    Study on the Role of Folic Acid and Compound Vitamin B in Reducing Stroke Recurrence

    Abstract Objective: To study the effect of folic acid and compound vitamin B on plasma homocysteine level and stroke. Methods: Enzymatic cycling assay was used to measure the plasma homocysteine levels in 134 patients with stroke. By screening, patients were randomly sub-divided into folic acid and compound vitamin B intervention group (n=67) and conventional treatment (n=67) group to observe the effect of intervention. Results: The plasma homocysteine level of intervention group reduced significantly (P=0.029). Recurrence rate of intervention group was significantly lower than that of the conventional treatment group (P=0.02). Conclusion: Increasing level of plasma homocysteine is a significant risk factor of stroke. Adding folic acid and compoun vitamin B can reduce the level of plasma homocysteine and play a key role in prevention and treatment of stroke.

    The killing effects of immune cells induced by Leukemia Stem Cell-derived Dendritic Cells on Leukemia Stem Cells from the same source

    Abstract Objective: To investigate the killing effects of stem cell-derived Dendritic Cells (DC) co-cultured with cytokine induced killer (CIK) on Leukemia Stem Cells(LSC)from the same source. Methods: Bone marrow mononuclear cells were isolated from CML-AP donors, CD34+CD38-CD44+ cells were sorted by flow cytometry, cultured and induced to DC. The mononuclear cells from the same doner were induced to CIK. DC and CIK were co-cultured to determined killing effects. The expressions of DC's and CIK's markers typical、P-gp resistance protein were detected by flow cytometry. The expressions of bcr/abl fusion gene of leukemia stem cells and DC were detected by fluorescence in situ hybridization (FISH). The cytotoxicity activity was measured by lactate dehydrogenase release assay. Results: DC and CIK showed typical immunophenotype. DC expressed P-gp resistance protein, and DC's bcr/abl fusion gene was positive. The killing effect increased with effector-target ratio. It was showed that under an effector-target ratio of 40:1 the killing effect was highest. At this level, killing rate can reach to (59.13±12.82)%. Conclusion: Leukemia stem cell-derived DC's functions were normal, and killing effects can be found when they were co-cultured with CIK.

    Study on the Relationship Between Serum E2 Levels, GnRH stimulation test and Morphologic changes of Ovary and Uterus in Girls with Precocious puberty

    Objective: To explore the value of ultrasound in diagnosing idiopathic central precocious puberty through study on the relationship between serum E2, FSH, LH levels and morphologic changes of uterus and ovary. Methods: Thirty-six girls with precocious puberty accepted GnRH stimulation tests and pelvic ultrasound examinations .The levels of E2, TSH, LH before injection and after 30, 60 minutes , and ovarian volume , uterine volume ,maximum follicular diameter were reviewed and analysed. Results: (1) Among the three types of ovaries (type1, homogeneous type ; type 2 , small follicular type ; type 3, large follicular type ), differences of the serum LH ,E2 levels and the ratio of LH∕FSH after GnRH 30 minutes and 60 minutes were significant (p<0.05).(2) The maximal follicular diameter was correlated to the serum LH ,E2,the ratio of LH∕FSH after GnRH 30 minutes and 60 minutes and the volume of ovary. (3) The volume of uterus was correlated to the serum LH,E2 levels and the ratio of LH∕FSH after GnRH 30 minutes and 60 minutes. (4) The volume of ovary was correlated to the serum LH levels and the ratio of LH∕FSH after GnRH 30 minutes and 60 minutes. Conclusion: (1)There was close relationship between LH levels and follicular development .It also proved that serum E2 levels was mainly decided by the development status of the follicles.(2)The volume of uterus was significantly correlated to E2 levels. Development of uterus was estrogen-dependent. (3)Judgement based on GnRH stimulation test and morphologic changes of uterus and ovary would be crucial for establishing the diagnosis of idiopathic central precocious puberty.

    The clinical significance of bone age index in the diagnosis of children sexual precocity

    Objective: To detect the correlation between bone age index (BAI) and index of GnRH stimulation tests for precocious children. Methods: 38 girls who had been diagnosed sexual precocity did GnRH stimulation tests and Bone age be determined by left hand and wrist X-ray, CHN method was used to determine the bone age, BAI equal to the bone age dividing the actual age, according to the clinical symptom and the results of laboratory, the 38 girls were divided into idiopathic precocious puberty (ICPP) group and peripheral precocious puberty (PPP) group. The correlation of BAI and index of GnRH stimulation tests were analyzed between the two groups. Results: the BAI and the LH peak 、FSH peak、LH/ FSH peak ratio had correlation in the idiopathic precocious puberty group ,but in the peripheral precocious puberty group the correlation was not existed. Conclusion: the higher BAI, the more possible to be central precocious puberty, the more necessary to do GnRH stimulation test.

    Medians for second-trimester Down syndrome screening in Tianjin

    Abstract Objective: To establish the medians of maternal serum alpha-fetoprotein (AFP) and free human chorionic gonadotropin (Free β-hCG) for second trimester Down syndrome screening in Tianjin district, in order to discuss its clinical medical value. Method: We measured serum level of AFP and Free β-hCG from 100,543 pregnancies ranged from 14 weeks to 20+6 weeks using time-resolved fluoroimmunoassay. The Medians Tool of the PE risk assessment software(LC3.0) was used to determine 61,881 pregnancies’ serum AFP and free β-hCG medians value, modified by median coefficients,and established the Weight equation. 38,662 pregnancies’ serum AFP and free β-hCG levels were adjusted by median shift to establish median values system for second-trimester Down syndrome screening .Other 13,001 pregnancies’ serum AFP and free β-hCG levels were measured. The screening results were calculated using the standard algorithm based on the Caucasian median value system and the Tianjin median value system. Patients with an increased risk of Down syndrome pregnancy (≥270) were offered genetic amniocentesis. Result: Medians from 13,001 pregnancies matched the established median formula. The Median factor of AFP and free β-hCG were 1.01 and 1.00respectively. The detection rate (DR)of the Tianjin median value system for second-trimester Down syndrome screening was 77.78%, false positive rate(FDR) was 5.33%, positive predictive value was 10.00‰,and the DR based on the Caucasian median value system was 77.78%, the FDR was 8.30%, positive predictive value was 6.49‰. Conclusion: Ethnic variance exists between the Caucasian and Tianjin populations. The accuracy of screening is increased by the use of race-specific medians.

    A study on Clinicopathologic Significance and Mechanism of Peritoneal Metastasis in Gastric Cancer

    Abstract Objective: To investigate the clinicopathologic significance of peritoneal metastasis (PM) in gastric cancer. We tend to illuminate the mechanism of peritoneal metastasis by performing the immunohistochemical staining of HIF-1a、FAK and MMP-9. Method: 164 gastric cancer samples with detailed follow-up data were collected. Immunohistochemical staining of HIF-1a、FAK and MMP-9 were performed for all samples. Result: PM was observed in 41 of the 164 GAC patients, especially in diffuse-type gastric cancer (DGC) (P=0.019). The Kaplan-Meier survival analysis showed that the survival duration of the PM-positive group was significantly shorter than that of PM-negative group (P=0.000). Cox proportional hazards model indicated that the PM and TNM stage were independent predictors of poor prognosis for GAC. The immunohistochemical expression of HIF-1a、FAK and MMP-9 were all higher in the PM-positive group than in the PM-negative group (P<0.05 for all). Conclusion: PM is an unfavorable prognostic indictor for gastric cancer which was associated with infiltrating depth and TNM stage. DGC tend to have peritoneal metastasis easily. Hypoxia might play a role in the formation of PM in gastric cancer.

    Analysis and significance of cell cycle phase in human glioma

    Objective: To explore the expression and significance of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 in human glioma, and study the correlation between parameters and clinicopathological features. Methods: Expression of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 were analyzed by S-P immunohistochemical analysis in 59 cases of gliomas(gradeⅠ ten cases, grade Ⅱ fifteen cases, grade Ⅲ fifteen cases, grade Ⅳ nineteen cases) and 37 normal brain tissues. Results: Expression of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 were not found in normal brain tissues. The positive expression rate of Cyclin D1, Cyclin A, Cyclin B1 and PHH3 in gliomas were 69.5%、69.5%、62.7% and 84.7% respectively. With the increasing of pathological grades, the mean labeling indexes for Cyclin D1、Cyclin A、Cyclin B1 and PHH3 in human gliomas increased accordingly. The difference of Cyclin D1、Cyclin A、Cyclin B1、PHH3 labeling index between four groups were significant(P<0.01). Conclusion: Cell cycle phase can be analyzed by immunohistochemical stain, and it is beneficial for tumor grading and helpful to predict the prognosis.

    The clinical reseatch of Glycated Hemoglobin(HbA1c) for the diagnosis of Diabetes Mellitus and Pre-diabetes.

    Objective To assess the validity of using fasting plasma glucose(FPG)and HbA1c for the screening of diabetes and pre-diabetes。Methods 457 subjects,( 297men,160 women,average age 61.4士13.1years) in our hospital underwent an oral glucose tolerance test(OGTT) for screening diabetes.HbA1c was examined at the same time.Based on 1999 WHO criteria,81 had normal glucose tolerance (NGT),7 had impaired fasting glucose(IFG),76 had impaired glucose tolerance (IGT),13 had IGT and IFG,280 had diabetes.Using a receiver oerating characteristic curve(ROC curve)to judge the optimal cut-point.Result 1.the optimal cut-point of HbA1c for DM was 6.45% that was associated with a sensitivity and specificity of 0.794 and 0.825 respectively.Area under the curve was 0.871(95%CI0.839~0.903).2. The cut-point of HbAlc for pre-diabetes was 5.85% ,which was associated with a sensitivity and specificity of 0.583 and 0.531 respectively.Area under the curve was 0.587(95% C1 0.504~0.671).3 When only used HbA1c to screening DM, the sensitivity and specificity are 0.794 and 0.825 respectively, the rate of misdiagnosis is 21.3%;When only used FPG to screening DM, the sensitivity is 0.646,the rate of misdiagnos is 35.3%;But when using FPG or HbA1c to screening DM,the sensitivitiy and specificity are 0.871 and 0.623 respectively,the rate of misdiagnosis is 12.9%。 Conclusions 1.Suggest the optimal cut-point of HbA1c for diabetes was 6.45% ,because it have the high sensitivity and specificity.2.The cut-point of HbA1c for pre-diabetes is 5.85%.But the sensitivity and specificity are lower,so it have little diagnose meaning.3.The detection of FPG and HbA1c may reduce erroneous misdiagnosis rate.

    Correlation between serum leptin and urine micro-albumin in patients with simple obesity

    Abstract Objective:To study the correlation between serum leptin (LEP) level and urine micro-albumin in patients with simple obesity,discuss the effects of leptin on kidneys . Methods:60 patients with simple obesity and 40 controls in this study. Height,body weight,waist circumference,hip circumference,systolic blood pressure, diastolic blood pressure,blood glucose, total cholesterol, triglyceride,blood urea nitrogen,serum creatinine, uric acid ,urine micro-albumin, urine creatinine as well as serum leptin were observed in each group and compared. Meanwhile, body mass index(BMI) , waist-hip ratio(WHR) ,mean arterial pressure(MAP) and the ratio of urine micro-albumin and urine creatinine(UACR) were counted. Results: In compare with control group,serum LEP and UACR in obese group were significantly increased(P<0.01). Serum LEP in obese group was in a positive correlation with BMI (r =0.650,P<0.001),WHR (r =0.617,P<0.001) and blood glucose (r =0.345,P<0.05),but had not significant correlation with UACR(r = -0.146,P>0.05). Conclusion: The level of serum LEP and UACR are related to obesity.But there is not significant correlation between serum LEP and UACR.

    Clinical analysis of the platelet parameters in 2109 cases of acute cerebral infarction

    Abstract Objective: To study the characteristics and clinical significance of platelet count (PLT)、mean platelet volume(MPV) and platelet distribution width (PDW) in patients with acute cerebral infarction.Methods: The parameters of blood platelet (PLT、 MPV、 PDW) were detected in 2109 patients with acute cerebral infarction and compared with the control group(480 healthy persons ).To analysis the characteristics of platelet parameters exposed in different risk factor for stroke, and then to compare the characteristics of platelet parameters in first cerebral infarctions and recurrent cerebral infarctions . Results: MPV in acute cerebral infarction was obviously higher than ones in control group(P<0.05), while PLT and PDW, there were no significant differences in all results(P>0.05). The difference of the groups that Pure cerebral infarction group, high blood pressure with cerebral infarction group, diabetes combined with cerebral infarction group, dyslipidemia with cerebral infarction group, has statistical significance (P < 0.05), PDW difference has no statistically significant(P > 0.05). MPV in the pure cerebral infarction was obvious lower than other groups(P < 0.01); MPV in Af with cerebral infarction group was higher than other groups, the difference was statistically significant (P < 0.05),its PLT is less than high blood pressure group, or the dyslipidemia group, statistical significance was found (P < 0.05), PDW difference were not significant with the other groups (P > 0.05); The differences of MPV, PLT, PDW in High blood pressure with cerebral infarction group, diabetes combined with cerebral infarction group, dyslipidemia cerebral infarction group, have no statistically significant (P > 0.05). The difference of PLT, PDW, MPV between recurrence of cerebral infarction and cerebral infarction group for the first time, have no statistical difference (P > 0.05) Conclusions: The increased MPV may be a independent risk factors of cerebral infarction, so it is important to detecte MPV for the prevention of acute cerebral infarction, especially for patiens with high blood pressure, diabetes, atrial fibrillation, and dyslipidemia , the early antiplatelet therapy should be advised.

    LVH and eGFR five-year follow-up in older hypertensive patients

    Objective To study change of LVH and eGFR in the process of therapy in the elderly patient with hypertension. Methods object of study were the elderly patients with hypertension hospitalized and the period of follow-up was five years or more than five years with comprehensive information including blood pressure, BMI, blood sugar, blood fat, blood creatinine, uric acid, urinary albumin/creatinine ratio, glomerular filtration rate, electrocardiogram and echocardiography . all patients would be divided into CKD group and NCKD group according to ACR and eGFR index. Candesartan was chosen mainly antihypertensive treatment measures to achieve the goal blood pressure during their hospitalization and the scheme was sustained during outpatient service.they were followed-up regularly.indexes related were compared at the same time points between groups, the same index were compared according to different followed-up time in the same group. Results (1) Compared between two groups, there were not statistically difference of corresponding systolic blood pressure and diastolic blood pressure, while there were statistically significance concerning difference of LVMI, LVH and eGFR at the same time point,(2) in comparison with the baseline state , except eGFR in NCKD group ,each index at various time point were improved with statistically significance ,including the systolic blood pressure, diastolic blood pressure, LVMI, LVH and eGFR, but the change with statistically significance did not appear among these indexes in the course of subsequent followed-up 4 years, there were not statistically difference concerning eGFR change in NCKD group in followed-up period of 5 years. Conclusion (1) The incidence of CKD and LVH were higher in the elderly hypertensive patients. (2) with blood pressure successful control, the LVH of patients was improved with glomerular filtration rate taken a turn for the better in CKD patients. The eGFR of patients with hypertension was a benign transformation process.

    The Detection of 16SrRNA Methylase Genes in Acinetobacter baumannii Isolates in Tianjin District and the Related Drug Resistance

    Abstract Objective To investigate the prevalence of 16SrRNA methylase genes and the rate of drug resistance in Acinetobacter baumannii isolates in Tianjin District.Methods A total of 152 strains of Acinetobacter baumannii were collected from clinical specimens in Tianjin District from August 1st,2010 to December 31st,2010.The primary antimicrobial susceptibility testing was performed by either Broth dilution or Kirby-Bauer Disk diffusion;Seven 16SrRNA methylase genes (armA、rmtA、rmtB、rmtC、rmtD、rmtE、npmA) were amplified by polymerase chain reaction(PCR) and sequenced.Results Polymyxin resistance rate (1.97%) among 152 strains is the lowest,followed are Cefoperazone/sulbactam(15.79%) and Levofloxacin(36.84%). The resistance rates to Aminoglycoside、Cephalosporin、 Carbapenem are above 55%.73 of the 83 resistance to Aminoglycoside strains were armA genes positive(87.95%).account for 48,03% of all the experimental strains. No rmtA、rmtB、rmtC、rmtD、rmtE、npmA were found.Except the Polymyxin,the rates of resistance to the rest 13 antibiaotics were significantly higher in armA gene positive strains than in armA gene negative strains.(P<0.05).Multidrug-resistance Acinetobacter baumannii (MDRAb) and Pandrug-resistance Acinetobacter baumannii(PDRAb) were also higher in armA gene positive strains (100% and 69.86% respectively) than in armA gene negative strains(21.52% and 11.39% respectively) .Conclusion The rate of 16SrRNA methylase genes (armA) were firstly reported in Tianjin district.armA gene is widely existed in Acinetobacter baumannii strains. No other six genes were found in Acinetobacter baumannii.The rate of drug resistance in armA positive strains were higher than in armA negative ones.The number of MDRAb and PDRAb are significantly larger in armA gene positive strains than in armA negative ones.Also armA positive strains are all MDRAb.Further research was needed to confirm wether the armA gene positive strains can be treated as MDRAb in clinical therapy by collecting more strains.

    Olfactory function assessed with magnetic resonance imaging and olfactory event–related potential

    Abstract Objective: Using olfactory event related potential (OERP) and magnetic resonance imaging (MRI) to evaluate olfactory function. Methods: Eleven healthy individuals with normal olfactory function (NL) and 11 patients with nasal polyposis (NP), 11 with posttraumatic olfactory loss (PT), and 11 with postinfectious olfactory loss(PI) were included in this study.Physical examination,nasal endoscopy, T&T olfactory testing, olfactory event related potentials and magnetic resonance imaging were performed in all patients. Results: T&T olfactory testing revealed that NL individuals had higher scores than patients with NP,PT, or PI. OERP testing were recorded in the 11 NL individuals and in 3 NP, 3 PT, and 4 PI patients. The OB volumes were higher in NL individuals compared with NP, PT, and PI patients. The OB volumes in PT patients were significantly lower than those from NP and PI patients. Conclusion: This finding suggests that these diagnostic tools provide information in terms of the clinical assessment of olfactory function.

    Construction of recombinant eukaryotic plasmids of HD5 and LL37 and their transfection and expression in human vaginal epithelial cells

    Objective:The eukaryotic expression vectors of human defensin 5 (HD5) and LL37 were built and transfected primary human vaginal epithelial cells. To observe the expression of HD5 and LL37 after transfection. Method: ① Total RNA was extracted from human vaginal epithelial cells using 1 ml of Trizol Reagent (Invitrogen Life Technologies, USA). The cDNA encoding the HD5 and LL37 were amplified through RT-PCR. Then inserted into pcDNA3.1(+)-EGFP , a expression vector. ② The vaginal epithelial cells from female vagina were culture primarily by Serum-Free Keratinocyte Medium and the tissue pieces culture method. These two kinds of recombinant eukaryotic plasmids were separately transfected or co-transferred into vaginal epithelial cells. At 6, 12, 24 and 48h the transfection efficacy was observed by fluorescence microscope,and the supernatant of every group was collected for determine the expression of HD5 and LL37 by ELSIA method. Result: This study constructed pcDNA3.1 (+) / HD5-EGFP and pcDNA3.1 (+) / LL37-EGFP eukaryotic expression vector, to achieve the expression of HD5 and LL37 in the vaginal epithelial cells. The highest expression of HD5 and LL37 protein in the cell culture supernatant was at transfected 24h by using ELISA method. Conclusion: We successfully constructed with HD5 and LL37 cDNA eukaryotic expression plasmid, and successfully transfected into human vaginal epithelial cells. On this basis, we can further study the antibacterial function of recombinant HD5 and LL37and vaginal epithelial cell innate immune system.

    Study on the Doppler spectrum waveform and value of arterac arcuatea uterus to women of childbearing age

    Abstract Objective:To establish the PW (pulsed wave Doppler) normal waveform and reference range by detecting of abdominal pulsed wave Doppler on uterine arcuate artery of normal women of childbearing age. Methods: ①PW waveform was observed: According to having flow imaging during diastole , PW will be divided into bimodal peak、single-peak shape.; ②PW blood flow index was measured and the PW waveform and reference range of uterine arcuate artery were obtained by statistical analysis. Results: (1) 95.2% of the PW waveform of uterine arcuate artery with flow imaging during diastole showed bimodal shape. 4.8% without flow imaging during diastole showed a single-peak shape.; (2) PW reference range of uterine arcuate artery: Vs:3.5~44.0cm/s; Vd:0~11.8cm/s; PI:0.7~2.2; RI: 0.6~1. Conclusion: waveform analysis and RI were reliable and convenient, which could be used as important reference.

    Effect of Edaravone on expression of caspase3 and pdcd5 protein in intracerebral hemorrhage rat

    Objective To investigate the effect of edaravone on the expression of caspase3 and pdcd5 protein following intracerebral hemorrhage (ICH) in rats. Methods 108 healthy adult Wistar rats were randomized into normal group, sham group ,ICH group,edaravon treat group .The later two groups were randomized into 8 subgroups respectively: 3h, 6h, 12h, 1d,2d,3d,5d,7d groups after ICH,each group 6 rats.ICH models were prepared by injecting 50μl of autogenous blood into the the right caudate nucleus of rats.The expression of caspase3 and pdcd5 protein was observed by immunohistochemistry respectively. Results Compared with hemorrhage group, the expression of caspase3 and pdcd5 protein of Edaravone treat group weakened after 6h、12h、1d、2d、3d、5dand 7d after ICH (p<0.05,p<0.01)). Conclusion The expression of caspase3 and pdcd5 protein following intracerebral hemorrhage were decreased by edaravone .The injury of intracerebral hemorrhage were protected by edaravone.

    Objective: To observe the inhibitory effect of Endostar combined with radiotherapy on transplanted CNE1 Nasopharyngeal Carcinoma(NPC) in BALB/c nude mice and explore the relationship of this inhibitory effect with the expression level of VEGF and apoptosis inhibiting protein-Survivin..Methods: The transplanted tumor model of human CNE1 nasopharyngeal carcinoma on nude mice was established to evaluate the antitumor activity of Endostar combined with radiation (Endostar+radiation,D group) treatment compared with monotherapy (Endostar,B group or radiation,C group).The growth characteristics of transplanted tumor were recorded. Transmission electron microscopy were used to observe the morphological changes of cell apoptosis. Real-time quantitative PCR was used to determine the effect on VEGF and Survivin mRNA expression level of of tumor tissues.Results: Significant on the tumor growth was found ,and more typical morphological changes were seen in cancer cells in D group. Level of VEGF in tumor tissue of D group was lower than that of C group ,B group or A group(P<0.05). And the level of Survivin in D group were much lower than in other three groups(P<0.05). The results of linear Correlation relevant analysis showed that there exist positive correlation between the inhibitory effect and the expression of VEGF and Survivin (r = 0.679, P < 0.01).Conclusion: Endostar could enhance the radiosensitivity of NPC transplantation tumor in nude mice to radiation.The mechanism may be down-regulating the expression of VEGF and Survivin.

    The Effect of Taurine on Train Metabolism in Rats Following with Severe Traumatic Brain Injury

    Abstract Object The purpose of this research is to observe the effect of taurine on energy metabolism in rats following with traumatic brain injury. Methods 24 rats were randomly divided into sham group, traumatic brain injury group (TBI) and taurine treating group,8 rats in each group. The groups of TBI and taurine were subjected to severe traumatic injury of left brain by lateral fluid pressure device. Then taurine group were injected traurin(200mg/kg)and the other groups were given saline through caudal vein immediately. Intercellular fluid in each group(each group contain 3 time points,1-3h, 22-24h, and 7d) in left brain tissue were collected by using the method of microdialysis, glucose, lactate, glycerol, pyruvate in the samples were measured using CMA600 auto biochemistry analyzer, and calculate the ratio of lactate and pyruvate. Results (1) The level of glucose in left brain tissue had no statistic differences in the three groups after traumatic brain injury; But the level of glucose were increasing significantly in TBT group with time goes.(2)The level of lactate was all higher in TBI group at the three time than those in sham group,and the differences had statistic significance, whereas ,the level of lactate decreased in TBI group with time goes. Taurine could markedly decrease the content of lactate in 24h, which had no significant difference to sham group.(3) After traumatic brain injury, the level of glycerol in damaged hippocampus had no statistic differences to sham group, but it decreased markedly in taurine group.(4)The level of L/P were all higher in TBI group at the 3 time point than those in sham group, and decreased with time goes. The level of L/P in taurine group were all lower than that in TBI group at 3 time point, but it was still markedly higher than that in sham group. Conclution It can appear metabolism function disorder and taurine may make an active effect on adjusting metabolism disorder after traumatic brain injury.

    Preparation and Propreties of PGA/β- Tricalcium Phosphate of Bone Tissue Engineering Scaffolds

    Abstract Objective:To fabricate porous PGA/β- Tricalcium Phosphate of tissue engineering scaffolds and test its properties. Methods: Used PGA andβ- Tricalcium Phosphate as materials, 1,1,1,3,3,3-Hexafluoro-2-propanol as solvent, sodium chloride as porogen(particle size :425–500μm), the PGA/β-TCP(in 1:1and 1:3 weight ratios ) scaffolds for tissue engineering are fabricated by solvent-casting particulate leaching and methods. The structure of the scffolds were observed using scanning electron microscopy (SEM). Their properties of physical and mechanics are tested in this paper. Result: The compressive strength of the 0.13gPGA/0.39gβ-TCP(B)group were obviously higher than that the compressive strength of the 0.13gPGA/0.13gβ-TCP(A)group P<0.001. The compressive strength of the A and B group after surgery were: 731.283±55.2140KPa, 173.833±36.1397KPa. Both A and B groups have a high porosity.(>85%). The opening porosities of PGA/β-TCP (1:1) scaffolds and PGA/β-TCP (1:3) scaffolds were 93.1±0.7% and 89.3±0.6%, respectively, which is a statistically significant difference . The SEM pictures showed that both scaffolds had irregular micro-pores .The average pore size of two groups were 300-400μm,the β-TCP particles were uniformly dispersed within the PGA matrix.The different between the two groups was that moreβ-TCP granules were present on the pore wall surface of group B compared to group A. Conclusion: The weight ratios (1:3) scaffolds materials can basically meet the requirements of materials for bone tissue engineering.

    The Establishment of Indivudal Human Lung Cancer Model by Xenotransplant Tumor Mass in Nude Mouse

    Abstract Objective:To establish clinically relevant individuation nude mouse models of human non-small cell lung cancer,which can be stably passaged,so as to lay a foundation for the future individualized therapy study. Methods: Tumor implanting group: BALB/C nude mouse subcutaneously injected in the back with diameter 2mm tumor derived from human operative specimens were killed when the tumor mass grown to 800mm3.This is the first generation.Then,to establish the second generation, BALB/C nude mouse were injected in the back with diameter 2mm tumor derived from first generation tumor mass.the same method redone to gain the third generation tumor. Cell lines implanting group: BALB/C nude mouse subcutaneously injected in the back with 1×106 human lung cancer cell lines A549 or H1795 were killed when the tumor mass grown to 500mm3.The tissue of primary human lung cancer 、first 、second 、third generation implanting tumor and the cell lines implanting group tumor were formalin-fixed、cut sections、HE stained,and then compared with the morphology by light microscope.Results: The tumor cells of the tumor implanting group not only retains the atypia of cancer cells, but also keep original arrangement structure; The cell lines implanting group tumor cells lose their arrangement structure, keeping only the atypia of the cancer cells. Conclusion: Using different surgical excision of lung cancer established human lung cancer tumor nude mice-model which have individual characteristics,this model can be useful for the future individualized therapy study in vivo.

    Linezolid versus vancomycin in the treatment of children with MRSA infections:Meta-analysis of randomised controlled trials

    Abstract Objective: A meta-analysis of randomized controlled trials on the efficacy and safety in children with MRSA infections treated with linezolid versus with vancomycin. Methods: The data were collected from the Medline database, and Chinese CNKI Database and the references of eligible studies were manually screened. Randomized controlled trials published in the English and Chinese literature comparing linezolid with vancomycin in children with MRSA infections were eligible for inclusion. Heterogeneity was examined by Chi-square test. Fixed effects model or random effects model were used to pool the data. Sensitivity analyses were used in the treatment course. Results: Three randomized controlled studies comparing linezolid with vancomycin were analyzed, focusing on the 588 children with MRSA infections. It was found by meta-analysis that, with respect to clinical treatment success, linezolid was as effective as vancomycin [89.72% vs. 85.71%, RR=1.05, 95% CI (0.98, 1.12), Z=1.49,P =0.14]. In the analysis with safety set, Significantly fewer linezolid-treated patients experienced drug-related adverse events than did vancomycin-treated patients [(0.17%vs. 0.40%,RR=0.49,95% CI (0.37,0. 65),Z=4.93,P <0.00001].Conclusion:Linezolid is as effective as vancomycin and more safety for treatment of patients with children infections caused by MRSA, although rigorously designed large sample size randomized double blind clinical trials are required to further demonstrate and support the conclusion.

    The Diagnostic Power of Diffrent Stage of Aldosterone to Rein Ratio in Diagnostic Primary Aldosteronism

    Abstract :Objective To evaluate the accuracy of aldosterone to rein ratio(ARR) in diagnosing primary aldosteronism(PA). Methods The aldosterone to rein ratio was determinned in 279 patients with primary aldosteronism and 70 patients with primary hypertension(PH),the patients were divided into 11 groups according to the range of 10. The numbers of PA and PH with diffent stage of ARR were determined,and the sensitivity,specificity,Youden index of ARR in diagnosing PA were caculated.The ROC curve was drawed. Results The value of ARR in 30~、40~、50~, the sensitivity,specificity,Youden index of ARR in diagnosing PA were 83.2%、82.9%、0.661,77.8%、88.6%、0.664 and 74.2%、92.9%、0.671,respectively. Conclusion The sensitivity,specificity,Youden index of ARR in diagnosing PA are pretty high when ARR in 30~60.

    The Expression of Bax、Livin and Survivin in Non-Small Cell Lung Cancer

    Objective: To investigate the expression of Bax, Livin and Survivin and relationship between their expression and clinicopathological factors in non-small cell lung cancer, in order to provide further theoretical basis for clinical diagnosis and treatment of lungcancer. Methods:The expression of Bax, Livin and Survivin protein was detected by immunohistochemistry two-step techniques (PV). Results:The rates of positive expression of Bax、Livin and Survivin were respectively 42.50%、80.00%,65.00%、10.00% and 50.00%、0% in the non-small cell lung cancer tissues and in the normal lung tissues. The rates of positive expression of Bax and Livin were 23.81%、63.16% and 85.71%、42.11% in patients with lymph node metastasis and in patients with no lymph node metastasis.The rates of positive expression of Bax and Livin were 55.56%、15.38% and 51.85%、92.31% inⅠ, Ⅱstage patients and Ⅲ, Ⅳstage patients.The positive expression rate of Survivin in patients with well- middle-differentiated (39.29%) was significantly lower than that in patients with poorly differentiated(75.00%).The differences were all statistically significant (P <0.05).There was negative correlation between the expression of Bax and Livin in the non-small cell lung cancer(r=-0.4294,P<0.05).There was no correlation between the expression of Bax and Survivin,and with no correlation between the expression of Livin and Survivin in the non-small cell lung cancer(P> 0.05). Conclusion: The increase of the expression of Bax in non-small lung cancer inhibited the invasiveness of tumors cell and tumors metastasi -s.Livin could be correlated with carcinogenesis of non-small cell lung cancer. The high-expression of Livin may enhanced the invasiveness of tumors cell and promoted tumors metastasis.Survivin could be correlated with tumors histologic -al differentiation degree and carcinogenesis of non-small cell lung cancer.There was negative correlation between the expression of Bax and Livin in the non-small cell lung cancer.It indicated that there could be the same pathway in the proceeding of non-small cell lung cancer. There was no correlation between the expression of Bax and Survivin, Livin and Survivin in the non-small cell lung cancer.It suggested that there could be a different pathway in the proceeding of non-small cell lung cancer.

    Effect of probucol on myocardial injury, inflammation and endothelial function in patients with unstable angina pectoris underwent percutaneous coronary intervention

    Abstract Objective:To observe the effect of probucol on myocardial injury, inflammation and endothelial function in patients with unstable angina pectoris (UAP) underwent percutaneous coronary intervention (PCI), and investigate the probable mechanism of probucol. Methods:107 patients with UAP randomly divided into probucol group (52 cases) and control group (55 cases). Standard medical therapies were given in all 107 patients. The CK-MB, cardiac troponin I (cTnI), high-sensitivity C-reactive protein (hs-CRP) and nitric oxide (NO) were measured in all enrolled subjects before PCI right time and at 6:00 in the next day morning after PCI. Results:The levels of hs-CRP, CK-MB and cTnI were elevated and the level of NO was decreaed significantly after PCI in control group (P<0.05). The levels of CK-MB and cTnI were elevated significantly (P<0.05), but there were no significant changes in the increased level of hs-CRP (P>0.05) and the decreased level of NO (P>0.05) after PCI in the probucol group. However, the ascended value of hs-CRP and the descended value of NO in the probucol group were significantly lower than those in control group (P<0.05), the ascended values of CK-MB and cTnI in the probucol group were higher than those in control group (P>0.05). Conclusions:Probucol therapy in more than 24 hours before PCI can significantly reduce the inflammatory reaction and improve endothelial function and prognosis in patients with UAP, but there is no evident function on reducing myocardial injury.

    The changes of Inflammatory Factors Due to ECMO Assisted Beating Heart CABG

    Objective: To investigate the impact of extracorporeal membrane oxygenation(ECMO) assisted beating heart coronary artery bypass graft(CABG) on inflammatory factors, and compare it with conventional coronary artery bypass graft (CCABG) and off-pump coronary artery bypass graft(OPCABG). Method: 45 patients with coronary artery disease undergo CABG were divided into three groups: CCABG group, ECMO assisted CABG group and OPCABG group, for each group 15 patients. The blood samples were collected from peripheral vein at the moment preanesthesia(T1) , 10 min after pump or ECMO starting 10 min(T2) , pump or ECMO stopped or surgery finished moment(T3) , 4th hour after surgery(T4) and 24th hour after surgery(T5) . Centrifuged blood samples for plasma. Observe the change patterns of inflammatory cytokines tumor necrosis factor-α(TNF-α), interleukin-6(IL-6) and interleukin-8 (IL-8) measured by enzyme-linked immune sorbent assay(ELISA). Result: The patient ages were significantly higher in ECMO assisted group than the other two groups(P<0.05). There was significant plasma level increase of inflammatory factors since pump or ECMO starting compared with preanesthesia (P < 0.05), and they had reached peak before 4th hour after surgery, kept a high level even at 24th hour after surgery. At the moment after surgery, the plasma level of TNF-α,IL-6 and IL-8 was obviously lower in both ECMO assisted group and CCABG group(P<0.05), but after 4h there was no significant difference among three groups(P>0.05). Conclusion: Though there is no significant superiority compared ECMO assisted beating heart coronary artery bypass graft with other surgery methods, the patient ages are obviously higher in ECMO assisted group, and it may be a new surgery pathway for ageing patients.
