    Clinical Significance of Syk and HER2 Expression in Gastric Cancer

    Objective: To investigate the expressions of spleen tyrosine kinase protein (Syk) and the epidermal growth factor receptor 2 (HER2) protein in gastric cancer, and their correlation with the occurrence and development of gastric cancer. Methods: The expressions of Syk and HER2 protein were detected by immunohistochemical method in 55 samples of gastric cancer and 20 samples of cancer adjacent tissue. The different histological types, TNM staging, lymph node metastasis, serous infiltration, distant metastasis and expressions of Syk and HER2 proteins in stomach cancer tissue were analyzed. Results: The expressions of Syk and HER2 proteins were 21.8% (12/55) and 29.1% (16/55) in 55 patients with gastric cancer respectively. The expressions of Syk and HER2 proteins were 85.0% (17/20) and 0 in 20 samples of cancer adjacent tissue. There was a lower expression level of Syk protein in samples of gastric cancer than that of cancer adjacent tissue (P<0.01), but the expression level of HER2 protein was higher in samples of gastric cancer than that of cancer adjacent tissue (P<0.01). There were significant differences in expressions of Syk and HER2 proteins between different histological stages, TNM staging, lymph node metastasis, serous infiltration, distant metastasis (P<0.05), but there was no significant difference between age and gender. The more poor differentiation and deep infiltrating appear the more lymph node metastasis and distant metastasis, higher TNM stages and higher HER2 positive expression were. There was no Syk protein expression in the low differentiation of gastric cancer. Conclusion: Detection of Syk and HER2 protein expression can be used to assess the malignant biological behavior and the prognosis of gastric cancer, providing a theoretical target for therapy of gastric cancer.

    Construction and Expression of pEGFP-C2-hG3BP-Domain (1-5) Recombinant Eukaryotic Plasmids

    Objective:To construct eukaryotic green fluorescent protein (GFP) expressing recombinant plasmids, pEGFP-C2-hG3BP-domain (1~5), which contain domain (1~5) fragments of human G3BP. Methods:The genes of G3BP fragments were amplified by PCR from the recombinant pEGFP-C1-G3BP plasmid and inserted into pEGFP-C2 fluorescent expressing vector with EcoRI and BamHI sites. These recombinant pEGFP-C2-hG3BP-Domain(1~5) plasmids were transfected into HeLa cells and the expression of green fluorescent fusion proteins was examined by fluorescence microscope and Western blotting assay. Results:The domain (1~5) fragments of G3BP were sequenced correctly and detected in the products of the restriction single/double enzyme digestion. The green fluorescent fusion proteins were also detected in the transfected HeLa cell by fluorescence microscope and Western blotting assay. Conclusion: These recombinant eukaryotic plasmids of pEGFP- C2-G3BP (1~5) were constructed successfully and expressed effectively.

    Significance of Serum Markers in Early Diagnosis of

    Abstract Objective: To explore the importance of the early diagnostic value of serum β- chorionic gonadotrophin (HCG),progesterone (P),carcinoembryonic antigen 125 (CA125) and pregnancy-associated plasma protein (PAPP)-A in ectopic pregnancy. Methods: The serum levels of β-HCG, P, PAPP-A and CA125 were measured in 100 cases of women with atypical ectopic pregnancy, 100 cases of women with normal interuterine pregnancy (nIUP) and control group.Patients were subdivided into >7 weeks suppressed menstruation group and ≤7 weeks group. Data was analyzed by SPSS13.0. Results: The serum levels of CA125, PPAPP-A, P and β-HCG were significantly higher in interuterine pregnancy group than those of ectopic pregnancy group. In interuterine pregnancy group there was a lower serum level of β-HCG in ≤7 weeks suppressed menstruation group than that of >7 weeks group (P<0.001). There was a positive correlation between days of suppressed menstruation and the levels of β-HCG and PAPP-A. In ectopic pregnancy group there were no significant differences in the serum levels of β-HCG, PAPP-A, P and CA125 between ≤7 weeks suppressed menstruation group than that of >7 weeks group (P>0.05). There was no correlation between the 4 serum marker and the days of suppressed menstruation. The diagnostic critical value was CA125≤34.925U/mL, diagnostic sensitivity was CA125:80% and the specificity was CA125:73.2% in ectopic pregnancy group. The diagnostic critical value was PAPP-A≤5.405mU/L, diagnostic sensitivity was PAPP-A:76% and the specificity was PAPP-A:73.2% in ectopic pregnancy group. The diagnostic critical value was β-HCG≤4761.4mIU/mL,diagnostic sensitivity was β-HCG92% and the specificity was β-HCG:81.3% in ectopic pregnancy group. The diagnostic critical value was P≤17.21ng/mL,diagnostic sensitivity was P:96% and the specificity was P:100.0%. in ectopic pregnancy group. Conclusion: The serum levels of β-HCG,P, CA125 and PAPP - A can be used as diagnosis markers of ectopic pregnancy, in which the serum level of P or P combined with β-HCG was the best.

    The Correlative Study between CT Pathological Classification and CT Manifestations in Acute Pancreatitis Involved Subperitoneal Space of Lesser Omentum

    Abstract Objective: To explore the correlation of manifestations of multi-slice spiral CT (MSCT) and Balthazar CT classification in acute pancreatitis (AP) involved subperitoneal space of lesser omentum. Methods: MSCT images of fifty-eight patients with acute pancreatitis proved by surgery and/or clinical and laboratory findings were retrospectively analyzed. CT examinations were performed on a 64-MSCT scanner in 19 patients and on a 16-MSCT scanner in 16 patients. The automatic high pressure injector was used to inject contrast material through forearm vein. The scan scope was set between the level of diaphragm roof and pelvic floor. After multi-planar reconstruction, the MSCT findings of AP involved subperitoneal space of lesser omentum were observed and the Balthazar CT classification was recorded. Results:(1)In fifty-eight patients, four cases belonged to C grade, eighteen cases belonged to D grade and thirty-six cases belonged to E grade by Balthazar's criterion. Hepatoduodenal ligament was involved in forty-five cases, and hepatogastric ligament was involved in fifty cases.(2)The hepatoduodenal ligament and hepatogastric ligament involvement showed edema, thickening, fluid collection and density increasing.(3)There were significant difference between the incidence rate of hepatoduodenal ligament and hepatogastric ligament involvement and Balthazar CT classification (P<0. 05). Conclusion: MSCT can sufficiently demonstrate the details of acute pancreatitis involved subperitoneal space of the lesser omentum. The hepatoduodenal ligament and hepatogastric ligament involvement can reflect the severity of acute pancreatitis.

    Study on the Risk Factors of Metabolic Syndrome

    Abstract Objective: To study the risk factors of metabolic syndrome by the cases of Tianjin policemen within different ages. Methods: 1250 policemen were divided into MS group and non- MS group by their clinical dates, and the changes of the levels of liver function, renal, lipid, heart rate, WBC, PLT, FIB and ESR which were analyzed. The policemen have been divided into two groups as less than 40 years old and over 40 years old group to study the difference of the risk factors of MS by the two groups. Result: MS group on alanine aminotransferase(ALT), alkaline phosphatase(ALP), γ-glutamyl transferase(gamma GT), total protein(TP), albumin(ALB), globulin(GLB),uric acid (UA), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), heart rate (HR), WBC and FIB were significantly higher than non-MS group, however, the high-density lipoprotein cholesterol (HDL-C) levels were significantly lower than non-MS group, the difference was statistically significant (P<0.05). The risk factors of the policemen under the age of 40 are serum alanine aminotransferase, UA, FIB, and HR. while the risk factors beyond the age of 40 are serum total protein, UA, gamma GT, WBC, and HR. Conclusion: FIB is high-risk factors for MS under the age of 40.

    Multi-Factor Analysis of Peripheral Arterial Disease in Patients with Type 2 Diabetes

    Abstract Objective: To explore the risk factors for the severity of angiographic peripheral arterial disease (PAD) in patients with type 2 diabetes (T2DM). Methods: Four hundred and fifty-five hospitalized T2DM patients were divided into four groups according to color Doppler ultrasound of lower limb, including 109 patients without stenosis (control group), 113 patients with stenosis<30% (mild stenosis group), 118 patients with 30%≤stenosis<70% (moderate stenosis group) and 115 patients with stenosis>70% (severe stenosis group). The risk factors were analysed in subjects of groups. Results: There were significantly higher values of age, course of disease, body mass index (BMI), waist circumference (WC), waist-hip ratio (WHR), waist-height ratio (WHtR), triglyceride (TG), total cholesterol (TC), low-density lipoprotein (LDL-C), systolic blood pressure (SBP) and glycosylated hemoglobin (HbA1c) in patients of severe stenosis group than those of other groups (P<0.01 or P<0.05). The level of high-density lipoprotein (HDL-C) was lower in patients of moderate stenosis group and severe stenosis group than that of mild stenosis group and control group (P<0.01 or P<0.05). There was a positive correlation between peripheral arterial diseases of lower limb and WHtR, HbA1c, course of disease, age,LDL-C,WC and SBP (P<0.01), but a negative correlation between peripheral arterial diseases of lower limb and HDL-C. Logistic regression analysis showed that values of WHtR, HbA1c and LDL-C were independent risk factors for the severity of angiographic peripheral arterial disease (P<0.01 or P<0.05). The increased level of HDL-C was the protective factor for the arterial diseases of lower limb (P<0.05). Conclusion: WHtR might be an easy and efficient anthropometric index for prognosis of PAD in patients with T2DM.

    The Relationship between Serum Uric Acid and the Vascular Complications in Patients with Type 2 Diabetes Mellitus

    Abstract Objective: To investigate the relationship between serum uric acid (SUA) and vascular complications in patients of type 2 diabetes mellitus (T2DM), and the possible mechanism thereof. Methods: The collected clinical data of 1040 patients with T2DM were retrospectively reviewed. According to SUA levels, 1040 patients of T2DM were divided into hyperuricemia (HUA) group (SUA≥420 μmol/L, n=117) and normal uric acid (NUA) group (n=923). The differences in clinical indicators and the incidence of vascular complications were compared between patients of two groups. Results: (1) The incidence rates of diabetic nephropathy (DN), hypertension and cerebrovascular complications were significantly higher in patients of HUA group than those of NUA group (P<0.01). (2) There were significant differences in body mass index (BMI), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and hemoglobin A1C (HbA1c) between HUA group and NUA group (P<0.05). (3)The multivariable non-conditional Logistic analysis showed that the values of BMI, TG, HDL-C and HbA1c were influencing factors of SUA. Conclusion: The incidence of vascular complications was higher in T2DM patients with hyperuricemia. Therefore, much attention should be paid to the risk factor of SUA.

    Analysis on the Antimicrobial-Susceptibility of Clinical Isolates of Shigella and the Mechanism of Fluoroquinolone-Resistance

    Abstract Objectives: To investigate the antimicrobial resistance characteristics of shigella in Tianjin, and the mechanism of fluoroquinolone-resistance. Methods: A total of 119 clinical isolates of shigella identified in Tianjin from 2009 to 2010 were analysed by K-B disk diffusion method for their antimicrobial susceptibility. The gyrA and parC in fluoroquinolone-resistant stains and plasmid mediated quinolone resistant (PMQR) determinants (qnrA, qnrB, qnrS, aac(6’)-ib-cr and qepA) were amplified and sequenced. Conjugation test was performed for PMQR determinant positive strains. The antimicrobial-susceptibility of E.coli J53AZR was compared before and after conjugation test. Results: The susceptibility rate of 119 shigella strains to nalidixic acid was the lowest (1.72 %), followed was ampicillin (2.52 %) and sulfamethoxazole (3.36 %). There were significant differences in sensitive rates to fluoroquinolone between shigella sonnei and shigella flexneri. Five isolates of Shigella flexneri were resistant to fluoroquinolone, in which 2 isolates were resistant to ciprofloxacin, 2 isolates were resistant to levofloxacin and 4 to double mutations (gyrA83,87 and parC80).There were 3 PMQR positive strains, including 1 strain qnrS,2 strains of aac(6’)-ib-cr. E.coli J53AZR showed decreased susceptibility to many antimicrobial after conjugation. Conclusion: Shigella clinical isolates show high resistance rate to many antibiotic,but low resistance rate to fluoroquinolone (<5%). The mechanisms of fluoroquinolone resistance in shigella include both sense mutations at target enzyme in quinolone-resistance determining regions and harboring plasmid mediate quinolone-resistance gene.

    Real-Time Three-Dimensional Echocardiographic Assessment of Left Ventricular Asynchrony in Pediatrics

    Abstract Objective: To investigate the clinical value of real-time three-dimensional echocardiography (RT3DE) in assessment of left ventricular systolic asynchrony in children. Methods: RT3DE was used to detect 55 normal children (control group), 10 patients with dilated cardiomyopathy (DCM group) and 22 patients with Kawasaki disease (KD group). The 16-segment myocardial systolic volume reached the lowest maximal difference (Tmsv16-Dif) and the standard deviation (Tmsv16-SD) were measured. To Values of Tmsv16-Dif and Tmsv16-SD were expressed as a percentage of the duration of the cardiac cycle (Tmsv16-Dif %, Tmsv16-SD %) to allow comparisons between patients with significantly different heart rate. Results: In control group, there was no significant difference in Tmsv16-Dif, Tmsv16-SD, Tmsv16-Dif % and Tmsv16-SD % between male and female patients. There was a significant difference in Tmsv16-Dif and Tmsv16-SD between different age groups, but no significant difference in Tmsv16-Dif % and Tmsv16-SD % between different age groups. The values of Tmsv16-Dif, Tmsv16-SD, Tmsv16-Dif % and Tmsv16-SD % were significantly higher in KD and DCM groups than those of control. The values of Tmsv16-Dif, Tmsv16-SD, Tmsv16-Dif % and Tmsv16-SD% were negatively correlated with left ventricular ejection fraction (LVEF) (r= –0.583, –0.565, –0.487 and –0.567, P<0.01). In control group, the RT3DE segmental time-volume curves showed in order, but they were disorder and the peak time-point spread in KD and DCM groups. Conclusion: Tmsv16-Dif% and Tmsv16-SD% of RT3DE can quantify global left ventricular mechanical dyssynchrony. RT3DE is a feasible and accurate method for analysis of left ventricular function.

    The Protective Effect of Ambroxol on Lung Injury in Children during Cardiopulmonary Bypass

    Abstract Objective:To investigate the protective effect of ambroxol on lung injury in children during cardiopulmonary bypass. Methods:Forty patients aged 4-9, scheduled operation of atrial septal defect (asd)\ventricular septal defect (vsd), were randomly allocated into two groups, ambroxol group (group M, n=20) and control group (group C, n=20). Ambroxol (4.5mg/kg in 50 ml of saline) was injected before cardiopulmonary bypass (CPB) in group M. The equal volume of normal saline was infused in group C. Blood samples were taken to detect white blood cells (WBC) before operation, after anesthesia (T1), 10 min after administration (T2), 5 min after CPB (T3), 1 h after ICU admission (T4), 2 h after ICU admission (T5), before decannulation (T6). Results:There were significant differences in R factors and time effect between two groups (P<0.001), and there were significant differences in T5 and T6 between two groups (P<0.01), which reached the highest level in T4 in two groups and then showed a downward trend. There was no significant difference in lung compliance (CL) in T1 and T5 between two groups (P >0.05). There were significant differences in values of WBC between two groups (P<0.001). There was a significant interaction between processing factors and time effects (P<0.01). There were significant differences in the peak inspiratory pressure (pip) and plateau pressure (Plat) between two groups (P<0.01), which showed a significant interaction between processing and time effects (P<0.01). Conclusion:Ambroxol has the protective effect on lung injury induced by CPB.

    Effect of Montelukast on IL-8 and TNF-α Level in Induced Sputum and the Pulmonary Function in Patients with Acute Exacerbation of Chronic Obstructive Pulmonary Disease

    Abstract Objective: To study the effect of montelukast on IL-8 and TNF-α level in induced sputum and the pulmonary function in patients with acute exacerbation of chronic obstructive pulmonary disease (COPD). Methods: Forty patients, aged from 65 to 70 years, with COPD and 20 health subjects were included in the study. Patients were randomly assigned to treatment group (n=20) and control group (n=20). Patients in two groups were given the conventional treatment, including antibiotic therapy, oxygen therapy, expectorant and bronchodilator, but patients in treatment group were also given montelukast sodium 10 mg orally before sleeping for 2 weeks. The levels of IL-8, TNF-α and the pulmonary function were observed before and after treatment. Results: The levels of IL-8 and TNF-α were higher in COPD patients than those in health subjects before treatment. The values of forced expiratory volume in 1 second (FEV1) and FEV1/ fast vital capacity (FVC) % were lower in COPD patients than those in health subjects. After treatment, the levels of IL-8 and TNF-α were lower in treatment group than those in control group. The value of FEV1 was higher in treatment group than that in control group. Conclusion: Montelukast can relieve the inflammation of airway, and improve the pulmonary function in patients with COPD.

    The Clinical Study of Interventional and Biteplate Treatment on Anterior Disc Displacement without Reduction

    Abstract Objective: To investigate the clinical effect of biteplate and interventional treatment of sodium hyaluronate on anterior disc displacement without reduction. Methods: Ninety-six patients with Anterior disc displacement without reduction were divided into three groups (32 patients in each group),group A (biteplate treatment only), group B (interventional treatment only) and group C (combination of biteplate and interventional treatment). Three months after treatment,the pain index and maximal mouth opening were compared between groups. Results: After treatment, the maximal mouth opening was increased and the pain index was decreased in patients of three groups. The improvement was statistically significant (P<0.05 or P<0.01). The change of maximal mouth opening was bigger in combined group than that of biteplate treatment only and interventional treatment only group. The pain index was significantly lower in combined group and interventional treatment only group than that of biteplate treatment only group (P<0.001). Conclusions: The effect is obviously better in combination group than that of biteplate treatment only group and interventional treatment only group. The effect of sodium hyaluronate on alleviating pain is better than that of biteplate treatment.

    The Correction of Malocclusion of Old Maxilla Fracture with Le FortⅠOsteotomy

    Abstract objectives: To study the possibility of Le FortⅠosteotomy correction for old maxillary fracture and to evaluate the clinical efficacy. Methods: Twenty-seven patients with old maxilla fracture were treated by Le Fort I osteotomy based on pre- and postoperative cephalometric and preoperative simulation and prediction (STO)analysis with RIF. Model surgery was performed on dental casts preoperatively. The pre-and postoperative measurements were compared with each other. Results: All cases healed well and had satisfactory occlusal function and facial form. The values of maxillary protrusion (SNA),upper jaw protrusion distance (sn-g)and the cutting edge of central incisor to the palatal plane points (U1-pp) were decreased after operation than those of before operation (P<0.05),but the value of N-ANS was increased(P<0.05). The values of G-Sn (//HP) and Ls-EP(⊥EP) were significantly decreased(P<0.05) after operation than those of before operation, but Cm-Sn-Ls and G-Sn(⊥HP) were increased compared with those of before operation (P<0.05). There was no recurrence after 3 - 6 months follow-up. Conclusion: Reconstruction of the maxillary with Le Fort I osteotomy is an effective method to treat delayed panfacial fractures.

    Expression of the Human Leukocyte Antigen G and Interleukin-10 in Human Non-Small Cell Lung Cancer

    Abstract Objective: To investigate the expression of the human leukocyte antigen G (HLA-G) and interleukin-10 (IL-10) in non-small cell lung cancer (NSCLC),and the correlation between expressions of them. Methods: The expressions of HLA-G and IL-10 were detected using immunohistochemical method in surgically resected specimens from 38 patients with NSCLC (patient group) and normal lung tissues from 12 patients without lung cancer (control group). Results: There were 28 positive expressions of HLA-G (74%) and 30 IL-10 protein expressions (79%) in specimens of NSCLCs, respectively. A positive correlation was found between HLA-G expression and stages of TNM (rs=0.443, P<0.01). No significance correlation was found between IL-10 expression and clinicopathological factors. There was no HLA-G and IL-10 protein expression in normal lung tissues. Furthermore, IL-10 protein expression was demonstrated in 25 (93%) of 28 HLA-G positive cases. There was a positive correlation between them (rs=0.424, P< 0.01). Conclusion: HLA-G and IL-10 form a vicious autocrine circle of immune response abrogation in cancer to help tumor evade immune surveillance.

    The Value of E-Tracking Technology for Evaluating Carotid Artery Elasticity Function in Active Smokers

    Abstract Objective:To explore the value of E-tracking technology (ET) in evaluating carotid artery elasticity function in active smokers. Methods:One hundred and thirty-two active smokers and 49 normal nonsmoking persons (group A) were divided into 3 groups by cigarette-smoking index (CSI= the number of cigarette-smoked each day × the year of cigarette smoking), including group B (CSI<400), group C (CSI=401-800) and group D (CSI=801-1200). ET was used to detect the intima media thickness (IMT), internal diameter of systolic (Ds) and internal diameter of diastole (Dd). The values of stiffness parameters (β), pressure-strain elastic modulus (Ep), arterial compliance (AC) and pulse wave conducting velocity (PWVβ) were calculated and analysed in 4 groups. Results:There were no statistical significances in age, body mass index (BMI), heart rate (HR), systolic blood pressure (SBP), diastolic blood pressure (DBP), pulse pressure (PD), IMT, Ds, and Dd between group A and other 3 groups. There were significant differences in β, Ep, AC and PWVβ between group A and other 3 groups (P<0.01), except for the group A and group B (P>0.05). And there were significant differences in β, Ep, AC and PWVβ between group B, group C and group D(P<0.05 or P<0.01). The values of β, Eρ and PWVβ increased with the increase of CSI, but AC decreased with the increase of CSI. Conclusion: ET technique can be used to access the vessel endothelial dysfunction accurately in active smokers; therefore, the tendency of arteriosclerosis could be detected in early stage.

    Abstract Objective:To investigate the association between serum level of osteopontin (OPN), disease activity and organ damage of systemic lupus erythematosus (SLE). Methods:Six-three SLE patients and 20 healthy, age- and sex-matched controls were enrolled in this study. According to the systemic lupus erythematosus disease activity index (SLEDAI),patients were divided into renal damage group (0-8 score,n=32) and without renal damage group (≥9 score,n=31). According to the interstitial pulmonary fibrosis index, patients were divided into interstitial lung disease (ILD) group (n=22) and without ILD group (n=41). Patients were also divided into heart damage group (n=25) and without heart damage group (n=38) according to the electrocardiogram,echocardiogram and cardiac enzyme results. Based on hematological results, patients were divided into with cytopenia group (n=45) and without cytopenia group (n=18). The serum level of OPN was detected by enzyme-linked immunosorbent assay (ELISA) in patients of groups. Results:The serum level of OPN was significantly higher in SLE group than that of healthy controls (P<0.05 or P<0.01). The serum level of OPN was significantly higher in patients with renal damage, lung interstitial disease, heart damage and cytopenia than that of patients without organ involvement in SLE (P<0.01). The serum level of OPN was significantly positively correlated with SLEDAI in SLE patients (r=0.982,P<0.01). Conclusion:OPN plays a role in the pathogenesis of SLE and is related to the disease activity and organ damage. OPN may serve as an active disease laboratory marker of SLE.

    Study of mRNA expression of glucocorticoid receptor in children nephrotic syndrome

    Abstract Objective:To observe the mRNA expression levels of GRα and GRβ as well as changes of various clinical indicators during the treatment of children PNS to evaluate the efficacy of glucocorticoids (GC). Methods:20 healthy children were selected randomly as control group(N group), 55 PNS kid patients are randomly divided into prednisone group(A group,n =17), triamcinolone group(B group,n = 20), methylprednisolone group(C group,n=18). After corresponding GC application, collect blood at 0th, 4th and 8th weeks; test the mRNA expression levels of the GR subtypes in PBMCs by semi-quantitative RT-PCR; record the response to hormone therapy and make follow-up prognosis. Results:At 0th week, the GRα of PNS group is lower than N group (P <0.05); the level of GRβis higher than N group (P <0.05). As the treatment time increases, statistically significant difference of GRαand GRβ were found amony three groups and time effects((P <0.05 or P<0.01).Differences in interaction between groups and time effect were only significant in level of GRβ(P <0.05).There were 6 cases of PNS children steroid-resistance; As the treatment continues, there were statistically significant difference of GRα and GRβbetween two groups and time effects(P < 0.05 or P < 0.01).Differences in interaction between groups and time effect were only significant in level of GRβ(P <0.05). Conclusion : Abnormities are found in the mRNA expression of GRα and GRβ of PNS children. The expression level of GRα and GRβ may evaluate GC effects and the steroid sensitive of PNS children at 8th weeks. The effect of methylprednisolone is best in the way of GR regulation.

    Chronic Intermittent Hypobaric Hypoxia on Inhibitory Effects of Paeonol in Ileum Spontaneous Contraction of Rats

    Abstract Objective: To investigate the effect of chronic intermittent hypobaric hypoxia (CIHH) on spontaneous contraction of rat ileum inhibited by paeonol in vitro, and the mechanism thereof. Methods: Sprague-Dawlay (SD) rats were randomly divided into normal oxygen group (CON) and CIHH treatment group (CIHH). Rats of CIHH group were exposed to hypoxia in a hypobaric chamber simulating 5000m altitude, 6 hours daily for 28 days. CON animals lived in the same environment as CIHH animals except hypoxia. Samples of rat ileum were prepared routinely. The ileum contractions were recorded using organ bath technique. Results: (1) Paeonol significantly decreased the amplitude, frequency, tension and activity of the ileam contraction in a dose-dependent manner both in CON and CIHH groups. With activity as criteria, IC50 was 65.13 mg/L and 135.74 mg/L. (2) CIHH significantly attenuated the inhibitory effects of Paeonol. (3) Paeonol inhibited acetylcholine-induced contraction of extracellular and intracellular calcium significantly in CON group. Paeonol inhibited the contraction of store-operated calcium channel (ROCC) and voltage-operated calcium channel. The inhibitory effect of Paeonol can be partly blocked by L-NAME, indomethacin and propranolol. (4) The acetylcholine-induced intracellular calcium release and ROCC-induced contraction were not blocked by Paeonol in CIHH, and which was not blocked by propranolol. Conclusion:CIHH reduced the inhibitory effects of Paeonol on rat ileum, which may be caused by the blocking of Paeonol induced ntracellular Ca2+ release, inhibiting of the ROCC and β- adrenergic receptor through CIHH.

    Abstract Objective: To investigate the effect of a disintegrin and metalloproteinase 17 (ADAM17)-siRNA on the invasion of human MCF-7 breast cancer cell by silence ADAM17 gene with siRNA. Methods: ADAM17-siRNA was transfected into MCF-7 breast cancer cells mediated by oligofectamine. ADAM17 mRNA and protein expressions were detected by real-time PCR and Western blot respectively, and the invasive ability of MCF-7 cells was studied by Transwell method. Results:ADAM17 mRNA and protein showed the high expression in MCF-7 cells, while it significantly decreased with ADAM17 siRNA treatment. Furthermore, the invasive ability of MCF-7 cells was markedly inhibited by ADAM17 siRNA. Conclusion:ADAM17 plays an important role in the invasion of MCF-7 breast cancer cells, and ADAM17-siRNA can inhibit the invasive ability of MCF-7 cell by blocking the expression of ADAM17.

    Effect of Maternal Lipopolysaccharide Stimulation on the Activation of Renin-Angiotensin System in New born Offspring of Rats HAO Xueqin College of Animal Science and Technology, Henan University of Science and Technology, Luoyang 471003, China Abstract Objective:To explore the effect of maternal exposure to lipopolysaccharide (LPS) on the activation of renin-angiotensin system in new born offspring of rats. Methods: Twelve Sprague-Dawley rats were randomly divided into two groups (n=6 for each grou),control group and LPS group. On the pregnant day 8, 10 and 12, rats of control group and LPS group were given intraperitoneally with normal saline 0.5 ml or LPS 0.79 mg/kg, respectively. The levels of renal rennin,angiotensin converting enzyme (ACE) mRNA and angiotensin Ⅱ type Ⅰ receptor(AT1-R) protein were detected by real-time PCR and Western blotting, respectively. The expression of angiotensin II in kidney was evaluated by immunohistochemical staining. Results:Compared with control group, the renin mRNA expression was significantly decreased,and the AT1-R protein expression was increased,in offspring of LPS group (P<0.01). There was no significant difference in ACE mRNA expression between two groups (P>0.05). The Angiotensin II-positive cells in kidney of LPS-treated offspring were lower compared with those of control group (P<0.01). Conclusion:Maternal lipopolysaccharide stimulation suppresses renal renin-angiotensin system in new born offspring of rats.

    Abstract Objective: To observed the Toll-like receptor 4 (TLR4) expression in the process of renal interstitial fibrosis (RIF), and therapeutically effect of fucoidan polysaccharide sulfate (FPS) thereof. Methods: Fifty-four rats were randomly divided into three groups,sham operation (SOR),unilateral ureteral ligation obstruction model group (UUO) and FPS treatment group (FPS). Rats were sacrificed in batches and specimens were collected at 2, 4 and 6 weeks after operation. The levels of serum creatinine (Scr), blood urea nitrogen (BUN) and HE stain were observed. The expressions of TLR4 and NF-ΚB in renal tissues were detected by immunohistochemical method. Results: There were no expressions or only slight expression of TLR4 and NF-ΚB in kidneys of SOR group. The renal fibrosis was found, and positive expressions of TLR4 and NF-ΚB in UUO group compared with that of SOR group (P<0.05). The expressions of TLR4 and NF-ΚB were gradually increased with extension of obstruction and the progress of the renal fibrosis. The expressions of TLR4 and NF-ΚB were decreased in FPS group than those of UUO group (P<0.05), which decreased levels of Scr, BUN and renal fibrosis. Conclusion: FBS alleviated renal fibrosis by regulating the expression of TLR4.

    Abnormal Serum Expression of the Angiopoietin-1/-2 in Aneurysmal Subarachnoid Hemorrhage

    Abstract Objective:To investigate the effect of the levels of serum Ang-1 and Ang-2 in aSAH. Methods: We carried out a free aSAH screening in Tianjin. Serum samples were collected as follows: 40 from normal subjects, 31 from patients with aSAH . All the patients were diagnosed by CT and 3D-DSA. Serum levels of Ang1, Ang2 and Tie-2 were tested and compared by enzymelinked immunosorbent assay (ELISA). Results: The levels of serum Ang-1 (t=-8.858, P=0.000), Ang-2 (t=-2.371, P=0.022), SBP (t=-8.844, P=0.000) and DBP (t=-5.316, P=0.000) of aSAH patients had significant higher levels , when compared with healthy subjects. The levels of serum Ang-1,Ang-2 and SBP in the patients of Hunt-Hess I~III were significantly lower than them of Hunt-Hess IV~V (Z=-3.928, P=0.000; Z=-3.657, P=0.000;Z=-2.127, P=0.034). Conclusions: These results suggest that serum Ang1 and Ang2 levels are promising clinically informative biomarkers of disease severity in aSAH.

    【Abstract】 Objective To investigate the morbidity of pulmonary arterial hy- pertension (PAH) in systemic lupus erythematosus (SLE) on the basis of pulmonary arterial pressure screening, and seek relational clinical index with PAH. Methods They have ordinary color Doppler echocardiography examination. According to the diagnostic criteria of PAH, divide them into PAH group and non-PAH group. Do statistical analysis between two groups, and contrast with previous literature. Results ①The morbidity of PAH in SLE is high to 25.8%.②The occurrence of PAH has positive relativity with Renault phenomenon, multiple serous effusions and interstitial lung disease(r is 0.318,.416 and 0.267 respectively, P <0.05). ③There’s statistic significance in decreasing of ALB, C3,PLT and increasing of GLO, IgG, CIC and RF in two groups(P <0.05). Conclusion SLE patients should have pulmonary arterial pressure general measure when initially diagnosed. The morbidity of PAH is obviously increased. When accompanied by Renault phenomenon, multiple serous effusions and interstitial lung disease etc, the morbidity of PAH in SLE may raise.

    The diagnostic value of DWI in diagnosis of ischemic brain lesions

    Abstract: Objective To explore the diffusion-weighted imaging (DWI) and apparent dispersion coefficient (ADC) value on the diagnostic value of brain ischemic infarction. Methods 78 cases of ischemic cerebral infarction cases according to the time, all groups doing MR T1WI, T2WI scan and DWI check, drawing ROI in the lesion and calculate ADC values through the software. Results The ischemic cerebral infarction lesions of hyperacute phase on conventional MRI examination showed no abnormalities, and DWI showed high signal, the average ADC values decreased to 0.512±0.07×10-3mm2/s, lower than that of normal brain tissue. The lesions of acute phase DWI showed high signal, the average ADC values decreased to a minimum of 0.423 ±0.13 ×10-3mm2/s. The lesions of subacute stage DWI showed low signal, average ADC values increased to 0.781±0.05×10-3mm2/s, close to the normal brain tissue. The lesions of chronic phase DWI showed obviously low signal, the average ADC values increased to 3.238±0.35×10-3mm2/s, was significantly higher than the brain tissue close to the cerebrospinal fluid. Conclusion DWI can effectively shows the hyperacute phase and the acute phase of ischemic cerebral infarction lesions, not only can be used for diagnosis, can also be used for the difference between the new and the old infarction, judge development period of disease.

    Objective To study the possible relationship between genetic polymorphisms of interferon and early response of interferon alpha Methods Clinical data from 145 patients in the original trial were analyzed. The SNP of INF-r gene intron at position1 +874 was determined by the restriction fragment length polymorphism analysis.all patients recieve interferon alpha treatment Results AA genetype account for 77.3% of early non-response patients, account for 42.4% of early part response patients, account for 43.1% of early complete response patients.AA genetype is associated with a lower early response rate compared with the other two genetypes. Conclusion There was association between IFN-r gene polymorphism and the early response of interferon alpha treatment in CHB patients. This study suggested the possibility that IFN-r gene polymorphism(intron 1 at position + 874) might be important in determining an individual’s efficacy of interferon alpha by higer expression or promting effect of host target cells

    Objective To investigate the relation of cellular immunological and humoral immunological indexs of the different iodine intake GD cases . Methods The clinical diagnosis GD cases were undertaked thyroid fine needle Cytology examination. Patients in GD group are divided into GD groupⅠand GD groupⅡ based on urinary iodine level. Detect the blood levels of FT3,FT4,TSH,TGAb,TPOAb,TRAb,TNF-α. Analyze the difference and dependablity of cellular immunological indexs and humoral immunological indexs of the different iodine intake GD cases .Results The positive rates of TGAb, TPOAb GDⅠgroup were higher than GDⅡgroup (P <0.05); The positive rates of TRAb GDⅠgroup were higher than GDⅡgroup (P >0.05); the TNF-α level in GDⅠgroup were higher than GD Ⅱ group, but the difference was not statistically significant (P <0.05). It was demonstrated by correlated analysis that FT3 was linear positive correlation with FT4、TRAb、TPOAb.,TNF-α was linear positive correlation with TRAb and TPOAb. Conclusion Iodine is important to development of Graves disease,it should be limited in patients with iodine excess.

    Side effect of capreomycin regimen on multi-drug resistant tuberculosis continuity for six monthes

    【Abstract】 Objective: To observe the side effect of capreomycin(CPM) and the safety of using CPM continuty for six monthes.Methods: Investigate the side effect of 20 multi-drug resistant tuberculosis patients who had used CPM continuity for six monthes.Results:All of the 20 patients had side effects,the rat was 100%. Frequency of the side effects was seleroma,hypokalemia,renal injury,Ⅷ cranial nerve's injury, Phlebitis.The side effects frequently appeared after 2-4 months of treatment. After rapid treatment and active observation ,all of them had the treatment for six monthes,and none of them had any side effects on the next six monthes.Conclusion:The side effects appeared commonly of the patients who had used CPM continuity for six monthes,and the hypokalemia was more serious.But capreomycin is a safe anti- tuberculosis drug for multi-drug resistant tuberculosis(MDR-TB).

    [Abstract]Objective To investigate the association between anterior ischemic optic neuropathy(AION) and lacunar infarction .Methods Review 79 eyes checked with visual sight,ocular fundus ,visual field ,electro-physiology,fundus fluorescein angiography and CT 0n 53 patients diagnosed as AION ,whom were treated in –patient department from mar.2007to jun.2010.Results All patients were diagnosed as AION.Among 53 patients ,47 of whom were checked with CT or MRI ,while 16 patients were diagnosed as lacunar infarction,the percent to 47 is 34%.Among 53 patients,43.4% which was 23 patients were with high blood pressure; 26.4% which was 14 patients were diabetics; Among those who diagnosed as lacunar infarction ,unilateral AION were 10 patients ,bilateral AION were 6 patients. Conclusions There are strong relativity between lacunar infarction and AION.All the patients with AION have to check with CT or MRI regularly.

    Study on Therapeutic Effect in Preventing Tubal Obstruction after Pelvic Inflammatory Disease

    Objective:To study the efficacy of anti-adhesion agent used in preventing tubal obstruction after acute Pelvic Inflammatory Disease(PID).Methods:The cervical and vaginal secretions of the patients with acute PID were collected for bacteria culture and test. All the patients received broad spectrum antibiotics. The patients recovered from acute PID were randomly divided into two groups, study group(n=30)and control group(n=30). The patients in the study group were given hydrotubation with regular drug and sodium hyaluronate for two times after menstrual period. The patients in each group were followed up on tubal patency exam after 1 month. Results:(1) In the PID patients the major etiological agents were eolibacillus, staphylococcus epidermidis, mycoplasma huminis (MH) and chlamydia trachomatis (CT). The combined infection rate was 64.8%,and 54.9% patients were concurrent infected with bacterial vaginosis(BV). (2) The tubal patency rate of study group after 1 month of treatment with sodium hyaluronate was 83.3%,which was significantly higher than 53.3% before treatment(P<0.05). After 1 month the tubal patency rate of study group treated with sodium hyaluronate was 83.3% which was significantly higher than 53.3% in the control group with statistical difference(P<0.05). Conclusions:(1)The etiological agents of PID always were combined infection. (2) Anti-adhesion agent could be effective to prevent tubal obstruction after PID.

    [Abstract] Objective:To investigate the distribution feature of malignant tumor in Yanbian Korean Minority National area.Methods :According to International Classification Diseases (ICD-9/ICD-10 ) , We analyzed a 9 -year (2001 to 2009) review of 8581 persons(8545 persons of Han and Korean minority nationality,36 persons of other nationalities )in the Affiliated Hospital of Yanbian University between the Korean minority nationality and Han. The standardized incidence rate in the period were caculated。Results: Variations in malignant tumor incidence patterns have been observed through different ethnicities as well as between males and females. Bing a male increased the risk of most types of cancer,and the prevalence rate of Korean minority nationality was significantly higher than that of in Han,Lung cancer and live cancer were identified with the highest incidence rates in Han and Korean minority nationality respectively among all malignant tumor. Conclusions: The main factors of compositionof malignant tumor and sexual difference in Yanbian are: Different dietary patterns 、physiological character and living habit.

    Abstract: Purpose To analyze autogenous bone graft harvested from circular saw plus plate internal fixation treated single segment cervical spondylotic myelopathy. Method Retrospective study of 40 cases form 2001-3 to 2010-3 in our hospital. Results All patiens were followed up for (28.6±3.8) months. The average operative time was 90min; the average blood loss was 80ml; the average hospital day was 8.4 days; the average preoperative JOA score was 10.2; the average postoperative JOA score was 16.4; fitness rate was 92.5%; the average postoperative improvement rate was 91.5%. Conclusions Maybe it’s a safe and effective approach using autogenous bone graft harvested from circular saw plus plate internal fixation for single segment cervical spondylotic myelopathy..

    Image diagnosis and analysis of esophageal leiomyoma

    Abstract:objective: We analyse 12 cases of typical esophageal leiomyoma in order to improve the understanding for it. Method: There are 12 cases of esophageal leiomyoma which have been proved by operation and pathological examination from 1997-2007. Results: In the 12 cases, 7 are males, 5 are females. The average age is 39. The course of disease is from 3 months to 3 years. All patients have the foreign matter sense after eating and the pain behind sternum. One of them occurred in the ingress of esophagus, 9 in the middle-under part, one in the upside, one in the underpart. The sizes of esophageal leiomyoma are from 0.5×0.5×l.0cm to18×3.0×2.0cm. The shape of esophageal leiomyoma is round and half-round for 10 cases , one looks like dumbbell, one case has pediculus. Conclusion: Most cases have the typical symptoms of the esophageal benign tumor, but some special cases need careful observation, for example, the esophageal leiomyoma with pediculus and the esophageal leiomyoma occurred in the ingress.
