    The effects and mechanisms of monoclonal antibody against CD44 (A3D8) on proliferation in sphere-forming cells from solid tumor of ovarian cancer

    Abstract Objective: To explore the effects and mechanisms of monoclonal antibody against CD44 (A3D8) on proliferation in sphere-forming cells from solid tumor of ovarian cancer. Methods: MTS assay was used to evaluate cell proliferation; flow cytometry was used to analyze cell cycle distribution; DNA damage was measured by comet assay; expression of CDK2 and cyclinA proteins after treatment with A3D8 was evaluated by western blot assay. Results: A3D8 could inhibit the proliferation of sphere-forming cells from solid tumor of ovarian cancer; the inhibition was in dose- and time-dependant manner; compared with treatment with DDP alone, the inhibition rate caused by A3D8 combined with DDP was increased significantly ;The percentage of cells in S phase significantly increased after treatment with A3D8;DNA was damaged after treatment with A3D8 for 24h; western blot analysis demonstrated that the expression of CDK2、cyclinA proteins was suppressed significantly. Conclusion: A3D8 could inhibit the proliferation of sphere-forming cells from solid tumor of ovarian cancer, and the inhibiting effect was probably carried out by arrest of cell cycle.

    Analysis of Chromosome X, Y and 18 in Cleavage Stage Embryos with Low Morphological Scores by the Fluorescent in situ Hybridization

    ABSTRACT Objective: To evaluate the aneuploidy status of chromosome X, Y and 18 in day 3 cleavage stage embryos with low morphological scores by the fluorescent in situ hybridization. Methods: The discarded embryos from IVF-ET cycles were collected for our research. Nuclei of all the blastomeres were fixed in which the chromosome X, Y and 18 were analyzed by FISH. Results: Fluorescent signals were successfully detected in 233 nuclei, in which 115 (49.4%) nuclei were normal euploidy of chromosome X, Y and 18, while 118 (50.6%) were abnormal. The aneuploidy rate of chromosome 18 and X/Y were 40.8% and 33.5% respectively, but the differences was not statistical significant. The trisome rate of chromosome 18 was statistical higher than that of chromosome X/Y (21.9%vs.12.0%, P<0.01). Conclusion: A majority of the day 3 cleavage stage embryos with low morphological scores were proved to be chromosomal abnormal. The general embryo morphology scoring criteria could predict the embryo euploidy status to some extent.

Methods The plasmid of p-LXSN-Akt1 which contains the whole Akt1 gene sequence and p-LXSN-Akt2 which contains the whole Akt2 gene sequence were used to transfect GES-1 cells with lipofectamine, The GES-1 cells which were transfected by the empty vector p-LXSN were grouped as the black vector ,the no transfected ones were the control. Then the positive cell clones were selected with antibiotics G418. The protein expressions of Akt1, Akt2 and Mmp-2 were measured by Western blotting analysis; and Transwell assay analyzed their invasion and metastasis capability. The expressions of F-actin were detected by Immunofluorescence. Results Cell modes which expresses Akt1 gene (Akt1 group) and which expresses Akt2 gene (Akt2 group) were obtained; Compared with the black vector group and control group, The protein expressions of Mmp-2 in Akt1 and Akt2 groups were increased significantly.(P<0.01; transwell assay showed Akt1 and Akt2 groups could significantly advance the invasion and metastasis of cells; the expressions of F-actin were increased corresponding1y in Akt1 and Akt2 groups. Conclusion Akt1, Akt2 gene could increase cell invasion and migration, which showed the promotion of malignant transformation of GES-1 cells .

    Study of comparing with two reference data for evaluation of maternal thyroid function during pregnancy

    Abstract Objective: To evaluate the differences and reliability, we apply trimester-specific reference data created by Yan (Yan’s criteria) for diagnosis thyroid dysfunction of women during pregnancy, compared with population-based reference data (Adults’ criteria) . Methods: Totle samples we studied were 893 pregnant women during the three trimesters. The consistency of serum TSH and FT4 were detected. Both Yan’s and Adults’ criteria were applied to diagnosis thyroid dysfunction. Consistency rate and Kappa value were used to evaluate the validity and reliability of two types of criteria. Results: The medians of serum TSH concentration during pregnancy in the 1st, 2nd, 3rd trimesters were 1.72,2.27,2.38 mIU/L respectively; and the FT4 were 14.24, 13.36, 13.49 pmol/L. The prevalence of subclinical hypothyroidism diagnosed by Yan’s criteria during pregnancy is 10.5% compared with 7.5% by Adults’ criteria. The differences between them are mainly found in the 1st and 3rd trimesters. The revalence of hypothyroxinemia diagnosed by Yan’s criteria during pregnancy isse 2.6% compared with 11.0% by Adults’ criteria. The differences between them are found particularly in the 2nd and 3rd trimesters. Conclusion: Trimester-specific reference data of thyroid hormones established by Yan was confirmed to be more validity and reliability than population-based reference data during the three pregnant trimesters. The rates of missed diagnosis of subclinical hypothyroidism and misdiagnosis of hypothyroxinemia during pregnancy were decreased due to use of Yan’s criteria which was demonstrated more valuable in clinical practice.

    The Relation of Hyperuricemia with the Presence and Severity of Angiographic Coronary Artery Disease

    Objective: To evaluate the association between hyperuricemia (HUA) and the presence and severity of angiographic coronary artery disease (CAD). Methods: The study group consisted of 647 consecutive patients undergone coronary angiography. The anthropometric measurements including serum uric acid for every patient were conducted. According to the results of angiography, the patients with or CAD were divided into two groups. The severity of CAD was measured in terms of the number of vessels with disease and Gensini score. Results: (1) In women, the HUA percentage was significantly higher in CAD patients than in non-CAD patients (P<0.01). (2) HUA was significantly associated with the presence of CAD only in women (OR = 2.022,95%CI: 1.098~3.723,P =0.023). However, after adjusting for age, the association was no longer significant (OR = 1.513,95%CI: 0.778~2.931,P=0.221). HUA was not an independent risk factor of CAD in both genders. (3) For both genders, the relation between HUA and the distribution of diseased vessel numbers was not significant in the CAD patients (P=0.57;P=0.30). For women the Gensini score was significantly higher in HUA patients than in non-HUA patients(P=0.04, and the rank correlation was found to be significant in the total, HUA and non-HUA groups (r= 0.181,P = 0.004;r= 0.291,P = 0.049;r= 0.138,P = 0.046) . Conclusion: In women, HUA was associated with CAD with statistic significance, but was not an independent risk factor for CAD. Particular attention should be paid in aged woman HUA patients in clinical CAD diagnosis.

    Analysis on the relationship between anemia and heart function in hospitalized patients with chronic heart failure caused by coronary heart disease

    Objective: To investigate the relationship between anemia and heart function in hospitalized patients with chronic heart failure caused by coronary heart disease. Methods : 1816 patients which were hospitalized between Dec 2005 and Jan 2007 in the second hospital of Tianjin medical university were analyzed. The retrospective indexes include anemia, heart function. Results: 1816 cases were enrolled and the distribution of gender has no statistical significance. Among these cases, 305(13.8%) were found to have anemia ,the proportion of which in male and female is 16.34%(151/925)and .28 % (154/891) respectively (p>0.05).The average age in anemia set is older than that of the non-anemia set (t=6.823,P<0.01).The proportion of anemia in over 60 years elders is higher than that of under 60 years elders(χ2=13.312,p<0.01) . With the progression of heart function classification, the value of hemoglobin has a lower gradually trend, the prevalence of anemia has a higher gradually trend. Between heart function classification and the value of hemoglobin take the negative relationship ( r=-0.192,p<0.05) .LVEDD of anemia set is higher than the non-anemia, IVS is lower. Anemia set has a majority proportion of LVEF(≤45%),while, LVEF in non-anemia set is mainly >45%.The proportion of E/A≤1 is more than E/A>1 in both sets and E/A≤1 is more in non-anemia set, these differences all have statistic significant. Conclusion: Coronary Chronic Heart Failure usually accompanies with anemia, and then anemia can make the degree of CHF aggravate, increase the hospital mortality and form vicious circle. Therefore while in the treatment of heart failure, we should actively correct anemia, this can significantly improve the prognosis of patients with Chronic Heart Failure.

    Correlation studies of 18F-FDG uptake with VEGF expression and MVD in neuroepithelioma

    Objective: To investigate the correlation of 18F-FDG uptake with vascular endothelial growth factor (VEGF) protein expression and microvessel density (MVD) in neuroepithelioma. Methods: 18F-FDG PET examinations were performed preoperatively in 41 patients with suspected neuroepithelioma. Immunohistochemical stain methods were used to demonstrate the situation of VEGF expression and quantitatively measure the MVD in tumors. According to the situation of VEGF expression, all the cases were divided into two groups including low VEGF expression group and high VEGF expression group. Mann-Whitney U test was used for comparing the difference between the two groups. Spearman correlation analysis was used for observing the correlation between T/WM and MVD. Results: T/WM in low VEGF expression group (n=24) and high VEGF expression group (n=17) ranged from 0.62~3.04 and 1.29~7.64, respectively. The difference of T/WM between two groups was statistically significant (P<0.05). The MVD of tumors were 9.76~94.52, which showed positive correlation with T/WM (P<0.05). Conclusion: The 18F-FDG uptake of neuroepithelioma are feasible for evaluation of microvessel angiogenesis in vivo and have instructive function for clinical treatment and assessment of prognosis.

    The Expression of her-2 in the breast cancer and its relationship with angiogenesis

    Objective To study the expression of her-2 in the breast cancer and its relationship with angiogenesis. Methods The expression of her-2 and MVD were detected in 98 cases of breast cancer and 56 cases of neighboring noncancerous tissue by the method of immunohistochemistry. Results The positive rates of her-2 in breast cancer was higher than that in the neighboring noncancerous tissue(P<0.05); the expression of her-2 was correlated with lymph node metastasis and the difference is significant (P<0.05);her-2 and MVD in poorly differentiated group was significantly higher than that in well-differentiated and moderate-differentiated group(P<0.05); her-2 was positively correlated with tumor size; her-2 expression increased with increased TNM staging(P<0.05).;CD105-MVD in her-2 positive tumors was significant higher than in her-2 negative tumors(P<0.05). Conclusions The Experssion of her-2 in breast cancer are up regulated, her-2 plays an important role in incidence, development and angiogenesis of breast cancer

    Construction and Identification of a Novel DC Vaccine

    [Abstract]: Objective: To construct a novel DC vaccine which can coexpressing suppressor of cytokine signaling 1 (SOCS1) inhibitor, high mobility group proteins B1 (HMGB1), and tumor-associated antigens. Method: First construct the plasmid (p/shS1-NY-IRES-sig-HMG B1)that coexpress SOCS1 inhibitor, HMGB1, and tumor associated antigens. Then p/shS1-NY-IRES-sig-HMGB1 was transfected into DC. PCR and western blot was used to verify the expression of the p/shS1-NY-IRES-sig-HMGB1. ELISA was used to analyze the variation of TNF-α、IL-12、IL-1 and IL-6. Result: The sequence of the plasmid coincided with the sequence that expected. NY-ESO-1 can be expressed correctly. HMGB1 can be express in the cytoplasm. SOCS1 can be silenced by SOCS1 shRNA plasmid shS. TNF-α、IL-12、IL-1 and IL-6 was up-regulated in the culture medium(p<0.05). Conclusion: Successfully construct a novel DC vaccine which can coexpressing SOCS1 inhibitor, HMGB1, and tumor-associated antigens.

    The Changes of Gastric Intestinal Metaplasia after Helicobacter pylori eradication

    Abstract Objective To investigate chronic long-term effect of gastric intestinal metaplasia after the eradication of Helicobacter pylori. Method the total 319 cases of intestinal metaplasia, including 182 cases of the eradication group and 137 cases remained Helicobacter pylori positive were analyzed the changes with the time. results Mild intestinal metaplasia in patients are significant difference compared with non-eradication group after 12~36 months and 36~70 months, the P values of comparison of two groups are P<0.05 ,P<0.01 respectively. There are a significant difference between two groups in patients with moderate intestinal metaplasia from 36 to 70 months after H. pylori eradication(P<0.05), Severe intestinal metaplasia did not changed within seventy months after H. pylori eradication (P> 0.05). Conclusion The patients with mild and moderate gastric intestinal metaplasia after eradication of Helicobacter pylori can receive long-term effect.

    Studying neutrophil adhesion molecule CD11b and CD64 expression on the value of early diagnosis of neonatal infection.

    [Abstract] Objective Investigating the value of the peripheral blood neutrophil adhesion molecule CD11b and CD64 expression in the early diagnosis of neonatal bacterial infection and evaluating its efficacy. Methods Sixty-three suspected neonatal bacterial infection cases who were hospitalized from May 2005 ~ May 2008 were recruited into the study. Recorded their clinical manifestations, blood culture, cerebrospinal fluid culture results and five non-specific indices were measured for each patient. The patients were divided into sepsis group (n = 33) and non-septisemic infection group (n =30).Fifteen hospitalized neonates with noninfectious diseases were enrolled as controls (n = 15). 1ml peripheral blood would be taken from each group patients within 24~48h after they hospitalized. The levels of peripheral blood neutrophil CD11b and CD64 were measured by flow cytometry. During the recovery period (treatment 1 week later) for sepsis group measured again. Results The levels of peripheral blood neutrophil CD11b in sepsis group were higher than those of non-septisemic infection group and control group (P <0.05 ). There were no difference between non-septisemic infection group and control group (P = 1.000). The expressions of neutrophil CD64 in sepsis group were significantly higher than those of non-sepsis infection group and the control group (P <0.05), non-septisemic infection group were higher than those of control group (P <0.05). The levels of CD11b and CD64 of the cases with septicemia significantly decreased on the restoration stage (P<0.05). Positive critical value of sepsis group were CD11b≥320MFI, CD64≥20MFI , which for diagnosis sepsis showed sensitivity and specificity were 72.73% and 75%, 94.74% and 80.49%,respectively. The expressions of neutrophil CD11b and CD64 have no statistics significance between Gram-positive bacteria and gram-negative bacteria infection. Conclusion The expression of peripheral blood neutrophil adhesion molecule CD11b and CD64 measured by the whole blood flow cytometry can be used as one of the laboratory index for early diagnosis neonatal sepsis. For the neonatal sepsis, the serial measurements of CD11b and CD64 expression have a high value on evaluated infection severity and observaed therapy reactions.Observed on the infection to determine the extent of the evaluation of treatment with a more a high clinical value.

    Clinical Analysis of Creatinine Clearance and Total Bilirubin Levels in Aged Patients with Coronary Heart Disease

    Abstract Objective: To explore the clinical significance of creatinine clearance(Ccr)and total bilirubin(TBIL)Levels in aged patients with coronary heart disease. Methods: According to coronary artery angiography (CAG), 784 subjects aged more than 65 years were divided into several groups. The creatinine clearance and total bilirubin Levels were compared. Results: With the increased number of involved branches, the level of Ccr was gradually decreased, the level of TBIL was gradually increased (P<0.05). Multi-Logistic regression analysis showed that Ccr and TBIL were independent risk factors of CHD. Conclusion: Ccr and TBIL may have clinical forecast value for CHD. It was important to improve renal and liver functions in intervention and prevention of aged patients with coronary heart disease.

    A comparison of effects of postoperative analgesia with sufentanil or flurbiprofen on cellular immune function

    Abstract:Objective To compare the effects of postoperative analgesia with sufentanil or flurbiprofen on cellular immune function. Methods Forty-five ASA ⅠorⅡ patients undergoing cholecystectomy were randomly divided into three groups with 15cases each: groupⅠPCIA with sufentanil 2ug/ml; groupⅡ PCIA with sufentanil 0.75ug/ml+flurbiprofen 1mg/ml and groupⅢ PCIA with sufentanil 0.5ug/ml+flurbiprofen 1mg/ml. PCIA was started with background infusion rate 2 ml/h, blouse dose 0.5ml, lockout interval 5 min. VAS scores were measured at 4,24h and 48h after operation respectively. Blood samples were obtained from peripheral vein at before operation, and at 1,24,72h after operation for determination of the plasma levels of CD3+、CD4+ and CD8+ in lymphocyte. CD4+ /CD8+ ratio was calculated. Results VAS scores of groupⅢ were markedly higher than that of groupⅠand groupⅡin postoperative 4,24h and 48h (P<0.05). Before anesthesia, there were no significant difference in T Lymphocytic cell subpopulation (CD3+,CD4+,CD8+,CD4+/D8+), among three groups(P>0.05). The level of CD3+,CD4+ and CD4+/D8+ were all decreased at postoperative 1h and 24h in three groups compared with baseline value, while the level of CD3+,CD4+ and CD4+/D8+ in groupⅡand groupⅢ were increased compare with groupⅠ(P<0.05). Conclusions Flurbiprofen can improve the cellular immune function in the patients after cholecystectomy through reducing the consumption of sufentanil for PCIA.

    Analysis on the correlation between atherogenic index of plasma and coronary heart disease as well as risk factors

    【Abstract】 Objective : To analyze the correlation between atherogenic index of plasma (AIP) and coronary heart disease (CHD), as well as its risk factors. Methods: We selected 348 residents in Dagang district, Tianjin. All participants were divided into CHD and control group according to the clinical and laboratory examinations. We compared the levels of plasma TG, HDL-C, LDL-C, AIP and other risk factors of CHD between these two groups and analyzed of the correlation between the presence of CHD, the risk factors of CHD and AIP respectively. Results: we found that the percentile of male, hypertension, diabetes mellitus (DM), smoking, stoke were significant higher in CHD than that of control group (P<0.05). Compared with control group, Age, BMI, systolic blood pressure (SBP), diastolic blood pressure (DBP), fasting blood glucose, TG, ln (TC), LDL-C, uric acid, AIP, HR were higher, and the level of HDL-C were lower in CHD group significantly (P<0.05). Total bilirubin, ln (ALT), ln(AST) had no significant differences between two groups. There were positive correlations among AIP and the percentile of hypertension, smoking, stoke, age, BMI, SBP, DBP, fasting blood glucose, TG, ln (TC), LDL-C and uric acid. But there were negative correlations between AIP and HDL-C. SBP, DBP, TG, HDL-C and LDL-C enter in the regression equation (P<0.05). Conclusion: The level of AIP is strongly correlated with the CHD and the risk factors. Combined with traditional CHD risk factors, it strongly predicts the presence of CHD, and can provide more information for the prevention, diagnosis and treatment of CHD.

    The correlation between GDF-9 expression in granulosa cells and poor ovarian response to controlled ovarian hyper stimulation

    Abstract Objective: To determine whether the expression of GDF-9 in granulosa cells was related to poor ovarian response to gonadotropin stimulation. Methods: RQ-PCR was performed to detect the levels of GDF-9 mRNA in granulose cells obtained from 70 patients undergoing IVF treatment, the relation was analyzed between the level of GDF-9 and poor ovarian response. Results: There were not significant differences with patient age, the serum levels of basal follicle-stimulating hormone and basal estradiol, years of infertility, dosages of gonadotropin in different ovarian response groups(P>0.05). The expression of GDF-9 mRNA in the poor ovarian response group was lower than those of moderate and high responders, being 18.64±8.35, 19.29±9.32 on mural and cumulus granulose cells, respectively(P<0.05). The GDF-9 expression was positively correlated with the number of follicles (r=0.508; 0.632, P<0.01). Conclusion: The lower expression level of GDF-9 in granulosa cells may be associated with poor response to controlled ovarian hyper stimulation.

    Retinopathy in preeclampsia and its associated factor

    Abstract Objective: Retrospectively study patients with preeclampsia and with gestational hypertension(GH) who delivered at term, the association of retinopathy with various clinical and laboratory manifestations and fetal birth weight was evaluated. Methods: Medical records of 60 patients with preeclampsia and 65 patients with GH who delivered at term were reviewed. Parameters analyzed were retinopathy, systolic and diastolic blood pressure (BP), fetal birth weight, and various biochemical parameters. Multivariable linear regression was used for statistical analysis. P<0.05 was considered significant. Results: Severity of retinopathy was found to be inversely related to fetal birth weight (P<0.05) and positively related to serum uric acid level (P<0.05) in the preeclampsia group. All other variables in the preeclampsia group including systolic BP (mean, 166.4mmHg; P>0.05) and diastolic BP (mean, 103.5mmHg; P>0.05) were found to have no significant association with retinopathy. In the GH group, none of the parameters analyzed were significantly associated with ocular changes. Conclusions: The severity of retinopathy in preeclampsia is directly related to the level of placental insufficiency and intrauterine growth retardation. The severity of retinopathy might be independent of systemic BP.

    Assessment of Left Atrial Function by Strian Rate Imaging in Patients with Hypertension

    Abstract Objective: To evaluate the clinical value of left atrial function in primary hypertensive patients by Strian Rate Imaging(SRI). Methods:89 patients with primary hypertensions were divided into groups of normal geometry (LVN,n= 40) and remodeling geometry (LVR, n= 49).41 healthy participants were also included.At four-chamber view,the strain rate curves was gained in the middle segments of each left atrial lateral wall by SRI.Strain rate parameters SRs were measured during left ventrial systolic phase,SRe and SRa were measured during left ventrial diastolic early phase and later phase;Peak velocity during left ventrial diastolic early phase and later phase (Ea and Aa) at the left ventricle lateral wall of annulus mitral valve were measured on Tissure Doppler imaging(TDI),then Ea/Aa were calculate. E Peak and A Peak velocity (E and A) of bicuspid were measured on pulsed wave Doppler(PW),then E/A were calculate.Left atrial AP diamind(LAD) were measured on 2D. Results: Compared with the control group, the SRs and SRe were reduced in two hypertensive Group,meanwhile the SRa was higher in LVN group(p<0.01);Ea/Aa were reduced in two hypertensive group,E/A were reduced in LVR group(p<0.01), LAD was higher in two hypertensive group and LAD in LVR was higher than that in LVN(p<0.01).Conclusions: The SRa、SRe and SRs can assess left atrial systolic and diastolic function.

    Endothelial Progenitor Cells Derived from Rat Bone Marrow Culture with Differential Adhesion Method

    Abstract Objective:Explore an economic and reliable, stable, high purity of endothelial progenitor cells in culture methods.Methods:Rat bone marrow-derived EPCs were separated by density gradient centrifugation and cultured,then plated on 25ml culture flask adherent culture with human fibronection, 48h no adherent cells were collected cultured for 7 days, and then identified by being observed the experirment of uptaking DiL-acLDL and combining FITC-UEA-1 by fluorescence microscope . Also Flow Cytometry was performed to analyze the expression of CD133、flk-1、VE-cadherin(CD144) and CD31 dynamically.Results:48h no adherent cells cultured,the cells were small and spindle or polygonal in shape.Large numbers of typical endothelial progenitor cells coloy-forming unites were found. Cells DiI-acLDL uptake and FITC-UEA-1 combining fluorescent staining positive rate(87.0±6.0)%.Flow cytometry to detect cell surface antigen CD133, flk-1, VE-cadherin and CD31 positive rates were 61.01%, 8.55%,8.20%,6.18%. Conclusions:Differential attachment method is a reliable, stable, high purity of endothelial progenitor cells culture methods. Provide methodological references for further research.

    A Long-term Neuroprotective Effect of Propofol Postconditioning on Ischemia-reperfusion Injury in Rats

    Abstract Objective: To observe the long-term effect of propofol on cerebral ischemia-reperfusion injury in rats. Methods: Middle cerebral artery occlusion models were performed in male SD rats. One hundred and forty-four rats were randomly divided into 4 groups with 36 rats in each group, which included S group, IR group, L group and P group. In the following 28 days, the modified neurological severity score, the percentage of cerebral infarct volume and Morris water maze test were detected respectively. Results: After a long-term observation,comparing with IR group, propofol improved the neurological impairment, reduced the cerebral infarct volume and increased the learning and memory ability. Conclusion: Propofol provides a sustained neuroprotective effect, can last for at least 28 days and this effect is not lipid-dependent.     Abstract Objective:Study the relationship between cisplatin resistance of ovarian carcinoma and PI3K/Akt singnal transduction pathway. Methods:Western blot was used to detect the expression level of PI3K and Akt in SKOV3 and SKOV3/DDP cells.The effection of LY294002 and DDP to expression of PI3K and Akt in Ovarian carcinoma cells were detected by Western Blot. MTT assay was used to analyze the synergism of LY294002 to DDP and the effection of LY294002 to control ovarian tumor cells growth .Results:The correspondence density of PI3K and Akt in SKOV3 cells were lower than that in SKOV3/DDP cells.LY294002 could reduce the expression of PI3K and Akt in SKOV3 and SKOV3/DDP cells. After interferenced by LY294002 40μmol/l, the sensitivity of SKOV3 cells to DDP increased 67.36% and the sensitivity of SKOV3/DDP cells to DDP increased 76.56%. Conclusion:The abnomal PI3K Akt/signaling pathways related to Cisplatin resistance of Ovarian carcinoma. Inhibiting PI3K Akt/signaling pathways could reverse ovarian cisplatin drug-resistant phenotype.

    The Effect of HMME-mediated Photodynamic Therapy on Streptococcus mutans of Cariogenic Bacteria and Caries Morphology

    Abstract Objective: To study the role of photodynamic therapy using hematoporphyrin monomethyl ether as photosensitizer in the prevention of dental caries by establishing the caries rat model. Methods: Wistar rats, whose molars were inoculated with Streptococcus mutans (S. mutans), were fed with caries - inducing diet 2000# in order to establish the caries rat model. These rats were divided into five groups, whose molars were treated respectively with 10 ng/L HMME-PDT (group A), 20 ng/L HMME-PDT (group B), 40 ng/L HMME-PDT (group C), 0.2 g/L sodium fluoride as positive controls (group D), and with 0.9%sterile physiological saline as negative controls (group E) once a week for four weeks and recorded the changes of S. mutans every week. The rats were then killed. The surface morphology of dental caries of the rats was observed by scanning electron microscope. Results: The colony count in each group shows no significant difference in the first and second weeks ( P<0.05). The colony counts of group B and C are significantly reduced compared with group E in the third week ( P<0.05). In the fourth week the colony counts of group A、B and C are significantly reduced compared with group E (P<0.05 or P<0.01 ). Meanwhile the colony count of group C is significantly lower than both group A and B (P<0.05). The tooth enamel of both group C and D are found to be damaged most slightly, and the tooth surfaces of the two groups are relatively smooth. Conclusion: PDT has anticaries effect in vivo. PDT is thought to be a promising method in the prevention of caries.

    The protective effect on intestine of propofol after hemorrhagic shock and resuscitation

    Abstract:Objective: To determine the protective effect on intestine of propofol after hemorrhagic shock and resuscitation by measuring the changes of intestinal SOD and MDA and plasma DAO levels. Method: Thirty Wistar rats were randomized into 3 groups: sham operation group, resuscitation group and propofol group. There were 10 rats in each group. To establish animal models of hemorrhagic shock and resuscitation. Following resuscitation, the resuscitation group was infused normal saline at 1ml/kg/h with infusion pump for 2 hours, the propofol group was infused propofol at 1ml/kg/h for 2 hours. Then sampling in each group. Results: The MDA and DAO levels in the resuscitation group and the propofol group were higher than those in the sham operation group (P<0.5),Propofol group was lower than resuscitation group(P<0.5) ; but SOD levels were lower than those in the sham operation group (P<0.5), Propofol group was higher than resuscitation group(P<0.5). Conclusion: The intestinal tissue was significantly damaged after hemorrhagic shock and resuscitation, propofol can reduce this damage by against the oxygen free radicals.

    Clinical study on 40 hypokalemic periodic paralysis cases

    Abstract Objective:To investigate the clinical features and OGTT results of 40 hypokalemic periodic paralysis cases. Methods:The clinical manifestations in 40 patients and OGTT results of 33 patients were analyzed and summarized. Results:40 patients had no clear hypoPP family history and they concurrent hyperthyroidism in 22 cases, subclinical hypothyroidism in 1 case and other 17 cases did not associated with any endocrine diseases after completing laboratory examinations .In comparison with the control,the OGTT results of 33 hypoPPs show a higher insulin level and insulin resistance. The deposition of immunoglobin on the muscle cell membrane in 16 cases was significantly increased. Conclusion:HypoPP can occur alone,also can concurrent hyperthyroidism. Hyperthyroidism and HypoPP can occur in a patient at the same time or in different periods . HypoPPs with or without hyperthyroidism have a higher insulin level and insulin resistance. Abnormal glucose metabolism and hyperinsulinemia may be one of the factors for disease.

    The Clinical and CT Presentation of Retroperitoneal Fibrosis

    Objective: To investigate the clinical and CT features of idiopathic retroperitoneal fibrosis (IPF). Methods: 7 cases of retroperitoneal fibrosis proved by clinicopathology were presented. The clinical and CT performance of the cases were analyzed. There were 5 males and 2 females, with an average age of 53.5 years. All the cases were idiopathic fibrosis. Plain CT scan and contrast enhancement CT scan were undergone in all cases. Results: The clinical performances of the cases were nonspecific. The first symptoms included abdominal and lumbar pain. Others symptoms included lower extremity swelling (4 cases), abnormal soft-tissue mass (3 cases) and urinary tract obstruction (3 cases).On CT study, well circumscribed masses were showed in 7 cases. The lesions could be hypodensity, inhomodensity or isodensity, and various enhancements by contrast administration. Conclusion: Retroperineal fibrosis is a rare disease with various manifestations on CT study. It should be diagnosed if the mass lesion occurs in retroperitoneal space complicating with hydronephrosis or ureterolysis.

    The Effect of Surgical Treatment in Children with Stage Ⅳ Neuroblastoma

    Objective: To summarize our data of surgical treatment, chemotherapy and radiotherapy for 48 cases of children with stage Ⅳ neuroblastoma, who with complete followed-up , further to investigate the effect of surgical treatment in the combined management for children with Ⅳ neuroblastoma. Methods: Form June 1995 to June 2009, 48 cases with stage Ⅳ neuroblastoma in our hospital were enrolled in this study, including 31 boys and 17 girls , who were divided into medium and high risk groups ( 10 and 38 cases for each group ) according to their age of onset. The comprehensive protocol included delayed tumor resection , intensive chemotherapy mainly composed of multiple cytotoxic drugs and local radiotherapy and so on. Results: After a follow-up ( range 2 to 110 months ),the 38 patients who underwent complete tumor resection ,had the median survival of 27.5 months. And the median survival of 10 patients who underwent partial tumor resection was 23.5 months. No significant difference was noted between them(P>0.05). And we found no significant difference in the CR rate ,relapse rate and survival condition between two groups (P>0.05). Conclusions: stage Ⅳ neuroblastoma should be taken combined therapy, including surgery, radiotherapy and chemotherapy , complete primary tumor resection has no significant effect on the CR rate and relapse rate and survival condition , thus the prognosis of patients with stage Ⅳ neuroblastoma undergoing surgical treatment depends on the effect of chemotherapeutics which inhibits the metastasis.

    Abstract: Objective To study the clinicopathological features and immunophenotype of primary rhabdomyosarcoma of the kidney. Methods 2 cases of primary rhabdomyosarcoma of the kidney were collected from 2000 to 2009 in the second hospital of Tianjin medical university.The immunohistochemical SP method was used to detect the expression of Vimentin、Myoglobin、Desmin、α-SMA、EMA、NSE、S-100.The clinical findings were analyzed and the hematoxylin and eosin-stained sections were reviewed. Results Primary rhabdomyosarcoma of the kidney account for 0.2% of malignant renal tumors. Both of the patients were male.One tumor was on the left and one on the right.The tumors with moderate pleomorphism composed of alternating fascicles of spindle cells with blunt-ended nuclei and eosinophilic cytoplasm.After SP staining, Vimentin showed positive or strong positive expression, but Myoglobin, Desmin was negative, EMA, NSEand S-100 expression in various degrees. Conclusion Primary leiomyosarcoma of the kidney is a rare malignant tumor of the kidney.Diagnosis of this tumor need to be differentiated from diverse malignant renal tumors by pathohistology、histochemical stain and immunohistochemical stain.

    Clinical Study of Recurrence Hepatitis C of Post Liver Transplantation

    Abstract Objective:Recurrence of hepatitis C in the graft is universal and may lead to chronic hepatitis in most patients and to cirrhosis in 20-30% of patients within 5-10 years of transplantation. Early identification of patients at a higher risk of rapidly progressive recurrent hepatitis post liver transplantation (LT) could help to tailor antiviral therapy. We retrospective studied the risk factors of recurrence of hepatitis C in the graft and wish to provide basis for prevention and treament of hepatitis C. Methods:We studied the before and early post-LT viral load and the histological and clinical outcomes of 44 consecutive patients (37 males,7 female, median age 57.84(39-76) years) ,whom were receiving liver transplantation in tianjin first central hospital from 2006 4th to 2009 4th. The immunosuppressive therapy(FK506+MMF+Pred),examing the liver function,FK blood concentration,HCV viraemia are convention,analysis the impactive factors of hepatitis C recurrence. Results:Hepatitis C recurred at histology in 31 of 44 (70.46%) patients. Brfore LT and early viral load after LT was higher in patients with rapidly progressive hepatitis C recurrence ((P<.01).Recipient older than 65 years have higher recurrent rate(P<.05). Antiviral therapy with interferon compared with untreated patients with higher HCV negative rate (P <.05). Cytomegalovirus infection and the use of immunosuppressive program has nothing to do with the recurrence Conclusion:The results emphasize the importance of high viral loads in the pre and early posttransplant period as an independent redictor of recipient outcomes. 2Interferon antiviral therapy is feasible for recurrence hepatitis C.
