    The Expression and Clinical Significance of DKK1, SFRP4and Wnt1in Cervical Squamous Cell Carcinoma

     [Abstract]   Objective  To investigate the expressions of DKK1,SFRP4and Wnt1in cervical squamous cell carcino-ma(SCC), and the clinical significance thereof.   Methods   There were76samples of cervical squamous cell carcinoma were included in SCC group and 36benign uterine resection specimens were control group (NC). The immunohistochemical method was applied to detect the expressions of DKK1,SFRP4and Wnt1in two groups.    Results   The expression of DKK1was significantly lower in SCC group than that in NC group (P<0.05). The expression levels of SFRP4and Wnt1were significantly higher in SCC group than those of NC group (P<0.05). There were significant differences in the expressions of DKK1,SFRP4and Wnt1between samples of different clinical staging, differentiation, sizes of tumor and lymph node metastasis (P<0.05). The expression of DKK1was negatively correlated with SFRP4and Wnt1in SCC group (P<0.05). The expression of SFRP4was positively correlated with Wnt1in SCC group (P<0.05).    Conclusion     The roles of SFRP4and Wnt1are synergistic interactions in the development of SCC. DKK1is an inhibiting factor of SCC

    [Abstract]   Objective  To analyzeKlebsiella pneumoniaeby DiversiLab system, providing scientific evidence for the
    control of nosocomial infection.  Methods  Eight strains of non-duplicated clinicalKlebsiella pneumoniaeisolated from the surgical ward in a hospital in2010were typed by rep-PCR-based DiversiLab system. The results were compared with those of pulsed-field gel electrophoresis (PFGE).   Results   Antimicrobial susceptibility test showed that7clinical strains were the same sensitivity to 11antimicrobial agents, except the strain K8-02. And these 7clinical strains were all multi-drug resistant. The result of PFGE showed that7strains were the same pattern. The result of DiversiLab also showed that7strains were the same pattern, and the strain K8-02was another pattern.   Conclusion   DiversiLab system is simple, quick, good repeatability and accurate, which is a kind of standardization and automation system. DiversiLab system and PFGE method could get the same result.

    The Expression and Significance of CRISP2Gene in Different Types of Spermatogenic Failure

    [Abstract]   Objective  To study sperm mRNA transcript expression of CRISP2gene in the semen of patients with
    different types of spermatogenetic failure. Methods  A total of150male infertility patients were divided into normal group,asthenozoospermia group and oligozoospermia group(n=50for each group). Total RNA was extracted from sperm cells. Re-al-time quantitative reverse transcription polymerase chain reaction (qRT- PCR) method was used to detect CRISP2gene mRNA levels in three groups.   Results  There was a significantly higher expression of CRISP2in normal group (10.281±2.173) than that of asthenozoospermia group (2.092±0.969, P<0.05). There was no significant difference in the expression of CRISP2between normal group and oligozoospermia group (9.420±2.794, P>0.05). The relative expression of CRISP2was significantly lower in asthenozoospermia group than that of oligozoospermia group (P<0.05).   Conclusion  The CRISP2 expression was significantly different in patients with different types of spermatogenic failure. The reduced expression of CRISP2may lead to the decreased sperm motility.

    Epidemiological Features and Relevant Risk Factors of Dry Eye among Police officers in Tianjin, China

    [Abstract] Objective To investigate the epidemiological features and relevant risk factors of dry eye among police officers in Tianjin. Methods Study subjects were selected through the stratified cluster sampling. After 1077 police officers in Tianjin, including 979 men and 98 women, filled out dry eye questionnaire, the dry eye tests were performed. The risk factors of dry eye were analyzed by the Logistic regression. Results The prevalence of dry eye was 13.74% among police officers in Tianjin. The prevalence of dry eye among policewomen (15.31%) is higher than that among policemen (13.59%). Relevant risk factors of dry eye included video display terminal use (OR=17.886), outdoor work (OR= 2.268) and female gender (OR = 1.493). Conclusion The prevalence of dry eye is high among police officers in Tianjin. Video display terminal use, outdoor work and female gender are risk factors for dry eye in police officers.

    [Abstract]   Objective   To investigate the effects of simvastatin on the expression of lectin-like oxidized low-density
    lipoprotein receptor-1(LOX-1) induced by oxidized low-density lipoprotein (ox-LDL) or Glucose in U937      macrophages  ,and explore the role of NF-κB in modulating of LOX-1expression.  Methods   U937macrophages were treated with PMA toinduce differentiation, which were co-cultured with 50mg/L ox-LDL or/and25mmol/L glucose. Pyrrolidine dithiocarbamate(PDTC) and simvastatin (1μmol/L or10μmol/L) were used to treat cells. The expression of LOX-1protein and NF-κB activity were detected by enzyme-linked immunosorbent assay technology. The expression of LOX-1mRNA was measured by RT-PCR.   Results   The expression of LOX-1was up regulated by ox-LDL, glucose and combination of both. The inhibitor of NF-κB PDTC suppressed this up-regulation. Simvastatin suppressed the expression of LOX-1induced by ox-LDL, and showed a significant effect in the higher concentration. There was no significant effect of simvastatin on the expression of LOX-1induced by glucose. The variation of NF-κB activity was similar to that of LOX-1expression.  Conclusion   Simvastatin suppressed the expression of LOX-1induced by ox-LDL, while no significant effect on the expression of LOX-1induced by glucose. The expression and regulation of LOX-1were related with NF-κB pathway.

    Expression and Phosphorylation of PKR and NF-κB p65in Cervical Lesions

    [Abstract]  Objective   To identify the significance of expression and phosphorylation of protein kinase R(PKR) and
    nuclear factor NF-κB p65in cervical lesions, and the effect of high-risk human papilloma virus(hsHPV) on expression and phosphorylation of R(PKR) and NF-κB p65.  Methods  A total of67patients with cervical cancer,149patients with cervical intraepithelial neoplasia (CINⅠ-Ⅲ) and15normal control were included in this study. The expression levels of PKR,phosphorylated PKR (p-PKR), NF-κB p65and phosphorylated NF-κB p65(p-NF-κB p65) were detected by immunohistochemical SP method in three groups.  Results  The positive expression rates of PKR and p-PKR in cytoplasm were significantly lower in hsHPV positive group than those in hsHPV negative group (27.2% and11.0%vs41.1% and21.1%,χ2=4.858and4.371,P<0.05). The positive expression rates of NF-κB p65and p-NF-κB p65in cytoplasm and nucleus were significantly higher in hsHPV positive group than those in hsHPV negative group (46.3% ,25.7% ,22.8% and12.5%vs32.6%,14.7%,11.6% and4.2%,χ2=4.345,4.048,4.729and 4.650 respectively,P<0.05). The positive expression rates of PKR in kytoplasm and karyon were significantly lower in NF-κB p65(+) group than those in NF-κB p65(-) group (25.5%vs38.0% and20.4%vs36.3%,χ2=3.898and4.396respectively, P<0.05). The positive expression rate of PKR in kyto?plasm was significantly lower in p-NF-κB p65(+) group than those in p-NF-κB p65(-) group (19.0%vs36.0%,χ2=4.462,P<0.05).  Conclusion  hsHPV may inhibit the expression and phosphorylation of PKR but promote the expression andphosphorylation of NF-κB p65. The expression and phosphorylation of NF-κB p65may inhibit the expression of PKR. Regulating effects of three may be associated with the generation and progression of cervical cancer.

    Timing of Invasive Mechanical Ventilation in Patients with Acute Left Heart Failure

    [Abstract]   Objective   To investigate the various occasions of invasive mechanical ventilation in patients with acute
    left heart failure.   Methods   Thirty-two patients with acute severe left heart failure were divided into two groups:17patients were treated with non- invasive mechanical ventilation (non- invasive mechanical ventilation group) and15patients were treated with invasive mechanical ventilation (invasive mechanical ventilation group). The respiratory rate (RR), arterial oxy?gen partial pressurep(O2), heart rate (HR) and mean arterial pressure (MAP) were observed after treatment for 0.5h,1h and3h in two groups of patients. The in-hospital mortality was compared between two groups.   Results   Ten patients of non-in?vasive mechanical ventilation group were in remission after0.5h,7cases without remission switched to endotracheal intubation for invasive mechanical. And 3of them were in remission in0.5~1h,4of them dead. Fourteen patients in invasive mechanical ventilation group were in remission after0.5h, one case was in remission after0.5~1h and no dead in this group.There were more patients in remission after0.5h in invasive mechanical ventilation group than those of non-invasive mechanical ventilation group (P=0.041). There were no significant differences in the number of patients in clinical remission after 0.5~1h and mortality between two groups. The values of RR and HR were decreased with the increasing treatment time in two groups. The values of p(O2) and MAP were increased with the increasing treatment time in two groups. There was a significantly lower RR at0.5h and1h in invasive mechanical ventilation group than that of non-invasive mechanical ventilation group (P<0.05). The values of p(O2) and MAP were significantly higher at 0.5h in invasive mechanical ventilation group than those of non-invasive mechanical ventilation group (P<0.05). The levels of HR at0.5h and3h were significantly lower in invasive mechanical ventilation group than those of non-invasive mechanical ventilation group (P<0.05).    Conclusion     Invasive mechanical ventilation should be used as soon as possible if non-invasive mechanical ventilation can not alleviate the symptoms in patients more than0.5hours.

    Correlation Analysis between EBV and Hp Infection and Gastric Cancer

    [Abstract]  Objective  To investigate the correlation between Epstein-Barrvirus (EBV) and H.pylori (Hp) infection
    and gastric cancer.  Methods  The EBV infection was examined by polymerase chain reaction (PCR) and in situ hybridization (ISH) in 100samples of gastric cancer tissue and82samples of gastritis tissue. The Hp infection was detected by PCR. The correlation between EBV and Hp infection and clinical and pathological features was analyzed in patients with gastric cancer.  Results  The positive rates of EBV and Hp were significantly higher in gastric cancer tissues than those in gastritis tissues (9.0%vs0and56.0%vs40.2%,P<0.05). There was significantly higher infection rate in EBV in cardia gastric can-cer than that of gastric cancer (16.3%vs2.0%,P<0.05). There were significantly higher infection rates in EBV and Hp in poorly differentiated carcinoma than those of well-differentiated carcinoma (15.7%vs2.0% and66.7%vs44.9%,P<0.05). There were no significant differences in EBV and Hp infection between gender, age, nationality and lymph node metastasis (P>0.05). There was no significant difference in Hp infection between different parts of gastric cancer. There was no correlation between EBV and Hp infection in gastric cancer tissues (r=0.137, P>0.05).  Conclusion  EBV and Hp infection aretwo independent factors in the development of gastric cancer, and both of them are associated with the malignant evolution of gastric cancer.

    Abstract: Objective To explore the reasons why critical hand,foot and mouth disease with pulmonary hemorrhage were cured successefully which was infected by Enterovirus 71,looking for the factors which influcing the prognosis of critical hand,foot and mouth disease. Methods A retrospective analysis of 28 critical hand,foot and mouth disease with pulmonary hemorrhage who were cured successefully was completed,which was about the treatment methods. Positive pressure ventilation had been used in all patients. According to the bedside EKG and UCG,dopamine and milrinone were used to improve circulation to those whose cardiac function weren’t obviously abnormal,dopamine and sodium nitroprussiate were used to improve circulation to those whose cardiac function were obviously abnormal,restriction of fluid volume,proper use of dehydrant,protection of cerebral cells,intravenous impulsion injection of glucocorticoid and γ-immunoglobulin,reasonable application of antibiotics,et al. Results 28 children were cured successfully,the days of hospitalization was 16-30,22 children had’t have any sequela in the near future,2children had the left upper limb with dropped muscle strength,2 children had the pair of lower limbs with dropped muscle strength,2 children had the left leg with dropped muscle strength and increased muscle tension. Conclusions The characteristic of critical hand,foot and mouth disease was acute onset,rapid progress. They were cured successfully because of the mechanical ventilation in first time,the reasonable used vasoactive drugs and comprehensive treatment. The damaged region of central nervous system is one of the most important factors which influencing the prognosis of hand,foot and mouth disease.

    Effects of MMP-3Gene Polymorphism in Restenosis after Percutaneous Coronary Interventions

    [Abstract]   Objective   To investigate the relationship between matrix metalloproteinase-3(MMP-3) gene promoter
    polymorphisms5A/6A and the restenosis after percutaneous coronary intervention (PCI).    Methods  A total of 437 patients with PCI were selected in this study. Patients were divided into mutant genotype group (5A/5A+5A/6A,n=136) and wild genotype group (6A/6A,n=301) according to MMP-3polymorphism. The angiography and clinic data were collected before and after coronary angiography in two groups of patients. The serum level MMP-3and genotype analysis were compared between two groups.    Results  There was no significant difference in the restenosis rate between two groups (42.2%vs33.1%,P>0.05). The restenosis degree was significantly higher in wild genotype group than that in mutant genotype group (56.28%±11.10%vs36.00%±10.17%,P<0.01). There was no significant difference in the serum level of MMP-3between two groups(13.38μg/L ±3.00μg/Lvs12.33μg/L±2.96μg/L,P>0.05). There was a higher restenosis rate in patients carrying 6A allele than that of patients carrying 5A allele (P<0.05). Carrying wild genotypes are risk factors for restenosis after PCI.   Conclusion   Patients carrying6A allele have significantly higher risk of resteonsis than patients carrying5A allele

    drug resistance and clinical uses analysis of pan-drug resistant Acinetobacter baumannii in the respiratory department

    Objective : To investigate the clinical medicine and drug resistance of pan-drug resistant Acinetobacter baumannii in the respiratory department in order to offer reference for the clinical therapeutics. Methods: The 283 strains of pan-drug resistant Acinetobacter baumannii were retrospective statistical analyzed about the result of sensitivity to antibiotics in the respiratory department from 2010 to 2012.The disk diffusion method was employed to detect the drug-resistance ratio. Results: Clinical infection of pan-drug resistant Acinetobacter baumannii had various degree of drug-resistance rate to fourteen different antibiotic agents,The highest rate of drug-resistance ratio was ceftazidime(93.4%)and the lowest of drug-resistance ratio to imipenem was 1.9%.Tne drug-resistance ratio of imipenem, cefoperazone/sulbactam, ampicillin/sulbactam , minocycline, amikacin and so on were low by comparision. Conclusion: Clinical infection of pan-drug resistant Acinetobacter baumannii had rather high drug-resistance rate to fourtheen different antibiotic agents in the respiratory department . More attention shoud be paid to provide theoretical and objective basis for clinical uses rationally.

    The Role of Expression of TGF-βand Smad Signaling Pathway andα-SMA in Patients with Diabetic Nephropathy

    [Abstract]  Objective  To investigate the mechanism of the pathogenesis of diabetic nephropathy (DN) bydetecting
    the expressions of transforming growth factor (TGF)-β, Smad proteins and α-smooth muscle actin (α-SMA) in kidney biopsyof patients with diabetic nephropathy (DN).   Methods  Twenty-eight patients with DN who underwent renal biopsy were col-lected as DN group. Ten subjects without DN who underwent nephrectomy were taken as control group. The expressions of TGF-β1, TGF-βRⅠ,TGF-βRⅡ,Smad2/3andα-SMA in renal tissues were detected by immunohistochemistry stain.  Results   (1)TGF-β1,TGF-βRⅠ,TGF-βRⅡ,and Smad2/3were expressed in the glomeruli and tubules of both DNgroup and control group, while the expressions of TGF-βand Smad proteins were significantly higher in DN group than those in control group. At the early stage of DN, TGF-βand Smad proteins were significantly expressed even though there was no remarkable lesions observed by light microscopy. There was no correlation between increased expression and the progression of DN. These proteins were not expressed after glomerulusand renal tubule fibrosis. (2) In control group, α-SMA was identified only in the vascular walls, glomerulus and renal tubules, while it was expressed in almost all parts of kidney in DN group.   Conclusion  TGF-βand Smad signals involved in the pathogenesis of DN, which may have similar pathogenesis with immune complex glomerulonephritis

    The Therapeutic Effects of the Cord Blood Stem Cell Transplantation in Patients with Severe Hepatitis

    [Abstract]   Objective   To study the effect of the cord blood stem cell transplantation therapy in patients with severe
    hepatitis.   Methods    Forty patients with severe hepatitis were randomly divided into control groups (n=20) and observed group (n=20). The patients in two groups were treated with the conventional liver protective therapy respectively, such as compound glycyrrhizin and glutathione. The patients in observed groups were performed with the cord blood stem cell trans?plantation therapy. After isolation and purification, the cells were transplanted into the liver via hepatic artery and then were transplanted via peripheral veins. The biochemical indices of albumin (ALB), prothrombin activity (PTA),alpha-fetoprotein (AFP)and clinical symptoms were detected at different times after therapy.   Results   After4-week therapy, the levels of PTA(%)and AFP(μg/L)were significantly increased in observed group compared with those of control group (53.56±5.15vs48.26±5.53,92.16±9.09vs43.57±11.33,P<0.01). After8-week therapy, the levels of alanine aminotransferase (ALT), totalbilirubin (TBIL) and AFP were significantly decreased in observed group than those of control group. The level of PTA was sig?nificantly higher in observed group than that of control group (62.61±4.35vs54.95±3.01,P<0.05orP<0.01). There were nosignificant differences in levels of ALB and blood ammonia between two groups (P>0.05). There were no serious complications and adverse reactions in two groups of patients. There was no significant difference in mortality between two groups (20%vs35%,χ2=0.818 P>0.05).   Conclusion   Cord blood stem cell therapy has a good effect on treating severe hepatitis, which is safe and no significant adverse reactions, and can be used as a new method for treating the severe hepatitis.

    [Abstract]  Objective   To investigate the relationship between phosphatidylinositol3- kinase catalytic subunitα
    (PIK3CA) expression and the incidence and different pathological grade of gastric cancer, and the mechanism thereof.   Methods   The expressions of PIK3CA, serine/threonine protein kinase (pAkt) and cell proliferation associated nuclear antigen(ki-67) in gastric carcinoma and adjacent tissues and normal gastric mucosa were detected by immunohistochemical method. The relationship between expressions of PIK3CA, pAkt and ki-67and tumorigenesis was analyzed. The expressions of PIK3CA, pAkt and ki-67in different pathological conditions of gastric tissues were analyzed. The relationship between tumor pathologic classification and differentiation were analyzed too. Results  There were significantly higher expressions of PIK3CA, pAkt and ki-67in gastric cancer (P<0.05), which were the highest in the poorly differentiated gastric adenocarcinoma (P<0.05). There were a positive correlation between expressions of PIK3CA, pAkt and ki-67and different pathological levels of gastric carcinoma and adjacent tissues and normal gastric tissues (P<0.05).  Conclusion   PIK3CA may be the initiating factor of PI3K/Akt signaling pathway, which induced phosphorylation of Akt and activation PI3K/Akt signaling pathway, promoting the proliferation, invasion and metastasis of gastric adenocarcinoma.

    Protective Effects of Panax Notoginseng on Retinal Ganglion Cells in Diabetic Rats

    [Abstract]   Objective   To explore protective effects and potential underlying mechanisms of panax notoginseng
    (PTS) on retinal ganglion cell (RGC) in diabetic rats.   Methods   SD rats were randomly divided into control group, diabetic group and treatment group. The diabetic model rats were induced by intraperitoneal injection of streptozotocin. Rats were given PTS50mg·kg-1·d-1in treatment group. One month later, the coexistence of nogo receptor and Brn3a (special marker of RGC) was observed by immunofluorescence staining double-labeled method. The expression of Nogo receptor was detectedby Western blot assay. The level of malondialdehyde (MDA) in retina was measured with detection kit. HE-staining was in?troduced to reveal the number of retinal RGC.   Results   A large number of Brn3a and Nogo receptors were co-existed in the retina. The Nogo receptor was exclusively expressed in RGC, which was up regulated in diabetic group compared with that ofcontrol group. The level of retinal MDA was increased and the number of RGC decreased in diabetic group than that of control group (P<0.001). Compared with diabetic group, there were decreased retina Nogo receptor, decreased level of MDA and increased number of RGC in treatment group (P<0.001).   Conclusion   PTS attenuates diabetes-induced loss of RGC,which may ascribe for down-regulation of retina Nogo receptor and decreased oxidative stress

Themodel of two-kidney one clip re?nal hypertensive rats was established. The tail arterial blood pressure was detected once a week. After20weeks, rats were sacrificed and specimens were collected. The left ventricular mass index (LVMI) was assessed. The myocardial ultrastructural changes were observed by electron microscope. Values of plasma renin activity (PRA), angiotensionⅡ (AngⅡ) and atrial natriuretic peptide (ANP) were measured by enzyme linked immunosorbent assay (ELISA).  Results   Compared with sham group, the levels of systolic blood pressure (SBP), LVMI, PRA, AngⅡ and ANP were significantly higher in2KIC group (P<0.01). Compared with2KIC group, values of SBP, LVMI, PRA and ANP were significantly lower in telmisartan group and amlodipine group (P<0.01), but the value of AngⅡwas significantly higher (P<0.01). The levels of SBP, LVMI, AngⅡand ANP were significantly lower in combined medication group than those of single drug medication group (P<0.01). There was no significant difference in the plasma PRA level between those groups (P>0.05). Results of myocardial electron microscopy showed that the left ventricular remodeling was significantly improved in combined treatment group.   Conclusion   Telmisartan and amlodipine can effectively improve the left ventricular remodeling induced by hypertension. There was more effective therapy using both medications together.

The expression of islet specific markers was tested by RT-PCR and immunofluorescence assay. RT-PCR/RFLP was used to test the imprinted genes IGF2parental expression in cells at different stages.   Results  Islet specific markers were expressed in differentiated cells, such as insulin, glucagon and C-peptide. PCR-RFLP showed that imprinted genesIGF2derived from embryonic stem cells were biallelic expression and loss of  imprinting . Conclusion   Gene imprinting status of IGF2was changed in differentiated cells in vitro.

    teins Bcl-2in CA1area of hippocampus in vascular dementia model rats. Methods  The experimental rats were randomly divided into sham operation group and vascular dementia model group. The model group was divided into five subgroups:1,2, 4, 8and12- w group (n=6for each group). The vascular dementia rat model was established by blocking four vessels.Learning and memory abilities were detected by Morris water maze. The expressions of Beclin-1and Bcl-2were detected by immunohistochemistry assay.   Results  The positive expressions of Beclin-1and Bcl-2were detected at1-w in CA1area of hippocampus in model rats, which reached the peak at4-w and began to decline in8-w, maintained the higher levels at12-w. There were significantly higher expressions of Beclin-1and Bcl-2at all-time points in model group than those of sham operational group (P<0.05orP<0.01). The positive expression of Beclin-1was positively correlated with Bcl-2at alltime points in CA1area of hippocampus in model group (r=0.809, P<0.05).   Conclusion   Results showed that autophagy and apoptosis occurred simultaneously and expressed the same trend in CA1area of hippocampus of vascular dementia model rats.

    Correlation Study of Metabolic Syndrom and Benign Prostatic Hyperplasia

    To explore the relationship between metabolic syndrome (MS) and benign prostatic hyperplasia (BPH) .Methods The patients of TURP from Octobour 2010 to September 2012 in our hospital were devided into two groups according to the definition of MS by IDF in 2005:MS group and without MS group. Data incloud age, body height, body weight, blood pressure, body mass index (BMI) and the international prostate symptom score (IPSS) were record. Fasting plasma glucose (FPG), TG, HDL-C, prostate specific atigen (PSA), prostatic volume (PV) and Qmax were measured and recorded. Results (1) The BMI , systolic blood pressure, diastolic pressure, fasting plasma glucose, triglyceride level were higher, while HDL-C level was lower in MS group than without MS group (all P<0.005) . (2) The PV and IPSS level were higher, while PSA and Qmax were lower in MS group than without MS group (all P<0.005) .Conclusions MS could increase IPSS and prostate volume,and reduce PSA. Obesity, hyperlipemia, and diabetes meuitus are risk factors of BPH.

    Risk factors of atrial fibrillation in patients with renal insufficiency

    Objective Analysis of hospitalized patients with atrial fibrillation (AF), renal dysfunction in patients with risk factors analysis. Methods Regression analysis of the clinical data of hospitalized patients(from January 2008 to December 2012 529 cases of patients) in the hospital diagnosis of atrial fibrillation , Atrial fibrillation whether renal insufficiency were divided into two groups, the clinical data were retrospectively analyzed to study possible risk factors for AF in patients with renal insufficiency. Results: Age, hypertension, heart failure and coronary heart disease history, the difference was statistically significant (P <0.05), while gender, smoking history, diabetes, rheumatic valvular disease, hyperthyroidism, cardiomyopathy, idiopathic AF, left atrial enlargement renal dysfunction incidence showed no significant difference (P> 0.05). Regression analysis: ≥ 60 years of age (P = 0.014, B = 0.036), hypertension (P = 0.024, B = 0.498), coronary heart disease (P = 0.034, B = 0.466), cardiac dysfunction (P = 0.000, B = 0.824). Conclusion: Age ≥ 60 years of age in patients with atrial fibrillation and renal dysfunction, hypertension, coronary heart disease and heart failure as the main factors, which elderly atrial fibrillation patients with renal insufficiency, pay attention to the control of their blood pressure and improved cardiac function,its secondary prevention of coronary heart disease in patients with coronary heart disease, is particularly important.

    Subjective and Objective Assessment of Quality of Life for Coblation-assisting Operation in Patients with Severe OSAHS

    [Abstract]   Objective   To evaluate the subjective and objective improvement of quality of life in patients with lowtemperature plasma- assisted modified uvulopalatopharyngoplasty (H- UPPP) combined with lymphoidectomy in radix lin?guae and coblation-channeling of the tongue(CCT) treating for severe obstructive sleep apnea hypopnea syndrome (OSAHS),and compare the surgical efficacy with that of H-UPPP.  Methods   A total of81patients with severe OSAHS were divided into treatment group (n=42) and control group (n=39). Treatment group underwent the low-temperature plasma-assisted HUPPP combining lymphoidectomy in radix linguae and CCT,and control group underwent H-UPPP treatment. The apnea hypopnea index (AHI), the lowest saturation of arterial oxygen (LSaO2) and Quebec sleep questionnaire (QSQ) and Epworthsleepiness scale (ESS) before operation and6-month after operation were recorded and compared between two groups.  Results   All of detection indicators were significantly improved after operation in treatment group (P<0.01). The total surgicalefficacy was significantly higher in treatment group than that of control group (83.3%vs12.8%,χ2=40.225,P<0.01). The total improvement rate of five dimensions in QSQ such as daytime sleepiness symptoms (76.2%vs25.6%), signs during the day(71.4%vs10.2%) and night (54.8%vs5.1%), emotion (54.8%vs2.5%), ability of social intercourse (50.0%vs2.5%) and thetotal score (57.1%vs7.7%) after operation were significantly higher in treatment group than those of control group. The severe indexes of ESS decreased to23.8% after operation in treatment group than those of control group (51.3%).   Conclusion    Low-temperature plasma-assisted H-UPPP combined with lymphoidectomy in radix linguae and CCT is effective in patients with severe OSAHS, which can also improve the quality of life postoperatively.

    [Abstract]    Objective  To explore the effects of cyclic tensile strain on the proliferation of cultured human osteoar -thritis chondrocytes.    Methods  The degenerated human cartilage came from a66-year-old patient with osteoarthritis. The degree of degeneration was assessed by pathological diagnosis. Under the sterile condition, degenerated chondrocytes werecultured by enzymatic digestion. The toluidine blue staining, safranin O staining, and immunofluorescence staining of colla-gen Ⅱ were used to identify the cultured cells. The third generation cells were seeded on the silicone rubber membrane carrier, using the EF3200mechanical tester equipped with BioDynamic bioreactor system, imposed a frequency of 0.25Hz, stain 0, 5%,10% and 15% load for 3h. The proliferation activity was detected by flow cytometry.  Results   The proliferative in-dex (PI) increased with the magnitude value of cyclic tensile strain.     Role of Kupffer Cells in High Fat Diet Induced Hepatic Insulin Resistance

    [Abstract]   Objective  To explore the role of Kupffer cells in high fat diet induced hepatic insulin resistance.      Methods   Eighty-four C57BL/6J mice were divided into two groups: normal chow (NC) group (7subgroups) and high fat diet(HFD) group (7subgroups). Glucose tolerance tests were performed at feeding time of1,2,4,8 ,  12and16weeks in one NC group and one HFD group. The mice livers at the same feeding time were obtained in other 6NC groups and 6HFD groups respectively. The expression levels of IRS1-ser307, Akt-ser473, S6K1-thr389and p-JNK were detected by Western blot as-say. The values of MCP-1,TNF-α,IL-1βand IL-10mRNA were examined by qRT-PCR.  Results  Compared with NC group, the impaired glucose tolerance was found from the first week in HFD group (P<0.05). The hepatic expressions of IRS1-ser307, S6K1-thr389(4-16weeks ) and p-JNK(2-16weeks ) increased and Akt-ser473(8-16 weeks)decreased in HFD group than those of NC group. The same results were gained by analysis of protein relative expression (allP<0.05). The hepatic pro-inflammatory factor MCP-1,TNF-αand IL-1βmRNA expressions were higher in HFD group than those in NC group during2-16weeks (allP<0.01). The anti- inflammatory factor IL-10mRNA was significantly lower in HFD group than that in NC group during4-16weeks (allP<0.01).   Conclusion   High fat diet maybe play a role in the hepatic insulin resistance by stimulating M1Kupffer cells to secrete pro-inflammatory factor and inhibiting M2Kupffer cells to secrete anti-inflammatory factor.

    [Abstract]   Objective   To investigate the effects of all- trans retinoic acid (ATRA)- intragastric- administration on
    the survival time of mouse skin allografts and the role of interleukin (IL)-23and IL-17thereof.   Methods    The skin trans-plantation of mice was done by DBA/2as donors and Balb/c as recipients. The recipients were divided randomly into three groups: control group, low-dose group and high-dose group. Mice of the corresponding groups were intragastrically administered corn oil, 10mg/kg ATRA and30mg/kg ATRA respectively from1day before the transplantation to the14th day after  the transplantation. The survival time of transplanted skin was observed after the operations. Skin grafts of mice were harvested for histopathological examination in three groups. The serum levels of IL-23and IL-17were measured by enzyme-linked immunosorbent assay (ELISA). The expression levels of IL-23, RORγt and IL-17mRNA in skin allografts were detected by real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (RT-PCR).   Results   Compared with control group, the average survival time of mouse skin allografts was significantly prolonged in low-dose group and high-dose group (P<0.05). The less lymphocyte infiltration and destruction of architecture were found in the skin biopsies. The serum expression of IL-23protein was lower (P<0.05), but no significant difference was found in two treatment groups. The serum expression levels of IL-17protein were reduced in turn in receptors of control group, low-dose group and high-dose group (P<0.05). The expression levels of IL-23, RORγt and IL-17mRNA in skin grafts were significantly lower in low-dose group and high-dose group than those of control group (P<0.05), but no significant difference was found in two treatment groups.   Conclusion   ATRA can effectively prolong the survival time of skin allografts, which may related with the inhibition of the expression of IL-23, RORγt and IL-17mRNA and the development of IL-23and IL-17protein

    apoptosis of hepatic stellate cells and the effects of H2S on expressions of collagen Ⅰand collagen Ⅲ mRNA in hepatic stel-late cells.  Methods   There were five groups of HSC-T6cells in this study includingcontrol group (DMEM medium containing10% fetal bovine serum), dimethyl sulfoxide (DMSO) group, sodium hydrosulfide (NaHS)group,SB203580(SB)group and SB + NaHS group. MTT method was used to detect the cell proliferation and inhibition rate of HSC- T6cells treated by SB203580and H2S. The apoptotic rate of HSC- T6cells was detected by flow cytometry with annexin V- FITC/PI double staining. RT-PCR was used to detect the expressions of collagen Ⅰand collagen Ⅲ mRNA in HSC-T6.   Results   The apoptotic rate of HSC-T6cells was significantly higher in SB group and SB+NaHS group than that of control group, and the rate was significantly higher in SB+NaHS group than that of SB group (P<0.05). There was no significant difference in the apoptotic rate of HSC-T6cells between DMSO and NaHS groups than that of control group. The expressions of collagenⅠand collagen Ⅲ mRNA were found in five groups of cells. There was a higher expression of collagen Ⅰand collagen Ⅲ mRNA in NaHS group than that of control group (P<0.05). The expressions of collagen Ⅰand collagen Ⅲ mRNA were significantly lower in SB group and SB+NaHS group than those of control group and NaHS group (P<0.05).   Conclusion     H2S activated P38MAPK signal pathway. And P38MAPK was specifically blocked by SB203580in HSC-T6cells, which inhibited the cell proliferation stimulated by H2S and promoted the apoptosis.

    [Abstract]   Objective  To investigate the effect of compound catechu anti- diarrhea ointment on the expression of
    aquaporin3(AQP3) in the colonic tissues of diarrhea rat model, and the mechanism thereof.   Methods   Forty SD rats were randomly divided into four groups: blank control group, model control group, positive control group and compound catechu anti-diarrhea ointment group. Rats were given6g blank matrix cream in model control group,2mL suspension of berberine in positive control group and compound catechu anti-diarrhea ointment in compound catechu anti-diarrhea ointment group, two times/d for 7d. The rat model of diarrhea was established by using senna intragastric administration. The water content of feces was measured. The expression of AQP3in colonic tissues was detected by immunohistochemistry assay.   Results   The water contents of feces were significantly higher in model control group (64.09±0.41) %than those of other three groups(F=53.879,P<0.05). There was no significant difference in the water content of feces between compound catechu anti-diarrhea ointment group (48.83±1.08)% and positive control group (46.87±2.19)%. The AQP3-positive cells were mainly expressed in the intestinal mucosa. The dyeing index was significantly lower in model control group (0.85±0.18) than that of other three groups (F=14.971,P<0.05). There was no significant difference in the dyeing index between compound catechu anti-diarrhea ointment group (1.30±0.18) and positive control group (1.37±0.14).    Conclusion   Compound catechu anti-diarrhea ointment can significantly reduce the water contents of feces, which may be related to the increased AQP3expression in colonic tissues.

    [Abstract]   Objective   To explore a novel quantitative detection method for the concentration of specific IgG (sIgG)
    in food intolerance, taking egg sIgG detection for the example.  Methods  A total of173patients underwent food allergen sIgG detection were included in this study, and 78healthy subjects were used as negative controls. The microtiter plates were coated with biotinylated bovine serum albumin (BSA) and linked with streptavidin. Then, the biotinylated egg antigen and specimen were successively added into the wells of plate.After washing, enzyme labeled anti-human IgG was added to establish an antigen indirectly coated liquid-phase reaction patterns of ELISA method. The concentration of the biotinylated allergens and enzyme labeled antibody were optimized and the reaction conditions were determined. This method was used to detect the sIgG in serum samples.  Results  The biotinylated egg white was selected as antigen, the optimal dilution rate was 1∶2000,and the most suitable enzyme labeled antibody dilution ratio was1∶12000in this method. The within-run and the between-run coefficients of variation were4.83%-8.55% and4.88%-7.93% respectively. The specificity is preferable, the species-crossed reaction rates with crab, cow milk and goat milk were <10%. There was a good correlation between the assay developed in this study and the food intolerance detection kit provided by United States BIOMERICA Inc ( =0.977X+8.45,r=0.961, P<0.05).   Conclusion   The detection method can be used to detect serum sIgG for egg intolerance patients with easy operation, highly accuracy and specificity. Furthermore, it showed the capacity of excellent repeatability and flexibility potential, which provided a good foundation for developing a kind of“personalized”random combination of food varieties ELISA kit.
