    An Experimental Study on Bovine Nucleus Pulposus Cells Labelled with PKH26 in Vitro

    【Abstract】 Objiective To investigate the application of PKH26 fluorescent labeling on nucleus pulposus cells isolated from bovine Coccyx disc. Methods Nucleus pulposus primary cells were isolated from the nucleus pulposus tissue detached from bovine Coccyx disc by enzymatic digestion, and observed under the inverted microscope. Safranin O, toluidine blue and type II collagen immunocytochemistry were stained for passage one generation cells. Nucleus pulposus cells were labeled with PKH26 fluorescence in accordance with the instructions, and labeled cells’ activity, fluorescence intensity at days 0, 14, 28 of culture and characteristics of proliferation were assessed. Results Isolated nucleus pulposus cells amounted to (1.56 ± 0.35) × 106/g. Under the inverted microscope, primary cells adhered at 4th day of culture, grew in groups, and cells covered the bottom of culture flask at 13th day, both primary cells and the P1 generation cells were chondrocyte-like morphology. The staining of safranin O, toluidine blue and type II collagen immunocytochemistry for P1 generation of nucleus pulposus cells showed positive result. Cell activity before and after PKH26 label showed more than 95%, the fluorescence intensity at 0d, 14d, 28d performed a decreasing trend, but 28d can still detect strong fluorescence, there were no significant differences in growth curve between before and after labeled cells (P > 0.05). Conclusion As the seed cells of tissue engineering, nucleus pulposus cells isolated from bovine Coccyx can reach a satisfactory number and maintain cartilage-like phenotype, and no changes shown in the biological characteristics after labeling. PKH26 labeled nucleus pulposus cells are suitable for the traceable cells in vivo study.

    Inhibitory Effects of Oridonin Combined with Gemcitabine on Pancreatic Cancer SW1990Cells

    Object: To investigate whether oridonin can enhance the antitumor effect of gemcitabine on pancreatic cancer SW1990 cells in vitro and explore the potential mechanism of its action. Methods: The pancreatic cancer SW1990 cells were treated with vehicle alone and various concentrations (10、20、40、80、160 μmol/L) of oridonin, followed by 24、48 and 72 h cell culture, Effects of oridonin on cell proliferation were determined by using a CCK-8 Kit; SW1990 cells were treated with oridonin (40 μmol/L) and gemcitabine(20 μmol/L) alone or together for 48 h, with the untreated cells used as the contro1; the survival rate of cells was detected by CCK-8 assay. Apoptosis induction was assessed by using Annexin V-FITC kit; Semi-quantitative RT-PCR was used to examine the changes of NF-κB mRNA and XIAP mRNA expressions. Results: Oridonin inhibited the growth of pancreatic cancer SW1990 cells in a dose- and time-dependent manner. Compared with the other groups, the cell survival rate of the combination group was significantly lowered (P < 0.05). oridonin combined with gemcitabine induced a higher percentage of apoptosis in pancreatic cancer cells than that of oridonin or gemcitabine alone(P <0.05).Moreover, the expressions of NF-κB and XIAP mRNA in pancreatic carcinoma cells were obviously down-regulated in combination group(P <0.05). Conclusion: Oridonin can potentiate the antitumor effect of gemcitabine on pancreatic cancer in vitro, which may be realized through the down-regulation of XIAP and induction of cell apoptosis.

    Assessing Radiosensitivity of Lung Cancer with the Expression of γH2AX

    [Abstract] Objective To observe the relationship between expression changes ofγH2AX and the radiosensitivity of lung cancer cells in vitro.Methods The radiosensitivity of lung cancer cell lines A549and SBC-3was measured by clone forming assay. The DSBs damage of lung cancer cell lines A549and SBC-3was determined by Western blot assay.Re? sults The clone forming rates of lung cancer cell lines A549and SBC-3were gradually decreased with the increased radia? tion dose. γH2AX expression was related to the cell radiosensitivity1hour and6hours after radiated. Conclusion The phosphorylated histoneγH2AX is a powerful tool to monitor DNA DSBs and to predict the radiosensitivity in lung cancer ra? diotherapy.

    The role of CXCL12/CXCR7 on the proliferation,adhesion and invasion of gastric cancer

    [Abstract] Objective To investigate the expression of CXCR7in gastric carcinoma tissues and the role of CXCR7 in tumor growth, adhesion and invasion of gastric cancer. Methods The expression levels of CXCR7, Survivin, CD44v6and matrix metalloproteinase (MMP)-3were detected by immunnohistochemistry method in gastric cancer specimens came from 160patients (gastric cancer group) and30specimens of normal gastric tissues (control group).Results The expression of CXCR7protein was significantly higher in gastric cancer group than that of control group (P<0.05). The positive CXCR7 staining was significantly different with tumor size, depth of invasion, lymph node metastasis and clinical staging (P<0.05). CXCR7expression was positively correlated with Survivin, CD44v6and MMP-3. Conclusion CXCR7might take part in progression of gastric cancer by anti-apoptosis (Survivin). CXCR7might take part in the lymph node metastasis of gastric cancer and mediate the adhesion and invasion by CD44v6and MMP-3.

    The specificity and efficiency of YFP labeled natural killer cell through Vav-Cre induced YFP reporter system in mice

    Objective Explore the specificity and efficiency of YFP labeled natural killer cells through Vav-Cre induced YFP reporter system in mice. Methods We crossed ROSA26R-YFP and Vav-Cre mice and screened their YFP and Cre gene double positive progeny by genotyping. Then we analyzed the specificity of YFP in hematopoietic cells from immune organs including lymph nodes, spleen, thymus and bone marrow through flow cytometry. Next we analyzed the percentage of YFP positive cells in natural killer cells from lymph nodes, spleen and bone marrow. Results We obtained 11 double positive mice among 17 offsprings by crossing ROSA26R-YFP mice with Vav-Cre mice. The percentages of YFP positive cells in the immune organs including lymph nodes, spleen, thymus and bone marrow are 73.87%±1.51, 56.07%±1.47, 86.17%±1.74 and 53.60%±3.56, there are significant differences compared with the corresponding negative control cell(0.27%±0.01, 1.33%±0.91, 0.11%±0.01, 0.29%±0.03),(P<0.01; double positive mice vs CTRL); nevertheless in non-immune organ kidney, cells were almost YFP negative(double positive mice 0.72%±0.43 vs CTRL0.92%±0.27; P>0.05). NK cells from lymph nodes, spleen and bone marrow of these double positive mice were 76.94%±0.84, 81.66%±1.18 and 88.92%±0.77 labeled by YFP (P<0.01; double positive mice vs CTRL). Conclusion YFP marked natural killer cells through Vav-Cre induced YFP reporter system in mice has strong specificity and high efficiency.

    The expression of NT in androgen independent prostate cancer animal model

    [Abstract] Objective To study the different expressions of neurotensin (NT) at gene and protein level in orthotopic model of prostate cancer .Methods The animal models of androgen dependent prostate cancer(ADPC,castrated for3days) and androgen independent prostate cancer(AIPC) were established by planting tumor tissue or undergoing surgical castra? tion. Affymetrix microarray technology was carried out to compare the gene expressions of NT. The result was verified by qRT-PCR. HE staining was used to observe the change of pathology. ELSIA and immunohistochemistry technology were fi? nally performed to detect protein expression of prostate-specific antigen(PSA) and NT in three different groups of prostate cancer tumor tissues.Results The expression of NT was5.10times higher in AIPC group than that in ADPC group. The ex? pression of NT was0.33times lower in castrated 3-day group than that in ADPC group. Results of RT-PCR and qRT-PCR showed that the expression levels of NT gene were1.41and7.27times respectively higher in AIPC group than that in ADPC group,but the expression levels of NT gene were0.78and0.46times respectively lower in castrated 3-day group than that in ADPC group (P<0.05). HE results showed that nuclear atypia and tumor structure in three groups. Immunohistochemistry and ELISA results showed that the values of PSA and NT were (0.48±0.03) and (0.031±0.008) μg/L in ADPC group; (0.17± 0.03) and (0.021±0.004)μg/L in castrated3-day group,and (0.87±0.02) and (0.042±0.010)μg/L in AIPC group. There were significantly lower expressions of NT and PSA in castrated3-day group that those of ADPC group (P<0.01). Conclusion In the transition from ADPC to AIPC, the over-expression of NT suggested that NT may be associated with prostate cancer progression. NT may be used as a new therapeutic target and specific diagnostic method of AIPC

    [Abstract] Objective To investigate the effect of construct theNotch1(NICD) eukaryotic expression vector on the proliferation and differentiation of rat bone marrow mesenchymal stem cells (BMSCs)in vitro. Methods Rat BMSCs were experimented as the object.NICDeukaryotic expression vector was constructed. pEGFP- N1- NICD expressing plasmids were used to transfect BMSCs. The study included control group (CON group), empty vector group (VEC group) and the trans? fection group (TRA group). After 48-hour transfection, BMSCs were observed for general morphology. The protein expres? sions of NSE, GFAP and Notch1were detected by real-time PCR and Western blotting assay respectively. The apoptosis, cy? cle distribution and cell proliferation were evaluated by flow cytometry and MTT assay.Results The DNA sequencing con? firmed that the pEGFP-N1-NICD recombinant plasmid was successfully constructed, and both VEC group and TRA group expressed green fluorescence after48-hour transfection. The relative expression levels of Notch1and GFAP mRNA and pro? tein were significantly higher in TRA group than those in VEC group and CON group (P<0.05), and there was no significant difference between VEC group and CON group. After48-hour transfection, the ratio of living cells was significantly lower in TRA group than that of CON group and VEC group, and the early apoptotic rate and late apoptotic rate were significantly higher in TRA group than those of CON group and VEC group(P<0.05). The late apoptotic rate was significantly higher in  VEC group than that of CON group. The proportion of G1/G0cells was significantly higher in TRA group than that of CON
    group and VEC group, but S and G2/M cells were significantly lower (P <0.05). The value of growth curve was gradually de?

    Effects of Ginsenoside Rg1on nNOS and iNOS Expressions in Rat Brain Tissue after Cerebral Ischemia Reperfusion

    [Abstract] Objective To investigate the effects of Ginsenoside Rg1on the expression of nitric oxide synthase (NOS) in brain tissue after cerebral arterial thrombosis in adult rats. Methods Thirty rats were randomly divided into the sham-operative group, cerebral ischemia-reperfusion group, Ginsenoside Rg1-L group, Ginsenoside Rg1-M group, Ginsen? oside Rg1-H group and nimodipine group (n=5for each group). The ischemia-reperfusion rat model was established by mid? dle cerebral artery occlusion. The neurological score after reperfusion was observed. The levels of nitric oxide (NO), neuronal NOS (nNOS) and inducible NOS (iNOS) were detected by nitrate reduction method and colorimetric method. The expressions of nNOS and iNOS after reperfusion were analyzed by immunohistochemistry and Western blot assay.Results (1) The neu? rological scores after cerebral ischemia were significantly lower in Rg1-L group, Rg1-M group and Rg1-H group than those of cerebral ischemia-reperfusion group(2.40±0.55,1.80±0.84, 1.60±0.89vs3.20±0.84,P<0.05). (2) Compared with those of model group, serum levels of NO and iNOS were reduced, and nNOS levels increased, in three groups of Rg1. (3) Compared with those of model group, the expression of nNOS was significantly increased,and iNOS expression was significantly re? duced, in three groups of Rg1. Conclusion The preventive effects of Ginsenosides Rg1on cerebral ischemia-reperfusion injury may be associated with the activation of nNOS and the inhibition of iNOS.

    Protective Effects of Active Ingredients of Notoginseng on Immunological Liver Injury

    [Abstract] Objective To investigate the protective effects ofactive components of notoginseng on immunological liver injury in mice. Methods Sixty Kunming mice were randomly divided into normal group (group A), model group (Group B), PNS group (Group C), total flavonoids group (Group D),and polysaccharidegroup (group E). C,D and E groups were given PNS,total flavonoids and polysaccharideorally,1/day for 14days. Then BCG was intravenous injected in B, C, D and E groups, and after26days lipopolysaccharide (LPS) was intravenous injected. Samples of eye venous plexus blood were collected, and serum levels of alanine aminotransferase (ALT), total superoxide dismutase (T-SOD) and Interleukin-4(IL-4) were detected. Organs index was calculated by checking pathological results of liver, spleen and thymus.Results Com? pared with group B, the thymus index and serum ALT levels were significantly decreased in C, D and E groups (P<0.01), but the levels of IL-4and T-SOD increased significantly (P<0.01). Pathological results showed that there were more serious inflammatory cell infiltration in liver, edema and necrosis of dot in C, D and E groups than those in group B. Conclusion The active components of notoginseng showed a significant protective effect on immunological liver injury

    Analysis of the Parameters of Serum Iron Metabolism in Viral Hepatitis

    [Abstract] Objective To investigate the relationships between parameters of serum iron metabolism and liver in? flammatory activity grades, and to find out the difference in parameters of serum iron metabolism between patients with hepa? titis B and patients with hepatitis C. Methods A total of166patients with viral hepatitis were enrolled in this study, in? cluding135case of hepatitis B and31cases of hepatitis C. The serum iron metabolism indexes including serum iron (SI), se? rum ferritin (SF), transferring (TRF), total iron binding capacity (TIBC) were collected, and the transferring saturation (TS), al? anine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL) and albumin (ALB) were calculated. Thirty-nine patients were selected from135hepatitis B patients by corresponding gender, age and liver function with31hep? atitis C patients. The indicators of iron metabolism were compared between two groups.Results There was a positive corre? lation between age , TRF and TIBC. There were positive correlations between SI, SF, TS and ALT, AST, TBIL respectively, and negative correlations between SF, TS and ALB. TRF and TIBC were negatively correlated with ALT, AST and TBIL, but positively correlated with ALB (rs=0.551,P<0.001). The value of SF was significantly higher in patients with hepatitis C than that of patients with hepatitis B. The levels of TRF and TIBC were significantly lower in patients with hepatitis C than those of patients with hepatitis B (P<0.05). There were no significant differences in SI and TS between two groups. Conclu? sion There was a significant correlation between parameters of serum iron metabolism and live inflammatory activity grades. The iron overload phenomenon is much common in patients with chronic hepatitis C than that of patients with chronic hepatitis B.

    The Relationship between Homocysteine and Carotid Intima-media Thickness in Brain Infarction Patients

    [Abstract] Objective To investigate the relationship between total homocysteine (tHcy) and carotid intima-media thickness (CIMT) in brain infarction patients. Methods Sixty patients with fasting plasma tHcy levels≤10μmol/L (nonHhcy group),60patients with fasting plasma tHcy levels >10μmol/L and≤15μmol/L (H1group), and60patients with fast? ing plasma tHcy levels >15μmol/L (H2group) were chosen in hospitalized patients with acute cerebral infarction. Values of CIMT were detected in three groups of patients. The clinical biochemical indicators including triglyceride (TG), total choles? terol (TC), high density lipoprotein cholesterol (HDL- C), low density lipoprotein cholesterol (LDL- C), fasting blood sugar (FBS), folic acid (FA), Vitamin B12 (VitB12) and glycated hemoglobin (HbA1c) were also detected.Results There was signifi? cant difference in CIMT between three groups (P<0.01). The value of CIMT increased in H2group [0.98(0.90, 1.05)mm] com? pared with that of non-Hhcy group [0.85(0.80, 0.95)mm]. The value of CIMT increased in H2group compared with H1group [0.98(0.90, 1.05)mm vs0.85(0.85, 0.95)mm], P<0.05). There were significant differences in tHcy, FA and VitB12 between three groups. Based on the log-transformed values of CIMT as the dependent variable, multiple stepwise linear regression showed significant associations of the following variables with increased CIMT: increasing age, the history of smoking, the history of diabetes, higher LDL-C and tHcy levels.Conclusion Brain infarction in patients with higher tHcy level often have lower levels of FA and VitB12, and increased CIMT. When the level of tHcy >15μmol/L, there is more significantly higher level of CIMT. The increased CIMT level was associated with some cerebrovascular risk factors in patients with brain infarction

    The Association between Serum Adiponectin Levels and Gene Polymorphism with Coronary Artery Disease in Patients with Essential Hypertension

    [Abstract] Objective To observe the association between adiponectin gene polymorphism, serum adiponectin lev? els with the incidence of coronary artery disease (CAD) and the severity of coronary artery stenosis in patients with essential hypertension (EH).Methods A total of414patients with EH (234cases with CAD and180cases without CAD) and185 control subjects were recruited in this study. Serum adiponectin levels were measured by enzyme-linked immunosorbent as? say (ELISA). Adiponectin single- nucleotide polymorphisms rs266729,rs7649121,rs1501299and rs3774262were geno? typed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results Serum adiponectin levels were significantly lower in EH with CAD group than those in control group and EH without CAD group. Adiponectin SNP rs7649121AT genotype reduced the risk of CAD compared with AA genotype (adjustedOR=0.566,95%CI0.346-0.925,P=0.023). Logistic regression analysis showed that age and LDL-C were risk factors of CAD, and adiponectin was the protective factor for CAD in EH patients. The severity of coronary artery stenosis was negatively related to the level of adipo? nectin. Adiponectin levels were not affected by the adiponectin gene polymorphism.Conclusion The decreased serum adi? ponectin level was the independent risk factor for CAD in EH patients, which was negatively related to the severity of coro? nary artery stenosis. Adiponectin SNP rs7649121may contribute to the risk factors of CAD in EH patients.

    The Relationship between Sclerostin and Bone Metabolism in Hyperthyroidism Patients

    Objective To compare the differences between serum sclerostin levels in hyperthyroidism patients and euthyroidism healthy people. To explore the relationship between sclerostin levels and bone metabolism markers, BMD in hyperthyroidism patients. Methods Analyze sclerostin levels in 60 hyperthyroidism patients and 30 euthyroidism healthy people. 60 hyperthyroidism patients were divided into groups according to the T-score of their BMD. The bone metabolism markers and BMD were measured and the correlation between sclerostin and them was analyzed. Results The sclerostin levels in the hyperthyroidism group were higher than that in the euthyroid group(P<0.01) . Among hyperthyroidism patients, the group with osteopenia or osteoporosis had a higher sclerostin, N-MID osteocalcin, total of type 1 collagen amino terminal extension of the peptide (tP1NP), beta collagen special sequence (P<0.05) and a lower parathyroid hormone (PTH) (P<0.05) compared to normal BMD group. Sclerostin was positively correlated with free triiodothyronine(FT3)and beta collagen special sequence and negatively correlated with PTH, total radius BMD, femoral neck BMD and lumber spine BMD in hyperthyroidism patients. Conclution Hyperthyroidism patients had a higher serum sclerostin levels, sclerostin is closely associated with abnormalities of bone metabolism.

    Clinical Analysis of Continuous Airway Positive Pressure Ventilation in Patients with

     Objective   To study the curative effect of continuous positive airway pressure (CPAP) on elderly patients with obstructive sleep apnea-hypopnea syndrome (OSAHS) complicated with diabetes mellitus. Methods  Eighty patients with OSAHS and diabetes mellitus were included and randomly divided into treatment group (n=40) and control group (n= 40). The control group was given conventional treatment including diabetes diet and hypoglycemic drugs. The treatment group was given Futong ST-25 continuous positive airway pressure (CPAP) ventilation besides the conventional treatment. The serum levels of fasting blood glucose (FBG), two-hour postprandial blood glucose (2 hPG), insulin, sleep apnea hypop? nea index (AHI), low oxygen saturation (LSpO2) and the longest apnea time were monitored before and after treatment in two groups. Results  After four-week treatment, values of FPG, 2 hPG, AHI and the longest apnea time were significantly lower in treatment group than those in control group (P < 0.01), but values of insulin, two-hour postprandial insulin and LSpO2 were significantly higher in treatment group than those in control group (P < 0.01). Conclusion   CPAP therapy can effec? tively decrease blood sugar level and improve AHI, LSpO2 and the longest apnea time in elderly patients with OSAHS compli? cated with type 2 diabetes.

    Study of Bacteriological Analysis and Treatment Efficacy for Surgical Site Infection after

    [Abstract] Objective  To investigate incision pathogenic bacteria after spinal surgery, and observe clinical effects of therapeutic methods. Methods  A total of 30 cases of early surgical incision infection (7 cases of superficial infection and 23 cases of deep infection) after spinal surgery were retrospectively analyzed. The basic data including clinical manifes? tations, diagnosis, treatment, etiology and follow-up results were analyzed. Results   It was found that the mean infection time after operation was 2-15 days in 30 patients. Thirty-three strains were isolated including 18 gram-positive cocci (54.5%, and Staphylococcus aureus account for 13, 39.4%), 15 gram-negative bacteria (45.5%, and Coli communior account for 6, 18.2%). The drug susceptibility test showed that gram-positive cocci were highly sensitive to vancomycin, rifampin, tei? coplanin and cotrimoxazole. No vancomycin- resistant Staphylococcus aureus were found. Gram negative bacteria were high? ly sensitive to imipenem (100%). Seven cases of superficial infection were cured after dressing. Twenty-three cases of deep incision wound infection were no recurrence of infection after treatment by deep wound debridement, and postoperative cathe? terization. Conclusion  The early wound infection after spinal surgery is usually due to Staphylococcus aureus. It is good to perform wound debridement, continuous perfusion drainage, and treatment with vancomycin for deep wound infection.

    [Abstract] Objective  To investigate serum DJ- 1 expression and its significance in ovarian cancer. Methods  The expression levels of DJ-1 protein were detected by double antibody sandwich method in 52 cases of ovarian tumors (9 cases of benign ovarian tumors, 5 cases of borderline ovarian tumors and 38 cases of ovarian cancer,ovarian tumor group and 20 non-ovarian tumors (control group). Results  The expression of DJ-1 protein was lower in the control group than that in ovarian tumor group (P < 0.05). The expression of serum DJ-1 protein was higher in ovarian cancer than that in borderline ovarian tumors and in benign ovarian tumors. The expression of serum DJ-1 protein was higher in borderline ovarian tumors than that in benign ovarian tumors (P < 0.05).The expression of serum DJ-1 protein was higher in poorly differentiated and moderately differentiated ovarian cancer than that in high-grade differentiated ovarian cancer (P < 0.05). No significant dif? ference was found in the expression serum of DJ-1 protein between different pathological and different histological types of ovarian cancer (P > 0.05). Conclusion  DJ-1 might be used as a serum marker in the diagnosis of ovarian cancer. It might be involved in the pathogenesis of ovarian cancer,and could predit the development and prognosis of ovarian cancer.

    Biliary Stenting in the Treatment of Large Common Bile Duct Stones

    Abstract: Objective To investigate the role and value of the biliary stent in the treatment of common bile duct huge stones. Methods 32 cases of common bile duct huge stones with oddi sphincterotomy underwent endoscopic stone extraction fails plastic biliary stenting, were followed up for 6 months. Results 32 patients were successfully placed biliary stents, postoperative jaundice, abdominal pain and other symptoms have been alleviated, six months after surgery underwent secondary ERCP, 32 cases of biliary stones in three cases of huge stones disappeared, 25 cases were reduced to varying degrees , 4 cases no obvious narrowing; Again will remove the residual stones, stone were successful, there are four cases still stone difficulties and continue to observe the drainage of biliary stent placement. The success rate of treatment of common bile duct huge stones by biliary stent is 87.5%. Conclusions The therapy of huge common bile duct stones by biliary stent is a simple, short operation time, fewer complications, results were satisfactory, safe and effective advantages.

    [Abstract] Objective To assess the curative effect of microscopic anterior cervical approach in cervical spondylotic diseases. Methods A total of 75 patients were selected, who underwent microscopic subtotal anterior cervical decompres? sion and titanium mesh to fusion fixation or microscopic decompression of resection of the cervical intervertebral disc and cage implant fixation in Tianjin People's Hospital since October 2011 to October 2012. The Cervical vertebra gap involved 1 level in 41 cases, 2 levels in 30 cases and 3 levels in 4 cases. Pateints followed up regularly after operation. The nerve func? tion was assessed using Japanese orthopaedic association scoring system (JOA) function was calculated to examine the im? provement rate and to assess the curative effect of microscopic anterior cervical approach. Results All patients were fol? lowed up. The nerve function recovery was excellent in 44 cases, was well in 25 cases, was general in 6 cases, and the ex? cellent and well recovery rate was 92%. There was no significant difference between excellent recovery rate and well recov? ery rate. There were significant differences in values of each cervical levels between before and after surgery in the last fol? low-up of patients (P < 0.05). Conclusion Microscopic operative treatment for cervical spondylotic is safe and effective, with less trauma and bleeding. After surgical decompression, the recovery is rapid in patients with early ambulation.

    [Abstract] Objective To study the effects of improved Wright-Giemsa staining. Methods To evaluate the morphological characteristics of the sperm of 40 cases of semen by both traditional and improved Wright-Giemsa staining; and to assess the staining effects of improved Wright-Giemsa staining on vaginal discharge. Results The defective rates of the middle and tail parts of sperm are respectively 7.02± 2.4% and 11.02±2.03% in improved staining group; while that is 5.48±2.8% and 8.05±2.56% in traditional staining group. There is statistical significance between these staining methods (p<0.05). There is no significant difference of the staining of normal sperm and sperm head between these two staining methods (p<0.05); however, improved staining group shows a much clear nucleus and acrosome staining than traditional methods. Improved Wright-Giemsa staining is better for sperm, vaginal discharge and Trichomonas vaginalis. More importantly, the axostyle and flagella are clearly stained with the improved methods. Conclusion Improved Wright-Giemsa staining is simple and can be used in clinical diagnosis.

    Objective To investigate the effects of different infusion rate of remifentanil infusion on spontaneous ventilation in children who received sevoflurane anesthesia. Methods: A total of 120 children ,aged 3-6 years,undergoing strabismus surgery were assigned to 4 groups at random(n=30):C group(administration of saline),L group(remifentanil 0.03μg?kg-1?min-1), M group(remifentanil 0.06μg?kg-1?min-1) and H group(remifentanil 0.09μg?kg-1?min-1).The mean blood pressure,heart rates and respiratory rates,tidal volime,minute ventilation,and endtidal CO2, endtidal SEV were recorded after laryngeal mask insertion(T1), an initial bolus dose of remifentanil(T2),10mins after remifentanil infusion(T3),15mins after remifentanil infusion(T4), laryngeal mask remove(T5) respectively.The adverse events and time of induction,maintainance and emergence were also recoded. Results: There was no significant difference in general description , MBP, HR and VT in four groups.There was no body movement and desaturation observed. The RR and MV of H group at T3 and T4 were lower than other groups significantly(P<0.05) while PETCO2 were higher. The RR and MV between M and L group were not significantly different but lower than that of C group. Conclusions: Remifentanil infusion at a rate of 0.03~0.09μg?kg-1?min-1 could depress spontaneous ventilation in children who received sevoflurane anesthesia,but rarely lead to desaturaton.The respiratory depression effect is mainly manifested by reduction of RR rather than MV,especially at the rate of 0.09μg?kg-1?min-1..It is a good option to choose the low or moderate dose of remifentanil infusion to keep spontaneous ventilation in children while avoiding severe respiratory depression according to the demand of operations.

    Effects of Serum sOX40L, MMP-9 in Patients with Acute Coronary Syndromes by Rosuvastatin with Different Doses

    [Abstract] Objective To explore effects of serum soluble OX40 ligand (sOX40L), matrix metalloproteinase-9 (MMP-9) in patients with acute coronary syndromes (ACS) by rosuvastatin with different doses. Methods 60 patients with ACS were evenly devided into 20mg rosuvastatin treatment group (20mg group) and 10mg rosuvastatin treatment group (10mg group) at random, and 20 healthy volunteers were randomly assigned to control group. Serum concentrations of sOX40L, MMP-9 among above three groups were measured and analyzed before and after treatment. Results After two weeks’ treatment with rosuvastatin, serum concentrations of sOX40L, MMP-9 in patients of two treatment groups decreased significantly compared with pre-treatment (P<0.01). Moreover, the levels of sOX40L, MMP-9 in patients of 20mg group were lower than those in patients of 10mg group after two weeks’ treatment with rosuvastatin(P<0.05). Conclusion Larger doses of rosuvastatin may decrease the degradation of extraceller matrix of coronary atherosclerotic plaque and inflammatory reaction, stabilize coronary atherosclerotic vulnerable plaque when being used to treat patients with ACS, and can play a important role in improving prognosis of patients with ACS.

    [Abstract] Objective  To explore the diagnostic value of combined detection of the liquid array technology, interferon (IFN)-γ and IFN-γ-inducible protein (IP)-10 in the rapid, accurate diagnosis and differential diagnosis of tuberculous pleural effusions. Methods Patients with transudative pleural effusions were divided into tuberculous pleural effusion group (n=52) and malignant pleural effusion group (n=38). The method of T-SPOT.TB was used to detect the number of effee tor T cells sensitized to Mycobacterium tuberculosis and spot forming cells (SFCs). The liquid array technology was used to detect the level of IFN-γ and IP-10. Logistic regression was used to analyze and compare the diagnostic value of the twomethod combination. Results The diagnostic sensitivity, specificity and the area under the ROC curve (AUC) of T-SPOT. TB were 90.38%, 84.21%, and 0.938 (95%CI: 0.867-0.978), respectively. The diagnostic sensitivity, specificity and AUC of combined detection of IFN-γ and IP-10 were 98.08%, 97.37%, and 0.995 (95%CI: 0.951-1.000), respectively. There was no significant difference in the diagnostic sensitivity and specificity between the two methods, and the diagnostic agreement for the two diagnostic methods was fine (Kappa=0.703). The difference of AUC between the methods was significantly differ? ent (Z=1.996, P <0.05). The method of combined detection of IFN-γ and IP-10 showed the larger AUC (AUC= 0.995). Conclusion The combined diagnosis meets the clinical needs of rapid, accurate diagnosis and differential diagnosis for tuberculous pleural effusion by simultaneously assaying the level of IFN-γ and IP-10 using the liquid array technology.

The IH is related to hypertension, atherosclerosis, coronary heart disease, arrhythmia, stroke, heart failure and sudden death and so on. IH from OSA can lead to metabolic dysregulation, endothelial dysfunction, systemic inflammation, oxidative stress and the change of nerve body fluids, which has been shown to increase the risk of cardiovascular diseases. This paper mainly describes the pathogenesis of IH leading to the various cardiovascular diseases.

    Recent advances on vasohibin family regulating angiogenesis

    Angiogenesis is regulated by the local balance between angiogenesis stimulators and inhibitors. Vasohibin family as a novel regulator of angiogenesis has impact on the process of various pathophysiology. Vasohibin-1 is the only one factor which has been found up to now that regulates angiogenesis by means of negative feedback as the mechanism of action. It is expressed in ECs to terminate angiogenesis. Subsequently one gene homologous to vasohibin-1 was found and designated as vasohibin-2. It is expressed in infiltrating MNCs or cancer cells to stimulate angiogenesis. Furthermore, one binding protein to vasohibin-1 and vasohibin-2 was discovered and renamed as small vasohibin-binding protein (SVBP). The analysis of the function of SVBP revealed that SVBP binds to vasohibin-1 within the cells, makes a heterodimer with vasohibin-1 and facilitates the secretion of vasohibin-1. This article reviews the construction features and biological activities of vasohibin family members and their effects of inhibiting pathology angiogenesis.

    【Abstract】Metabolic syndrome (MS) and breast cancer are common diseases of women. Triple negative breast cancer (TNBCs) is one type of breast cancer which is of much attention in recent years. Important components of MS include central obesity, high blood sugar, high triglycerides and low high-density lipoprotein (HDL-C) increased the incidence risk of TNBCs. Common biomarkers of MS which are insulin, adiponectin and leptin play an important role in the occurrence and development of breast cancer especially TNBCs. Insulin-like growth factor-IImRNA binding protein 3 (IMP3, an oncofetal protein) may be TNBCs’ new invasive cancer biomarkers. In this paper, the progress of research on the relationship between MS and TNBCs will be reviewed.
