    Molecular Detection and Analysis of Resistance in Mycoplasma Pneumoniae in Children Hospitalized with Pneumonia in Tianjin

    [Abstract] Objective  To study the characterization and molecular mechanisms of macrolide-resistant mycoplasma pneumoniae (MP) in children hospitalized with pneumonia in Tianjin. Methods  A total 237 of alveolar lavage fluid specimens were collected from children with pneumonia hospitalized in Tianjin children’s hospital from October 2010 to December 2011. The DNA of them was extracted. The genomic fragments of the strains were amplified by using polymerase chain reaction (PCR) method with specific primers. The positive strains were selected randomly to be amplified with nest PCR method. The amplifiable production was electrophoresised and purified and the 23 S rRNA gene sequences were sent to sequencing. The results of sequencing were compared with the sequence of MP M129 to detect the mutations. Results  Among 237 specimens,129 samples were MP positive after detecting by PCR with the primers. The positive rate was 54.43% (129/237). Of the 50 strains that were selected to sequence, 4 strains and the standard strains were completely same. The other 46 strains showed A-to-G transition at position 2063. There was no other site of mutation. The percentage of macrolide-resistant was 92% (46/50). Conclusion  The macrolide-resistance in mycoplasma pneumoniae is serious in Tianjin. The major molecular mechanism is A-to-G transition at position 2063 of the 23 S rRNA domain Ⅴ.

    The predict effect of pregnancy plasma protein A and macrophage inhibitory factor 1 on miscarriage in the early pregnancy period

    Abstract Objective: To investigate the predict effect of pregnancy associated plasma protein A(PAPP-A) and macrophage inhibitory cytokine-1(MIC-1) on miscarriage in the early pregnancy period. Methods: From July 2010 to July 2011, we collected gestation maternal serum at 6-12 weeks of pregnancy and tracked the outcome of pregnancy. During observation period, all of 20 pregnant women appeared miscarriage (miscarriage group). Their serum levels of PAPP-A and MIC-1 were compared with 120 normal pregnant women (control group). Results: PAPP-A and MIC-1 level in the miscarriage group were lower than in the control group at 6-12 weeks of pregnancy (P <0.05). In the multivariate logistic regression analysis, PAPP-A (OR, 95% CI: 0.79-0.84) and MIC-1 (OR, 95% CI: 0.83, 0.77-0.90) was negatively correlated with miscarriage. Conclusion: In the early stage of pregnancy, PAPP-A and MIC-1 could predict miscarriage effectively. When the two indicators appeared a low level, clinicians should take appropriate treatment to ensure gestation.

    The Effects of Fructus Corni Polysaccharides on the Proliferation of Human Cervical Cancer Cells and Expressions of Survivin and Bcl-2

    [Abstract] Objective  To explore the inhibitory effect of fructus corni polysaccharides on human cervical cancer cells and expressions of Survivin and Bcl-2 in HeLa cells. Methods  MTT assay was used to determine the cell proliferation. Flow cytometry and immunohistochemistry SP methods were used to detect the apoptosis and protein expressions of Survivin and Bcl-2 in HeLa cells. Results  There was a significant inhibitory effect of fructus corni polysaccharides on the proliferation in HeLa cells. The inhibitory degree was correlated with different concentrations and different action times of fructus cornipolysaccharides. The apoptosis rates of HeLa cells were 10.1%, 21.6% and 28.9% for fructus corni polysaccharides concentrations of 6.25 g/L, 12.5 g/L and 25 g/L. The expressions of Survivin and Bcl-2 in HeLa cells were significantly lower in fructus corni polysaccharides treatment group than those of control group. Conclusion  Fructus corni polysaccharides showed significant inhibitory effects on the proliferation in HeLa cells, which also decreased Survivin and Bcl-2 expressions and induced the cell apoptosis.

    [Abstract] Objective  To detect the differently expressed genes of Streptococcus mutans (S. mutans ) cultured in 1.0% glucose and 0.2% glucose. Methods  S. mutans was cultured in 1.0% and 0.2% glucose. The genetic expression of S. mutans under different conditions was detected using gene chips built previously. Results  In 1.0% glucose environment, expressions of 57 S. mutans genes significantly changed compared with 0.2% glucose. Forty-one genes associated with sugar transport and energy metabolism, virulence, pathogenesis, DNA metabolism and regulatory functions were significantly up-regulated. While 16 genes encoding some competence proteins and enzymes related to cellular processes were significantly down-regulated. Conclusion  The environmental glucose affects multiple gene expressions of S. mutans.

    [Abstract] Objective  To study the effects of different types of glucocorticoid (GC) on the serum cortisol (COR) and adrenocorticotropic hormone (ACTH) in patients with non-frequently relapsing nephrotic syndrome (PNS). Methods  Sixty patients with non-frequent relapses PNS were randomly divided into prednisone group (Ⅰgroup), triamcinolone group (Ⅱ group) and methylprednisolone group (Ⅲ group). Each of them was treated with standard glucocorticoid therapy. The serum levels of cortisol and ACTH were observed before treatment (A phase), the end of remission phase (B phase), the day after using 1 mg·kg-1·d -1 prednisone (C phase), the day after using 0.25 mg·kg-1·d -1 prednisone (D phase) and 4 weeks after the end of glucocorticoid therapy (E phase). Results There were significant differences in the serum COR levels in the B phase between three groups (P < 0.05). There were also significant differences in the serum COR levels of Ⅰgroup between A phase and B phase, C phase and D phase (P < 0.05 orP < 0.01), and there was significant difference between B phase and D phase (P < 0.05). There were no significant differences in the serum COR levels between D phase and A phase of Ⅱgroup, and between C phase and A phase of Ⅲ group (P > 0.05). There were significant differences in the serum ACTH levels between B phase and A phase of three groups (P < 0.05). The serum ACTH levels were higher in D phase than those in A phase of Ⅰ group and Ⅱgroup. The serum ACTH level was higher in C phase than that of A phase in Ⅲgroup (P < 0.05). The serum ACTH level was still higher in E phase than that of A phase in Ⅰgroup, but the level returned to A phase from E phase in Ⅱ group and Ⅲgroup. Conclusion  Triamcinolone is the most obvious impact on the serum COR level. Prednisone affects the smallest and is the slowest recovery. Methylprednisolone shows the fastest recovery. The inhibitory effect of prednisone on pituitary function is longer than that of triamcinolone and methylprednisolone.

    Experimental research on antibechic, expectorant and antiasthmatic effects of extracts from Clausena lansium(Lour.) Skeels leaves and its active fractions

    [Abstract] Objective  To study the antibechic, expectorant and antiasthmatic actions of Clausena Lansium (Lour.) Skeels (CLLSL) leaf extracts. Methods (1)Sixty mice were randomly divided into six groups: normal control group, pentoxyverine group, petroleum ether extraction group, dichloromethane extraction group, ethyl acetate extraction group and liquid extraction group. After corresponding drug administration for seven days, cough models were induced by concentrated ammonia in mice. The latent phase and the cough frequency per two minutes were recorded in mice.(2)Another sixty mice were randomly divided into six groups: normal control group, ammonium chloride group, petroleum ether extraction group, dichloromethane extraction group, ethyl acetate extraction group and liquid extraction group. After corresponding drug administration for seven days, the concentration of tracheal phenolsulfonphthalein was detected by 5% phenolsulfonphthalein solution intraperitoneal injection.(3)Seventy mice were randomly divided into seven groups: normal control group, model group, dexamethasone group, petroleum ether extraction group, dichloromethane extraction group, ethyl acetate extraction group and liquid extraction group. The asthmatic mouse models were established by ovalbumin(OVA)intraperitoneal injection at the 1th, 7th and 14th day, and atomization inhalation was given from 18-22 days. The asthmatic mice were treated with corresponding medicines respectively before atomization inhalation. The total leukocyte number and different cell counts in bronchoalveolar lavage fluid (BALF) were observed. Results  (1) Compared with normal control group, the latent phase was significantly longer and the cough frequency per two minutes was significantly reduced in petroleum ether extraction and dichloromethane extraction groups (P <0.01). (2) Compared with normal control group, dichloromethane extraction, ethyl acetate extraction and liquid extraction could significantly increase the concentration of phenolsulfonphthalein of mouse trachea (P <0.01). (3) Compared with model group, the total leukocyte number, eosinophile granulocyte count and neutrophil count in BALF of asthmatic mice were significantly decreased in petroleum ether extraction group (P <0.01). Conclusion The extracts of CLLSL have antibechic, expectorant and antiasthmatic effects.

    The Evaluation of the Michigan Neuropathy Screening Instrument in the Diagnosis of Diabetic Peripheral Neuropathy

    [Abstract] Objective  To investigate the value of the Michigan Neuropathy Screening Instrument (MNSI) in the diagnosis of diabetic peripheral neuropathy (DPN). Methods  A total of 267 patients with diabetes mellitus underwent the measurements of MNSI and neural electrophysiological test. All of the participants were divided into three groups based on MNSI score: 112 patients in group A (score 0-2.4), 88 in group B (score 2.5-4.4) and 67 in group C (score≥4.5). The differences of gender, age, history of diabetes and neural electrophysiological test results were compared between three groups. The stepwise logistic regression was used to analyze the effect of MNSI questionnaire for the DPN. The relationship between the MNSI questionnaire, exam score and the index mentioned above was analyzed. Results  (1) The patients in group A were much younger and had a shorter history of diabetes than those in group B and group C. Compared with the other two groups, patients in group C got significantly higher perception of non-waveform in the neuroelectricity test (P <0.05). (2) There were significantly higher values, including the amplitude of the motor nerve of the posterior tibial nerve, the nerve conduction velocity of the sensory nerve and motor nerve, in group A than those of group B and group C. The diagnosis rate of DPN was significantly lower in group A than that of group B and group C (P <0.05). (3) The questions of 1, 2, 11 and 14 in MNSI were the risk factors for DPN. (4) The MNSI questionnaire was positively correlated with age and history of diabetes, and negatively correlated with the amplitude of motor nerve of posterior tibial nerve and sensory nerve conduction velocity of both legs.The MNSI score was positively correlated with age, history of diabetes and body mass index, and negatively correlated with the amplitude of motor nerve of posterior tibial nerve and sensory nerve, motor nerve conduction velocity of both lower limbs (P <0.05). Conclusion  Regard the MNSI questionnaire 4 and exam score 2 as a cut-off point, higher score will get a high risk of DPN, and the nerve conduction will be worse.

    Preparation of lidocaine hydrochloride-loaded transactivation transcriptional activator conjugated polymetric liposomes and permeation through mouse skin in vitro

    [Abstract] Objective  To prepare the lidocaine hydrochloride-loaded transactivation (TAT) conjugated octadecyl quaternized lysine modified chitosan polymeric liposomes (LID-TAT-PLs) and to study its in vitro skin permeation characters in mice. Methods  LID-TAT-PLs were prepared by reverse-phase evaporation method. The concentrations of lidocaine hydrochloride were determined by HPLC. The isolated mouse skin and Franz diffusion cell were used to evaluate the characters of permeation of LID-TAT-PLs, LID-PLs and lidocaine hydrochloride injection (LID-IJ). Results  The permeation rate and cumulative permeation amount were significantly higher in LID-TAT-PLs group than those of LID-PLs and LID-IJ groups (P <0.05). Conclusion  TAT can significantly enhance the transdermal permeation of drugs in mice. The LID-TAT-PLs can be prepared via a relatively simple method.

    The study on the mechanism of Ulinastatin for treatment of lung injury induced by severe acute pancreatitis in rat

    [Abstract] Objective  To evaluate the effect of ulinastatin on the level of surfactant protein A (SP-A) in bronchoalveolar lavage fluid (BALF) in lung injury induced by severe acute pancreatitis (SAP) in rat model, and protection mechanisms of ulinastatin thereof. Methods Sixty SD rats were randomly divided into 3 groups: control group (n=20), SAP model group (n=20) and ulinastatin group (n=20). The SAP model was induced by retrograde infusion of 5% sodium cholate into the bile-pancreatic duct. Twenty-four hour after operation, the left lung’s BALF liquid was collected and the level of SP-A in BALF liquid was detected by ELISA method. The BALF liquid was differentially counted by using microscope. The wet-to-dry lung weight ratio was recorded. Results The value of wet/dry lung weight ratio and the level of neutrophil granulocytes in BALF were higher in SAP group than those of control group and ulinastatin group. The wet/dry lung weight ratio and the neutrophil granulocyte content in BALF were significantly higher in ulinastatin group than those of control group (P < 0.05). The levels of SP-A and macrophages in BALF were significantly lower in SAP group than those of control group. The level of lymphocytes was significantly higher in SAP group and ulinastatin group than that of control group (P < 0.05). There were no significant differences in total cell number of BALF between three groups. Conclusion The level of SP-A in BALF was lower in lung injury induced by SAP. Ulinastatin can protect the SP-A of BALF in SAP rats.

    The study of clinical efficacy of radiofrequency thermal ablation of targets in intervertebral disc disease

    Abstract Objective:To explore the clinical efficacy of radiofrequency thermal ablation of targets in treatment of intervertebral disc disease.Methods:After Collecting the clinical data of 130 patients comforming to the indication through CT and MRI,sixty-five patients(A group) were treated by radiofrequency thermal ablation of targets,anather sixty-five patients(B group) were cured by conservative treatment and anlysed the general clinical data and excellent and good rat in three monthes after treatment between two groups.Results:In the A groups’s patients,forty-nine patients gained excellent effects,eleven patients obtained good effects,four patients had poor effects and one cases had no effects after three monthes of the treatment of radiofrequency thermal ablation of targets with the excellent and good rate of 92.3%. In the B groups’s patients,twenty-seven patients gained excellent effects,sixteet patients obtained good effects,fourteen patients had poor effects and eight cases had no effects after three months of conservative treatment with the excellent and good rate of 66.2%.There were significant difference in treatment of intervertebral disc disease between radiofrequency thermal ablation of targets and conservative treatment(P<0.05).Conclusions:Radiofrequency thermal ablation of targets for treatment of intervertebral disc disease is a safety and efficacious treatment in the field of minimally invasive.

    Preoperative enteral nutrition support treatment on the respiratory function of esophageal cancer patients with low pulmonary function

    Abstract Objective:To explore the preoperative enteral nutrition support treatment on the respiratory function of esophageal cancer patients with low pulmonary function. Methods:Choose 2011 1 to 12 56 patients with low pulmonary function and esophageal cancer, according with the maximum voluntary ventilation(MVV) was less than 0.5 and the ratio of the first one second to forced vital capacity (FEV1) in the forced expiratory volume (FVC)was less than 0.7,were randomly divided into nutrition group (28patients)and control group (28 patients). The nutrition group patients were given enteral nutrition supportment and the control group patients were given equal amount of calorie daily diet.Both groups were given the same routine care. After two weeks compared two sets of patients with pulmonary function and arterial blood gas.Results: The values of FVC, FEV1, MVV, arterial partial pressure of oxygen(PaO2), arterial partial pressure of carbon dioxide(PaCO2) were significantly improved after treatment, the difference was statistically significant (P < 0.05), but FEV1/ FVC difference was not statistically significant (P > 0.05)。 The control group index was also improved, but the difference was not statistically significant (P > 0.05). Compared with the control group the values of FVC、FEV1、MVV、PaO2 and PaCO2 were significantly improved in the nutrition group (P < 0.05), but no significant difference in the level of FEV1/FVC between two groups of patients(P > 0.05). Conclusion:Preoperative enteral nutrition support can significantly improve the low pulmonary function with esophageal cancer patients of respiratory function, to provide perioperative treatment the necessary supportment.

    Expression of collagen, MMP-2,MMP-9,TIMP-2 and TIMP-1 in patients with direct and indirect inguinal hernia

    [Abstract] Objective  To investigate the expressions of collagen, MMP-2, MMP-9, TIMP-2 and TIMP-1 in patients with direct and indirect inguinal hernia, and explore the pathogenesis of the inguinal hernia. Methods Herniorrhaphy was performed in 46 patients with inguinal hernia (19 direct and 27 indirect), and the expressions of collagen, MMP-2 and TIMP-2 in the transversalis fascia were examined by using Masson stain and Western blot. The serum levels of MMP-2, MMP-9, TIMP-2 and TIMP-1 were examined by using ELISA method. Results  The irregular and disorder arrangement of collagen were found in patients with direct hernia, and the regular arrangement of collagen was found in patients with indirect hernia. There was a significantly lower level of collagen in the transversalis fascia in patients with direct hernia than that of patients with indirect hernia(P < 0.05). There was a significantly higher serum level of MMP-2 and the higher level in the transversalis fascia in patients with direct hernia than that of patients with indirect hernia(P < 0.05). The levels of TIMP-2 and TIMP-1 were significantly lower in patients with direct hernia than that of patients with indirect hernia(P < 0.05). Con clusion  The disordered expressions of collagen, MMP-2, TIMP-2 and TIMP-1 might be associated with the development of direct inguinal hernia.

    [Abstract] Objective: Observation of methylprednisolone combined with montelukast sodium in treatment of capillary bronchitis curative effect.Methods: 56 cases of infants with bronchiolitis were randomly divided into two groups, 28 cases in each group, the control group was given conventional treatment, the treatment group was treated with routine therapy plus methylprednisolone intravenous, oral montelukast sodium.Results: the treatment group total effective rate 92.9%, significantly higher than that of control group 67.9 ﹪.Group therapy in relieving asthma, dyspnea, wheezing and moist rale decreasing time was better than the control group.Conclusion: methylprednisolone combined with montelukast sodium in the treatment of bronchiolitis effect is good, can be used as bronchiolitis is a safe, effective treatment scheme.

    A clinical observation of VSD treatment of a large area of soft tissue trauma of the four limbs in less-developed area

    Summary Background:With a large area of soft tissue trauma were difficult to repair,it not only need a long time for treatment,but also so expensive.As a result,their economic burden was increased for those patients who lived in less-developed area.Nowadays,vacuum sealing drainage technique for the repair of various wounds have been to applicate widely. Purpose:Discuss on vacuum sealing drainage treatment of a large area of soft tissue trauma and efficacy in less-developed area.Method:The 74 patients were divided into VSD group 40 cases and control group with 34 cases.Those in control group were given routine dressing change after debridement .Those in VSD group were covered with VSD dressing and continued negative pressure suction after debridement.We removed the VSD dressing 5~10 days later.Then evaluated and appraised the curative effect according to the comparison of two groups of the wound cleaning time、two stage operation time、the wound area after treatment 、frequency of dressing changes、skin effect、application of antibiotic cost、the total duration of hospitalization and so on.Result:As a result,the indicators in VSD group were better than the control group.The differences between the two group were statistically significant.(p<0.05~0.01)In conclusion,the VSD treatment of a large area of soft tissue trauma is more effective than the traditional method.The VSD treatment will develop well in less-developed area because it reduces medical costs significantly.

    The Diagnostic Value of Bronchoalveolar Lavage to Pneumocystis Carinii Pneumonia

    【Abstract】 Objective To evaluate the diagnostic utility of bronchoalveolar lavage(BAL)in solid organ transplant (SOT) recipients with Pnuemocystis carinii pneumonia(PCP) . Methods 117 bronchoscopys with BAL were performed in 114 SOT hospitalized recipients with suspected pulmonary infections in our hospital. Cytological smear and polymerase chain reaction (PCR) were done to detect P carinii in lavage specimens. Results A definite P carinii etiological diagnosis by BAL was confirmed in 36 patients. P carinii infections alone accounted for 36 episodes, mixed infections by P carinii and CMV accounted for 6 episodes, mixed infections by P carinii and bacterium accounted for 4 episodes. 31 episodes were observed in period 2, 5 episodes in period 3. Conclusion BAL is an extremely useful tool for PCP diagnosis in solid organ transplant (SOT) recipients. The procedure is safe and the results obtained by the technique can guide specific antimicrobial therapy.

Methods The clinical feature,radiological characteristics,laboratory diagnosis and treatment of one case of pulmonary nocardiosis. Results The disease has a nonspecific clinical feature and radiological characteristic may present cavitary lesions.The most precise diagnosis was bacteriological test.The treatment must last a long course. Conclusion Immunosuppression is the most important predisposing factor for pulmonary nocardiosis. Sulfonamides are still the drugs of first choice and linezolid appears an effective option for the treatment of nocardiosis. Key words: Nocardia infections; lung diseases;diagnosis;drug therapy

    [Abstract] Objective  To evaluate the association of cyclin-dependent kinase 5 regulatory subunit-associated protein 1-like 1 (CDKAL1) gene rs7756992 polymorphism and type 2 diabetes mellitus (T2DM). Methods The case-control studies of CDKAL1 rs7756992 polymorphism and T2DM were collected and meta-analysis was performed by RevMan 5.0 software. Results A total of 11 documents concerning 32 604 diabetics and 23 939 controls were included in this study. The summaryORs(with 95%CI)under allele contrast and recessive model were 1.18 (1.13, 1.23) and 1.34 (1.25, 1.44) , respectively. Subgroup analysis showed the correlation between the CDKAL1 gene rs7756992 polymorphism and incidence of T2DM in European and Asian populations. Conclusion In Asian and European populations,the allele G is the risk factor to T2DM under the two heredity models.

    Analysis on Factors Influencing the Pregnancy Rate of Intrauterine Insemination with Husband Sperm

    [Abstract] Objective  To investigate the various factors that affect pregnancy rate following intrauterine insemination (IUI) with husband sperm. Methods  A total of 3 998 IUI cycles were retrospectively evaluated. The correlation between pregnancy rate and following factors were analyzed including female age and duration of infertility. Results  A total of 3 093 infertile couples were treated with 3 998 cycles of IUI. Five hundred and forty-seven cases conceived the clinical pregnancy during the study. The pregnancy rate was 13.7% per cycle. There were significant differences in female age, duration of infertility, IUI cycles, ovulation protocol, progressive sperm quantity, timing of IUI and diameter of the leading follicular. There were no significant differences in different types of infertility, different causes of infertility, tubal patency, endometrial thickness and endometrium type (P > 0.05). Results of non-conditional stepwise regression analysis showed that the increase of female age and number of IUI cycles were risk factors of IUI outcome. Meanwhile, the stimulating protocol (HMG group and Le? trozole group), progressive sperm quantity (5×106-10×106group) and tubal patency were protective factors of IUI outcome. Conclusion  The female age, the number of cycles, ovulation protocol, progressive sperm quantity and tubal patency are of great importance in the pregnancy rate of IUI cycles.

    Influence of chronic inflammation on angiogenesis in mouse malignant melanoma

    [Abstract] Objective  To investigate the influence of chronic inflammation on the growth and angiogenesis in mouse transplanted melanoma. Methods Sixty inbred C57BL mice were randomly divided into three groups,control group (C),inflammation + tumor group (I+T) and tumor group (T). The subcutaneous balloon was made on the mouse back, and the suspension of B16 melanoma cells was injected into the groin areas in both I+T group and T group. Either complete Freund adjuvant (CFA) or physiological saline was injected into the subcutaneous balloon of mice respectively. No intervention measures were taken in C group. Tumor tissues were stained with CD31 immunohistochemistry and the microvessel density (MVD) was counted. Enzyme-linked immunosorbent assays (ELISA) was used to detect the serum contents of interleukin (IL)-6, IL-17, IL-23 and vascular endothelial growth factor (VEGF) in three groups. Results  The tumor volumes and weights were significantly larger in I+T group than those in T group from 15 days to 27 days after model establishment (P < 0.05 orP < 0.01). The values of MVD in tumor tissues were significantly higher in I+T group than those in T group 9, 15, 21 and 27 days after model estab lishment (P < 0.01). The serum levels of IL-6, IL-17, IL-23 and VEGF were significantly higher in I+T group and T group than those of C group (P < 0.01). The serum levels of IL-6, IL-17, IL-23 and VEGF were significantly higher in I+T group than those of T group except for the 9-day after model establishment (P < 0.05 orP < 0.01). Conclusion The chronic inflammation induced by CFA can promote the development of mouse melanoma. And tumor angiogenesis could be promoted by some cytokines secreted by inflammatory cells.

    Promoter Methylation and Protein Expression of Apoptosis Protease Activating Factor-1 Gene in Esophageal Squamous Cell Carcinoma

    [Abstract] Objective  To investigate the promoter methylation of apoptosis protease activating factor-1 (Apaf-1) and its association with the protein expression of enhancer of zeste homolog 2 (EZH2) in esophageal squamous cell carcinoma (ESCC). Methods  The methylation-specific PCR (MSP) method was used to examine the methylation status of Apaf-1 in 128 ESCC patients, and immunohistochemistry method was used to detect the protein expression of Apaf-1 and EZH2 in tumors and corresponding normal tissues. Results  The frequencies of Apaf-1 methylation in ESCC tumor tissues (52.3%, 67/ 128) were significantly higher than those in normal tissues (4.7%, 6/128) (P < 0.01), which was associated with TNM stage. The positive protein expression of Apaf-1 was significantly decreased in ESCC tumor tissues (41.4%, 53/128) compared with that of paired normal tissues (94.5%, 21/128) (P < 0.01). The positive protein expression of EZH2 was significantly increased in ESCC tumor tissues (72.7%, 93/128) than that of paired normal tissues (12.5%, 16/128) (P < 0.01). The protein expressions of Apaf-1 and EZH2 were associated with the TNM stage and pathological differentiation in ESCC patients (P < 0.01). The methylation status of Apaf-1 in ESCC was negatively correlated with the positive protein expression. The protein expressions of Apaf-1 and EZH2 were negatively correlated in ESCC. Conclusion  Promoter hypermethylation of Apaf-1 and the higher protein expression of EZH2 may play important role in esophageal squamous cell carcinoma.

    Research of the treatment of lipoxin A4 to preeclampsia rats model

    [Abstract] Objective  To investigate the effects of lipoxin (LX) on the plasma nitric oxide (NO) concentration in preeclampsia rats, and make an experiment foundation for the treatment of human preeclampsia. Methods Thirty pregnant rats were randomly divided into three groups: normal group (saline), preeclampsia group (endotoxin) and LX group (endotoxin + LX). The experimental rat model of preeclampsia was established with small dose of endotoxin. LX was tail injected intravenously in the day that endotoxin was injected in LX group. The same volume of normal saline was tail injected intravenously in saline and endotoxin groups. Levels of blood pressure, urine protein and LX were detected on 8, 15 and 18 days after successful Modeling. The body weights of newborn rats were measured after operation on the 20th day of pregnancy. The NO concentration of abdominal aorta in mother rats was detected by nitrate reductase enzyme method. The level of LX was detected by double antibody sandwich method. Results  The endogenous lipoxin level was increased with the gestational age. The level of LX on 8 days of pregnancy was decreased in endotoxin group than that of saline group. The body weights of newborn rats and NO levels of pregnant rats were significantly higher in LX group than those of endotoxin group. There were significantly lower levels of blood pressure and urine protein in LX group than those of endotoxin group (P < 0.05). Conclusion  Lipoxin may be potentially useful in treatment of preeclampsia in rats.

    Diagnosis and treatment and prognosis of urachal carcinoma

    [Abstract] Objective:To probe into the diagnosis,trestment and prognsis factors of urachal carcinoma. Methods:The clinical data,diagnosis,therapeutic regimen and the results of the prognosis of 6 cases of urachal carcinoma treated from 2007 to 2011 were review and analyzed.Results: Of 6 cases, 5 cases were treated by extensive partial cystectomy;1 case received radical cystectomy and modified cutaneous ureterostomy.Follow up 6~52 months,with an average of 24 moths. 2 cases died after 6, 23 months as distant metastasis,and the other 4 cases were tumorfree survival. Conclusion:Urachal carcinoma is a rare and highly malignant with poor prognosis. Surgical excision being the primary trestment.It is unclear whether radiation or chemotherapy are of any benefit to patients with urachal carcinoma,but some tumors may respond to cisplatin and/or 5FU based chemotherapy. Although the prognosis of this disease was bad,the extensive partial bladder excision can cure the majority of urachal cancers whose stages are low grade.Tumor stage and margin status were the strongest predictors of survival.
