    Effects of ZEB2 3′UTR Transfection on Proliferation ,Invasion and Migration of Human Gastric Epithelial Cell GES-1

    [Abstract] Objective To explore the effects of zinc finger E-box binding protein (ZEB)23′UTR gene transfection
    on proliferation, invasion and migration in human gastric epithelial cell line GES-1. Methods The synthetic ZEB23′UTR
    and miR-200b micmics were transfected into GES-1cell line by lipofectamine2000. We set up control grop, the mutation group and ZEB23′UTR group. Real-time quantitative PCR was performed to evaluate the expression levels of miR-200a/b/c and ZEB1/ZEB2mRNAs after transfection.And then we set up control group, ZEB23′UTR group, ZEB23′UTR+negative control group and ZEB23′UTR+miR-200b micmics group. The protein expression levels of ZEB1, ZEB2, matrix metallopro?teinases (MMP) 2/9and proliferating cell nuclear antigen (PCNA) were detected by Western blot assay. The invasion and migration capability were analyzed by transwell assay and wound healing test. MTT assay was used to detect the proliferation ability.Results Compared with control group and mutation group, the expressions of miR-200a/b/c were significantly decreased, especially for miR-200b. And the expressions of ZEB1/ZEB2were significantly increased at both mRNA and protein levels after transfected with the ZEB23′ UTR, enhancing the capability of migration,invasion,and proliferation (P<0.05). Compared with ZEB23′UTR group, the capabilities of proliferation,invasion and migration were significantly lower in combined group.Conclusion ZEB23′ UTR can increase the ability of cell proliferation, invasion and metastasis through regulating the levels of miR-200a/b/c, and then influence the regulation of transcription of the target gene, which could lead to malignant transformation of GES-1cells.

    Study of the regulation of BDNF/TrkB signal pathway in hippocampal neurons by different isoforms of TrkB

    Objective Brain derived neurotrophic factor(BDNF) and its specific tyrosine kinase receptor B(TrkB) are highly correlated to epilepsy. This study mean to explore the regulation of BDNF/TrkB signal pathway in hippocampal neurons by different isoforms of TrkB. Methods The cells were divided into 14 groups.The immunofluorescent technique was used to identificate the hippocampal neurons.Epileptiform discharges were detected by electrophysiological techniques.Western blot assay was used to determine the protein expression of TrkB and p-TrkB of each group. Results Compared with control group, the p-TrkB/TrkB value in control + BDNF group was higher. In epilepsy + BDNF group, the p-TrkB/TrkB value was lower than control+BDNF group, but higher than epilepsy group.The p-TrkB/TrkB value of epilepsy + ALLN + BDNF group was lower than epilepsy + BDNF group. The difference of the p-TrkB/TrkB value between epilepsy + ALLN + BDNF group and epilepsy + ALLN group was not significant.The p-TrkB/TrkB value in epilepsy + anisomycin + BDNF group was higher than both of epilepsy + anisomycin group and epilepsy + BDNF group. Conclusion BDNF can activate BDNF/TrkB signal pathway.The increased expression of TrkB.T can inhibit the activation of BDNF/TrkB signaling in status epilepticus.The change of TrkB.FL protein levels cannot inhibit the BDNF/TrkB signal pathway.

    Verification of differential expression genes after CacyBP/SIP unclear translocation in colon carcinoma cell lines

    [Abstract] Object To verify the genes screend by the PCR chip of cell cycle. Methods The colon cancer cells were randomized into two groups. The test group recoved the stimulation of gastrin,while the control group not .The method of western blot were used to detect the expression of Cacybp/SIP before and after the stimulation of gastrin. Verification of differential expression of CDK8, CKS2 by using real-time quantitative PCR. Results It is found that before the stimulation of human gastrin, CacyBP/SIP is located and expressed in the cytoplasm, and then in both cytoplasm and nucleus after the stimulation of human gastrin. The results of real-time quanntitative PCR of CDK8 and CKS2 gene were consistent with those of microarray detection , The expressions of CDK8 and CKS2 were up-regulated. Conclusion The stimulation of human gastrin can lead to the nuclear translocation of CacyBP/SIP. The results of microarray are reliable and differentially expressed genes screened through gene chip deserve futher study.

    The Analysis of Abnormal Gene Expression in Mammary Epithelium for Early Diagnosis of Breast Cancer

    Abstract: Purpose: Abnormalities always exist in breast epithelial tissues of normal histologic feature; thus, investigation of these differential expressed genes plays an important role in understanding of breast cancer development and in early diagnosis of this disease. Methods: Microarray technology provides a powerful tool to detect a large number of genes at the same point in time, which can be used to identify abnormal gene expression. In this study, we used bioinformatics tools to establish a model for early diagnosis of breast cancer, and screen differentially expressed genes by using signal pathway enrichment analysis. Results: The best prediction model was derived from the combination of differential genes enriched from KEGG and BioCarta database; the number of differential expressed genes in three random created prediction models was reduced from 22 to 7, 14 to 3 and 18 to 4, however, the prediction accuracy was consistently with the model established from all of the differentially expressed genes, and the average accuracy of all models is 96.3%. Conclusion: By this means, the prediction model can be simplified with the prediction accuracy unchanged, and thus facilitate the model apply to early diagnosis and prevention of breast cancer.

    Objective: to investigate the inhibiting lysyl oxidase (LOX) gene expression of laryngeal cancer cell-Hep-2 proliferation, invasion and influence of radiotherapy sensitivity.Methods: LOX interference plasmids were transfected into cells, The detection of LOX mRNA and protein expression were used by RT-PCR and Western blot., Ki-67,PCNA, MMP-2/9 protein expression were assied by western blot; flow cytometry (FCM) detection of apoptosis in Hep-2 cells. Results after LOX interference plasmid were transfected into Hep-2 cells, LOX protein and mRNA expression levels were cut down; Western blot results showed that LOX, Ki-67, PCNA, MMP-2/9 protein expressions were typically lower than the negative control group and control group; the apoptosis rate of transfection cells combined with radiotherapy was significantly higher than the control and the negative control group. Conclusions Downregulation of LOX expression can inhibit Hep-2 cells’ proliferation and invasion, enhanced radiosensitivity, the mechanism of this action may be related to the downregulation of Ki-67, PCNA, MMP-2/9 protein expression.In a word, this could provide a new target for clinical threapy, improving the effect of laryngeal squamous cell carcinoma.

    Abstract Objective:To study the functional roles of 1,25 (OH) 2D3 inducing osteogenic differentiation of the dental papilla stem cells.Methods:The dental papilla stem cells were isolated and cultured in medium supplemented with different concentrations of 1,25 (OH) 2D3. MTT assay was used to detect cell growth, and flow cytometry to detect cell cycle. Western blot was used to detect protein expression levels of receptor activator of nuclear factor-k B ligand (RANKL), osteoprotegerin (OPG) and vitamin D receptor (VDR). After siRNA silencing VDR expression, RANKL and OPG protein level were detected. Results:MTT and flow cytometry results showed that 1,25 (OH) 2D3 have no obvious effect on dental papilla stem cell proliferation. Western blot results showed that RANKL, OPG and VDR protein expression level were in a dose correlation with 1,25 (OH) 2D3 concentration. When VDR expression was silenced by siRNA, the protein expression levels of RANKL and OPG were decreased as vary degrees. Conclusion:1,25 (OH) 2D3 affects the osteoblast differentiation process of the dental papilla stem cell by adjusting the VDR expression.

    Objective:To explore sunitinib in highly metastatic hepatocellular carcinoma cell lines MHCC97-H in vitro cytotoxicity, as well as focal adhesion kinase (Focal Adhesion Kinase, FAK) expression levels.Methods:To observe the morphological changes of cells stained using Giemsa, to monitor highly metastatic hepatocellular carcinoma cell lines MHCC97-H inhibition by MTT assay,to detect the protein expression of FAK before and after treatment by Western-Blot. Results:Sunitinib on hepatoma cell lines MHCC97-H inhibition, Switzerland 400 times Giemsa staining can be observed under the light microscope chromatin condensation, nuclear fragmentation and apoptotic bodies and other typical apoptotic morphology. 48h drug inhibition rate the most obvious, 2.5, 5 and 20umol / L concentration inhibition rate were (32.9 ± 1.5)%, (49.2 ± 2.3)%, (63.8 ± 2.8)% and (58.9 ± 3.4) %, the group differences were statistically significant. Western-Blot results showed that different concentrations of sunitinib after 48h, FAK protein levels were reduced in 85.51%, 72.09%, 57.61%, 62.02% of the control group, the differences were statistically significant (P<0.05). Conclusion:Sunitinib on hepatocellular carcinoma to inhibit and induce apoptosis, and can reduce the expression of FAK.

    Systematic and Endothelial Inflammation Status in Intermittent Hypoxia and Emphysema Rat Model and Level of Endothelial Progenitor Cells in Peripheral Blood

    Objective To establish the rat overlap syndrome (OS) model of intermittent hypoxia (IH) and emphysema, explore systematic and endothelial inflammation status, and observe the change of endothelial progenitor cell (EPC) level. Methods Overlap exposure was IH exposure on the base of pre-existing emphysema which was caused by 16 weeks of smoke exposure. 60 male Wistar rats were randomly divided to four groups with 15 per group according to the exposure conditions as follows: normal oxygen control group (A); IH group (B); emphysema group (C); OS group (D). After exposure, ELISA method was used to detect values of tumor necrosis factor alpha (TNF-α) and interleukin (IL)-6 in plasma and in the endothelium of right common carotid artery. Real-time-PCR assay was used to analyze RhoA mRNA level in the endothelium of right common carotid artery. Flow cytometry was used to detect EPC level. We also obtained tissue of right carotid artery for counting the percentage of intima-media thickness (IMT) in the all wall (C-IMT%). Results D group had highest values of TNF-α, IL-6, RhoA mRNA and C-IMT% among all groups. The quantity of EPC were highest in D group among all groups (all P < 0.05). Conclusion OS rats had more serious vascular endothelial injury than emphysema or IH rats. Meanwhile, the repair capacity of EPC for endothelium was the worst, which increase the risk of cardiovascular diseases.

    Syk kinase Signaling is Required for the Inflammasomes Activation during Listeria monocytogenes Infection

    [Abstract] Objective The aim of this study is to clarify the role of syk kinase in inflammasomes activation during Listeria monocytogenes (LM) infection. Methods Murine peritoneal exudates macrophages pretreated with inhibitor of syk, P38MAPK, or PI3K kinase were infected with LM for 24h, the protein level of IL-18 in the supernatant was detected by ELISA kit, the activation condition of key molecule ASC in the infected-macrophages cytoplasm was observed under fluorescence microscope. Furthermore, the syk protein kinase phosphorylation level at different time point during LM infection were detected by western blot. Results IL-18 protein level produced by macrophages in the presence of syk inhibitor was significantly reduced compared with that in macrophages without pretreatment. Meanwhile, the percentage of ASC-speck formation in the cytoplasm of syk inhibitor pretreated-- macrophages infected with LM diminished apparently. Furthermore, we found that the phosphorylation level of syk could be detectable at 5min postinfection, which confirmed the critical role of syk in the inflammasomes activation upon LM infection. Conclusion Syk kinase signaling is required for the inflammasomes activation upon Listeria monocytogenes infection in murine macrophages.

    Effects of Kangnao liquid on expression of Pi3kmRNA and AktmRNA with Focal Cerebral Ischemic-reperfusion Injury in rats

    Objective To observe the therapeutical effect of kangnao liquid in rats with focal cerebral ischemia-reperfusion(I/R) and to investigate the expression of Pi3kmRNA and AktmRNA in rats.Methods 180 healthy male SD rats were randomly divided into 6 groups:sham operated group,model group,The high-,middle- and low-dose kangnao liquid groups and Nimodipine group.Rats in drug treatment groups were administrated with kangnao liquid in accordance with 24g.kg-1.d-1 and 12g.kg-1.d-1 and 6g.kg-1.d-1 and with Nimodipine in accordance with 1mg.kg-1.d-1.Rats in model and sham groups were treated with distilled water with the equal volume.Continuously administrated for 7 days, the animal models of middle cerebral artery occlusion (MCAO) were established by a monofilament method from right internal carotid artery.Neurological evaluation was performed 24h after reperfusion.The In situ hybridization was used to investigate the expression of Pi3kmRNA and AktmRNA in rats on 12h, 24h, 48h, 72h and 168h after ischemia for 2h.Results Compared with model group,kangnao liquid groups had significantly improved neurological responses.The expression of Pi3kmRNA and AktmRNA at different time points of brain tissue were significantly improved.Nimodipine group had also improved neurological responses and the expression of Pi3kmRNA and AktmRNA,but it was lower than high- and middle-dose kangnao liquid groups.Conclusions Kangnao liquid can treated the rats with cerebral ischemia-reprefusion injury and protect nerve cells of cerebral infarction areas.The protective effect of kangnao liquid may be associated with that kangnao liquid can incresse the expression of Pi3kmRNA and AktmRNA.

    [Abstract] Objective To study the effect of Rimonabant for cannabinoid receptor 1 andα-SMA of C57 mice with experimental hepatic fibrosis,Analysis the mechanism of action of rimonabant in Liver fibrosis progression . Methods thirty C57 mice were randomly divided into three groups, i.e. normal control group, model control group, model rimonabant group. There were 10 mice in every group. C57 mice with experimental hepatic fibrosis were induced by intraperitoneal injection of 10% CCl4. Mice were sacrificed at the end of six weeks , liver tissue were Specimened. The expression of CB1 andα-SMA in liver tissue of mice was observed by immune histochemical technique. The score of fibrosis stage (S) in liver tissue were also analyzed. Results The expression of CB1 andα-SMA and the score of fibrosis in model groups and model rimonabant group were significantly higher than those in normal control group (P<0.05), the expression of CB1 andα-SMA and the score of fibrosis in model groups were significantly lower than those in model rimonabant group (P<0.05), while Correlation analysis Indicate that there were correlative obvirously in the score of CB1、α-SMA and the fibrosis in each experiment group(P <0.05) .Conclusion The activation of CB1 can promote the formation of liver fibrosis, Rimonabant can Inhibit the proliferation and activation of HSC by inhibit expression of CB1, so have effect of anti hepatic fibrosis.

    Objective: To explore the effect of the different concentrations of magnesium ions on rat vascular smooth muscle cell (VSMCs) calcification. Methods: VSMCs were obtained from rat aortic, and identified by immunocytochemistry, then randomly divided into control group, high phosphorus group, magnesium intervention group (the group was settled three subgroups according to the concentration of magnesium ions based on based on the high phosphorus medium, namely 1 mmol/L, 2 mmol/L and 3 mmol/L).Calcification staining, calcium content and alkaline phosphatase activity were measured and the expression of Cbfα1 mRNA was detected by RT-PCR after stimulating 7 days. Results: Compared with the control group, the remaining groups had calcium deposits, and they were higher than the control group, the calcified nodules gradually reduced with the increasing magnesium ion concentration in the intervention group, In addition to the calcium content in the intervention group was no difference with the high phosphorus group, the intervention group 2 and 3 were lower than the high phosphate of calcium content (P<0.05), compared with the control group, ALP activity and Cbfα1 mRNA expression were no difference in the intervention group 2, the remaining groups were higher than the negative control group (P<0.05). ALP activity and the expression of Cbfα1 mRNA were gradually decreased with the increasing magnesium ion concentration in the intervention group, and were lower than the high phosphorus group (P<0.05). Conclusion: Magnesium can reduce calci?cation and osteoblastic transdifferentiation in vascular smooth muscle cells in a certain extent, which may be achieved by reducing the expression of Cbfα1 in VSMCs Cbfα1.

    microRNA-21 regulates cardiac remodeling by promoting proliferation and differentiation of fibroblast after myocardial infarction

    Objective To investigate the role of microRNA-21(miR-21) on cardiac fibroblast proliferation and differentiation in the mouse model of myocardial infarction. Method Male C57BL/6 mice underwent ligation of the left coronary artery to produce models of myocardial infarction (MI). Echocardiographic assessment and histological evaluation were performed after ligation. The expressing levels of miR-21 were measured by quantitative real-time PCR in the various myocardial tissues. The in vitro experiment was divided into control group, blank group and miR-21 minic group. In cardiac fibroblasts stably overexpressing miR-21 by transfection of miR-21 minic, proliferation was assessed by immunostaining for 5-ethynyl-2’-deoxyuridine (EdU). Western Blot was used to detect the expression of α-SMA and Smad7 in the cardiac fibroblasts. Results The expression of miR-21 was significantly increased in border area compared to remote areas in MI group than that of sham group (P<0.01). There was a higher expression of miR-21 in miR-21 minic group than those of control group and blank group (P<0.01). The EdU positive rate was significantly higher in miR-21 minic group than those of control group and blank group (P<0.01). Overexpression of miR-21 in cardiac fibroblasts increased α-SMA level, while Smad7 level, a target gene of miR-21, was significantly decreased. Conclusion Overexpression of miR-21 in cardiac fibroblasts disrupts TGF-β signaling pathway by reducing the expression of Smad7, which promotes the proliferation and differentiation of cardiac fibroblast, and finally regulates cardiac remodeling after myocardial infarction.

    Effect of 1,25-dihydroxyvitamin D3 on alleviating the stress response in severely scalded mice

    Objective To explore the effect of 1,25-dihydroxyvitamin D3 on insulin resistance and inflammatory response in mice with severe burns under stress. Methods 130 mice were randomly divided into: the healthy group (n = 10), the experimental group I (n = 40), the experimental group II (n = 40), the control group (n = 40). Each mouse was scalded in the experimental group I ,the experimental group II and control group to 30% of the total body surface area (TBSA) III °. Mice in experimental group I received 1,25-(0H)2VitD3 1μg?kg-1 with 0.6 mL peanut oil by gavaging at 8 o'clock every other day.At the same time and by the same way, mice in experimental group II received 1,25-(0H)2VitD3 4μg?kg-1 with 0.6 mL peanut oil,and mice in control group received only 0.6mL peanut oil. FBG, FIns, serum TNF-α concentrations and wound tissue NF-κB positive rate were determined respectively on day 1, day3, day7 and day 14 after burn . Results (1) At each time point of the experimental groupⅠ, the experimental groupⅡand control group, the means of HOMA-IR , Serum TNF-α concentrations and wound tissue NF-κB positive rate were higher than those in healthy group. (2) The means of HOMA-IR of scalded mice in experimental group Ⅰand experimental groupⅡ were lower than those in control group at the same time, and the means of experimental group Ⅱ were lower than those in the experimental group Ⅰ (P <0.05); at the different time points ,the means of HOMA-IR of scalded mice in same group were the highest at day 3, and then gradually decreased (P <0.05). (3) The means of Serum TNF-α concentrations and wound tissue NF-κB positive rate of scalded mice in experimental group Ⅰand experimental groupⅡ were lower than those in control group at the same time, and the means of experimental group Ⅱ were lower than those in the experimental group Ⅰ (P <0.05); at the different time points ,the means of Serum TNF-α concentrations and wound tissue NF-κB positive rate of scalded mice in same group were gradually decreased (P <0.05). Conclusion 1,25-dihydroxyvitamin D3 can reduce insulin resistance and inflammatory response in mice with severe burns under stress.

    Abstract: Objective To investigate the effect of microRNA-1 (miR-1) on H9C2 cardiomyocytes. Methods MiR-1 mimic was transfected into the cultured H9C2 cell line was defined as miR-1 group. Cells transfected with random miRNA fragement was negative control group. Normal control cells had no treatment. The MTT and flow cytometry method were used to test cell vitality and apoptosis rate,while the mRNA and protein expression level of Bcl-2 were detected by real time PCR and western blot methods. Results Compared with the normal control group, negative control cells had no changes. The application of miR-1 mimic can significantly increase the miR-1 level and apoptosis rate, reduce cell vitality and Bcl-2 expression level. Conclusion miR-1 mimic can up-regulate miR-1 level, inhbit proliferation and induce cardiomyocyte apoptosis .

    Clinical significance of change of plasma protease C1 inhibitor in Latent autoimmune diabetes in adults

    Objective To examine levels of Plasma protease C1 inhibitor (SERPING1)of patients with latent autoimmune diabetes in adults(LADA)and explore their clinical significance. Methods The levels of SERPING1 in plasma were detected and compared among LADA, type 1 diabetes (T1DM), type 2 diabetes (T2DM) and healthy people. The correlation between the levels of SERPING1 in plasma and other clinical indicators such as age, disease course, glycosylated hemoglobin(HbA1c), fasting blood glucose (FPG), 2h postprandial plasma glucose(2hPG), Fasting c-peptide(FCP), 2h postprandial C peptide(2hCP), and Glutamic Acid Decarboxylase Antibody(GADA)were analyzed. Multi-factor regression analysis and receiver operating characteristic (ROC) were used to evaluate the predictive effect of SERPING1 in LADA at the early stage. Results The level of SERPING1 in LADA was significantly higher than that in the other groups except T1DM(P<0.05). There was remarkable relation between SERPING1 and FCP、HbA1c、FPG、2hPG, while no significant correlation was found between SERPING1 and the other clinical indicators(P>0.05).FCP was involed in the regression equation(P<0.05),and it was the main influences on the levels of plasma SERPING1. ROC indicated that it would be not enough to estimate LADA by SERPING1 alone. SERPING1 along with other indicators would great improve the diagnostic efficiency. Conclusions SERPING1 along with other indicators will be useful for identifying LADA from T2DM at the early stage.

    Objective To improve the detection sensitivity of serum sIgE by improve the quality of shrimp coated antigen. Methods The shrimp protein extracts were prepared and those >55KD proteins were separated by Gel Filtration Chromatography. The separated protein components were separated by SDS–PAGE and using Western blot and ELISA to evaluate whether the detection sensitivity of serum sIgE could be improved by Purified antigen.Result Those >55KD proteins ccould be successfully separated by Gel Filtration Chromatography. The SDS-PAGE of those >55KD proteins showed 10 identifiable bands. Dot-pot immunoassay showed that >55KD proteins could be improved the density spots of the weak serum. Meanwhile, the result of ELISA showed that 92.9% detection value of serum sIgE increased by coating effctive antigen. Conclusions The sensitivity of detection for serum sIgE might be improved by effective protein components of shrimp as coated antigen.

    End-stage Liver Disease Score Model in The Evaluation of Chronic Liver Failure Application Effect of Artificial Liver Treatment

    Objective: To investigate the value of model for end -stage liver disease (MELD) score, MELD with incorporation of serum sodium (MELD-Na) score and integrated MELD (iMELD) score for evaluation of prognosis of chronic liver failure. Methods :A total of 159 consecutive patients with chronic liver failure were included in the study and divided into two groups (death group and survival group) according to the prognosis. The levels of total bilirubin (TBIL), serum creatinine (Cr), prothrombin time (PT), PT international normalized ratio (INR), Serum sodium (Na), age, MELD, MELD-Na and iMELD were calculated respectively and the comparative analysis was performed. Areas under the receiver operating characteristic curve (AUC-ROC) of MELD, MELD-Na and iMELD were used to assess the prognosis in patients with chronic liver failure. Results: The values of TBIL, INR, MELD, MELD-Na and iMELD were significantly higher in death group than those in survival group (P < 0.01). The serum level of Na+ was significantly lower in death group than that of survival group (P < 0.01). The mortality of liver failure was higher in patients with the increased scores of MELD, MELD-Na and iMELD. The area under curve (AUC) values generated by the ROC curves was no difference respectively (P > 0.05) for MELD score (AUC=0.691), MELD-Na score (AUC=0.690) and iMELD score (AUC=0.674) . The cut-off scores of three systems were 25.8 (MELD), 31.0 (MELD-Na) and 53.5 (iMELD) respectively, which could discriminate higher and lower mortality accurately. Conclusion: MELD, MELD-Na and iMELD score can predict liver failure patients undergoing artificial liver comprehensive medical treatment combined short-term clinical outcomes.

    Aggressive natural killer cell leukemia—one case report and review of literature.

    Objective To improve the diagnosis and therapy of aggressive natural killer cell leukemia. Methods The characteristics of aggressive NK cell leukemia and its treatment were discussed with review of literatures. Results After combination chemotherapy which contained l-asparaginase and methotrexate,the patient would have a good result.Conclusion As aggressive NK cell leukemia is rare, its diagnose should include clinical mainfestation,cell morphology,immunophenotype,gene rearrangement.Treatment with joint asparaginase and (or) methotrexate scheme is given prior to hematopoietic stem cell transplantation ,which may prolong survival period.

    Percutaneous transforaminal endoscopic spine system in lumbar discectomy for disc herniation

    Objective To analyze the value of percutaneous transforaminal endoscopic spine system (TESSYS) in lumbar discectomy for disc herniation. Methods Thirty one patients of lumbar disc herniation were treated with percutaneous transforaminal endoscopic discectomy and followed 6-12 months, included 27 men and 4 women, with a mean age of 39 years (range, 18-58 yrs). The involved level included L34 at 2 cases, L45 at 21 cases and L5S1 at 8 cases. Local anesthesia was used in all the patients, and targeted puncture was performed under fluoroscopic guidance. The foramen of involved level was enlarged gradually with four trephinations, and the working canal was introduced transforaminal into the canal. Then the herniation was exposed and removed with full endoscopic technique, including the loosen nucleus pulposus. The nerve root and thena was exposed and released adequately. Results The procedure was evenly carried out in 27 cases, with discetomy and nerve root release. For the first case of far lateral herniation and the second case, the nerve root was not exposed. Two cases converted to posterior microendoscopic discetomy, included one case of large herniation, and one woman who complained intraoperative pain. The patients could walk in 1-2 days after operation, with obvious relieve of leg pain. One recurrence at 2 weeks was treated conservatively, A the final follow-up, the visual analuge scale of leg pain decreased from 8.1±1.9 to 1.1±0.9, and the ODI decresed from 31±8 to 3.4±3. According to MacNab scale, excellent results were obtained in 25 cases and good results in 6 cases. Conclusion The percutaneous endoscopic TESSYS is a good technique for lumbar discectomy. Excellent or good results can be obtained with this minimal invasive technique, while a learning curve exists.

    【Abstract】 Objective: To investigate the soft tissue changes and the relationship with the hard tissue in the class III malocclusion adult patients treated with premolar extraction. Methods: Thirty-four adult patients with skeletal class III malocclusion who met the including criteria were included into this retrospective study. Lateral cephalographs of pre and post treatment were traced.Relevant measurements of pre and post treatment were analyzed. Results: After orthodontic treatment,the values of U1-SN(°) was descreased 5.22°, the values of L1-MP(°)was descreased 13.86°,the angle of MP/FH was increased 1.78°averagely.The SL line as a reference,the retraction of upper incisor was 2.41mm,the retraction of lower incisors was 6.49mm significantly;the retraction of upper lips was 1.34mm,the retraction of lower lips was 3.82mm,the values of Ns-Sn-Pos was increased 3.36°, the values of H angle was descreased 2.96°,after orthodontic treatment(P<0.05 or P<0.01). Relevant measurements of chin were non-significanty. The retraction of upper incisor was the only measurement positively correlated with upper lip retraction. The retraction of upper incisor and lower incisor were correlated with the lower lip retraction;the upper lip retraction was correlated with lower lip retraction. Conclusions:Soft tissue profile improved significantly after four premolars extraction.The changes of soft tissue profile was moderate correlated with hard tissue.

    Relationship between Ambulatory Arterial Stiffness Index and Target Organ Damage in Primary Hypertensive Patients

    Objective To investigate the correlative factors of ambulatory arterial stiffness index (AASI) and the relationship with target organ damage (TOD) in primary hypertensive patie- nts. Methods A total of 330 hypertensive inpatients were included in the study and divided into two groups according to the AASI:low AASI group (n=167) and high AASI group(n=163).AASI was calculated obtained from 24-hour ambulatory blood pressure monitor (ABPM). The clinical data were collected including general information ,the data of ABPM, coronary lesions,left ventricular mass index (LVMI), estimated glomerular filtration rate(eGFR) and ankle brachial index(ABI) to compare between the groups.Results The levels of age(64.91±9.70vs59.12± 10.00) ,the proportion of diabetes(33.8%vs14.8%), the proportion of non-dipper(65.6% vs43.7%), 24 hours pulse pressure (PP)(65.27±11.31 vs56.06±10.51),24 hours diastolic blood pressure standard deviation(DBPSD)( 9.64±2.47 vs8.31±2.31), LVMI(125.74±29.65 vs107.69±23.23), coronary lesions(1.78±1.10 vs1.27±1.07) and the proportion of periphreral vascular disease(27.3% vs16.4%) were higher in the high AASI group than those in the low group ( P<0.01) ,the levels of eGFR(85.31±20.31 vs99.67±17.76) were inverse.In the correlation analysis,AASI was positively associated with coronary lesions (r=0.235), LVMI(r=0.168), periphreral vascular disease(r= 0.167),negatively with eGFR(r=-0.187).In multiple linear regression analysis model,independent predictors of AASI were age,diabetes, PP, DBPSD,non-dipper.Conclusion AASI correlated with age, diabetes,PP and blood pressure variability, non-dipper. The higher level of AASI may relate to the development of TOD in primary hypertensive inpatients.

    [Abstract] Objective To discuss the clinical efficacy between colonoscopic metal stenting and ileus tube catheterization in the therapy for left colorectal cancer with acute obstruction. Methods Clinical data of 80 cases applied metal stents and ileus tube to treat with left colonic cancer of our hospital from august,2011 to december,2013 was retrospectively analyzed.The 80 cases who met the diagnostic criteria were devided into metal stent group (40 cases) and ileus tube group (40 cases). The two groups were carried on by colonoscopy and X- line. The metal stent group were treated by metal stent and the ileus tube group were treated by ileus tube. After obstruction disappear (7~10d), two treatment groups were all implemented colorectal cancer radicalresection and anastomosis.To compare the technical operation, improvement of obstruction and effect of surgical treatment. Results Technical comparison in two groups , The success rate of the metal stent group was 87.5%, the success rate of ileus tube group was 97.5%,P>0.05.Both of two groups did not appear developed complications.But the operation time and cost of metal stent group were more than ileus tube group. Obstruction relief in two groups, Obstruction of remission rate of metal stent group was 100%,the other group was 95%,P>0.05,while 2 cases in ileus tube group were emergency operated because of failure of obstruction relief.Besides, both the time of obstruction ease and the difference between c - reactive protein of metal stent group were superior to ileus tube group. After obstruction disappear, two treatment groups were all implemented colorectal cancer radicalresection and anastomosis, both of two groups did not appear anastomotic leakage,and there were not difference in operative time, infection rate and hospitalization time. Conclusion Both of two methods can treat with left colonic cancer safely and effectively,be implemented colorectal cancer radicalresection and anastomosis,and reduce the incidence of perioperative complications.But in Health Economics ,the ileus tube is better ,while in obstruction relief aspect, the metal stent is better.     【Abstract】Objective:To assess the relationships between bile reflux gastritis(BRG) and Helicobacter pylori infection in children. Methods: BRG group include 258 cases of BRG and control group include 1749 cases of mucosal inflammation without bile reflux in children.Underwent a comparative analysis of Helicobacter pylori infection rate on the two groups. According to disease classification in BRG, the 258 cases were divided into three groups and underwent a comparative analysis of Helicobacter pylori infection rate. According to different reflux indexing in BRG, the 258 cases were divided into three groups and underwent a comparative analysis of Helicobacter pylori infection rate. Results: Compared to control group,the infection rate of BRG group were higher. The infection rate of three groups according to different disease classification was not statistically significant. The infection rate of three groups according to reflux indexing was statistically significant. Conclusions: Helicobacter pylori infection is one of the causative factors leading BRG in children.And Helicobacter pylori infection increased the degree of bile refluxing.

    Comparative study of microscrew implant for orthodontic anchorage and extraoral arch anchorage in patients with maxillary protrusion

    [Abstract] Objective:To investigate the clinic application of the treatment of maxillary protrusion malocclusion with micro implant anchorage. Method : 32 patients with maxillary protrusion were divided into two groups randomly with 16 patients in each group, whose ages ranged from 18 to 25 years. All patients were treated with extraction. Micros crew palatal implant was used in the cases of experimental group as orthodontic anchorage,and traditional anchorage composed of extraoral arch used in the control cases. The cephalometric films before and after treatment were measured , Statistical methods were utilized to analyze the morphological changes of facial profile and hard tissues in both groups. Results:The  U1-NA(mm) and U1 – SN(o) average after treatment were less than that before treatment in the experimental group(P<0.01)and the controls group(P<0.01), and U1 - L1(mm) average after treatment were more than that before treatment(P<0.05)in the experimental group(P<0.05)and the controls group(P<0.05). The U6-PtPNS(mm) average after treatment were more than that before treatment(P<0.01)in the controls group Conclusion:Treatment of patients with maxillary protrusion need with strong anchorage, can be used implants palatal side micro screw type .

    [Abstract] Objective To explore the influence of different glass-fiber post lengths and the content of fiber on teeth fracture resistance, and investigate the optimal glass-fiber post length and the content of fiber when apply fiber post restorative treatment.Methods Forty-two intact single canal mandibular premolars were collected from orthodontic patients.All the teeth were endodontically treatment after cut off the crowns from the 2mm above cemento—enamel junction.Divided the mandibular premolars randomly into three experimental groups (A, B, C) 14teeth a group.The experimental groups were post lengths of 10mm(1onger than 1/1 clinical crown length),8mm(1/1 clinical crown length)and6mm (shorter than 1/1 clinical crown length) respectively with 3# fiber Post drill.Each experimental group was averagely divided into two subgroups (A1, A2, B1, B2, C1, C2 ).Group A1, B1,and C1were restored with the 42% glass fiber post, group A2, B2, C2 with 75% glass fiber post.And then,all of them were restored with resin cores and metal crowns.Fracture resistance was measured with universal testing machine and recorded the root fracture pressure data and the mode of all the teeth.Results Fracture resistance of group A of restoration was significantly lower than that of B group and C group(P<0.05).Fracture resistances of group B was significantly lower than that of C group (P<0.05).Conclusion Within a certain range, the longer the length of pile, tooth fracture resistance is greater when we use them to restore dental defects.The fracture resistance of fiber content75% is better than the fiber content42%.

    A Clinical Applicationg to Restoration of Short Coronal Cracked Molar with Cutting Titanium Inlay Crown

    Objective: To estimate the clinical effect of cutting titanium inlay crown in the restoration of short cracked molar. Methods: Cutting titanium inlay crown were used to restore 45 short cracked molars. The cases were followed up for 3 years. Results: The success rate of the restoration was 93.3% in total. The effects of other 42 cutting titanium inlay crowns were satisfactory, the marginal sealing and the retention of the inlay crowns were acceptable, the secondary caries and perirhizoclasia had not been found. Conclusion: Not only retention but also resistance of a short cracked molar can be improved by means of cutting titanium inlay crowns.

    In vitro Study of Four Endodontic Irrigation Protocols on Smear Layer Removal in Curved Canals

       [Abstract] Objective To research effective endodontic irrigation protocols on smear layer removal from curved canals.Methods Forty mandibular first molars with curved mesial root canal were selected,decoronated. Mesiobuccal root canal was instrumented to F3 using handed Protaper system,and randomly divided into four groups according to final irrigation protocols. They are group A:syringe irrigation;group B:#30K File agitation;group C:passive ultrasonic irrigation (PUI);group D:combination of #30K File agitation and PUI. Ultrasonic file was located within 0.5mm coronally from start of the curvature. All root canal were mesio-distally split,snicked at start of the curvature and 2mm apically from start of the curvature ,and divided into three sections.They are Ⅰ,Ⅱ,Ⅲfrom coronally to apically. Smear layer of each section of root canal was examined under scanning electron microscope(SEM). Result Group D presented small amounts of scattered or thin smear layers and obtaind the lowest score at each section of root canal wall. At sectionⅠandⅡ,there was not significant difference for smear layer score between Group D and Group C ,and they were both lower than Group A,B(P<0.05); At section Ⅲ, Group D showed lower score than Group A,B,C, while Group B,C were lower than Group A (P<0.05). Conclusion When ultrasonic file was located coronally from start of the curvature, to apply K File which was the same size as dimension of apical portion preparation to agitate and PUI can remove smear layer from curved canals efficiently.

    [Abstract] Objective To analyze the antihypertensive drug patterns of outpatients from one Grade 3A hospitals of TianJin and analyze the gap and deficiency comparing with the guidelines of china to improve the control rates of hypertension. Methods In this retrospective survey of a Grade 3A hospital ,A total of 154262 hypertension electronic prescriptions of outpatients were enrolled and varied by gender, age, seasons, drug kinds, high blood pressure level group for statistics and analysis, as the same time ,the common used patterns and the condition of combination therapy were counted. Results 1.Of the patient, most patients were receiving calcium channel blockers(52.3%), followed by angiotensin receptor blockers (34.0%), beta-blockers (25.9%), angiotensin- converting enzyme inhibitors (12.1%), fixed-dose combination(11.0%) and diuretics (1.4%).2. Combination therapy were less than monotherapy(43.9% vs 56.1%)and there were some prescriptions which the Guideline of China do not recommend (more than 4. 4%).3.The combination therapy of stage 3 hypertension was higher than stage 1or stage 2 hypertension(44.5% vs 37.7% P<0.01); The combinations rate of Cardiology was the highest(P<0.01); the elderly patients tend to use more combination therapy than non-eldly patients(P<0.01).3.The prescriptions in summer were lowest than other seasons either alone or in combinations (P <0.01).Conclusion The combination therapy and the diuretic use were insufficient. These findings indicate the existence of a gap between clinical practice and the Guideline.

    Relationship between TGFβ1 gene polymorphism and liver cirrhosis after hepatitis B virus infection: A meta-analysis in Chinese population

    Objective To evaluate the relationship between genetic polymorphism of transforming growth factorβ1 (TGFβ1) and susceptibility of liver cirrhosis after hepatitis B virus infection in Chinese population. Methods CBM, VIP, CNKI, Wanfang technological periodical full-text databases and Pubmed from set up to July, 2013 were electronically searched to identify case-control studies on the relationship between genetic polymorphism of TGF β1 promoter 509 site, codon 869 site and liver cirrhosis after hepatitis B virus infection. The data were quantitatively analyzed by RevMan 5.1 software after assessing the quality of included studies. Results 6 case-control studies were selected for Meta-analysis based on our inclusion and exclusion standards. The results of meta-analysis showed that the pooled OR value for liver cirrhosis among Chinese patients after hepatitis B virus infection with T allele of TGFβ1 gene at promoter 509 was 1.02 (95%CI: 0.67~1.54), the pooled OR values for patients with TT and CT genotype were 0.80 (95%CI: 0.36~1.78); Patients with C allele of TGFβ1 gene at codon 869 was 1.05 (95%CI: 0.69~1.62), the pooled OR values for patients with CC and CT genotype were 0.98 (95%CI: 0.48~2.00). No significant publication bias was found. Conclusion The genetic polymorphism of TGFβ1 at promoter 509 and codon 869 shows no association with susceptibility of liver cirrhosis after hepatitis B virus infection in Chinese population.

Ultrasound microcapsular contrast agent as a drug transporter is a new type of disease targeted treatment strategies. It has provided a high-targeted, an effective and a safety gene therapy for prostate cancer, and brings a new hope for late-stage prostate cancer patients. This article aims to summarize the recent progress in using the microbubble technology in prostate cancer.
