    Study on antimicrobial resistance and Staphylococcal cassette chromosome mec genotype of 79 methicilin-resistant Staphylococcus aureus

    Abstract Objective: To investigate antimicrobial resistance and Staphylococcal cassette chromosome mec(SCCmec) genotype characteristics of methicillin-resistant Staphylococcus aureus(MRSA) isolated from one teaching hospital, TianJin. Methods: mecA gene was detected by PCR, SCCmec genotype by multiplex PCR. Antimicrobial susceptibility of penicillin, oxacillin, ampicillin, ciprofloxacin, moxifloxacin, cefazolin, chloramphenicol, lincomycin, gentamycin, ceftriaxone, erythromycin, Rifampicin, tetracycline, vancomycin, teicoplanin, sulfamethoxazole complex, linezolid, tigecycline was detected by microdilution broth method or Kirby-Bauer method. Inducible macrolide-lincosamide- streptogramin B(MLSB) resistance was identified as a D-shaped inhibition zone by the clindamycin-erythromycin double-disk test. Results: All of the 79 MRSA harboured SCCmec type Ⅲ. They had high resistant rate to vary kind of antibiotics, but the resistant rate to chloramphenicol and sulfamethoxazole-trimethoprim were relatively lower (3.80%、10.13%). All of the testing strains were susceptible to glycopeptide antibiotics, linezolid, Tige- cycline.7 strains are D-zone test positive. Conclusion: MRSA isolated from clinic source had multiple resistance to commonly used antibiotics. All of the isolates contained SCCmec type Ⅲ. Inducible MLSB resistance was found in erythromycin-resistant, lincomycin-sensitive strains.

    Distributions of Ganglia and Cajal cells in Congenital Imperforate Anus

    Objective: To evaluate the distributions of ganglia and cajal cells in congenital imperforate anus and to discuss clinic significance. Methods: HE staining and immunohistochemical analysis of congenital imperforate anus samples from patients with congenital imperforate anus were done for ganglia, myenteric plexuses, NSE and c-kit. Results: HE staining showed us that decreases proportion of ganglia and myenteric plexuses in the anus samples with CIA. A significantly higher proportion of myenteric plexuses was more c-kit and NSE positive expression in low-position imperforate anus than that in superior position imperforate anus (P<0.05).Conclusion: The postoperative defacation disorders may be related to the decrease of interstitial cells of cajal and ganglia in the myenteric plexuses.

    The Effect on Blood Glucose by Different Alimentary Reconstruction following Radical Surgery on Gastric Cancer Patients with Type 2 Diabetes

    Objective:To assess the effect of different alimentary reconstruction on glucose in gastric cancer patients complicated with type 2 diabetes. Methods:Retrograde analyzing 55 cases of gastric cancer with type 2 diabetes from 2004 to 2008.These patients were divided into three groups: distal gastrectomy with BillrothⅡanastomosis(n=25),total gastrectomy with esophago-jejunum Roux-en-Y anastomosis(n=19), distal gastrectomy with BillrothⅠanastomosis(n=11). Results:Comparing to the third group,the blood glucose level of the former two groups after surgery showed a steady and sustained descent(P< 0.05).The effective rate was 72.00% and 73.68% respectively.However, there was no difference in the control of diabetes between the former two groups(P>0.05). Conclusions:BillrothⅡanastomosis or Roux-en-Y anastomosis is an effective method for the patients of gastric cancer with type 2 diabetes.

    Construction and Identification of fusion expression vector of the genes of EGFP and hTIMP1

    Abstract: Objective To construct the fusion expression vector of the genes of enhancement green fluorescent protein (EGFP) and human tissue inhibitor of metalloproteinases1 (hTIMP1). Methord After extracting mRNA from normal human myocardial tissue, open reading frame of hTIMP1 was obtained by nested-PCR. Then the PCR products was cloned into the pEASY-Blunt Simple vector. After identified by sequence analysis, the hTIMP1 ORF gene was cutted by two enzymes and inserted into pEGFP-C1. Then the recombinant plasmid was transformed into the competent cell of E.coliTOP10, and the correct transforments was screened to be identified. Results The PCR products was about 624bp. We compared the sequence of the hTIMP1 ORF gene which inserted into the pEASY-Blunt Simple vector with the ones reported in Genbank, the sequence was not changed. The recombinant plasmid pEGFP-hTIMP1 was also confirmed correct by identification of PCR and double enzymes’ digestion. Conclusion: We constructed fusion expression vector of the genes of EGFP and hTIMP1 successfully and laid primary basis on further reserches.

    Phenytoin toxicity in both epileptic and non-epileptic patients

     Objective To study the clinical features ,diagnosis and treatment of phenytoin (PHT) toxicity in both epileptic and non-epileptic patients, a brief review of PHT toxicity also made. Method Retrospective study of 14 cases of acute PHT toxicity admitted to our hospital from 1988 to 2008,analysis were made of their clinical feature, cause of toxicity ,medication of antiepileptic drugs, blood concentration of PHT and prognosis. Results Cerebellar syndrome is the cardinal clinical feature, some of them manifest the extra-pyramidal syndrome in both epileptic and non-epileptic patients. History of taking antiepileptic drugs is straightforward and immediate diagnosis and treatment can be done timely in epileptic patients; in contrast, ambiguity of medications history made the diagnosis and treatment delay and wasting unnecessary examinations in non-epileptic patients. All 3 cases of non-epileptic patients are uninformed ingestion of PHT, in order to control symptoms of asthma they take anti-asthma compound with brand name “fei-bao”,which main component is PHT, their clinical presentation is episode ataxia. Prognosis of acute PHT toxicity is favorable. Conclusion First report of acute PHT toxicity in patients with asthma taking PHT compound. High index of suspicion of PHT toxicity should be raised whenever patients with unexplained cerebellar syndrome are encountered, no matter they are epileptic and non-epileptic. It is inadvisable to use PHT as fist line medication in treating non-epileptic disorders. A brief review discuss the pharmacokinetics of PHT susceptible to toxicity and the mechanism and risk factors of PHT toxicity.

After These molecules and their surname combing with the acceptor, in different stages of T cells immune response wich plays an important role in regulating, involving the immune system cells and humoral immune regulation of positive negative aspects. Correct understanding of these pathways for autoimmune diseases will provide a new prospect of treatment.

Research is needed to better understand the mechanisms of bone loss with TZDs .

    Pathological and imaging analysis of primary orbital intraconal tumors

    ABSTRACT Objective :To analyze the pathologic types of orbital intraconal tumors and their image characteristics . Methods: The clinical and image characteristics of 43 patients verified by post operation pathological examinations were studied retrospectively.The patients then underwent operation based on the clinical diagnosis and the findings on the imaging modalities .Results: There were 24 cases of cavernous hemangioma , 7 cases of neurilemmoma , 6 cases of hematolymphangioma ,5 cases of venous hemangioma ,and one meningeoma. In all patients, different tumors have different image characteristics, so it is helpful for diagnosis in combination with CT and color doppler imaging(CDI).Conclusions: The common orbital intraconal tumors are cavernous hemangioma, neurilemmoma, hematolymphangioma and venous hemangioma. Combined clinical characteristics with CT and CDI can be useful for making correct diagnosis of orbital tumors.

    Differential proteomics analysis of cerebral cortex in experimental congenital hypothyroidism neonatal rat.

    Objective:To find the expressive proteomics changes in experimental congenital hypothyroidism neonatal rat cerebral cortex,, which provide valuable clues to clarify the pathogenesis of CH-induced brain developmental disorders. Methods:Neonatal hypothyroidism rat was induced by administation of propylthiouracil (PTU) solution to the dams by gavage(1%PTU,2.5ml/d) on embryonic day 13.Cerebral cortex were collected from the control and hypothyroidism neonatal rats.2-DE analysed protein expression diversities between the euthyroid and hypothyroidism neonatal rat cerebral cortex.RIA analysed serum FT3,FT4 level of each groups. Results:Compared with control group,body weight and serum FT3,FT4 level in CH neonatal rat were lower.Stable 2-DE maps of normal and CH neonatal rat were constantly obtained.Maps were analyzed by software.Eight differential protein spot were found,three differential protein spot were highly expressed in CH group,two differential protein spot were highly expressed in control group,three protein spot present only in CH group.Conclusions:Differential protein expression in CH neonatal rat cerebral may play critical roles in CH.

    The analysis of Electrocardiographic changes in patients with acute insular infarction

    Objective To observe Electrocardiographic changes in patients with acute nonlacunar MCA territory infarction, and analyze the influence of acute insular infarction on ECG. Methods ECG data of 205 patients with acute nonlacunar MCA territory infarction was analyzed retrospectively. Results Electrocardiographic changes in insular infarction group of 59 cases which manifested as sinus tachycardia(7cases, 11.86%), ectopic beat(9cases, 15.25%), ST segment elevation(4cases, 6.78%), T wave changes (13cases, 22.03%) was more than in non-insular infarction group. The difference had statistical significance(P <0.05 or P <0.01). The difference of electrocardiographic changes between patients with left and right insular infarction had no statistical significance(P >0.05). Conclusion The effect of acute cerebral infarction on cardiovascular autonomic function may be mainly correlated with insular lesion. Patients with acute insular infarction are proned to have electrocardiographic changes.

    Correlative Study of Advanced Glycation End Products ( AGEs) in Diabetic Rats and Periodontitis in Rats

    Abstract objective:The goal of this study was relevance of AGEs in rat serum and periodontitis severity by detecting AGEs of diabetic periodontitis in the rat analysising the serum content of AGEs .Methods :Two-month-old male wistar rats (n=32)were randomly divided into four groups, the control group (N group), periodontitis group (P group), diabetic group( DM group), diabetic periodontitis group (DP group). Diabetic rats were induced by intraperitoneal injection of streptozotocin ( STZ) . Periodontitis rats were initiated by ligating floss around maxillary molars and High-sugar soft diet . Observation of rats in each group: body weight, food ,water, urine output, activities, periodontal index (periodontal pocket depth, gingival inflammation). Four and six weeks later, animals were sacrificed and the specimens were prepared for histological analysis. serum AGEs were measured in ELISA method. Results: Ligation in rats after four weeks and six weeks, there is more food and more drink urine to reduce the symptoms of the relative weight, blood glucose is greater than 16.7mmol / l.Rats in each group the value of periodontal pocket probing depth comparison between the two groups DP group and DM group, DP group and P group, DP group and N group, P group and DM group, P group and N group the difference were statistical significance (P <0.05).Serum AGEs in the DP group and N group, DP group and P group, DM group and P group, DM group and N group, statistically significant difference (P<0.05). Conclusion : It is relevant that Serum AGEs in diabetic rats and the severity of periodontitis.

    Effects of Atorvastatin on proliferation and expression of COX-2 protein in hepatic cancer cells

    【Abstract】 Objective To investigate the effect of atorvastatin on the proliferation , cell cycle and COX-2 protein expression of human hepatocellular carcinoma (HepG2) cells in vitro.Methods  The human hepatocellular carcinoma HepG2 cells were employed during logarithmic growth phase and atorvastatin were administrated and the final concentrations were 0, 0.1μM,1μM,10μM,100μM respectively. Proliferation of the cells was detected by cell counting and MTT assay ,cell cycle was measured by flowcytometry (FCM), COX-2 protein expression was detected by immunocytochemistry. Results 1 Atorvastatin suppressed growth of HepG2 cells in a dose- and time-dependent manner. There was no significant difference between 0.1μM group and 0μM group. 2.Cell cycle analysis revealed that atorvastatin caused a dose-dependent increase of cells in G0/G1 phase of the cell cycle. At the same time the cells in S and G2/M phase were decreased.However, apoptosis in HepG2 cells was not obvious. 3.Atorvastatin inhibited COX-2 protein expression in a dose-dependent manner. Conclusion Atorvastatin inhibited HepG2 cells growth in vitro, which may be related with its inhibitory effect on COX-2 protein expression and arresting HepG2 cells at G0/G1 phase of cell cycle.

    The Study of Mesenchymal Stem Cells Heterogenic Transplanted to Improve Liver Damage

    Objective To comparative study the effect of mesenchymal stem cells (MSCs) and mononuclearcells(MNCs) from mice bone marrow in liver damage of carbon tetrachloride (CCl4)-treated rat, and to provide a new method for bone marrow stem cells transplantation to care liver disease. Methods Bone marrow was collected from male BALB/c mice under sterile condition. MNCs were isolated, and MSCs were cultured, purified and identified by using flow cytometry. Female Wistar rat were challenged with CCl4 to make liver damage models. The rat with liver damage were randomly divided into MSCs transplanted group, MNCs transplanted group and liver damage control group. MSCs or MNCs were infused immediately through the tail veins after CCl4 treated in these cells transplanted mice. After six weeks, we assessed liver damage and mice cell engraftment. Results According to flow cytometry assay, MSCs were positive for CD29, CD44 and CD90, while negative for CD34 and CD45; Liver function and Histological examination suggested that liver damage recovery was much better only in these MSCs transplanted mice; PCR analysis revealed that mice Y-chromosome-positive cells only can be founded in these MSCs transplanted rat. Conclusions Mice MSCs can be isolated, cultured and expanded in vitro. Transplanted to CCl4-treated rat, they can improve liver damage.

    Relationship between personality traits and compliance of orthodontic adults

    Abstract Objective: To study the influence of personality traits on compliance of orthodontic adults.Method:Ninety orthodontic adults aged twenty to twenty-nine were classified into the group. The subjects were assessed by using the Eysenck Personality Questionnaire-Revised, short Scale for Chinese (EPQ-RSC) and clinical compliance evaluation(CCE) form.To study the different influence of different personality traits on compliance.Results:The intro-extroversion personality has significant different on oral hygiene of orthodontic adults(P<0.05). The other two Eysenck Personality don’t have significant differents on compliance(P>0.05). Conclusion:The extroversion personality predicted good orthodontic compliance on oral hygiene.

    Biological Effects of the Chitosan Thermosensitive Hydrogel on Human Gingival Fibroblasts Cells in Vitro.

    Objective::To evaluate the the biological effects of the chitosan thermosensitive hydrogel on human gingival fibroblasts (HGFs) and provide information about local application of the chitosan thermosensitive hydrogel on periodontal diseases. Methods:Human gingival fibroblasts were cultured in medium containing the chitosan thermosensitive hydrogel at different concentrations,MTT colorimetric assay were used to evaluate the proliferation and ALP kit was used to evaluate the ALP activity of the GFCs. Results:On the early cultures ,there was no significant difference between the 4 groups and the negative control group and each different concentrations of the chitosan thermosensitive hydrogel about the promotion on GFCs growth and proliferation(P>0.05). After 48h, the promotion of each different concentrations was better than those of control group,through the statistic analysis,there is statistical difference between the 4 groups and the negative control group(P<0.05),there is no statistical difference between the the 25 times group and the 0.2 times group, 5 times group and the 1 times group (P>0.05).There is statistical difference between the the 25 times group and the 5 times group after 48h(P<0.05). Conclusion:The results suggest that the chitosan thermosensitive hydrogel could promote the proliferation of the GFCs, local application of the chitosan thermosensitive hydrogel for treating periodontal diseases is feasible.

    Risk Factors for Deep Vein Thrombosis after Operations for Fractures

    Abstract Objective To analyze the clinical risk factors for deep vein thrombosis(DVT) after operation for fracture.Methods The data of 515 patients with fracture undergoing operations in a hospital in Tijin between December 2003 and January 2005 were consecutively studied. Color Doppler ultrasonography(American ACUSON128/XP) was used to detect DVT of bilateral lower extremities in all patients in 7 days after operation.25 clinical factors were examined by univariate and multivariate logistic regression analysis in order to analyze their influence on DVT after surgery.Result There were 394 male(76.5%)and 121female(23.5%)patients with a mean age of 40 years,ranging 14-79 years.The prevalence of DVT after surgery was 11.3% among the patients(58/515).Logistic regression analysis demonstrated a definite association of time rate of thrombin,smoking,body mass index, lactic dehydrogenase,phosphorus with odds ratio of24.015, 2.802,1.113, 1.007 and 0.100 respectively(P<0.05).Other clinical factors had no statistical significant association with DVT(P>0.05).Conclusion Smoking,body mass index, time rate of thrombin,lactic dehydrogenase are risk factors for DVT after operatins for fractures while phosphorus is protecting factor for DVT after operatins for fractures.

    The relationship and analysis with vascular cognitive impairment not dementia and risk factors

    Objective: To investigate the relationship between the vascular impairment not dementia and the risk factors, this will facilitate the diagnosis and therapy as earlier as possible. Methods: Collect the patients who have the risk factor of ischemic cerebrovascular disease from the ward or the clinic in Tianjin general hospital from April 2009 till August 2009.The patients were divided to 2 groups. The 2 groups are VCIND group and the control group. All the patients have been detected the blood pressure and completed some related chemical and image examinations. Then compare the clinical data between the 2 groups. Result: There are variations in statistics between these two groups in the comparison of rations of diabetes mellitus, hypercholesteremia, hypertriglyceridemia, hyperhomocytinemia, major artery stenosis and white matter lesion. There is also variation in statistics of the score of MoCA between single risk factor group and the control group respectively. We divided all the patient into high risk group and low risk factor group by the score of ESSEN, we can find the variation in statistics by the comparison of MoCA between this 2 group. Conclusion: There are apparent correlation between these risk factors (except hypertension) and VCIND. Any single factor can lead to VCIND. The cognition will be damaged much more severely if the risk factors is more.

    【Abstract】 Abstract Objective:To investigate the biological effects on hepatoma cell Hep3B of p62 by use of p62 siRNA. Methods: First,Three siRNA sequences specifically targeting p62 (named as Si-h-GTF2H1_001, Si-h-GTF2H1_002 and Si-h-GTF2H1_003) were selected,the best one which silent p62 was Si-h-GTF2H1_002.The siRNAs were transfected into hepatoma cell Hep3B with Lipofectamine 2000.Then,the exprement were classified four groups: 1,blank control group;2,siRNA group;3,Ncontrol(NC)group;4,Lipofectamine(Lip)group.The expression of p62,xpd,p53,cyclinD1 were detected by RT-PCR and Western blot.The cell cycle were examined by MTT.Results : Contrast with blank control group , NC group, Lip group ,the expression of p62 mRNA was decreased significantly in siRNA group(p<0.01);the expression of xpd mRNA and p53 mRNA were decreased obviously;but the expression of cyclinD1 mRNA was enhanced obviously.The trends of protein was consistent with mRNA.MTT results showed that cell proliferation increased in siRNA group. Conclusion: p62 gene may initeract with xpd,p53 and cyclinD1 to inhibit cancer cell,and possibly regulate cell proliferation through DNA damage checkpoint.
