen-2A1 and aggrecan, but not versican, were increased at 12%, and decreased at 18%, whereas catabolic genes: MMP-3, MMP-13, ADAMTS-4 and ADAMTS-5, were increased at 18%. Interestingly, mechanical stretch induced a magnitude-dependent increase in ADAMTS-4 synthesis. Furthermore, the effect seems to be finely tuned by stretching-triggered activation of distinct mitogen-activated protein kinase cascades. Specifically, an ERK1/2specific inhibitor, U0126, significantly inhibited stretching-induced ADAMTS-4 expression, whereas the inhibitors of p38 and JNK, SP6000125 and SB203580 only slightly decreased stretching-induced ADAMTS-4 expression. ConclusionThese results indicate that different magnitudes of mechanical stretch influence the biological behavior of intervertebral disc cells, which profoundly affects intervertebral disc degeneration.

Objective Brain metastasis was the main reason for the death of non-small cell lung cancer (non-small cell lung cancer, NSCLC). This study retrospectively analyzed the clinical efficacy and prognostic factors of arterial interventional therapy combined with epidermal growth factor receptor (epidermal growth factor receptor, EGFR)and tyrosine kinase inhibitors (tyrosine kinase inhibitors, TKI) erlotinib (erlotinib) Method Form January 2009 to January 2012,45 NSCLC patients with brain metastasis confirmed by pathology or cytology were collected in our study. They all underwent infusion chemotherapy two-six cycles by selective bronchial artery or intracranial arterial. Erlotinib was used simultaneously or sequentially with the infusion chemotherapy. We assessed the clinical efficacy every two cycles or when the disease got progressed. Results All the patients received at least two cycles of treatment. The median number of cycles was 3(range 1-6). The results were as follows: complete remission (CR) 7 cases (15.56%), partial remission (PR) 12 cases (26.67%), stable (SD) 16 cases (35.56%), progression (PD) 10 cases (22.22%).The objective response rate (ORR, CR + PR) and disease control rate (DCR, CR + PR + SD) was 42.22% and 77.78% respectively. 45 cases of patients with progression-free survival (PFS) time of 7-29 months, the median PFS time was 11.00 months, the overall survival time was 9-44 months, the median OS was 17.00 months. Univariate analysis showed that patients with low PS score has longer PFS than those with higher PS score, patients with low PS score has longer OS than those with higher PS score. Conclusion Arterial interventional therapy combined with erlotinib has got a better short-term effect and prolonged the survival time but with mild side effects. It would be a useful treatment for NSCLC patients with brain metastasis.

Abstract: [Objective]To investigate the feasibility of U100plus laser in treating stone incarcerated in gallbladder. [Methods]52 cases of stone incarcerated in gallbladder were dealt with U100plus from Sep 2011 to Oct 2012. Diameter 0.5cm laparoscopy under general anesthesia under direct vision crawl gallbladder,Gallbladder open a small incision at the bottom,,then explored in it. The mirror end put ordinary clear plastic tube used to expose and fixed stones. Consecutive crushed stones under direct vision, and then draw the gravel into the plastic tube out.The fiber choledochoscope see bile pouring from the cystic duct Or placed by the operation holes Choledochoscope the observed zebra guidewire C-arm to the guide wire can be confirmed easily enter the common bile duct cystic duct patency. After confirming that no residual stones with 4-0 absorbable suture continuous suture catcher gallbladder incision.[Results] 52 cases in this group were successful surgery, including 12 cases of gallbladder wall edema, anti partial bile leakage after gallbladder nearby drainage tube placed 2-4 days after color Doppler ultrasound examination no abnormal drainage tubes were removed. The mean operative time was 40min, the average amount of bleeding in surgery 2ml, after one day can get out of bed, the average length of stay of four days. Follow-up of 6 months to 2 years and 6 months, no special discomfort, color Doppler ultrasound examination stone recurrence. [Conclusions] The U100 Plus laser crushed stone is the treatment of gallbladder incarcerated stones safe and effective method.

Diagnosis and treatment of bladder hemangioma(a report of 12 cases and a review of literature)
    2014, 42 (3):  282-283 .
    Abstract ( 647 )   PDF (291KB) ( 2852 )
Abstract Objective:To investigate the clinical feature, diagnosis, treatment and prognosis of the bladder hemangioma. Methods:The clinical data of 12 cases with bladder hemangioma was retrospectively analyzed and discussed in combination with relevant literature. All patients with bladder hemangioma have been followed-up successfully. Results:10 patients were treated with partial cystectomy, with 2 patients treated with TUR-BT. All patients were diagnosed the bladder hemangioma by postoperative pathology. During the time of following-up from 4 months to 6 years, there were no recurrence and metastasis to be found in all cases. Conclusion:The urinary bladder hemangioma with good prognosis is benign tumor occurred rarely in clinic. Although the initial diagnosis of bladder hemangioma could be made in combination with medical imaging, the final diagnosis depends on pathological examination. Treatment options should rely on the factual situations, and partial cystectomy is the first choice for the treatment of bladder hemangioma.
