    A Preliminary Study of 18F-FDG and 11C-PIB PET in the Differential Diagnosis of Alzheimer's Disease and Frontotemporal Lobar Degeneration

    [Abstract] Objective   To investigate the value of N-[11C] methyl-2 - methylamino-phenyl-6 - hydroxy - benzo-thiazole (11C-PIB) and 18F-deoxyglucose (FDG) positron emission tomography (PET) in the differential diagnosis of Al-zheimer's disease (AD) and frontotemporal dementia (FTD). Methods   Ten patients who were difficult to diagnose for AD and FTD underwent 11C-PIB and 18F-FDG PET combined brain imaging. Eighteen age-matched healthy elderly controls un-derwent 18F-FDG PET. SPM8 software was used for FDG scan image data analysis. The 2-sample t-test was used in the study and P < 0.001 was considered statistically significant. The 55~60 min PIB scans were classified as positive or negative by visual analysis. PIB scans were rated as“PIB-positive”if tracer binding was deemed greater in cortical gray matter than in white matter, and as“PIB-negative”if only nonspecific white matter binding was observed. Results   After undergoing amy-loid and FDG PET scans, 5 typical AD images were distinguished. FDG imaging pattern presented as focal cortical hypome tabolism in bilateral parietotemporal association cortexes and the posterior cingulated gyrus, the precuneus. PIB was positive. There were two typical FTD images, showing the significant regional reductions in rCMRglc in bilateral frontal, the cingualte gyri, insulae and the subcortical structures. PIB was negative. Three patients, who underwent 18F-FDG brain imaging, were still difficult to identify. 11C-PIB brain imaging prompted 2 AD patients and 1 FTD patient, which were confirmed after 5 to 6-month follow-up. Conclusion   PIB and FDG PET could provide dual functional information in the differential diagnosis of AD and FTD, especially the lower specificity in rCMRglc in some cases, PIB imaging was helpful.

    Effects of Azithromycin on Recurrence or Relapse of PrimaryNephrotic Syndrome in Children

    Abstract Objective   To explore the intervention effect of Azithromycin in recurrence and(or) relapse on the glucocorticoid reduction course of primary nephrotic syndrome in children. Methods   One hundred and seventy-nine patients who had a relapse/ recurrence of nephrotic syndrome were randomly divided into treatment group and control group. Treatment group patients were treated with azithromycin 10 mg/kg each day, 3 days for a course, and the patients of control group were given non-macrolide antibiotics, oral or intravenous. The clinical and laboratory data [age, gender, weight, repeated/recurrence condition, the time of NS, the daily dosage of GC, the total time of GC cumulative application, 24 h urine protein quantitative (24 h - Upro), blood total protein (Tp), albumin (Alb), cholesterol (Tcho), urea nitrogen (Bun), serum creatinine (Scr), immune globulin (IgA, IgG, IgM, IgE), complement (C3, C4), blood mycoplasma IgM antibody positive ratio, etc]were analyzed by single and multiple regression analysis. Results  No significant difference was found in clinical and laboratory data between treatment group and control group and between Azithromycin effective group and Azithromycin ineffective group. There were significant differences on the efficient of treatment group (75.28%) and control group (34.44%). Urinary protein qualitative degree and the length days of urine protein before the treatment was different between the Azithromycin effective group and Azithromycin ineffective group. The non-conditional logistic regression analysis of treatment group revealed that the higher efficiency appear in the cases with the shorter duration days of Urinary protein before treatment. Conclusion  Azithromycin is effective in the early stage of PNS relapse/ recurrence children. Azithromycin maybe used as a steroid-sparing medicine. Azithromycin will provide a new treatment option to the PNS children with relapse or recurrence.

    [Abstract] Objective  To investigate the changes and significance of serum visfatin, omentin and tumor necrosis factor (TNF)-α in newly diagnosed type 2 diabetic patients (T2DM) with hypertension. Methods  One hundred and six patients with newly diagnosed type 2 diabetes mellitus were divided into two groups, 54 T2DM patients without hypertension (T2DM group) and 52 T2DM patients with hypertension (DM+H group). The values of blood pressure, height, body mass, blood lipids and uric acid were detected in two groups, and oral glucose tolerance test and insulin challenge test were performed as well.Blood samples were collected to determine the fasting blood glucose and blood glucose concentration 120 min after glucose administration, insulin, visfatin, omentin and TNF-α. The body mass index (BMI) and homeostasis model assessment insulin resistance index (HOMA-IR) were also measured. Results   (1) The levels of fasting blood glucose and serum visfatin and TNF-α 120 min after glucose administration were significantly increased in DM+H group than those of T2DM group, but the level of omentin was significantly decreased in DM+H group (P < 0.05). (2) The levels of visfatin and TNF-α were positively correlated with systolic blood pressure, waist circumference, fasting insulin and HOMA-IR, but the level of omentin was negatively correlated with diastolic blood pressure, waist circumference, fasting insulin and HOMA-IR. (3) The multivariate linear regression demonstrated that the level of fasting insulin was influencing factors of fasting and glucose-stimulated visfatin, but systolic blood pressure mainly affected fasting visfatin. The level of fasting omentin was mainly affected by HOMA-IR and diastolic blood pressure, but waist circumference, HOMA-IR and serum uric acid were variables of glucose-stimulated omentin. Values of BMI and fasting insulin were influencing factors of fasting and TNF-α 120 min after glucose administration respectively. Conclusion   The levels of serum visfatin, omentin and TNF-α are significantly changed in newly diagnosed type2 diabetic patients with hypertension, which may be involved in the pathological process of hypertension through many ways.with diastolic blood pressure, waist circumference, fasting insulin and HOMA-IR. ③Multivariate linear regression demonstrated that the levels of fasting insulin were influencing factors of fasting and glucose-stimulated visfatin, but systolic blood pressure mainly affected fasting visfatin; Fasting omentin were mainly affected by HOMA-IR and diastolic blood pressure, but waist circumference, HOMA-IR and serum uric acid were variables of glucose--stimulated omentin.; BMI and fasting insulin were influencing factors of fasting and 120min TNFα respectively.     Study on Plasma Oxidized Low-density Lipoprotein Levels in Patients with Acute Cerebral Infarction and the Effect of Supplement Therapy with Probucol

    [Abstract] Objective   To investigate the levels of ox-LDL in patients with cerebral infarction,and to evaluate the effect of the supplement therapy with probucol on the plasma concentrations of ox-LDL. Methods    ox-LDL, NO and eNOS concentrations were measured in subjects of cerebral infarction normotensive group, cerebral infarction with hypertension group and health control group, each of which had 60 cases. Patients in acute cerebral infarction normotensive group and hypertension group were randomized to divided into intervention group and nonintervention group respectively. Patients in the intervention group had been treated with probucol for 12 weeks. Then ox-LDL, eNOS, NO were measured after 2 weeks and 12 weeks of supplementation. Results  The levels of ox-LDL in plasma were raised significantly in patients with acute cerebral infarction (P<0.05), as well as the levels of eNOS and NO in serum were reduced obviously (P<0.05). The ADMA and Hcy levels in cerebral infarction hypertension group were higher than those in cerebral infarction normotension group (P<0.05), while the eNOS and NO levels were lower (P<0.05). With supplementation of probucol for 2 weeks and 12 weeks, the level of ox-LDL were decreased significantly (P<0.05). Conclusion  The present study demonstrated that ox-LDL which inhibited eNOS, reduced NO production, eading to endothelial dysfunction, was regarded as a risk fator of cerebral infarction. With supplementation of probucol, levels of ox-LDL were able to be decreased significantly, and sequential improvement of the endothelial function, which was propitious to anti-artherosclerosis. These findings provided a novel target for pharmacotherapy of cerebral infarction.

    Characteristics of serum Ghrelin secretion and pathogenesis mechanisms in obese and non-obese patients with polycystic ovarian syndrome

    [Abstract] Objective   To compare the relationship between secretory characteristics of serum ghrelin and mechanism of metabolic syndrome in obese and non-obese patients with polycystic ovarian syndrome (PCOS). Methods   Based on the body mass index (BMI), 60 PCOS patients were divided into two groups, 30 cases in obese group(BMI≥25 kg/m2)and 30 cases in non-obese group (BMI<25 kg/m2). Thirty infertile patients with tubal factor and normal menstruation were the control group. The serum levels of ovarian sex steroid hormones were detected by fully automatic chemiluminescence method,and the experiment of sugar metabolism was carried out in three groups. The serum ghrelin level was detected by enzyme linked immunosorbent assay (ELISA). The correlation between ghrelin level and biochemical metabolic indexes was analyzed by simple correlation analysis. Results  (1) The serum ghrelin level was significantly lower in obese group (1.84±0.46) and non-obese group (2.13±0.52) than that of control group (2.52±0.78,P < 0.05),and the level was significantly lower in obese group than that of non-obese group. (2) The values of fasting insulin (FIN), homeostasis model assessment insulin resistance index (HOMA-IR), and serum INS levels of 2 h and 3 h oral glucose tolerance test (OGTT) were significantly higher in obese group than those of non-obese group and control group (P < 0.05). In non-obese group, the serum values of LH and LH/FSH were markedly higher than those of obese group and control group. (3) The serum ghrelin level was negatively correlated with FIN and HOMA-IR (r=-0.542 and -0.503,P < 0.05). In non-obese group, the serum ghrelin level was negatively correlated with testosterone (r=-0.568,P <0.05). Conclusion   The serum ghrelin levels may play a significant role in pathogenesis mechanisms in PCOS patients,especially in obese ones.

    Inflammatory Stimulation of Porphyromonas Gingivalis Lipopolysaccharide on Human Gingival Fibroblasts

    [Abstract] Objective To explore the effect and mechanism of Porphyromonas gingivalis (P.g) lipopolysaccharide (LPS) on the secretion of inflammatory cytokines in human gingival fibroblasts (HGFs). Methods  HGFs were separated and cultured. Tumor necrosis factor-α and interleukin-1β in the HGFs were examined by enzyme-linked immunosorbent assay at 6 h and 12 h after stimulation with different concentration of LPS (1 μg/ml, 10 μg/ml). Meanwhile, the expressions of TLR2, 4 were examined by real-time PCR. Analysis of variance (ANOVA) statistical analysis was performed with SPSS10.0 software package. Results   Compared with the control group (without LPS stimulation), the expression of TLR2 and secretion of inflammatory cytokine (TNF-α、IL-1β) in HGFs were enhanced obviously at 6 h, 12 h after stimulation of LPS in P.g LPS concentration- and time-dependent manner (P<0.05). The expression of TLR4 was enhanced obviously at 6h after stimulation of P.g LPS in P.g LPS concentration-dependent manner (P<0.05). However, the expression of TLR4 showed no difference between LPS-stimulated group and non-LPS-stimulated group. Conclusion    P.g LPS stimulates HGFs to secrete inflammatory cytokines mainly through TLR2, 4 at early stage. After that, the expression of TLR2 plays an important role in inflammatory reaction induced by p.g LPS in HGFs.

    In vitro Influence of Different Root Canal Filling Systems on the Fracture Resistance of Roots

    Abstract Objective  To evaluate the influence of different root canal filling systems on the fracture resistance of roots. Methods    68 single canal premolars were selected. The crowns were removed at the cementoenamel junction. And then they were randomly divided into four groups.The root canals of groupA, B and C were instrumented to F3 with rotary Protaper system using crown-down technique. Group A root canals were obturated with master gutta-percha cone and Cortisomol sealer by using cold lateral compaction.Group B root canals were obturated with gutta-percha and Cortisomol sealer by using warm vertical compaction. Group C root canals were obturated with RealSeal by using cold lateral compaction.Group D root canals were not instrumented or obturated. Then placed all the specimen in a universal testing machine and loaded each specimen to fracture. For each specimen,we recorded the force to fracture and the fracture mode of the teeth specimen using an optical stereomicroscope. Results  The force required to fracture the roots of Group D was significantly higher than that required to fracture the roots of all other groups. The force required to fracture the roots of Group C was significantly higher than that required to fracture the roots of Group A and B. In the Group A and B,the ruptures were mainly occurred in the dentine and sealer interface. In the Group C, there was a predominance of mixed failures.Conclusion   Teeth obturated with RealSeal were more resistant to fracture than those obturated with the other two systems.

    The Influence of Liver Inflammation and Bilirubin on the LSM in Patients with CHB

    [Abstract] Objective  To investigate the influence of serum indexes and the liver inflammation at histology on the diagnostic value of FibroScan for liver fibrosis in patients with CHB. Methods  A total of 70 cases of patients with CHB were enrolled in this study. These patients were admitted to the Tianjin Second People’s Hospital, from January 2011 to November 2011.All of the patients underwent liver biopsy and liver stiffness measurement (LSM) by FibroScan at the same time. Meanwhile, the serums of the patients were collected to detect alanine aminotransferase(ALT), aspartate aminotransferas(AST), γ-glutamyl piptidase(GGT), alkaline phosphatase(ALP), total bilirubin(TBIL), direct bilirubin(DBIL), total bile acid(TBA), albumin(ALB), albumin /globular protein (A/G), and cholinesterase(CHE). The influence of serum indexes, the liver inflammation at histology on the LSM was analyzed. Results   LSM was positively correlated with age, the grade of liver inflammation at histology, the stage of liver fibrosis at histology, ALT, AST, GGT, ALP, TBIL, DBIL and TBA while it was negatively correlated with ALB, A/G, and CHE when taken by correlation analysis. LSM was only correlated with TBA and age when taken by multiple regression analysis. During the period of S01, LSM was mainly affected by ALT, especially when ALT≥400u/L (P<0.01). During the period of S234, there was a positive correlation between LSM and the grade of liver inflammation at histology (r=0.439,P=0.013). And LSM were affected by the levels of TBIL, the abnormal of TBIL could lead to the rise of LSM. Conclusion   In order to properly illustrate the liver fibrosis taken by FibroScan in CHB patients, the degree of liver inflammation and TBIL should be taken into consideration and it is also suggested to be combined with liver biopsy when necessary.

    Effects of Hispidulin on Inhibiting Lung Carcinoma A549 Cell Line

    Abstract   Objective  To investigate the effects and the mechanism of hispidulin on the proliferation of lung carcinoma A549 cell line. Method   After human lung carcinoma cell line A549 was treated with different concentrations of hispidulin, the cellular proliferation was detected by MTT assay. The flow cytometry was used to determine the change of cell cycle in A-549 cells. Western blot was employed to detect the expressions of cyclin D1, CDK4, p21 and p27 in A-549 cells. Immunocytochemistry was used to detect the expression of proliferation cell nuclear antigen in A-549 cells. Result  The proliferation of lung carcinoma cells was inhibited significantly by hispidulin. The cell viability values were (57.30±3.92)%, (43.91±4.32)%, (17.82±2.73)% and (13.20±2.46)% respectively after treatment with the concentrations of hispidulin 15、30、60、120μmol?L-1 for 72h, which were significantly lower than those (96.30±2.08)% in control group (P﹤0.01). Treatment with hispidulin (15 and 30 μmol.L-1) for 24h induced cell cycle arrest at G1/S phase in A-549 cells. Treatment with hispidulin (15、30 and 60μmol.L-1) for 24h in A-549 cells reduced the expression of cyclin D1 and CDK4 proteins and increased levels of p21 protein. The positive expression of proliferation cell nuclear antigen was significantly decreased in A-549 cells treatment with hispidulin (15、30 and 60μmol.L-1) than those of control group (P﹤0.01). Conclusion  Hispidulin can inhibit the proliferation of A-549 cells through changing cell cycle, which may be related to the up-regulated level of p21 and down-regulated levels of cyclin D1 and CDK4.

    Abstract   Objective   The purpose of this study was to determine whether a time variation could be observed in the occurance of acute myocardial infarction (AMI) in older patients in nearly three years. Methods    A retrospective analysis of patients diagnosed with AMI between 2009 and 2012 in the department of Cardiology of The Second Hospital of Tianjin Medical University (n=908 patients) was carried out. We calculated the number of AMI cases day by day. Combined with the general clinical data ,we analysis that whether a seasonal or monthly or weekly variation could be observed in the occurance of AMI in the elderly .Results    The occurance of AMI in older people increases year by year(p<0.01).A peak period of the incidence of AMI was recorded during winter, while minimum number of events were found during summer. Significant difference was observed between the number of events each season (p<0.01).And the monthly occurrence of AMI was highest in January and lowest in June (p<0.05). But the variation was only found in the patients which had coronary heart disease history or ST segment elevated in ECG or had poor heart function (P<0.05). The cicadian rhythm of AMI morbidity was found, and the rhythm has no relation with seasons and months (p<0.01) . The time peak period of AMI morbidity was found in the morning. Conclusion   Results of our study reveal that the incidence of AMI is on rise year by year and shows a characteristic rhythm with respect to seasons and months .Based on the results ,we can formulate prevention measures.

    Relation of obstructive sleep apnea hypopnea syndrome with SYNTAX Score

    [Abstract] Objective To investigate the relationship between obstructive sleep apnea syndrome (OSAHS), SYNTAX(SX)score and coronary heart disease risk factors. Methods A total of 67 patients with OSAHS were divided into three groups included mild group (n=17), moderate group (n=16) and severe group (n=34) according to Chinese Society of Respiratory Diseases. The coronary angiography was performed and SXscore was measured in three groups. Results There were no significant differences in age, gender, weight, height, hypertension,and the proportion of smokers and diabetes between three groups (P>0.05). There were also no significant differences in renal function, blood lipid, left ventricular diastolic diameter and left ventricular ejection fraction between three groups (P>0.05). There was a significant difference in SXscore between three groups (P <0.01), and which increased with the elevated apnea hypopnea index (rs=0.326,P=0.007). Conclusion There is a positive correlation between the severity of OSAHS and serious and complex coronary artery lesions, which has partly guiding role for OSAHS patients with coronary heart disease taking revascularization therapy.

    Affect of percutaneous coronary intervention on the instability of respiratory regulation in OSAHS patients

    Objective  To investigate whether percutaneous coronary intervention(PCI)causes instability of the respiratory regulation in patients of coronary heart disease with obstructive sleep apnea-hypopnea syndrome(OSAHS). Method   The author conducted a prospective self-control study in which 41 cases were involved by admission order who suffered coronary heart disease with OSAHS, and follow-up treated with PCI. Polysomnograph, echocardiogram and serum test performed not only ahead of PCI but also 3 days and 1 month after PCI respectively. All patients had never been treated with constant positive airway pressure (CPAP) ventilation or any type of operation during the study. Result   There was no significant change in echocardiogram and serum test outcome of the patients throughout our study (P>0.05). The central sleep apnea index(CSAI), the mixed sleep apnea index(MSAI) and the micro-arousal index (MAI) had increased significantly in 3 days after PCI(P<0.01), and then decreased to preoperative levels 1 month later(P<0.01). Conclusion  PCI may provoke transient instability of central respiratory regulation, and increase sympathetic activity, which caused more CSA, MSA and micro-arousal events during sleep periods.

    The Study of Pre- and Post-Operative Nasal VolumeChanges in Patients with Severe OSAHS

    Objective   To investigate pre- and post-operative of nasal volume in severe OSAHS patients without nasal disease. Method  This study included 102 healthy volunteers as control group and 46 severe OSAHS patients without nasal disease as observation group, all patients were underwent coblation-assisting modified uvulopalatopharyngoplasty combined with coblation-channeling of tongue(H-UPPP & CCT). The nasal volume from 0 to 5cm,2 to 5cm(V5,V2-5),the minimum cross-sectional area(MCA) and the distance of the minimal cross-sectional area to the nostril(DCAN) were measured by Acoustic rhinometry(AR), while snoring index(SI), apnea-hypopnea index(AHI), the lowest oxygen saturation(LSaO2) and the average oxygen saturation(SaO2) were recorded by Polysomnography(PSG) before operation and 6 months after operation respectively. Result  Compared with normal group, the preoperative V5 and V2-5 decreased in observation group(P<0.01). V5 and V2-5 in observation group increased after operation(P<0.05), but with no significant difference compared with normal group. SI, AHI, LSaO2 and SaO2 were all improved in observation group post-operative(P<0.05). Conclusion  Compared with normal group,the volume of the nasal cavity in severe OSAHS patients without nasal disease significantly reduced. And while improving the upper airway collapse by H-UPPP & CCT, nasal capacity can be enlarged, and nasal ventilation can be improved.

    Research on the impact of chronic rhinosinusitis to COPD

    Abstract Objective  To investigate chronic sinusitis impact on chronic obstructive pulmonary disease (COPD) patients quality of life and the influence of acute aggravating. Methods   For the 101 patients who met the inclusion criteria in hospital history collection, In the next early morning 6h extraction fasting venous blood samples 20ml,determination of erythrocyte sedimentation rate、 C reactive protein,、blood coagulation items;then we calculated the CAT score, reviewed the acute exacerbations of 1 year before admission hospitalization, recorded the time of hospitalization of each patient enrolled in the study .The patients were divided into two groups : simple COPD group and group of COPD combined with chronic rhinosinusitis, according to the results of paranasal sinus CT scans.Then the data were analyzed through SPSS software(Version 17.0).Results   Sinus CT scan examination showed 101 COPD patients with chronic rhinosinusitis incidence was 64.1%(62/101). 62.9% (39 / 62)of 62 patients with chronic sinusitis had nasal symptoms, while 37.1%(23/62)patients without any nasal symptoms. Maxillary sinus was the most frequently involved 93.55%(58/62), followed by the ethmoid sinus30.65%(19/62). CAT scores and the acute exacerbation frequency increased more significantly in the group of combined with chronic rhinosinusitis, than the group of only COPD ( P < 0.05 ), but the two groups when compared to the ESR and CRP, FIB and length of hospital stay indifference, compared with no difference in the occurrence of different classification of lung function in COPD patients with chronic sinusitis.. Conclusion  Chronic rhinosinusitis were the most common upper respiratory tract complications of COPD,and it influents the quality of life and acute exacerbation of the patient. Unfortunately the clinical diagnosis rate is too low,which calls for more attentions.

    [Abstract] Objective  To investigate the protective effect of miRNA-21 in myocardial ischemic postconditioning, and to explore the interaction effects in the mechanism of myocardial protection with JAK2-STAT3 pathway in ischemic postconditioning. Methods   The healthy adult male Wistar rats (body weight 240~280g) were randomly divided into five groups:Sham group, schemia-reperfusion injury group (I/R group), Ischemic postconditioning group (PostC group), schemic postconditioning + antagomiR-21 treatment group (PostC + antagomiR-21 group), and Ischemic postconditioning + AG490 group (PostC + AG490 group). In situ terminal deoxynucleotidyl transferase labeling (TUNEL) staining was used to analysis myocardial apoptosis index; Western-blot was used to detect total STAT3 (t-stat3) and phospho-STAT3 (p-stat3) protein levels.miRNA-21 levels were detected by quantitative reverse transcriptase–polymerase chain reaction (qRT-PCR). Results   Compared with the I / R group after reperfusion,ischemic postconditioning significantly reduced the apoptosis index, dramatically increased the expression levels of miRNA-21 and markedly up-regulated the stat3 phosphorylation levels. AntagomiR-21 significantly weaken the protective effect of ischemic postconditioning. PostC + antagomiR-21 group compared with PostC, STAT3 phosphorylation levels were observably lower. Compared with postC group, STAT3 phosphorylation levels and miRNA-21 were significantly lower in PostC+AG490 group. Conclusion   miRNA-21 play a regulatory role in myocardial ischemic postconditioning. STAT3 and miRNA-21 in post-processing adaptive mechanism there may be some kind of loop regulatory mechanism. But the reciprocal of the detailed regulation mechanism is unclear, pending further study.

    The Effects of VEGF and bFGF on Ovarian Tissue Transplantation in Mice

    Objective   To investigate the effects of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) on the follicular survival and angiogenesis after ovarian tissue heterotopic transplantation by mouse model. Methods   Ovarian tissues isolated from 18-day-old ICR outbred mice were transplanted into subcutaneous part of nuchal region of adult female mice. They were divided into four groups: A) injected with 10μl DPBS as control; B) injected with 10μl of bFGF (50μg/ml); C) injected with 10μl of VEGF (25μg/ml); D) injected with 10μl of bFGF (50μg/ml) + VEGF (25μg/ml). We injected subcutaneously around the grafted ovarian tissue once a day for four days. The grafts were retrieved 7 days after transplantation. The number and morphology of follicles were observed by H.E. staining and vascular density by IHC staining of anti-CD31 antibody. Results   The number of primordial follicles in group B and group C (17.25 ± 2.55; 18.88 ± 2.42 respectively) was significantly higher than that in group A (13.25 ± 2.49) (P < 0.01), while there was no statistical difference between group B and group C; the number of primordial follicles in group D (22.50 ± 3.42) was higher than that in group B and group C (P < 0.05). Compared with the control group, the vascular density in group B and group C increased, significantly higher than group A (P <0.05); but there was no statistical difference between group B and group C; vascular density in group D were statistically higher than that in group B and group C respectively (P < 0.05). Conclusion   Intervention with VEGF and / or bFGF promoted the angiogenesis after ovary tissue transplantation, while reducing the loss of follicles. That benefits improving the quality of ovarian tissue survival after transplantation.

    Abstract   Objective   Exploring biological effects of stathmin silencing on nasopharyngeal carcinoma 5-8F cell line. Methods    Cell proliferation and cell cycle were analyzed by MTT experiment and Flow Cytometry. Cell tumorigenicity was analyzed by nude mouse xenograft experiment. Results    The cell proliferation inhibition rate of stathmin-silenced 5-8F cells was significantly higher than that of negative and blank control group cells (p<0.01). The cell cycle of stathmin-silenced 5-8F cells was significantly arrested in phase G2 compared with that of negative and blank control group(P<0.05). The transplanted tumor weight of stathmin-silenced 5-8F cells in nude mice was (0.32±0.21)g,significantly lower than that of negative control group (1.10±0.40)g and blank control group(1.52±0.30)g(P<0.05). Conclusion   Stathmin-silencing inhibits malignant phenotype of nasopharyngeal carcinoma 5-8F cell line.

    [AbstractObjective   To observe effects of atorvastatin on endothelial progenitor cells (EPCs) and neural function in rats with traumatic brain injury.  Methods  Thirty adult male Wistar rats were randomly divided into sham group,treatment group and control group,10 for each group. The treatment group and the control group adopted hydraulic combat to produce traumatic brain injury (TBI) model. Rats in sham group underwent the same surgical procedure without being exposed to percussion injury. Rats were treated with atorvastatin (orally, 1 mg·kg-1·d-1) starting 1h after TBI for 14 consecutive days in treatment group. Rats were given saline orally in control group. Circulating EPCs were measured at 1, 4, 7, 14 and 21 days after TBI. The mNSS test was conducted at 1, 4, 7, 14 and 21 days after TBI. Morris water mass(MWM)was performed at 21-25days after TBI. The percentage of target quadrant was recorded in three groups.  Results  The neural function of rats was impaired by hydraulic combat. The mNSS scores were significantly lower in treatment group at 14 and 21 days after TBI compared with those of control group (P < 0.05). The percentage of target quadrant was significantly higher in treatment group than that of control group at 24 and 25 days after TBI (P < 0.05). Conclusion   Atorvastatin can regulate circulating EPCs and promote the recovery of neural function in rats with traumatic brain injury.

    Objective   To establish a model of mouse continuing endotoxemia thrombocytopenia (TCP) induced by lipopolysaccharide (LPS) and observe the change of mean platelet volume (MPV) and thrombopoietin (TPO) in this model. Methods   C57/BL mice were randomly divided into four groups, group of LPS 5mg/kg low dose of continue injection (LL-c group), group of LPS 5mg/kg low dose of single injection (LL group), group of LPS 50mg/kg high dose of single injection (HL group), group of physiological saline injection (C group). The blood platelet count(PLT) and MPV were detected before injection as base lines,and then observe variation of PLT ,TPO ,MPV and mortality. Results   The mouse PLT was significantly lower in LL-c group among seven days than basic line (P<0.01),which was continued in the base by about 30%. The PLT were low and then high in HL and LL group. The MPV was continued significantly higher in LL-c group than basic line (P<0.01),which was augmentation and then diminution in HL group and LL group.The TPO level of LL-c group in the seventh day was higher than C group,that has significance(P<0.05).The mortality of LL-c group ,LL group and HL group were 25%(5只)、0%(0只)、70%(14只). Conclusion   The model of continuing endotoxemia TCP can be induced by consecutive low dose LPS intraperitoneal injection,the mechanism and therapy of which was futher to study.

    Objective  To investigate the effect of nourishing kidney and activating blood recipe of traditional Chinese medicine on the expression of Nephrin in renal tissue of mice with adriamycin nephropathy(ADRN) induced by adriamycin(ADR) Methods  The focal segmental glomerulosclerosis( FSGS) change of nephropathy model was induced by injection of adriamycin into the tail vein.After 1 week,Twenty four male rats were randomly divided into normal control group,ADR model group and treatment group.The level of body weight,right renal weight and urinary albumin creatinine ratio (UACR) were measured,Furthermore, the morphological changes of renal tissue were observed under microscope.The mRNA and protein expressions of Nephrin in renal tissues were determined by RT-PCR and immunofluorescence respectively. Results  There was a significantly higher level of body weight and lower level of UACR in normal control group and treatment group than that of ADR model group (P < 0.05). There were intensy expression of Nephrin in glomerular mesangial region in normal control group and treatment group compared with in ADR model group.There were significantly higher expressions of Nephrin mRNA in those of normal control group and treatment group than in ADR model group .(P <0.05) There were no significant differences in Nephrin mRNA between normal control group and treatment group( P>0.05)Conclusion  The method of nourishing kidney and activating blood recipe of traditional Chinese medicine attenuates Nephrin decrease in FSGS change nephropathy model which is closely related with the reduction of proteinuria.

    Effects of HTK and Perfadex Solution on the Function of Pulmonary Artery Endothelium

    Objective  To study the effect of HTK and Perfadex solution on the function of pulmonary artery endothelium. Methods   Fifty-four porcine pulmonary rings in 2mm long were randomly divided into three groups. Control group (n=18): Incubation in Krebs-Henseleit (KH) at 4℃ for 4 hours; group A (n=18) or B (n=18): Incubation in HTK or Perfadex solution at 4℃ for 4 hours. Endothelium-derived hyperpolarizing factor (EDHF)-mediated relaxation (percentage of -7.5logM U46619 precontraction) induced by bradykinin or calcium ionophone A23187 in the present of indomethacin (7μmol/L), L-NNA (300μmol/L) and oxyhemoglobin (20μmol/L)were measured in the organ chambers. Results   Vasoconstriction induced by U46619 is no significant difference among three groups(P>0.05). Compared with control group, the relaxation induced by bradykinin or A23187 was no significant difference in group A or group B. Conclusions  Endothelium-derived hyperpolarizing factor (EDHF) plays an important role in endothelium-mediated relaxation of porcine pulmonary artery. EDHF-mediated relaxation showed similar performances when the lung preserved with HTK or Perfadex solution.

    Objective   To evaluate the effect of probucol, folic acid and vitamin B12 combined medication on vascular endothelial dysfunction induced by hyperhomocysteinemia in rats. Methods   Seventy male SD rats were randomly divided into seven groups. Each group had ten rats: normal control group, Hhcy group, low-dose- probucol group, middle-dose- probucol group, high-dose- probucol group, folic-acid-vitamin B12 group, folic-acid-vitamin B12 and probucol group(combination treatment group). The normal control group was fed on plain feedstuff for twelve weeks. Other groups were fed on homomethionin feedstuff for twelve weeks .And then from the eighth week added six kinds of intervention measures for four weeks,such as water, probucol(three kinds of dose), folic acid and vitamin B12,folic acid and vitamin B12 and probucol. The plasma concentrations of the homocysteine(Hcy) , oxidation low density lipoprotein(OX-LDL),nitric oxide(NO)levels were measured at the beginning ,the eighth week and the twelveth week. Hcy were measured by high pressure liquid chromatography analysis (HPLC). OX-LDL and NO were measured by enzyme-linked immunoassay (ELISA) method. Results   After adding high concentrations of methionine feedstuff, the level of Hcy was significantly higher than that normal control group (P<0.05) while the level of OX-LDL increased and the level of NO decreased. After four week intervention, the levels of Hcy, OX-LDL and NO in groups added the three different doses probucol, and added the folic acid and vitamin B12 had statistically differences compared with normal control group(P<0.05). The plasma levels of Hcy and OX-LDL in the combination treatment were lower than other intervention groups(P<0.05) while the level of NO was more obviously higher(p<0.05). The Hcy, OX-LDL and NO levels in the combination treatment group had not statistically significant (P > 0.05) compared with normal control group. The levels of Hcy, OX-LDL and NO had no statistically differences (P >0.05) in groups added the three different doses probucol of rats . Correlation analysis showed Hcy levels were positively correlated with OX-LDL levels(P<0.05). Conclusion   The present study demonstrated that OX-LDL interacted with Hcy, leading to endothelial dysfunction possiblely . With probucol,folic acid and vitamin B12 combined medication, the level of Hcy and OX-LDL could be decreased more significantly, meanwhile the level of NO increased , and thus the endothelial function was improved, which may slow down the development of atherosclerosis.

    Abstract   Objective  To identify and locate adult stem cells in ectopic endometrium and to find its markers using the 5-bromo-2-deoxyuridine (BrdU) labeling method in mouse model of endometriosis. Method  The female nude mice were pulse labeled by BrdU for 5 consecutive days, 24h prior to the endometriosis model was established. The label-retaining cells(LRCs) were identified by getting BrdU labeling rate by immunohistochemistry on 4h,1w, 3w, 5w, 6w,7w,8w,9w when the last injection was finished. The expression level of CD34 and CD146 on LRCs were detected by double staining of immunohistochemistry. Result  The maxium BrdU labeling rate was 69.89±0.78% and 73.27±1.06% for epithelial and stromal respectively. After a 6-week and an 8-week chase, 2.07±0.19% epithelial cells and 4.63±0.72% stromal cells retaind BrdU labeling respectively, which were considered to be epithelial and stromal LRCs.Both the epithelial LRCs and the stromal LRCs were mainly located in the boundary of the lesions and the mice tissues where the lesion was implanted. The positive expression rate of CD34 and CD146 were 19.32±0.03% and 14.76±0.03% respectively. CD34 did not co-expressd on LRCs, CD146 co-expressed on a few stromal LRCs. Conclusion  Two kinds of stem cells were identified successfully in ectopic endometrium by label-retaining cell approach, which were epithelial and stromal LRCs located in the boundary of the lesions and the mice tissues where the lesion was implanted. It seems that CD34 was not the marker for ectopic endometrial stem cells, whether CD146 can be the candidate marker for ectopic endometrial stem cells or not by combining with other markers deserving further research.

    The occurrence of dysbacteria in hemodialysis and influencing factor

    Abstract  Objective  To study the incidence rate of dysbacteria after antibiotics applied in hemodialysis patients,expectation to approach the possible correlation between clinic index and dysbacteria. Methods  Those who have taken three more months hemodialysis were the HD group ,all of them needed antibiotic therapy. While the people who need antibiotic therapy but not accompanied by renal or digestive disease were allocate to control group. Recording their clinic index immediately after hospitalization , and the incidence rate of dysbacteria after antibiotics applied.Results   The incidence rate of dysbacteria was 34.5% in HD group while it was 18.1% in the control group; Among these clinic index age,creatinine and SGA is the factor that could impact dysbacteria, regression coefficients were 0.037(p<0.05),0.003(p<0.05) and -1.152(p<0.001)respectively and OR was 1.307,1.003 and 0.316 . Conclusion   It is true that micro-ecological has been changed in hemodialysis, the incidence of dysbacteria is more often than the control group after antibiotics applied , age,creatinine and SGA is the factor that could impact dysbacteria,SGA is the protect factor.

    The Risk Factors of Mid and Long-term Patency Rate for Grafts after Off-pump Coronary Artery bypass Grafting

    Abstract  Objective To analyze the mid and long-term patency rate for grafts after off-pump coronary artery bypass grafting and the risk factors of patency rate. Methods  Seven thousand five hundred and twenty-one patients were underwent CABG in Tianjin Chest Hospital, one hundred and thirteen cases underwent OPCABG were followed up, among them, fifty-four cases were followed up by searching the coronary artery angiography and fifty-nine cases were followed up by searching dual Source CT. The patients were 70 males and 43 females with a mean age of 63.02±8.83 years at the time of surgery. Preoperative coronary angiography revealed one-vessel disease in 5 patients, two-vessel disease in 13 patients, three-vessel disease in 95 patients, and left main coronary artery disease with or without peripheral coronary artery disease in 32 patients.Eighty-six patients had high blood pressure, 42 patents had diabetes mellitus,26 patents had hyperlipidemia,36 patents had old Myocardial Infarction,14 patents had old cerebral infarction,40 patents had a smoke by now and 31 patients were older than 70 years. The mean following up period was 27.01±18.59 months and the average number of distal anastomoses per patient was 2.96±0.86. Internal thoracic arteries (ITA) were used in 75 and saphenous veins were used in 260 . Calculate the patency rate for arterial graft and the patency rate for saphenous vein grafts respectively and analyse the risk factors of mid and long-term patency rate for grafts after off-pump coronary artery bypass grafting. Results  The patency rate of all grafts after coronary artery bypass grafting was 83.0%;and 98.0% for internal thoracic arteries; the one year and five years patency rate was 98.6% and 88.5% for ITA; the patency rate was 78.1% for saphenous vein grafts, and the one year and five years patency rate was 85.9% and 61.3% for saphenous vein grafts; the patency rate with angina group was 78.9% and 92.2% for angina-free group, there were differences between the two groups(P=0.007. Conclusion   The ITA grafts have a better long-term patency than saphenous vein grafts;The follow -up period after OPCABG、the average number of distal anastomoses and the female were independent risk factors of patency rate for grafts after CABG; Graft lesion after CABG is an important factor leading to postoperative recurrent angina pectoris.

    Primary Biliary Cirrhosis-Autoimmune Hepatitis Overlap Syndrome in 49 Cases Analysis

    Objection  To summary and analyze the clinic characteristics features and associated extrahepatic autoimmune diseases of patients with primary biliary cirrhosis -autoimmune hepatitis overlap syndrome(AIH-PBC OS). Methods   To analyze retrospectively the clinic characteristics and pathologic features of 49 patients that were diagnosed as PBC-AIH OS and been followed-up when been treated in digestive department of general hospital during Jan.2000 to Jan.2012. Results   These patients were most female with age of 51-60 years old, and the mean age of onset was 57.2±8.9 years old. The levels of serum ALT, AST, GGT and TBIL were increased in various degree in all the patients, while serum GLO/IgG was inceased in 79.6% patients. Positive rate of ANA,AMA and SMA was 98.0%,89.8% and 6.1% respectively. When been diagnosed, 38.8% of the patients had been developed to hepatic cirrhosis, 57.9%(11/19)of them at the stage of decompensation . Besides, pathologic stage was 3,4 in 16 (66.7%)patients with PBC and 1,2 stage in 8(33.3%)ones. Among them, there were 20 (40.8%)patients had extrahepatic autoimmune diseases at the same time, 16 cases (32.7%) had two kinds of autoimmune disease, 4 cases (8.1%) with three kinds of autoimmune diseases. Abdominal ultrasonic wave showed abnormality in 93.9% of patients,the abdominal lymph nodes were enlarged in 91.8% of the patients. Conclusion  Most the patients with PBC-AIH OS had been diagnosed as hepatic cirrhosis at the first time, so as to make early diagnosis and treatment. The symptoms were various and complicated so that it’s significant to expand the screening of incorporated autoimmune diseases. Abdominal lymph nodes enlargerment found by imaging examination may had guiding sense to diagnosis.

    Observation of Compound Musk Injection in Treating Children with Viral Encephalitis

    Objective   To investigate the efficacy and safety of compound musk injection on viral encephalitis in children . Methods   113 cases with viral encephalitis in children were randomly divided into two groups: the control group (53 cases) were treated by conventional therapy and the treatment group (60 cases) were treated with compound musk injection of 0.5mL/kg, intravenous infusion, daily 1 in addition to the conventional therapy. A course of teatment of both groups were 7 days. Results  The efficiency of the treatment group and the control group were 96.67% and 71.70%. The difference was highly statistically significant (P<0.01). As for the disappearance time of clinical symptoms and syndromes, the treatment group was significantly shorter than the control group. The difference was statistically significant (P<0.01 or P<0.05). The adverse incident relevant to drugs in two groups was no occurred at the course of therapy. Conclusion   The effect of compound musk injection on viral encephalitis in children is good and safe.
