    The study of the copy number variations in the two sisters with the reproductive abnormalities

    Purpose Through the study of the copy number variations in the two sisters with reproductive abnormalities, we discussed the influence of the genome copy number variations and environmental pollutant---benzene on the reproductive abnormalities. Materials and Methods The current study centered on a family, including two sisters with the reproductive abnormalities after working with benzene for two years, their brother, and their sons. The peripheral blood was collected; then Karyotyping, cytokinesis-block micronucleus and array comparative genomic hybridization were performed, as well as the four typical genes of PSGs in the common lost region of two sisters’ chip results were analyzed by real-time polymerase chain reaction. Results There are no abnormalities on the chromosomal G-bands of the five people. The micronuclear rates of the two sisters are higher than those of the other three subjects and normal range. The results of the gene-chips revealed that the two sisters have a common lost region in 19q13.31. The results of real-time PCR and chips were consistent with each other. Conclusion PSGs play a role in normal pregnancy; person with haploid is more vulnerable by environmental factors; benzene as an environmental pollutant seriously affects human health.

    Effect of Different Frequency of Intermittent Hypoxia on Proinflammatory Cytokines and Adipokines in 3T3-L1 Adipocytes

    Abstract Objectives: To detect the levels of IL-6, TNF-α, leptin and adiponectin on the supernatant of the adipocytes, which was treated by different frequency of intermittent hypoxia. methods: Differentiated 3T3-L1 adipocytes, was randomly divided into 8 groups including: four groups which were treated by different frequence intermittent hypoxia (IH) and their sham control (SC) groups which were treated by normal oxygen. Collected the supernatant after finishing exposure, the levels of TNF-α, IL-6, leptin and adiponectin were detected by ELISA. Results: The levels of TNF-α and IL-6 of intermittent hypoxia groups were all significantly higher than the control group(P<0.01), and the levels of TNF-α and IL-6 of IH3 group were significantly higher than other intermittent hypoxia groups(P<0.05) . The leptin levels of intermittent hypoxia groups were all significantly higher than the control group(P<0.01), and the leptin. level of IH3 group was significantly higher than other intermittent hypoxia groups(P<0.05).The adiponectin levels of intermittent hypoxia groups were all significantly lower than the control group(P<0.01), and the adiponectin level of IH3 group was significantly lower than other intermittent hypoxia groups(P<0.05). Conclusions: Intermittent hypoxia was able to induce the secretion disorder of TNF-α, IL-6, leptin and adiponectin in cultured adipocytes. It was indicated that adipocytes’endocrine dysfunction was involved with insulin resistance (IR) in OSAHS patients.

    Nanog Expressed in Esophageal Cancer Cell-line and Sensitized to Cisplatin

    [Abstract] Background and purpose:Esophageal squamous cell carcinomas (ESCC) chemotherapy drug resistance is very common, the cancer stem cells theory suggested that this was due to existing the esophageal cancer stem cells which is resistant to traditional chemotherapy. This study is testing stem cell gene Nanog expression in ESCC cell lines TE-1 and its Downregulation by RNAi Sensitizes cells to Cisplatin. Methods:In this study, we detected the expression of Nanog in the ESCC cell lines TE-1 by immunofluorescence. Then we used fluorescence labeled small interfering RNA (siRNA) to block Nanog expression while evaluating the effect of Nanog siRNA by RT-PCR and western blot , and the combined effects with Cisplatin in ESCC cell line TE-1. Results:Nanog is Highly Expressed in ESCC Cell-line TE-1and the main expression in the nucleus.The Nanog expression at the protein and mRNA level in Nanog siRNA-transfected group was reduced by 85% as compared with that in the negative control group. The cells exposed to Nanog siRNA in the presence of Cisplatin showed a significant decrease in IC50 (4.03μg /ml)compared with control siRNA (5.17μg /ml)and mock transfection (5.26μg /ml), (p < 0.05). Conclusion:The results showed that Nanog is highly expressed in ESCC cell-line TE-1 and Nanog siRNA efficiently decreased Nanog expression . Treatment with Nanog siRNA in combination with cisplatin enhanced chemosensitivity.

    Study on reliability and validity of Pittsburgh Sleep Quality Index among civil servants in Tianjin

    Objective: To assess the reliability and validity of Pittsburgh Sleep Quality Index (PSQI) and investigate the sleep quality status among civil servants in Tianjin. Method: The participants were selected randomly from civil servants in Tianjin by using stratified cluster sampling method and investigated with PSQI, 3 610 valid samples were obtained. The reliability and validity of PSQI was evaluated by calculating the split-half reliability, Cronbach’s alpha coefficient, construct validity and discriminate validity. Result: The average score of PSQI is 4.53±2.96, the incidence of sleep disorders is 13.82%. Mean difference of PSQI score between different age, education, marital status, family income groups is statistically significant, while no difference is found between gender groups. The split-half reliability of PSQI is 0.881, Cronbach’s α coefficient is 0.746. Factor analysis produced three components which explained 52.91% of total variance. Most items have a higher factor loading in one of the three factors, while relatively lower in other factors. Conclusion: The incidence of sleep disorders is rather high in Tianjin civil servants, PSQI has good reliability and validity.

    Relationship of Serum Uric Acid Level With the Presence and Severity of Angiographic Coronary Artery Disease

    Objective: The study was designed to determine the relationship between serum uric acid (UA) level and the presence and severity of coronary artery disease (CAD). Methods: A Total of 251 patients who underwent coronary angiography were included in this study according to the inclusion criteria. Patients were divided into CAD group and Non-CAD group according to the results of angiography. The severity of CAD was measured in terms of the number of vessels with disease. Results:1. The serum uric acid level was higher in CAD patients than that of Non-CAD patients (p<0.001). Logistic regression analysis showed that the serum uric acid level was an independent risk factor of CAD (OR=1.007, 95%CI:1.002-1.011, p=0.004). 2. Although the serum uric acid level of one-, two-, three-vessel disease was higher than the non- vessel disease(p<0.05), among the three group there was no significant association. The same results were found in women, there was no significantly different. in men. 3. A significantly higher percentage of CAD patients was found in UA levels in the range of quartile 3 and 4 than that of UA levels of quartile 1 and 2 (p<0.05). Conclusion: In conclusion, UA level was found to be associated with the presence but not with the severity of CAD. Particular attention should be paid with the UA level in clinical CAD diagnosis.

    The effect of fasudil to right ventricular systolic function assessed by Speckle Tracking Imaging in patients with Pulmonary Arterial Hypertension

    Objective To quantitatively analyse the peak systolic strain(S) and peak strain rate(SR) of the right ventricular free wall and ventricular septum in patients with pulmonary arterial hypertension (PAH) using speckle tracking imaging(STI) and discuss the effect of Rho kinase inhibitor Fasudil to right ventricular systolic function in PAH patients. Methods The PAH group, consisted of 17 PAH patients, who were diagnosed by echocardiography(PASP >35mmHg). A gender and age-matched control group of 20 healthy volunteers were assessed with echocardiography. To measure the inner diameter of right atrium(RA),right ventricle(RV),main pulmonary artery(MPA),right ventricular wall thinkness(RVWT) and right ventricular ejection fraction (RVEF). Select high frame rate images recorded from the apical four-chamber view (frame rate≥60Hz), peak myocardial systolic strain(S), strain rate(SR) were measured respectively in basal and mid segments of the free wall of right ventricle and ventricular septum. 3. To compare the differences in above parameters between the PAH group and control group. To compare the differences in above parameters in patients before and after fasudil therapy. Results 1. Compared with the control group, RA,RV,MPA and RVWT in the PAH group were increased (P <0.01), RVEF was reduced in the PAH group(P <0.01). In PAH group,S of basal and mid segments of right ventricular free wall and basal, mid segments of septum were lower than those in control group(P <0.01), SR of basal and mid segments of free wall and basal segment of septum were lower than those in control group(P <0.01 or P<0.05).2. The patients who got treatment of Fasudil 60mg,bid,have a reduction in PASP, RA, RV(P <0.01), and have improvement in RVEF(P <0.05). S and SR of basal, mid segments of free wall were increased(P <0.01 or P <0.05),S of basal, mid segments of septum were increased(P <0.05).Conclusions S and SR of basal, mid segments of right ventricular free wall measured by STI can be used to evaluate the improvement of right ventricular systolic function after fasudil therapy sensitively and exactly. It is a beneficial addition of conventional parameters which are used to evaluate the systolic function of right ventricle.

    Expression and Significance of FHIT and Survivin in sinonasal inverted papilloma

    Abstract Objective:To investigate the expression and significance of FHIT and it’s relationship with Survivin in sinonasal inverted papilloma (SNIP) . Method:Immunohistochemical method was used to determine the expression of FHITand Survivin in 50 cases of SNIP,20 cases of sinonasal squamous cell carcinoma (SCC) and 20 cases of normal sinonasal tissues,to analyse the result depending on clinical stages and recurrence . Results:(1)FHIT was expressed in 15 of 50 (30%) cases of SNIP,the diffences with normal sinonasal tissues have statistic significance(P <0.01) , the diffences with SCC have not statistic significance(P>0.05), and the expression is not relative to the clinical stages and the age. (2) Survivin was expressed in 40 of 50(80%) cases of SNIP, the diffences with normal sinonasal tissues have statistic significance(P<0.01) , he diffences with SCC have not statistic significance(P>0.05) ,and the expression are increasing with the clinical stages and the age. (3) The inactivation of FHIT and the activation of Survivin in SNIP are distinct correlation(P<0.01). Conclusion:The inactivation of FHIT and the activation of Survivin may play an important role in the pathway of progression of SNIP and SCC. They may be identified as a new therapeutic target .

    Abstract Objective:To investigate the expression of Livin proteins and Smac proteins in the bladder transitional cell carcinoma (BTCC) and their correlation. Methods:The expression of Livin proteins and Smac proteins were assessed in 42 cases of the BTCC tissues and 18 cases of peritumorial normal mucosa tissue by SABC Immunohistochemistry,and their relationships with clinical pathological features were analyzed.Results:The expression of Livin proteins and Smac proteins in the BTCC were 73.8%(31/42) and 28.6%(12/42),while in 18 cases of peritumorial normal mucosa tissue,respectively, 0 and 77.8%(14/18).Significant differences of Livin proteins and Smac proteins expression were found between BTCC and peritumorial normal mucosa tissue groups(P<0.01);and both were significantly correlated with recurrence(P<0.05);but they didn’t correlate to sexualities, ages, histological grades, clinical stages, the number of tumors. And the expression levels of Livin proteins and Smac proteins showed a negative correlation in the BTCC (rs=-0.462, P<0.05).Conclusion:The relatively high expression of Livin proteins and the low or not expression of Smac proteins may lead to the occurrence and recurrence of the BTCC, and both maybe play an important role in the cell apoptosis of the BTCC.

    The study about expression and significance of CD4 + CD25 + T cell in different liver tumor

    Objective To study the distribution of CD4 + CD25 + regulatory T cells (Treg) in normal liver tissue,the liver focal nodular hyperplasia(FNH),atypical adenomatous hyperplasia of the liver(AAH) , hepatocellular carcinoma (HCC) and side of cancer. We will evaluate its the role in tumor occurrence and development. Methods The expression of treg cells were measured in 90 cases with double ELISA method of immunohistochemical staining. Analysis of its expression and significance in hepatocellular carcinoma.Rusult Normal liver tissue, FNH group were not found in the expression of Treg cells. AAH and HCC group have the expression of Treg cells. Treg cells of HCC group was signifieantly higher than in AAH group(P=0.000), The expression of Treg Cells was no significant difference between different age and sex and preoperative AFP value (P> 0.05); It is a relationship between treg cells number and the size of tumor, Treg cells of Enveloped group was signifieantly higher than in no capsule group,Comparison between normal and upper groups of Preoperative HBV-DNA was statistically significant difference.(P=0.007).Conclusion The increase of Treg cells favor occurrence and development of the tumor. It is plays a negative regulatory role in the immune of the tumor. Remove or reduce the Treg cells of the liver micro-environment may increase the effect of tumor immunotherapy.

    Clinical pathology review of lung primary signet-ring cell carcinoma

    Abstract Objective: to discuss the clinical pathology features and biologic characters of lung primary signet-ring cell carcinoma. Methods: to observe the histopathology features of 8 cases of lung primary signet-ring cell carcinoma,immunohistochemistry staining with cytokeratin pan (CKpan), cytokeratin 7 (CK7), cytokeratin 20 (CK20), vascular endothelial growth factor(VEGF) and thyroid transcription factor-1 (TTF-1)and special staining with AB and PAS in the all cases. Results: patients with lung primary signet-ring cell carcinoma present with cough and expectoration, and clinical symptoms are deficient with specificity. Nodule masses are present in periphery of lung by imaging, which have no unclear border and the diameters are 2.5-5.5cm. tumor cells are moderate size, round, and contain considerable mucin, which push against cell nucleus creating a signet ring cell appearance under light microscope. These special stains with AB and PAS are all positive. The immunohistochemical stains with CKpan and carcinoembryonic antigen (CEA) are all positive, 7 of 8 patients with CK7, TTF-1 and VEGF are positive, 1 of 8 patients with CK20 is positive.Conclusion: lung primary signet-ring cell carcinoma is scarce, high malignancy, tending to spread and metastasis. The final diagnosis depends on pathology and immunohistochemistry stain, TTF-1, CK7 and CK20 may help differentiate primary and metastatic signet-ring cell carcinoma.

    Changes of Hyoid Bone Position following Orthodontic Treatment in UCLP Patients.

    Objective:To compare the hyoid bone position in patients with the Unilateral Cleft Lip and Palate(UCLP) following the orthodontic treatment. Methods: 20 UCLP patients with crossbite of anterior teeth were included in this study. There were 15 male and 5 female patients,with average age 18.60±4.72 years. Lateral cephalogram were taken for each patient before and after treatment. 6 measurements were carried out to find the changes of the hyoid. The paired t test was used to evaluate the changes of the hyoid bond. Result: H-PTRper(mm) and H-Sper(mm) increased significantly(P<0.05)after the orthodontic treatment. Other measurements had no significant changed after treatment. Conclusion: The anteroposterior position of the hyoid bone was significantly backward after orthodontic treatment There were no significant changes of the vertical position and the axial inclination of the hyoid

    Study on the Relationship of Serum Bilirubin and Uric Acid Level with Carotid Plaque in Patients of Coronary Heart Disease

    Objective: To observe the relationship of carotid plaque with serum bilirubin and uric acid in patients of coronary heart disease(CHD). Methods: One hundred and eighty-four patients with CHD were enrolled in our study. All patients were examined for carotid intima-media thickness (IMT) by carotid artery ultrasound technology. According to the IMT, we divided the subjects into plaque group including eighty-two cases(IMT≥1.3mm) and non-plaque group including one hundred and two cases(IMT<1.3mm). The general conditions including age, sex, history of hypertension , smoking ,etal and laboratory parameters including the levels of total serum bilirubin, direct bilirubin, indirect bilirubin , uric acid, etal were recorded. All clinical data were statistically analyzed and processed. Results: The levels of total bilirubin and indirect bilirubin were lower in plaque group patients than those in non-plaque group patients, while the levels of uric acid were higher in plaque group patients than those in non-plaque group patients. All of these differences had statistical significance (P<0.05), Pearson correlation analysis indicated that the level of total bilirubin and indirect bilirubin negatively and the level of uric acid positively related with carotid plaque, the result of logistic step regression analysis showed that indirect bilirubin and uric acid were the independent risk factors of carotid plaque. Conclusion: Carotid atherosclerosis associated with abnormal serum bilirubin and uric acid in CHD patients. Interfering with the generation of bilirubin and uric acid may prevent cardiocerebrovascular incidents in CHD patients.

    【Abstract】 Objective: To study the characteristics of insulin resistance and pancreatic β- cell function in pregnant women with different abnormal glucose tolerance conditions,reveal the difference of insulin resistance and pancreatic β- cell secretion in abnormal glucose metabolism course Methods: Collected 504 patients with normal body mass index 、regular antenatal care and 50Ggct(+)in our hospital , without diabetes history and other pregnancy complications,who carried out oral glucose torelance test (OGTT) and insulin release test. Classified into normal glucose tolerance (NGT) , isolated impaired fasting glucose ( GIGT0) , isolated impaired glucose tolerance after drinking glucose water 1 hour(GIGT1)、2 hour(GIGT2)、3 hour(GIGT3)and Gestational diabetes mellitus (GDM) groups based on OGTT result according to ADA. HOMA-IR 、HOMA-β、and △PI/△PG of different groups were statistically analyzed by SPSS10.0. Results : 1、The trend of different time points in abnormal glucose metabolism course were not same. 2、Compared to NGT group , HOMA-IR was increased by 36.4%、22.5%、19.7%、6.4% and 39.9% respectively in GIGT0、GIGT1、GIGT2、GIGT3andGDM。3、The level of HOMA-β was decreased by 11.7%、6.5%、7%、1.4% and 12.3% respectively in GIGT0、1、2、3andGDM;△PI/△PG ratio was decreased in GIGT0、1、2、3 and GDM,but the diversity had statistical significance in NGT and GDM group only(P<0.05)。Conclusion: Patients with abnormal glucose metabolism not only have stronger IR compared with normal pregnancies ,but also have significant decline in pancreatic β- cell function. GIGT is actually a heterogeneous metabolic disorder,GIGT3 was resembles that of NGT, andGIGT0、GIGT1 of GIGT were seem as GDM ,both conditions are characterized by increased of glycemia, insulin resistance, and decreased β- cell function.So we should strengthen the administration to abnormal glucose metabolism patients,intervention as early as possible ,in order to decreased the complications .

    Inhibitory effect of berbamine on proliferation of retinoblastoma HXO-RB44 cells

    Objective: To study the effect of berbamine (BBM) on the proliferation and cell cycle of retinoblastoma HXO-RB44 cell . Methods: HXO-RB44 cells were cultured and passaged. HXO-RB44 were treated for 24,48 and 72 h with BBM at concentrations of 2,4,8,16 and 32μg/ml,respectively.The proliferation of HXO-RB44 cells was detect by MTT. HXO-RB44 were treated with BBM at concentrations of 4, 8, and 16μg/ml,and collected at 24h . Flow cytometry was used to detect the cell cycle using PI labeled. Results: Berbamine could obviously inhibit the proliferation of HXO-RB44 cells compared with the control group (P < 0.05 or P < 0.01) in a time- and dose-dependent manners.The IC50 value at 24,48 and 72h were 25.26, 10.94 and 6.25μg/ml, respectively. G0/G1 phase of control group and BBM treated group were (70.60±1.70)%, (46.43±0.81)%,(46.03±0.29)% and(42.77±2.44)%, respectively. S phase of control group and BBM treated group were (18.90±1.28)%, (20.53±2.50)%, (17.93±0.55 )% and (19.10±2.60)%, respectively. G2/M phase of control group and BBM treated group were (10.48±0.97)%,(33.07±1.64)%,(36.03±0.64)% and(38.13±0.35)%,respectively.There was a significant difference in G0/G1 and G2/M compared with the control group (P <0.01).Conclusion: Berbamine can inhibit the proliferation of HXO-RB44 cell and induce cell cycle arrest in G2/M phase ,which suggests that berbamine may be an effective anti-retinoblastoma drug.

    Primary Research of Immunization Therapy on Drug-resistance Breast Cancer Bearing Mice Model (continued)

    Objective Our former mice model research showed that after CIK cells were co-cultured with Dendritic cells (DC) loaded with multidrug resistance tumor antigen, the tumor were inhibited most on the same tumor bearing mice model. So we further research into the anti-tumor effect on tumor bearing mice model. Methods DC and CIK were respectively cultured from peripheral blood mononuclear cells (PBMC) derived from healthy individuals. MCF-7/ADR, a kind of breast cancer cell line expressed multidrug resistance, was prepared to obtain the antigen lyses. CIK was co-cultured with DC pulsed or unpulsed by the above antigen lyses (AP-DC+CIK and DC+CIK). So there were AP-DC+CIK, DC+CIK and CIK groups, NS was used as control group. The tumor-bearing mice were injected by intravenous with the above 4 groups’ effector cells. This study researched on the expression of MDR1, the apoptosis of tumor cells and the expression of Bcl-2 and Bax associated with apoptosis. Results After CIK co-cultured with DC, the anti-tumor effect of CIK cells were increased. The AP-DC+CIK group had the highest apoptosis index, and the lowest MDR1 expression. In above 4 groups, the AP-DC+CIK group had the highest Bax and lowest Bcl-2 expression. But the NS group was on the contrary, which meaned the lowest Bax and highest Bcl-2 expression. The difference between each two groups was significance. Conclusion After CIK cells were co-cultured with DC loaded with multidrug resistance tumor antigen, the quantity of CD4+T was increased obviously, which could induce the tumor cells apoptosis through activating Fas/FasL apoptosis pathway.

    Objective: Producing the Carboxymethyl chitosan thermosensitive hydrogel,to research the proliferation of the Carboxymethyl chitosan thermosensitive hydrogel on mouse lung fibroblasts (L929),moreover, probe into its tissue compatibility through animal experiments which can provide the fundamental experiment proof for further clinical application.Methods: Carboxymethyl chitosan interworked with glycerol phosphate to a preparation of the Carboxymethyl chitosan thermosensitive。Using MTT colorimetric assay to determine the evaluation on proliferation of L929 cells at different concentrations of carboxymethyl chitosan thermosensitive hydrogel.To take the transplant carboxymethyl chitosan temperaturesensitive gel in mice’s subcutaneous tissue, and kill them separately on the third, 7th and 14th day.And make regular tissue slice, then HE dye and observe the inflammatory response on the surrounding tissue of gel, finally evaluate the tissue biocompatibility and degradability of the gel of each group.Results: After 24h,the each concentration of extract groups on L929 cell have a certain role in promoting.In 48h,72h,96h,each experimental group as compared with negative and positive control group showed a more obvious role in promoting cell proliferation,in which 0.4 times leaching liquor have the cell-multiplication function obviously (P <0.05);On the 3dth after histology injecting, the experimental group is mainly in the acute inflammation.With the time of inflammatory cells in each experimental group gradually decreased to disappearit descriptions that carboxymethyl thermosensitive hydrogels have good biocompatibility.Conclusion:The results suggest that the Carboxymethyl chitosan thermosensitive hydrogel could promote the proliferation of the L929 and have good biocompatibility., local application of the chitosan thermosensitive hydrogel for treating periodontal diseases is feasible.

    Combined Effects of Hyperhomocysteinemia and Hypercholesterolemia on genomic DNA methylation in Wistar rats

    Objective: To study the combined effects of hyperhomocysteinemia and hypercholesterolemia on genomic DNA methylation and DNA methyltransferase activity in aortic tissue of Wistar rats.Methods: 44 healthy clean level male Wistar rats were 2x2 factorial design randomized into four groups: negative control group, high-homocysteine group , high-cholesterol group,High homocysteine mixed of high cholesterol group. The rats in negative control group were fed a normal chaw, and the other three groups were fed a chaw formula as designed.The rats were fed for three months. Heart blood was then drawn for detection of serum homocysteine and cholesterol; aortic genomic DNA was extracted for detection of genomic DNA methylation levels; and aortic nucleoprotein was extracted for detection of DNA methyltransferase activity.Results: Hyperhomocysteinemia combined effects of hypercholesterolemia increased serum homocysteine level, and there was significant statistics difference (P<0.01) ,make the performance for synergies,but there was no significant difference in rats total cholesterol (TC)、triglyceride (TG)、low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) levels by hyperhomocysteinemia combined effects of hypercholesterolemia (P>0.05). Hyperhomocysteinemia combined effects of hypercholesterolemia increased aortic genomic DNA methyltransferase activity ,and there was significant statistics difference(P<0.01) ,make the performance for synergies, and promoted aortic genomic DNA demethylation (P<0.01) ,make the performance for synergies effect.Conclusion: Hypomethylation and High-methyltransferase activity induced by hyperhomocysteinemia and the combined effects of hypercholesterolemia is one of the important mechanisms for the development of atherosclerosis.

    The protective effect of Liangge San on intestinal I/R injury

    Objective: To observe the protective effect of Liangge San on intestinal I/R injury. Method: A total of 168 Wistar rats were randomly divided into 5 group: normal group, sham operation group, model group, Liangge San therapeutics group and Liangge San prevention and cure group. The I/R model was made by blocking the superior mesenteric artery, and Liangge San was used to intervene. Chiu’s score, D-lactic acid content of janitrix blood plasma, MDA content and SOD activity of intestinal homogenate, apoptotic intex, the expression of the apoptotic protein Bax in the small intestine cells were measured after 45 min of ischemic and 3h、12h、24h、48h、72h after reperfusion. Results: After I/R, the intestinal morphology was significantly injuried, Chiu’s score and D-lactic acid contend of blood rised. The intestinal MDA content in model group increased and SOD activity decreased. The apoptotic index and the expression of Bax in intestinal mucosal cells of model group were obviously higher than those in other gropps. After the intervention of Liangge San, Chiu’s score and D-lactic acid content were lower than those in model group. MDA content decreased and SOD activity increased. The apoptotic index and the expression of Bax all dropped. Conclusions: Schemic reperfusion injury can destroy the function of intestinal barrier, Liangge San can protect the intestinal mucosa through cleaning the oxygen free radical.

    Expression and Significance of VEGF,vWF in Vascular of Metabolic syndrome young rats

    Abstract:Objective:To investigate the effect of the expression of VEGF 、vWF on vascular wall of Metabolic syndrome young rats . Methods:Four-week-old SD rats (50 g body weight) were randomly divided into 3 groups, routine diet group (NC), high fat diet group (FC) and high fat + high salt diet group (FSC). The values of body weight, abdominal circumference, blood pressure, visceral fat weight, plasma lipids, were measured 4 weeks after experiments. The levels of fasting plasma glucose and insulin were measured, and the insulin resistance index was calculated. HE stain and immunohistochemistry were used to detect the pathologic change and the infiltration cells with positive expression of VEGF,vWF in the abdominal aorta, Use elisa method to detect serum VEGF,vWF. Results: The values of body weight, abdominal circumference, blood pressure, visceral fat, plasma glucose and insulin level were significantly increased in the FSC group than that of the NC group. There were increased dyslipidemia and insulin resistance in the FSC group compared with those of the NC group. The abdominal aorta and serum express VEGF、vWF of FSC group were significantly increased than that of the NC,FC group. (p<0.01). Conclusion: The rat model of metabolic syndrome was successfully established by high fat+salt diet in juvenile SD rats. The high expression of VEGF、vWF may be related to the occurrence and development of vascular pathology and physiological changes of metabolic syndrome in children.

    [Abstract]Objective: To construct the mouse bone marrow hematopoietic stem cells with chimeric MHC-I gene. To explore the mechanism of it inducing T lymphocyte response to allogeneic dendritic cells. Methods: mouse bone marrow hematopoietic stem cells were collected with the lymphocyte separation medium (ficoll), by density gradient separation. Identified hematopoietic stem cell (HSC) were infected by the constructed lentiviral vectors. After infection, the molecular chimerism was successfully constructed. The ability of molecular chimerism reduce T lymphocyte response to allogeneic dendritic cells was observed through MLC. Results: Using ficoll, the Balb/C mouse bone marrow HSCs were successfully separated. The rate of CD34+ cells was up to (34.64±2.76)%. 3 days after lentivirus infection, H2-K1 gene expression at mRNA level in Balb/C mouse hematopoietic stem cells increased 657-fold than that of the non-infect group, increased 666-fold than that of the control group. The H2-K1 expression at mRNA level between control group and non-infect group had no significant difference (P>0.05). Flow cytometry analysis indicated that H-2Kd expressed in 98.17% cells. The result of MLC demonstrates that the index of stimulation to T lymphocyte significantly decrease. (P<0.01) Conclusion: The lentiviral vector carrying C57 mice`s MHC-I gene H2-K1 was successfully constructed. Balb/C mouse bone marrow hematopoietic stem cells can be successfully infected by the lentiviral vector. After infection, a stable and long-term expression of H2-K1 was achieved and H-2Kb protein was expressed. The molecular chimerism could reduce T lymphocyte response to allogeneic dendritic cells.

    Abstract Objective: To explore the flourence of pericentric inversion on spermatozoa chromosome. Methods:Fluorescence in situ hybridization(FISH) with TelVysion 7q、TelVysion 7p probes on 1000 spermatozoa for a 46,XY,inv(7) (p13q11) carrier. Results:The FISH analysis using the specific probes for the pericentric inversion indicated high disequilibrium(25.2%),while the normal men with 1.2%. Conclusion: Pericentric inversion is associated with a high risk of recombination sperm.It is difficult for chromosomally unbalenced embryos to implantation.Even if the abnormal embryo is conceived,it is tended to be miscarriaged at the early pregnancy.To improve the rate of pregnancy and reduce abortion,it is very important for couples with men heterozygous for this inversion to proceed preimplantation genetic diagnosis(PGD).

    Detection and correlation analysis of sperm DNA fragmentation and level of MDA in seminal plasma among infertile males with varicocele

    Abstract: Objective To detect the sperm DNA fragmentation and level of MDA in seminal plasma and explore their correlation among infertile males with varicocele. Methods The sperm chromatin dispersion test was employed to detect the sperm DNA fragmentation in 52 infertile men with varicocele and 24 healthy fertile male; Content of malonddialdehyde in seminal plasma was measured by reaction of thiobarbiuric acid; At the same time, the classical semen parameters,their including sperm concentration, progressive motility and sperm morphology were evaluated by computer assisted sperm analysis. The differences in seminal parameters between infertile groups and control group were compared, and the correlation of the sperm DNA fragmentation and level of MDA in seminal plasma was explored in varicocele group. Results Men with varicocele had lower ejaculate volume, sperm concentration,progressive motility and morphology than men in the control group. Sperm DNA fragmentation and level of MDA in seminal plasma among infertile males with varicocel were significantly higher than those in control group.The perm DNA fragmentation was positively correlated with the level of MDA in varicocele group. Conclusion The increased sperm DNA fragmentation and level of MDA in seminal plasma may be related to the infertile males with varicocele. The increased level of MDA in seminal plasma may be one of the causes of increased sperm DNA fragmentation in infertile males with varicocele.

    Analysis of the risk factors of acute renal injury after off-pump coronary artery bypass grafting

    to study the incidence and risk factors after off-pump coronary bypass grafting(OPCABG). Methods: the clinical data of 537 patients of our hospital undertaking OPCABG between June 2009 and December 2009 were retrospectively analyzed. According to the relevant domestic AKI diagnostic criteria, divided the patients into AKI group with AKI and control group without AKI, the incidence and the risk factors of postoperative AKI were counted and analyzed. Results: AKI with an incidence of 14.7 % (79/537) occurred in 537 patients. Single factor analysis showed that there were significant statistics differences among advanced age, hypertension ,preoperative serum creatinine, number of grafts, mechanical ventilation time,ICU time and postoperative low heart output. Logistic multi-factor regression analysis showed that advanced age, hypertension, preoperative serum creatinine, number of grafts , mechanical ventilation time and postoperative low heart output were independent risk factors of AKI. Conclusion: AKI was one of the most common and serious complications after OPCABG, which should cause clinical high attention. Advanced age, hypertension, preoperative serum creatinine, number of grafts, mechanical ventilation time and postoperative low heart output were independent risk factors of AKI.

    Abstract Objective: To investigate the expression of Antibiotic Peptide LL-37and HD5 in the vaginal fluid of vulvovaginal candidiasis(VVC)and recurrent vulvovaginal candidiasis(RVVC). Methods:Human vaginal fluid from 31 VVC patients, 27 RVVC patients and 30 healthy women ( the control group)were collected respectively.Enzyme-linked immunosorbent assay ( ELISA)was applied to detect the levels of antibiotic Peptide LL-37and HD5 which were infectded by candida albicans that were found by culturing in the CHROMagar. Results: Both the LL-37and HD5 were found in the VVC group、RVVC group and the control group. The levels of LL-37and HD5 of VVC group were higher than the control group.The level of LL-37 of RVVC group was higher than the control group. The level of HD5 had no difference between the RVVC group and the control group. The levels of LL-37 and HD5 had no difference between the RVVC group and the VVC group. Conclusions: The level of LL-37 may play an important role in the pathogenesis of VVC and RVVC. The level of HD5 may play an important role in the pathogenesis of VVC , while may had no relationship with RVVC.

    【Abstract】Objective: To Study the correlation between lesions of deglutition center and acute Cerebral Infarction by OCSP classification of stroke. Methods: The clinical data of 848 patients into groups in order with acute cerebral were retrospectively analyzed. Cerebral infarction by OCSP classification of stroke were divided into the following 4 groups: TACI for the whole anterior circulation infarction, PACI for the partial anterior circulation infarction,POCI for the posterior circulation infarction and LACI for the lacunar infarction. Cerebral infarction by water swallow test were divided into the following 2 groups: dysphagia and non- dysphagia. Data related were collected and analyzed through statistics. Results: Dysphagia incidence of acute Cerebral Infarction by OCSP classification of stroke were significantly different(P<0.05). Dysphagia incidence on TACI 、POCI –type and PACI 、LACI-type were significantly different(P<0.05). Dysphagia incidence on TACI 、PACI 、LACI-type and brain of One-tailed or Double side were significantly different(P<0.05). Dysphagia incidence on POCI-type and brain of One-tailed or Double side weren′t significantly different(P>0.05). Dysphagia incidence on TACI-type and brain of One-tailed were significantly different(P<0.05).Dysphagia incidence on PACI 、LACI-type and brain of Double side were significantly different(P<0.05). Conclusion:Dysphagia incidence on TACI 、POCI –type of OCSP classification of stroke were high. There were relevant TACI-type of OCSP classification of stroke and lesions of brain of One-tailed. There were relevant PACI、 LACI-type of OCSP classification of stroke and lesions of brain of Double side.Lesions of brain of One-tailed or Double side can lead dysphagia of POCI-type of OCSP classification of stroke.

    A Study of CD64 Surface Expression on Neutrophils in Children with Connective Tissue Diseases

    Abstract Objective: To study the clinical value of CD64 surface expression on neutrophils in children with connective tissue diseases(CTDs). Methods: 73 children were studied including normal controls (n=30), cases with bacterial infection (n=27) and CTDs (n=16). All patients in CTD group (11 cases of juvenile systemic lupus erythymatosus and 5 cases of juvenile dermatomyositis) were active rheumatic diseases with systemic infections excluded clinically. CD64 surface expression on neutrophils was measured quantitatively using QuantiBRITE flow cytometry technique. SAS System for Windows V8 was used to perform statistical analysis. Results: The mean antigen-per-cell of CD64 on neutrophil surface in normal controls was 1434±529, without difference between two genders(1300±494 vs 1556±547,p>0.05). The neutrophil CD64 level was 1391±576 in patients with CTDs, showing no significance when comparing to that of normal controls (p>0.05). Compared with normal controls and patients with CTDs, the expression of CD64 increased significantly in patients with bacterial infection (4995±2636, p<0.0001). Conclusion: CD64 surface expression on neutrophils upregulates in patients with bacterial infection, however it does not elevate in active diseases of CTDs in children. It might become a potential indicator for distinguishing bacterial infection and the flare of connective tissue diseases.

    Comparison of curative effects of three treating methods for distal radius fracture

    Abstract Objective To explore the proper method for the treatment of the distal radius fracture. Methods The data of 87 patients with the distal radius fracture were collected and analyzed retrospectively. The patients were divided into three groups and classified with the AO method. The surgical treatment comprised the closed reduction and the application of the splint, the closed reduction and percutaneous pin fixation, and the open reduction and the internal fixation. .Result The date of 87 patients were followed up, and the curative effect was evaluated by the Dienst system. The excellent and good rate was 42.8% in closed reduction and the application of the splint,54.5% in the closed reduction and percutaneous pin fixation, and 86.5% in the open reduction and the internal fixation. Conclusion Compared with the closed reduction and the application of the splint,plate internal fixation is better in treatment of distal radius fractures

    Clininal use of 131I of thyroidectomy dose in the metastases of the differentiated thyroid carcinoma

    【Abstract】Objective: To evaluate the clinical value of 131I of thyroidectomy dose in the metastases of the differentiated thyroid carcinoma patients who were cut off most part of thyroid. Method: Twenty three differentiated thyroid carcinoma patients who were suspected metastases, including 17 cases of papillary cancer, 6 cases of follicular cancer , all were cut off most part of thyroid before 4 months to 35 months, measured the thyroglobulin levels in the blood serum, took ultrasound or CT check, took 131I- WBS using diagnostic dose, then gave patients 131I of thyroidectomy dose, took 131I- WBS again after one week, compared the results with the previous results. Result: 23 cases of patients, with thyroglobulin levels arising in 20 cases, ultrasound found recurrence in 4 cases, CT found metastases in 15 cases, including lymph node of neck, lungs, skeletons and brain. 131I- WBS of diagnostic dose found abnormal uptakes in 5 cases of patients, including 4 positions in lymph node of neck,3 positions in the lungs, while, 131I- WBS of thyroidectomy dose found abnormal uptakes in 21 cases of patients, including 16 positions in lymph node of neck and mediastina, 8 positions in lungs, 5 positions in skeletons and 2 positions in brain.,Conclusion: The positive rates of 131I- WBS of thyroidectomy dose in differentiated thyroid carcinoma is obviously superior to that of diagnostic dose. 【Key words】thyroidectomy; 131-iodine; differentiated; thyroid carcinoma;metastases
