    Construction and Expression of pERFP-C1-hTudor-SN-SN (1~4) Recombinant Eukaryotic Plasmids

    Abstract Objective:To construct eukaryotic red fluorescent protein(RFP) expressing recombinant plasmids, pERFP-C1-hTudor-SN-SN(1~4), which contain SN((1~4) fragments of human Tudor-SN. Methods:The genes of Tudor-SN fragments were amplified by PCR from the recombinant pSG5-Tudor-SN-flag plasmid and inserted into pERFP-C1 fluorescent expressing vector with EcoRI and BamHI site. These recombinant pERFP-C1-hTudor-SN-SN(1~4) plasmids were transfected into HeLa cells and the expression of red fluorescent fusion proteins was examined by fluorescence microscope. Results:The SN(1~4) fragments of Tudor-SN were sequenced correctly and detected in the products of the restriction single/double enzyme digestion. The red fluorescent fusion proteins were observed in HeLa cell after the transfection. Conclusion:These recombinant eukaryotic plasmids of pERFP-C1-Tudor-SN(1~4) were constructed successfully and expressed effectively.     摘要:目的:学习的分配和整合的多药耐药基因的鲍曼不动杆菌流行在天津,分析与整合和多药耐药的鲍曼不动杆菌创制的关系。方法:55多药耐药鲍曼不动杆菌收集了3个在天津医院,药物sensitivy试验用KB测试完成后,整合酶基因进行了PCR,检测临床分离的关系是由多基因组分析了打字。结果:47株Ⅰ类整合酶基因阳性,其中23载可变区,Ⅱ,Ⅲ类整合酶基因均为阴性。 aacA4在可变区和aadA1卡带是主要原因,导致氨基糖苷类阻力。 3个克隆株,成立于55多药耐药鲍曼不动杆菌的多基因组打字。结论:Ⅰ类整合酶基因存在多药耐药鲍曼不动杆菌广泛。 aacA4在可变区和aadA1卡带是主要原因氨基糖苷类resistance.All株原因可能是由多基因簇打字的流行病学研究,控制院内感染和抗生素useness供应的证据类型。 关键词:鲍曼不动杆菌,整合,集群打字

    Significance of TIMP-1 and TIMP-2 expression in uterine cervical squamous cell carcinoma

    [Abstract] Objective: To investigate the expression of tissue inhibitor of metalloproteinase-1 (TIMP-1) and TIMP-2 in uterine cervical squamous cell carcinoma and the relationship between these levels and the clinicopathologic characteristics of cervical cercinoma. Methods: The expression of TIMP-1 and TIMP-2 were detected by immunohistochemical MaxvisionTM methods in 46 cases of cervical squamous cancer, 34 cases of carcinoma in situ of uterine cervix (CINⅢ), and 20 cases of normal cervical epithelium (NCE). The correlation between the expression TIMP-1, TIMP-2 and clinical stages, pathological grades, lymph node metastases in cervical squamous cancer were analyzed. Results: The expression rate of TIMP-1 in uterine cervical squamous cell carcinoma, carcinoma in situ of uterine cervix and normal cervical epithelium were 54.3%(25/46), 73.5%(25/34), 20%(4/20), respectively, and that of TIMP-2 were 60.9%(28/46), 50.0%(17/34), 20%(4/20). The positive rate of TIMP-1 and TIMP-2 in the cervical carcinomas group and CINⅢ group were obviously higher than that in NCE group(P<0.01). The protein expression of TIMP-1 and TIMP-2 did not correlate with tumor stage or histological grade(P>0.05). The positive rate of TIMP-2 in lymph node metastasis was higher than that in without lymph node metastasis(P<0.01), but there was no significant difference for TIMP-1(P>0.05). Conclusion: TIMP-1 and TIMP-2 may play a key role in invasion and lymph-node metastasis of in uterine cervical squamous cell carcinoma.

    Construction of Umarked htrA-Deficient Mutant of Streptococcus mutans with Cre/loxP System

    Objective: To construct the Streptococcus mutans(S. mutans) htrA-deficient mutant and to remove the antibiotic resistance marker with the Cre/loxP site-specific recombination system. Methods: The DNA fragment containing htrA was amplified by PCR and cloned into the pGEM-T-Easy TA cloning vector. Then, a kanamycin resistance cassette(loxP-Km-loxP)was inserted into this recombinant plasmid and replaced a part of the htrA gene, yielding homologous recombination plasmid pIB△htrA-Km. Electrotransformation of S. mutans cells with pIB△htrA-Km resulted in isolation of kanamycin resistant S. mutans transformants. One such mutant was transformed with a cre expression plasmid(pCrePA). The kanamycin resistance gene was then excised. The pCrePA was then easily eliminated at nonpermissive temperature, resulting in a markerless mutant strain carrying a deletion at the htrA loci, which was verified by PCR and DNA sequencing. Result: The deletion of a part of htrA and the kanamycin resistance gene was confirmed by PCR analysis and DNA sequencing. There was a loxP at this loci. Conclusion: A htrA-deficient mutant of S. mutans was constructed and the antibiotic resistance marker was deleted successfully, which can help to further study the role of htrA in the pathogenesis of S.mutans.

    Research of type 2 diabetes prevalence in old age population in Tianjin

    Abstract Objective To investigate the prevalence of type 2 diabetes and its risk factors in old population in Tianjin. Methods select the target population of urban and rural areas and meet the residents live 5 years and above. A cross-sectional study was conducted in this research. Using the multi-phasic stratified cluster sampling, we selected 4325 residents who are more than 60 years old in Tianjin. We collect information of risk factors throughout face-to-face questionnaire, fasting blood glucose and testing physical examination. Finally, unconditional multivariate logistic regression model was used to explore the risk factors for T2DM in old population. Results The prevalence of type 2 diabetes based on the criterion of FBG≥6.1mmol/l was21%(standardized prevalence:19.88%);The prevalence of type 2 diabetes which based on the criterion of FBG≥7.0mmol/l is 16.43% (standardized prevalence:15.55%).And both of the prevalence in female was higher. The difference was statistically significant(χ2=13.12、13.05,both P<0.01).Upon the examination, the prevalence of type 2 diabetes which based on the criterion of FBG≥6.1mmol/l in male and female and the prevalence of type 2 diabetes in male which based on the criterion of FBG≥7.0mmol/l in urban was higher than that in rural, and the difference was statistically significant(χ2=48.90、4.59、52.96,P<0.01、P<0.05、P<0.01).The results of multivariate logistic regression showed that the risk factors for type 2 diabetes development in old population may be weight change, family history of diabetes, waist, hypertension(OR=1.105,3.642,1.038,1.476,all P<0.01). While the protective factors may be intake of more fresh vegetables, fruit, dairy(OR=0.292,0.497,0.663,0.707,0.681,0.883,all P<0.05). Conclusions The prevalence of type 2 diabetes in old population in Tianjin is similar to the level of the other big cities in china. Analysis of influencing factors are mainly changes of body weight, family history, waist and hypertension may be the risk factors, intake of more fresh vegetables, fruit, and dairy may be the protective factors.

    The application of real-time three-dimensional echocardiography in the diagnosis of ventricular septal defect

    Abstract Objective:To evaluate the diagnostic potential of Real-time Three-Dimensional Echocardiography (RT3DE) in Ventricular Septal Defects (VSD). Methods:102 patients with VSD were diagnosed by RT3DE and Two-Dimensional Echocardiography (2DE), and the results were compared with surgical findings. Results:The defects were delineated objectively from many unique directions and angles by RT3DE that would not be available with 2DE, even simulated intraoperative visualization. The RT3DE views of VSD including the anatomical shape, positon, size and spatial relations could be appraised in their nature. The VSD maximum diameters obtained from RT3DE was 1.02±0.14 cm, from 2DE was 0.86±0.28 cm, from surgical findings was 1.05±0.36 cm. There was no statistical difference between measurements made by RT3DE and those obtained at surgery, there was a statistical difference between 2DE and RT3DE or surgical findings. Conclusion:Compared with the 2DE, RT3DE could display the true stereoscopic views of VSD, and provide better quantitative evaluation of the size, and lead to an improved understanding of the pathophysiology of VSD. RT3DE has important applied potentiality in clinical field.

    [Abstract] Objective To describe the vessels and biliary anatomical vatiations of the right liver living donor. Methods Surgical anatomy of vessels and biliary tract structures in 90 right lobe grafts of a single surgical team were reevaluated by reviewing the results of preoperative assessment.The data were analyzed in relation to surgical management of anatomical variations. Results Portal vein (PV): 94.4% had classic anatomy, 5.6% had a trifurcated PV. Hepatic artery (HA):The hepatic artery anatomic variations were very common. Biliary tree: 66.7% had normal anatomy; 20% had a trifurcated biliary anatomy; in 12.2% the right anterior bile duct or the right posterior bile duct opened into the left bile duct; 1.1% was of type 4. Hepatic veins:In 70.0% of the donor,the middle hepatic vein and the left hepatic vein have common trunk;Most(68.8%) veins of S4 import into the left hepatic vein;The incidence of right inferior hepatic vein was 26.6%,which were larger than 5mm.Conclusion The anatomic variations of the liver are very common, detailed and precise assessment of vascular and biliary anatomy is vital for appropriate surgical management.

    The relationships between serum C-reactive protein (CRP) and serum uric acid (SUA) levels in elderly patients with type 2 diabetes

    Objectives:To explore the relationships between C-reactive protein (C-RP) and serum uric acid (SUA) levels in elderly patients with and without type 2 diabetes. Methods:All the subjects, more than 60 years old, were divided into three groups, 55 diabetic patients with normal blood pressure (DM group) , 61 diabetic patients with hypertension(DMG group), and 54 subjects without diabetes and hypertension as control(NC group) . The serum C-RP、SUA、FPG、WBC、TG、TC、HDL-C、LDL-C were assayed, and the BMI、WC、SBP、DBP were determined . Results Levels of C-RP、SUA、TG、LDL-C、FPG 、BMI、WC、SBP、DBP in the DMG and DM group were significantly higher than that in NC group(P<0.01, respectively), and WBC and TC levels in DMG and DM group were significantly higher than the NC group(P<0.05, respectively); levels of C-RP、SUA and FPG in DMG group were significantly higher than those in DM group(P<0.01); TC、LDL-C in DMG group were significantly higher than those in DM group(P<0.05). Conclusions :In elderly type 2 diabetic hypertension group, the level of C-reactive protein and serum uric acid increased significantly, which was higher than the control group. Among the three groups, there are positive correlation between CRP and SUA (r = 0.335, P < 0.01).     The expression of IL-23p19mRNA in RA synovial tissues and RA synovial fibroblasts

    Abstract Objective: Discuss the secretion source and the diagnostic of IL-23 in RA patients, find out the pathway of IL-23 involved in RA pathogenesis. Methods: 6 Synovial tissues of RA and 6 Synovial tissues of OA were obtained during synovectomy and joint replacement surgery. RASF and OASF was extracted by enzyme from synovial. RT-PCR was used to detected the IL-23p19mRNA, IL-1βmRNA and GAPDH mRNA. Results: ①IL-23p19mRNA is positive in all of the 6 RA synovial tissues and 6 RASF, 4 OA synovial tissues and 4 OASF is positive. In all the positive samples, RA is significant higher than OA (p<0.05). ② All of the 6 RA synovial tissues is positive in the expression of IL-1βmRNA, which is correlated to the expression of IL-23p19mRNA(r=0.900, p<0.05), All of the 6 RASF is positive in the expression of IL-1βmRNA, and it is also correlated to the expression of IL-23p19mRNA in RASF(r=0.883, p<0.05). Conclusion: IL-23 is involved in the pathogenesis of RA, SF can secrete IL-23. IL-23 may be a verify of RA and OA. The expression of IL-23p19mRNA in RA synovial tissues and RASF is correlated with IL-1βmRNA illustrate that IL-1β maybe a pathway of IL-23 involved in RA pathogenesis.

    Study of the Effect of Stroner Neo-MinophagenC and Dexameth on the Biological Properties of Dermal Papilla Cells 【Abstract】Objective To observe the effects of Stroner Neo-MinophagenC (SNMC) and dexameth on the activity of dermal papilla cells (DPCs). Methods Dermal papillas were isolated from human scalp hair follicles by digestion with dispase and collagenase D and cultured. α-smooth muscle actin (α-SMA) was detected by immunohistochemical staining to identify DPCs. Effect of different concentration of SNMC and dexameth (0.5~100ng/ml) on the cell proliferation of DPCs were measured by MTT. Results Human dermal papillas were isolated obtained by "two-step enzymatic digestion". α-SMA was expressed in DPCs by immunohistochemistry method. The results of MTT showed that, 10,20,50μg/ml of SNMC could significantly promote the proliferation of DPCs (P <0.05), the effect of the proliferation was dose-dependent within concentration of 0.5~20μg/ml. 2.5μg/ml of dexameth could significantly promote the proliferation of DPCs (P <0.05), the effect of the proliferation was dose-dependent within concentration of 0.5~2.5μg/ml. Conclusion The mechanism of SNMC and dexameth may also induce hair growth by regulating the activity of DPCs in addition to anti-inflammator,immune regulation and suppression.

    Alteration of transmural dispersion of repolarization and its relationship to ventricular arrhythmia in patients with ST segment elevated acute myocardial infarction

    [Abstract] Objective To study the alteration of transmural dispersion of repolarization (TDR) and its relationship to ventricular arrhythmia or automatic nerve system dysfunction in patients with ST segment elevated acute myocardial infarction. Method 129 STE-AMI cases which were not capable being revascularized(AMI group) and 36 cases of control(control group) were enrolled randomizedly. The values corrected TDR(TDRc) between AMI and control groups, and TDRc recorded at the time point of in-charged, the 2th day and the 3th day were compared. The correlationship between standard deviation of NN intervals (SDNN) and TDRc were probed; Patients in AMI group were divided into malignant ventricular arrhythmia(MVA) group and non-MVA group according to Holter outcomes, and the TDRc of the 2 groups were also compared. Result (1) TDRc of AMI group was much higher than the corresponding values in control group (P<0.001), whereas there was no remarked difference among the TDRc values at different time-points of AMI group(P>0.05).(2) Significant negative correlationship was found between TDRc and SDNN in AMI group(r=-0.258, P< 0.05);(3) TDRc of MVA group was higher than those in non-MVA group(P<0.01).(4) there were 2 cases who died of ventricular febrilation in MVA group while no dead occurred on non-MVA group. Conclusion TDRs of STE-AMI cases are significantly elevated and sustain a high-level during the first 3 days at least; In comparison with non-MVA group, the MVA group was demonstrated with higher TDRc and cardiovascular risk; The automatic nerve system may contribute to the enlargement of TDR of non-AMI patients.

    摘要:目的:探讨经纤维支气管镜吸痰结合支气管肺泡灌洗术(BAL)在危重症合并重症肺感染中的应用价值。方法:将84例危重症合并重症肺感染患者随机分为治疗组、对照组,每组42例,治疗组除常规予以抗生素、化痰、湿化气道、营养支持、促进排痰外,均采用纤支镜吸痰结合BAL。对照组予以常规治疗而未使用纤支镜吸痰及BAL,将两组临床疗效及两组体温、白细胞计数、C反应蛋白进行比较。结果:治疗组治疗前后呼吸频率(RR)、心率(HR)及血气分析主要指标:酸碱度(PH)、动脉血氧分压(PaO2 )、动脉血二氧化碳分压( PaCO2 )、经皮血氧饱和度(SaO2)有明显好转,差异均有统计学意义(P <0.05);治疗组治愈26例,显效9例,好转4例,无效3例,有效率92.9%。对照组治愈16例,显效9例,好转5例,无效12例,有效率71.4%。两组比较有显著性差异(P<0.01),治疗组与对照组体温、白细胞计数及C反应蛋白比较有显著的临床意义(P<0.05)。结论:经纤支镜吸痰结合BAL可提高重症肺感染患者的治愈率,是一种安全、简便、而有效的方法,具有推广和应用的价值。

    Clinical analysis of mesenteric venous thrombosis:a report of 48 cases

    Object To assess early diagnosis and therapeutic management of mesenteric venous thrombosis (MVT). Methods The clinical data of 48 inpatients treated in our hospital from May 2000 to April 2010 was reviewed and incident cause,clinical features,diagnosis and treatment were analysied retrospectively. Results There were 32males(66.7%) and16 females(33.3%),the age ranged from 25 to 80 years old with a mean age of 51.23 years.19 cases (39.6%)were treated by emergency operation,6 cases(12.5%) needed surgical intervention.23 cases(92.0%) were cured,2 cases(8.3%) were died after operation.23 cases(47.9%) were cured with non-surgical management. 23 cases (100%)were cured.After follow-up one to sixty months,6 cases(12.5%)had MVT recurrence.one died. Conclusion :The risk factor of mesenteric venous thrombosis are DM, pancreatitis,cardiac,hypercoagulation, oral contraceptive,liver disease.Early diagnosis and anticoagulant thrombolytic therapy, permit radical surgery in time are key to improve the cure rate.

    Study on different methods in primary cultures of human dental pulp cells in vitro

    Objective To compare the effects of different methods and explore a simple primary culture technique of human dental pulp cell in vitro. Methods: Pulp tissue was separated from healthy young human teeth extracted for orthodontic purpose. The pulp explants were cultured by the tissue explants method, tissue-explants collagenase /dispase digestion method and modified tissue culture techniques respectively. The cell culture efficiency and cell morphology were observed and evaluated by phase-contract microscopy. Immunocytochemical staining method was used to characterize the dental pulp cell lineage. Results: The cultured cells appeared to be fibroblast-like cells.Anti-vimentin was positive and anti?cytokeratin was negative by immunohistochemical stain. The primary dental pulp cells cultured by the tissue explants method could reach confluence after 4 weeks and the culture successfulness was 26.7%. The culture successfulness of dental pulp cells obtained by collagenase /dispase digestion method was only 13.3%. Dental pulp cells can be obtained by the modified tissue culture without floating tissue fragments and the success rate could reach to 80%. Conclusion: Modified tissue culture techniques solved the difficulty of adherence of tissue fragments, simplified the procedure, and kept a high success rate. The modified tissue culture established in this study is an effective method for the primary culture of human dental pulp cells in vitro.

    The Effect of the Conditioned Medium from Macrophages on Insulin Function in Skeletal Muscle Cells

    Abstract Objective: To explore the effect of conditioned media (CM) from nomoxia- and hypoxia-treated macrophages on insulin-regulated glucose transporter 4 (GLUT4) in skeletal muscle cells. Methods: The macrophages were cultured under normoxia or hypoxia condition, respectively. Then conditioned media were collected and used to incubate C2C12GLUT4myc skeletal muscle cells. The levels of GLUT4myc on the cell surface were measured by an antibody-coupled absorbance assay. The mRNA expression of TNF-α in macrophages was determined by real-time PCR. The levels of TNF-α in the media of macrophages were determined by ELISA. Results: The increase of insulin-stimulated GLUT4myc translocation was significantly impaired under incubation with CM from both nomoxia- and hypoxia-treated macrophages (P<0.01). Both mRNA and protein expression of TNF-α were significantly higher in CM from hypoxia-treated macrophages than nomoxia-treated ones (P<0.05). But there was no difference between two CMs on insulin function in skeletal muscle cells. Conclusion: Both CM from nomoxia- and hypoxia-treated macrophages directly induce insulin resistance in skeletal muscle cells. CM from hypoxia-treated macrophages does not worsen insulin resistance.

Methods Flag to mark mouse IL-2 and join the eukaryotic cell promoter CMV, recombinant plasmid pFB-CMV-IL-2 and to the restriction enzyme digestion and PCR identification; the mouse IL-2 was cloned into the baculovirus expression vector pFastBacDual in the pFB-CMV-IL-2 was transformed into baculovirus shuttle vector containing the DH10Bac Competent Bacmid bacteria, the clones were recombinant baculovirus vector Bacmid-pFB-CMV-IL-2, extracted plasmid ; with liposome transfection method Bacmid-pFB-CMV-IL-2 transfected Sf9 insect cells packaging the virus, harvested recombinant baculovirus, will build a virus transduction of BHK cell culture, using Western Blotting Detection of IL-2 protein. Results success in BHK cells of rat recombinant IL-2 protein. Conclusion The experiment using recombinant baculovirus in mammalian cells successfully expressed recombinant mouse IL-2 protein.

    Abstract Objective: To investigate the effects of rat bone marrow mesenchymal stem cells on liver regeneration and repair after 80% hepatectomy. Methods: BMSCs were isolated and purified by adherent method, identified by surface markers via flow cytometry. 36 rats model of 80% hepatectomy were randomized into 2 groups: experimental group contains 16 rats with BMSC injection via their portal veins and control group contains 20 rats with saline by the same way. Using biochemical methods to test liver function, the flow cytometry and immunohistochemistry to detect cell cycle and proliferation cell nuclear antigens. Results: ① We isolated, cultured and proliferated BMSCs successfully in vitro. ② Survival rate after operation: survival rate was 93.7% in the experimental group and 75% in control group. ③ Liver tissue display that the hepatocyte necrosis and inflammatory cells proliferation and cellular edema, ballooning degeneration markedly reduced significantly in experimental group compared with the control group. ④ 1std, 3th d, 7th d after operation, the levels of ALT and AST recover significantly in experimental group than in the control group, the difference was statistically significant (P <0.05). ⑤ The S phase of cell cycle was significantly higher in experimental group than in the control group (P <0.05). ⑥ Remnant liver immunohistochemistry showed that PCNA expression was significantly higher in the experimental group than in the control group (P <0.05). Conclusion: BMSCs can improve significantly rat liver regeneration and repairment after 80% hepatectomy.

    Study of synthesis and radiosensitizing effect of pyridylacryl acid derivative

    Objective:To study the toxicity and radiosensitizing effect of N-(3-(3-pyridyl)acryl)-2-aminobutyric acid on hyperoxic and hypoxic Hela cells. Methods:Synthesized N-(3-(3-pyridyl)acryl)-2-aminobutyric acid with 3-pyridylacrylic acid and 2-aminobutyric acid ethyl ester hydrochloride. The survival rate and IC20 of the compound was determined by MTT assay. Sensitizing enhancement ratio(SER) was measured using colony formation test. Result:N-(3-(3-pyridyl)acryl)-2-aminobutyric acid was synthesized for the first time, the total yield was 78%. IC20 of the compound on hyperoxic and hypoxic Hela cells was 1.66 mmol/L and 1.32 mmol/L separately measured by MTT assay and SER was 1.44 and 1.84 separately by colony formation test. Conclusion:N-(3-(3-pyridyl)acryl)-2-aminobutyric acid is easy synthesized and with low cytotoxicity and radiosensitizing effect on Hela cells.

    Effect of Low Bone Metabolic Rate on the Stability of Immediate Loading Micro-implant Anchorage of Rats.

    [Abstract] Objective To study the effect of low bone metabolic rate on the feasibility and stability of immediate loading micro-implant anchorage, the experiment was conducted. Methods 20 healthy female Wistar rats were randomly divided into two groups, namely hypothyroid group and control group. Each group has 10 rats. 1ml/100g 0.1% propylthiouracil was instilled to each rats of hypothyroid group according to their body weight for 28 days. The rats in control group were stilled distilled water in the same manner. After that the venous blood from inner canthus was collected and T3, T4, TSH were detected by radioimmunoassay. Then micro-screws were implanted and 100g force was loaded to all rats for after 28 days. Tetracycline and calcein was marked to the rats after loaded 16 days and 24 days, respectively. All rats were killed after loaded 30 days. The mobile space was measured by X-ray. Super-hard nail tissue sections containing plant was observed under fluorescence microscopy and light microscopy. The bone mineral deposition rate and bone contact ratio was calculated. Results It showed that the level of T3 and T4 in hypothyroid group was lower than that of in control grou, but the level of TSH was contrary. The mobile space, bone mineral deposition rate and bone contact rate were lower in low bone metabolism group than those of in control group. Conclusion Immediate loading micro-implant anchorage has feasibility and validity.

    The effect of L-alanine on the Insulin Secretion of clonal beta cell INS-1E Cell

    Abstract: Objective: To explore the dose- and glucose-dependency of L-alanine induced insulin secretion from INS-1E cells. Materials and Methods: Passage INS-1E cells incubated in the RPMI 1640 for 2 days, then the cells were incubated in the Krebs-Ringer buffer(KRB) with 0.1%BSA for 30 minutes. After preincubation, the cells were incubated for 60 minutes in the modified KRB containing glucose and alanine according to the protocol. After the incubation the medium was collected for insulin assay. Results: Alanine at concentration of 0.1-20mM potentiated the glucose (16.7mM) induced insulin secretion in a concentration dependent manner with a maximal effect obtained at 20mM. Glucose at concentration of 1.1mM--25mM enhanced insulin secretion in a concentration dependent manner with a maximal effect obtained at 25mM. 10mM L-alanine with 1.1mM—3.3mM glucose did not enhance insulin secretion; 10mM L-alanine with 6.7mM--25mM glucose enhanced insulin secretion in a glucose concentration dependent manner and the maximal effect obtained at 10mM L-alanine plus 25mM glucose. Conclusion: The present study shows that L-alanine can enhance glucose-induced insulin secretion of INS-1E cells, The higher glucose, the more insulin secreted.

    Expression of gial fibrillary acidic protein in Brain Tissue of Newborn Rats with Hypoxic-Ischemic Brain Damage by Progesterone Intervention

    Abstract Objective: To observe the effect of progesterone on the expression of gial fibrillary acidic protein in brain tissue with hypoxia-ischemia brain damage in neonatal rats, and discuss the protective mechanism of progesterone hypoxic-ischemic encephalopathy of neonatal rats. Methods : Twenty four 7-day-old neonatal rats were randomly divided into three groups: sham-operated group, hypoxia-ischemia group and pretreatment group. Rats in hypoxic-ischemic group and pretreatment groups were subjected to left common carotid artery ligation, then were exposed to 80mL/L oxygen and 920mL/L nitrogen gas in 37℃ closed container for up to 2.5h to establish HIE model. Progesterone was injected intraperitoneally into the pretreatment groups rats respectively at 30 minutes before hypoxia, Solution was injected into the first two groups. All rats were killed at the 24 hour after operations, immunohistochemistry staining and RT-PCR were used to examine the expression of GFAP in brain. Results: GFAP was expressed in every group rat. The number of GFAP positive staining cells and the expression of GFAP in hypoxic-ischemic group were significantly higher than that in the sham-operated group. The number of GFAP positive staining cells and the expression of GFAP in pretreatment group were significantly lower than that in the hypoxic-ischemic group(P<0.01). Conclusion: The hypoxic-ischemic brain damage may induce significant increase of GFAPexpression in brain tissue of neonatal rat. Progesterone can inhibit the raising of astroeyte induced by hypoxia-ischemia brain damage, so it plays protective role by Anti- hypoxic-ischemic brain damage.

    The clinical investigation of the risk of malignancy index in patients with ovarian masses

    Abstract :Objective To explore the clinical value of the risk of malignancy index in patients with ovarian masses. Methods Choosing 30 cases of malignant ovarian tumor patients and 201 cases of benign ovarian tumor patients and computing the risk of malignancy index(RMI)based on serum CA125.menopausal status and ultrasound score. The risk of malignancy index quantity before laparotomy and their pathology after laparotomy were analyzed retrospectively sensitivity,specificity,negative predictive value,positive predictive value was used for calculation.Results The sensitivity and specificity of RMI for ovarian neoplasm was 80% and 93.5%,respectively its positive predictive value was 64.9%while its negative predictive value was 96.9%。Conclusion The serum of CA125 combined with ultrasound can significantly improve the sensitivity and specificity of malignant ovarian tumor.The combined detection may discover the ovarian neoplasm early and it is good for evaluating the character of the ovarian masses.

    Abstract: Objective: To study the mechanism of multidrug resistance of human glioma. Methods:The human glioma cells U251 and multidrug resistance cells U251/Dox were cultured in vitro. Accumulation and efflux ability of Rhodamine123 (Rh123) in U251 and U251/Dox cells were determined by flow cytometry;Multidrug resistance gene 1(mdr1) mRNA expression level in U251 and U251/Dox cells were measured by semi-quantitative reverse transcription and polymerase chain reaction (RT-PCR). Expression of P-gp was detected by immunohistochemistry. Results:MFI of U251 and U251/Dox cells were different;The mdr1 mRNA level in U251/Dox cells increased significantly. The expression of P-gp in U251 cells was significantly lower than U251/Dox cells. Conclusion: Human glioma develop cross-resistance to multiple tested cytotoxic agents and the possible molecular mechanism is involved with mdr1 mRNA overexpression and influence the function of P-glycoprotein.

    The clinical value of noninvasive positive pressure mechanical ventilation in acute left cardiac dysfunction

    Abstract Object To investigate the feasibility of bi-level positive airway pressure (BiPAP) mechanical ventilation applied in curing acute left cardiac dysfunction (ALCD). Methods Forty-two patients with ALCD were divided randomly into 2 groups: noninvasive positive pressure mechanical ventilation group in which the patients got the treatment of BiPAP mechanical ventilation, and invasive mechanical ventilation group. Both two groups all accepted the same routine drug therapy. Heart beats, arterial pressure, arterial blood gas analysis, and clinical symptoms before and after the treatment were compared between the two groups. Results Satisfied effects were obtained in both groups, and there was no significant difference between the two groups in heart beats, arterial pressure, arterial blood gas analysis, and clinical symptoms before and after treatment (P>0.05). Conclusions Patients with ALCD can obtain the benefit of early application of IPPV in improving clinical symptoms, redressing hypoxemia, and prompting heart function.

    The efficacy of different therapy time of sequential compression devices for proventing deep venous thrombosis following replacement

    Objective Assessing the effectiveness of different therapy time of sequential compression devices (SCD) for proventing deep venous thrombosis following arthroplasty Methods 111 patients undergoing primary arthroplasty from December 2007 to December 2009 were divided into two groups randomly. In the observation group, 56 patients received SCD after operation, therapy time one hour, intermission 6 hour. In the control group, 55 patients received SCD after operation, continous therapy for 24 hour. From the Second day, therapy time one hour, intermission 4 hour. DVT formation of two groups was diagnosed with colour ultrasonography at 7~10 days post-operation. Results In the observation group, three cases of DVT(5.36%) were found, one case in total hip replacement ( THR), two cases in total knee replacement (TKR). In the control group, two cases of DVT(3.64%) were found, one case in THR, one cases in TKR. The incidence of DVT in two groups had no difference (χ2=0.109, P>0.05). In the observation group, 11cases of cutaneous ecchymosis of surgical leg(37.93%) were found on the first day after surgery. In the control group, 3 cases of cutaneous ecchymosis of surgical leg(10.00%) were found on the first day after surgery. Comparing the incidence of cutaneous ecchymosis , the observation group was higher than control group (χ2=6.365, P<0.05).Conclusion Different therapy time of SCD had same effectiveness for proventing deep venous thrombosis following arthroplasty. The therapy of intermission 6 hour reduced incidence of cutaneous ecchymosis.

    【Abstract】 Objective  To explore the diagnostic significance of mammography in non-palpation breast lesions (NPBLs) . Methods 85 patients with non-palpable breast lesions were randomized into A group (to examined with sonography,42 cases) and B group (to examined with mammography,43 cases ).To get all the breast neoplasms’ biopsy through punctura or on operation and have the pathological diagnosis.Then to analyze the result. Result The breast carcinoma’ s detection rate of group A was lower than that of pathological diagnosis(68.75% VS 95.83%,P < 0.05),but the breast cancer’ s detection rate of group B has no significant difference with pathological diagnosis(86.27% VS 96.08%,P > 0.05). To examined the breast lesions with mammography has more accuratation (90.2% VS 72.92%),sensitivity(89.8% VS 71.74%),breast cancer’ s positive rate (89.8% VS 71.74%)and cut edge’s negative rate(98.04% VS 85.42%)(P < 0.05). Conclusion Mammography and sonography , especially mammography which has some superiority , play an important role in detection , diagnosis and treatment of non-palpable breast cancer in early stage.

    Expression and Significance of Serum ECP in Wheezing Children Under 5 Years of age

    Abstract: Objective To explore the expression and clinical significance of serum eosinophil cationic protein (ECP) and peripheral blood eosinophile count(PBEC) in wheezing children under 5 years old. Methods Children with recurrent wheezing under 5 years old had been randomly enrolled including 43 boys and 10 girls. They were divided into 2 groups : wheezing group Ⅰ with atopic high risks (n=27)、wheezing group Ⅱ without atopic high risks (n=26).Children without infectious diseases such as hernia and renal calculus had been enrolled randomly as normal control group(n=20)inculding 11boys and 9girls.After taking 3ml venous blood and centrifuging, the level ECPof in the serum was measured with PharmaciaUniCAP system, PBEC measurement was performed in the meantime by means of eosine staining . Results The serum levels of ECPand PBEC in wheezing groupⅠ were significantly higher than those of wheezing group Ⅱand the control group, the difference was statistically significant (P <0.05, P <0.01), while wheezing group Ⅱ group and the control group had no significant difference (P > 0.05); serum ECPand PBEC have positive correlation (r=0.786,P<0.01). Conclusion The level serum of ECPand PBEC have an intimate relation with atopic high risks .The detections of them can be used as laboratory index to distinguish wheezing children with different clinical phenotype, early intervention and treatment should be given to the children who have a tendency to persistent wheezing.

    Abstract Objective: To discuss morphological character of central serous chori-oretinopathy under 3D-Optical coherence tomography(OCT),and correlation with visual acui-ty.Methods: 46 patients(50 eyes)were checked with 3D-OCT, heights、ranges and values of detachment in macular areas were measured ,and correlation analysisof statistical software SPSS 13.0 was used to calculate correlation between the results abo-ve and visual acuity.Results: There was negative correlation between height of detach-ment in macular areas and visual acuity(r=-0.78,P=0.00),but no correlation between ranges、values and visual acuity(r=-0.39,P=0.26),(r=-0.09,P=0.80).Conclusio-ns: Heights of detachment in macular areas were the main factor infl-uenced visual acuity,high resolution 3D-OCT can get images approaching to hi-stopathology of ophthalmology in CSC affected eyes ,so as to get clinical implications in analyzing diagnosis、Differential Diagnosis 、degrees of disease and were most valuable in supervison after the therapy.

    Abstract: Evolutionarily conserved Notch signaling plays an important role in developmental processes and adult tissue homeostasis by regulating cell fate determination, proliferation, differentiation and apoptosis. There are four Notch receptors and five ligands in mammals. Notch intracellular domain (NICD) translocates to the nucleus after being cleaved and activate the transcription of Notch target genes. Notch2 is the predominant regulator of endochondral bone formation. The conditional deletion of Notch2 caused caused an accumulation of hypertrophic chondrocytes in the growth plate, resulting in severe skeletal malformations. The differentiation of mesenchymal cells into cells of the osteoblastic lineage is regulated by bone morphogenetic proteins (BMPs), Wnt and Notch Signaling. Notch signaling inhibit osteoblast differentiation by arrest of osteoblastic cell differentiation at different stages of maturation. Notch signaling negative regulates osteoclasts through RankL/Rank/OPG system.
