    Influence Of GDNF To The Expression Of Matrix Metalloproteinase 9 In Pancreatic Carcinoma

    Abstract: Objective To investigate the influence of Glial Cell Line–Derived Neurotrophic Factor(GDNF) to the expression of Matrix Metalloproteinase 9(MMP-9) in pancreatic carcinoma. Methods Make use of RT-PCR 、Western blot and Elisa to investigate the effect of GDNF to the expression of MMP-9 mRNA and protein in MIA PaCa-2 and Capan-2 human pancreatic carcinoma cell line. Results We found that GDNF can up-regulate the expressions of MMP-9 mRNA and its protein in a dose-dependent manner in RT-PCR and Elisa experiments. Conclusions GDNF has the effect to stimulate the expression of MMP-9 and thereby strengthen the anility of invasion in panereatic carcinoma. Key Words: Pancreatic Carcinoma; GDNF; MMP-9; Expression

    The Role of XPD in The Hepatoma Cells Growth and Its Relationship with ERG Gene

    Abstract Objective:To investigate the effects of transfected XPD gene on the growth of hepatoma cell SMMC-7721 and the expression of ERG gene. Methods: The XPD gene was transfected into SMMC-7721 with Lipofectamine 2000. There were four groups in this study including SMMC-7721 - pEGFP- N2-XPD group(XPD group), SMMC-7721-pEGFP-N2 group (N2 group), Lipofectamine group (Lip group ) and blank control group. Reverse transcriptase PCR and Western blot were used to measure the expressions of XPD and ERG at mRNA and protein levels,respectively.The cell prolifera tion was assessed by MTT assay and the cell apoptosis was examined by flow cytometry . Results: Compared with N2 group, Lip group and blank control group, the expression of EGR mRNA and protein decreased significantly in XPD group (P < 0.01), in contranst the expression of XPD mRNA and protein were enhanced obviously in XPD group (P < 0.01). The proliferation of SMMC-7721 cells was markedly inhibited and the apoptosis of SMMC-7721 cells was increased obviously after transfection of XPD(P < 0.01). Conclusion: The wild-type XPD could inhibit the expression of ERG ,decrease the proliferation of SMMC-7721 cells and enhanced the apoptosis of SMMC-7721 cells in vitro.

    [Abstract] Objective To explore the prevalence, cause and clinical features of Sj?gren’s Syndrome complicated with pulmonary arterial hypertension. Methods The data of 61 cases in my department with the diagnosis of pSS during November 2010 to April 2011 were collected. All the cases take ultrasonocardiograph. Results In this study, the prevalence of pSS-PAH is 16.4%. The occurrence of pulmonary arterial hypertension is associated with Raynaud’s phenomenon, dry cough, chest tightness or chest pain, progressive dyspnea, heart palpitations, interstitial lung disease or disease active. Conclusion pSS patients without clinical symptoms, they also should do UCG, which is important to early diagnosis and treatment.

    【Abstract】 Objective The aim of the study was to determine the function of matrix metalloproteinase-2,-9(MMP-2,-9)and matrix metalloproteinase-8(MMP-8)in carious development,comparison observations on MMP-2,-9 and -8 were made between healthy and carious teeth and to evaluate the function of MMP-2,-9 and -8 in carious development. Methods The expression of MMP-2,-9 and-8 were analyzed by enzymography, immunohistochemistry and the Western blot . Results MMPs were expressed in odontoblast of healthy and carious teeth, correlated with increased ? expression of MMP-2, MMP-9 and MMP-8. Conclusion The odontoblast is one of the source of MMP-2,-9 and-8.     Clinical significance of dyslipidemia in patients with systemic lupus erythematosus

    Objective: To investigate the changes of serum lipids in patients with systemic lupus erythematosus and the correlation between this change and the activity of SLE and renal involvement.Methods: Blood lipid levels were analyzed by routine chemistry. Disease activity was estimated using the Systemic lupus Erythematosis Disease Activity Index (SLEDAI). Anti-dsDNA antibody was measured by indirect immunofluorescence (IIF) and renal involvement in SLE was monitored. Results:The serum level of TG was higher in SLE group than control group, and the serum level of HDL was significantly lower in SLE group than control group, the difference was significant(P<0.01). The level of LDL in the two groups was not significant. The serum level of TG was higher significantly in active SLE group than inactive SLE group(P<0.01), and the level of HDL was lower significantly in active SLE group than inactive SLE group(P<0.05). The level of LDL in the two groups was not significant. TG was positively related to the SLEDAI score (r=0.53, P<0.05), and HDL was negatively related to SLEDAI score (r=-0.62, P<0.01). The level of TG was significantly elevated in positive anti-dsDNA group than negative anti-dsDNA group (P<0.01). The level of HDL was significantly lower in positive anti-dsDNA group than negative anti-dsDNA group (P<0.05). The level of LDL in the two groups was not significant.The level of TG was higher in SLE patients with renal involvement than without renal involvement, the level of HDL was lower in SLE patients with renal involvement than without renal involvement (P<0.01). The level of LDL in the two groups was not significant. Conclusion Dyslipoproteinemia with high levels of TG and low levels of HDL correlates with disease activity in patients with SLE. Also dyslipoproteinemia was related to renal manifestations in SLE, and thus it maybe serve as markers of more severe disease.

    The expression and clinical significance of IGF-1 and COX-2 in endometrial cancer

    Abstract Objective:With the aim to further investigate the etiology of endometrial cancer, tissue chip technology was used to evaluate the expression of Insulin-like growth factor-1(IGF-1) and cyclooxygenase-2(COX-2)in endometrial cancer tissues. Furthermore, the association between their expression and the clinicopathological features of this malignancy was also examined. Methods:The tissue chip contained 150 cases endometrial cancer examples, 54 atypical and 84 normal endometrial examples were also contained.Immunohistochemistry SP method was applied to evaluate the expression of IGF-1 and COX-2 in the tissue chip. Results:(1)The expression of IGF-1 and COX-2 in endometrial cancer were significantly higher than that in the normal examples(P<0.05).(2)The expression of IGF-1 was positively associated with myometrial invasion(P<0.05).No association was found between the expression of IGF-1 and histological grade, FIGO phase and lymph node metastasis. The expression of COX-2 was found positively associated with histological grade(P<0.05).No association was found between the expression of COX-2 and myometrial invasion, FIGO phase and lymph node metastasis.(3)The effects of IGF-1 and COX-2 on the development of endometrial cancer were synergistic(P<0.05). The expression of them was positively associated. Conclusion:With the tissue chip to evaluate the expression of IGF-1 and COX-2 in endometrial cancer ,help to earlier diagnosis of endometrial carcinoma and its pre-cancerous,comprehensive evaluation its biologic behavior, and predict invasion of biology indexes, to judge the degree malignant to have the important meaning.

    Three-dimensional Finite Element Analysis for the Influence of Micro-implant Diameter on Micro-implant and Surface of Mandibular

    Objective: To analyze the stress distribution at micro-implant-bone interface and the displacement of micro-implant based on different endosteal section diameters, and for choosing orthodontic anchorage micro-implant and optimize design of micro-implant. Methods: The finite element models of micro-implant with different endosteal section diameters and mandible were established by Pro/E; the elements were divided by Hypermesh and the results were calculated by Ansysl0.0. A force of 1.96 N was applied on the top of the implant parallel to the mandible surface to analyze the differences of the micro-implant displacement and that of the force on the micro-implant-bone interface. Results: With the increasing of endosteal section diameter, both the maximum Von-mises stress on micro-implant-bone interface and the maximum displacement in micro-implant decreased. The maximum Von-mises stress focused on the neck and tip of micro-implant. The Von-mises stress of micro-implant on z axis is the smallest, and the stress curved is the flattest at the endosteal section diameter of 1.5mm than that of other sizes. The tendence of the contour map area for mandible surface stress is 1.7mm>1.6 mm>1.4 mm>1.5 mm .Conclusion: The influence of the micro-implant diameter in-bone on the micro-implant is significant. The damage on the mandible and the displacement of the micro-implant are both the smallest in the 1.5mm case.

    The effect of Emodin on the human renal tubular epithelial to mesenchymal transdifferentiation induced by human albumin

    Objective To explore the effect of emodin on the transdifferentiation of human renal tubular epithelial cell to mesenchymal induced by human albumin. Methods Human kidney proximal tubular epithelial cell line (HK-2) was stimulated by human albumin(5mg/ml)in vitro. Morphological changes of the HK-2 cells were observed by an inverted phase contrast microscope. The expressions of α-SMA, E-cadherin, FNmRNA were detected by reverse transcription polymerase chain reaction(RT-PCR). Results Albumin up regulated α-SMA, FN mRNA expression in HK-2 cells in a time-dependent manner and E-cadherin mRNA was down-regulated. Emodin repressed the effect of albumin on them when using a certain concertration emodin. Conclusion Emodin inhibit the transdifferentiation of human renal tubular epithelial cell to mesenchymal induced by human albumin., slow down the progression of renal interstitial fibrosis.

    Outcomes in Patients with Type 2 Diabetes Mellitus Undergoing Percutaneous Coronary Intervention

    Objective:To compare outcomes of the diabetic and nondiabetic patients with coronary artery disease (CAD) undergoing successful percutaneous coronary intervention (PCI) ,and to investigate the relationship between type 2 diabetes mellitus(T2DM)and outcomes of CAD. Methods: The 1356 patients undergoing successful PCI in our hospital enrolled in the study, were separated into diabetic group (DM, n=292) and nondiabetic group (NDM, n=1064). Using the SYNTAX Score to evaluate the outcomes of CAG, then all the patients were divided into high, medium and low three groups. They had been followed up within 12 months. The clinical data were recorded including age, gender, history of hypertension, family history , the biochemical indicators including blood lipids, fibrinogen, and the echocardiography parameters. The endpoints were main adverse cardiovascular and cerebrovascular events (MACCE), including all-cause deaths, myocardial infarction (MI), target vessel revascularization (TVR) and stroke. All clinical date were statistically analyzed and processed. Results: Compared with the NDM group, the levels of FIB, uric acid in DM group was higher, while the mean left ventricular ejection fraction (LVEF) was lower. The mean SYNTAX score in DM group was higher than that in NDM (P<0.01); from the low group to the high group of SYNTAX score, the proportion of DM were gradually increased (P<0.01). During the follow-up the proportion of MI, death and total MACCE in DM were all significantly higher (all P<0.01). The proportion of TVR in DM was higher, and the proportion of stroke was slightly lower, while there were no difference in two groups (all P>0.05). Conclusions: The CHD patients with T2DM had a higher proportion of MACCE. It suggested that clinicians need to emphasize the control of blood glucose and blood lipid in the secondary prevention of the CHD patients.

    [Abstract ] Objective To contrast the changes of serum calcium, phosphorus and iPTH levels through using of the low flux dialyzer with high flux dialyzer alternately or continually.To approach an effective and feasibility method preventing from hyperphosphataemia and high iPTH level. Methods To choose 40 MHD cases in our hospital who were used the Sureflux-130G low flux dialyzer more than three months. The patients were called the low flux hemodialysis group ( LFHD ), as their own control group. The patients 40 of casses were divided into high flux dialysis A and B two groups ( HFHD-A, HFHD-B ),20 cases in each group. HFHD-A group: to use of 130G and 150U dialyzer alternately; HFHD-B group: to use of 150U dialyzer continually. First, to detection serum BUN, Scr, Hb, Alb, Ca, P, iPTH levels before and after treating with 130G, 150U dialyzer single and contrast their solute reduce rate .Then to measure above mentioned observations after the therapy for 3 months and observe the relieve dugree of Skin pruritus Results After single use of 130G, 150U dialyzer, serum BUN, Scr, UA, Hb, Alb, Ca decrease rate, no significant difference ( P> 0.05 ); serum P, iPTH levels decrease rate showed significant difference ( P < 0.05 ), HFHD group was superior to LFHD group. After the therapy for 3 months, HFHD-A and HFHD-B groups serum P, iPTH levels decreased significantly comparing with LFHD group, showed significant difference ( P < 0.05 ), the serum P removal effect became more remarkable in HFHD-B group(P<0.01). With HFHD for 1 months, skin itching symptoms that have improved significantly, HFHD-A, HFHD-B two group skin itching relieving degree respectively: 45.5%, 66.7%; effective:72.7%,91.7%; invalid: 27.2%, 0.83%. 3 months after the show: effect: 63.6%, 83.3%; efficiency reached 100%. Skin itching relieving degree of group HFHD-B was significantly higher than that of HFHD-A group. Conclusion It can reduce the serum phosphorus, iPTH levels effectively after use high flux dialyzer alternately or continually. It was important clinical significance to remission secondary hyperparathyroidism, reduce cardio-cerebro-vascular complications decrease mortality in MHD patients.

    Evaluation of The Biliary Ranson Score in Predicting Severity and Prognoses of Gallstone Acute Pancreatitis

    Objective: To evaluate the ability of the Biliary Ranson score in predicting the severity of gallstone acute pancreatitis and its prognosis by comparison with APACHE II score and Ranson score. Methods: 328 patients of gallstone pancreatitis admitted into Wuxi People’s Hospital from January 2005 to December 2010 were studied retrospectively. The APACHEⅡ score were calculated in the first 24 hours after admission, so was the Ranson and the Biliary Ranson score within the 48 hours after admission. Receiver operating characteristic curve analysis was used to evaluate the ability of Biliary Ranson score and other scoring systems in predicting the severity of gallstone pancreatitis and the occurrence of pancreatic necrosis, mortality and organ failure. Results: Among 328 patients,severe acute pancreatitis(SAP) was identified in 72 patients and mild acute pancreatitis(MAP) in 256 patients. The gender and age between MAP and SAP were of no statistical difference. The scores were correlated significantly between every two systems, respectively. The Biliary Ranson score is more useful in predicting SAP than APACHE II score and Ranson score, and can perform better in predicting pancreatic necrosis in the cases of SAP compared to APACHE II score. The cutoff value of Biliary Ranson score in predicting SAP was 2 in which the sensitivity, specificity, positive predictive value(PPV), negative predictive value(NPV) were 81.94%, 75.78%, 48.8% and 93.7%, respectively. Conclusions: The Biliary Ranson score is a more valuable score than other scores in predicting the severity of gallstone pancreatitis.

    Clinical investigation of recurrent ovarian epithelial cancer in Ⅲ stage

    Objective:To discuss the possible causes of recurrent ovarian epithelial cancer in Ⅲ stage Methods:A total of 85 ovarian epithelial cancer patients in Ⅲ stage were allocated to recurrent group (31patients) and non-recurrent group (54 patients). CA-7000 automated coagulation analyzer was used to examine the level of plasma fibrinogen (FIB), plasma prothrombin time(PT), thrombin time(TT), activated partial thromboplastin time(APTT), PLT was measured by Sysmex KX-21; CA125 was measured by electro-chemiluminescence, volume and quality of ascites, FIGO substages, histological type and grades, size of residual tumor during operation, pelvic or abdominal lymph node resectable or not and CA125 level of plasma 2 months after operation. Univariate analysis and multivariate logistic regression analysis were used to differentiate between recurrence and non-recurrence patients. Result:In univariate analysis, there was significant difference in the level of PT and TT, the rate of positive peritoneal cytology, the rate of ≥2cm residual tumor, the rate of lymph node unresected and the rate of CA125 not declined to normal 2 months later after operation between patients of recurrence and non-recurrence(P<0.05). Multivariate analysis showed that ≥2cm residual tumor, the rate of positive peritoneal cytology, the rate of lymph node unresected and CA125 not declined to normal 2 months later after operation were important dangerous factors for recurrence of patients in Ⅲ stage(P<0.05). conclusion:In patients of ovarian epithelial cancer in Ⅲ stage, residual tumor, peritoneal cytology, lymph node resected or not and CA125 level 2 months after operation were important prognostic factors of recurrence.

    The expression of apoptotic factors and collagen type I & III by recombinant adenovirus Ad-hBNP in rats’ hearts with chronic heart failure

    Objective: To observe the expression of apoptotic factors and collagen type I & III by recombinant adenovirus Ad-hBNP in rats’ hearts with chronic heart failure. Methods: Thirty CHF rats defined by echocardiography at 12 weeks post abdominal aortic constriction were randomly divided into Ad-hBNP group (2.5×1010 VP/mlNS Ad-hBNP 1 ml/weekx4, n=14), Ad-Track group (n=8), placebo group (NS, n=8), 10 sham-operated rats served as control group. After 4 weeks treatment, the mRNA expression of Bcl-2, Bax, Caspase-3 and collagen type I & III were detected by RT-PCR. Results: The mRNA expression of Bcl-2 and Caspase-3 were significantly decreased in the Ad-hBNP group [2.70±1.86, 0.75±0.30] than those in the Ad-Track group[(0. 49±0.02, P<0.01), (0.43±0.26, P<0.05)] and in the NS group [(0.95±0.17, P<0.05), (0.37±0.16, P<0.05)]. The mRNA expression of Bax in the Ad-hBNP group [0.86±0.17] was significantly increased than the sham group [0.70±0.07,P<0.05], but was decreased than the Ad-Track group and the NS group. The mRNA expression of collagen type I & III were also decreased in the Ad-hBNP group than the the Ad-hBNP group. Conclusion:Exogenous hBNP affected the mRNA expression of apoptosis factors in the rats’ hearts with chronic heart failure , also reduced the mRNA expression of collagen type I & III, and maybe attenuated remodeling in CHF rats.

    Reversal mechanism of Tetradrine on human pancreatic carcinoma multi-drug resistance cell line SW1990-GEM Abstract Objective To investigate the reversal effects of Tetradrine (Tet)on human pancreatic carcinoma multidrug resistance cell line SW1990-GEM and the mechanism involved in this reversal. Methods Immunocytochemistry was used to test the expression of P-glycoprotein(P-gP) coded by the multidrug resistance-1 gene of SW1990-GEM cells. The inhibitory effects of Tet on the proliferation of SW1990-GEM cells were evaluated by MTT assay, IC50 and reversal fold were computed. The effects of noncytotoxicity of TET on Rhodamin123 accumulation in SW1990-GEM cells was analyzed by Flow Cytometry. The effects of Tet on the expression of P-gP was detected by Western Blot. Results The expression of P-gP in SW1990-GEM cells were higher than that of SW1990 cells. Noncytotoxicity of Tet possessed reversal effects on SW1990-GEM cells. Tet increased significantly intracellular chemotherapeutical drug accumulation in SW1990-GEM cells. The resistance index of SW1990-GEM cells to Gem was 217.91, when added 1.5 mg/L Tet, the resistance index was 24.32. The reversal folds was 8.96. Tet did not diversify the expression level of P-gp in SW1990-GEM cells. Conclusion Tet significantly reversed MDR of SW1990-GEM cells to enhance accumulation of chemotherapeutical drug in cells and increase its antitumor function.This mechanism seems Tet directly inhibited transport function of P-gp, but it did not alter the protein expression level of P-gp.

    Evaluation of MRI in the Circumferential Resection Margin of Rectal Cancer

    Objective: To explore the reliability of MRI in the assessment of whether rectal cancer patients have invasive carcinoma on circumferential resection margin (CRM). Methods: Examine 7 corpses partial sectional specimens (specimen group), 10 healthy volunteers (compare group) and 49 rectal cancer patients (patient group) using MRI. Then dissect the corpse specimen and perform total mesorectal excision to the patient group. Compare the MRI result of the compare group and the specimen group, and measure the distance between mesorectal soft tissue nodules, irregular streaks shadow and mesorectal fascia of patient group MRI result. It is CRM (+) when this distance > 5mm, and it is CRM (-) if the distance < 5mm, compared with pathological findings. Result: The mesorectum fascia of both specimen group and patient group is shown on both T1WI and T2WI as a low-signal-intensity linear structure, lymph node, blood vessels, and lymphatic vessels are all shown. The MRI result is consistent with the anatomy result. Preoperative pathology results in patient group showed 22 positive CRMs, 27 negative CRMs. CRM judgment by MRI was consistent with the pathology results in 45(91.8%) of 49 patients.4 cases were misjudged: 2 patients with false positive results and 2 patients with false negative result. Conclusion: MRI is a relatively reliable method in evaluating the status of CRM.

    Abstract Objective:To observe the effectiveness of arsenic trioxide(AS2O3)associated with all-trans retinoic acid (ATRA )and chemotherapy in treating acute promyelocytic leukemia(APL).Method:69 cases of APL was provided randomly two groups,37 cases in treating group with AS2O3+ATRA+chemotherapy,32 cases in control group with ATRA+ chemotherapy.To observe the effectiveness and side effect. Result:The first induce complete remission(CR)is 96.6% in the treating group,CR is 87.5% in the control group. The bad reaction related to the treatment no significant difference in two groups. Conclusion:Arsenic trioxide(AS2O3)associated with ATRA and chemotherapy had highter curative effect ,The combination is security.

    Abstract Objective To evaluate whether the way of intubation by intubated laryngeal mask airway (ILMA) and stylet optical system (SOS) could take advantage of intubation by macintosh laryngeal scope (MAC) in reducing cardiovascular response.Methods Sixty undergoing orbital plastic surgery patients, ASA gradeⅠor Ⅱ, airway evalutated normal, were randomly divided into three groups: Macintosh group (MAC, n=20), intubated laryngeal mask airway group (ILMA, n=20) and stylet optical system group (SOS, n=20), and all the patients of three groups received intubation by MAC, ILMA, SOS respectively after induction intravenously. Record the patients’ IBP, HR and BIS at time point 5minutes after arriving (baseline value, TBL), immediately before intubation (TBI), immediately after intubation (TAI), 1-5min after intubation (T1-T5); The blood samples of left radial artery were taken for determination of serum NE concerntration at the moment before intubation (baseline value, TBL), 1 min after intubation (T1), 3 min after intubation (T3). Results Inter-group comparison: SBP, DBP and HR of group SOS at time point T1 were lower than that of group MAC (P<0.05) and group ILMA, but no significant difference was found between group ILMA and group MAC. There is also no significant difference of plasma NE at time point TBI (P>0.05), but at time point T1 AND T3 Plasma NE concentration of group SOS is the lowest among the three groups.. Conclusion Endotracheal intubation could cause cardiovascular response compared to intubation by Macintosh laryngeal scope, intubation by ILMA could not decrease the level of cardiovascular, but SOS could decrease the cardiovascular response.     [Abstract] Objective:To investigate the effect of high sensitivity C reactive protein(hsCRP) and matrix metalloproteinase 9(MMP-9) on the patients with unstable angina pectoris treated with Atorvastatin. Methods:106 patients with UA were divided into 2 groups,50 patients in the treatment group were treated with Atorvastatin 40mg every day for therthy days,in addition to the conventional therapy. 56 patients in the control group were treated with the conventional therapy.The detection was performed to test the hsCRP and MMP-9 in two groups.Results:① Two groups before treatment do not have difference on total cholesterol(TC),low density lipoprotein- cholesterol(LDL-C),high density lipoprotein- cholesterol(HDL-C).② The treatment group after treatment have a significant difference on TC,LDL-C,HDL-C(P<0.05).③ To compare 2 groups before treatment and the control group after treatment, hsCRP and MMP-9 have no difference.④ HsCRP and MMP-9 in the treatment group after treatment decreased significantly(P<0.05,P<0.01).Conclusion: HsCRP and MMP-9 in patients with unstable angina pectoris treated with Atorvastatin have a significant decrease.The data suggested that Atorvastatin can inhibite inflammation,stable plaque and prevent fracture of plaque in addition to regulate lipid.

    A Clinical Study on the Lactate Clearance Rate Assessing Prognosis of Patients with Acute Severe Alcoholic Intoxication

    Objective The present study attempts to investigate the correlation between the prognosis and the lactate clearance rate in patients with acute severe alcoholic intoxication. Methods Eighty patients suffering from acute severe alcoholic intoxication with initial serum lactate levels after admission to ICU greater than 2.0 mmol/L underwent detection of serum lactate six hours later to calculate the lactate clearance. The APACHE II score and mortality were recorded. The eighty patients fell under survival group (n=76) and death group (n=4), or high lactate clearance rate group (n=68) and low lactate clearance rate group (n=12). The correlation of lactate clearance rate with prognosis was analyzed. Results After the experiment no significant differences in APACHE II score and the initial blood concentration of lactate were found (P>0.05). The lactate clearance rate of the survival group was (17.24±8.08)%, which was significantly higher than that of the death group [(0.85±4.45)%, P <0.01]. The mortality of the high lactate clearance rate group was 1.47%, which was significantly lower than that of the low lactate clearance rate group (25%, P <0.001). Conclusion The lactate clearance rate can be used for early evaluation of the prognosis of the patient with acute severe alcoholic intoxication.

    Detecting the expression and significance of hCTR1 in cervical carcinoma and CIN by using tissue microarray

    Abstract:Purpose To investigate the expression and clinical pathological significance of hCTR1 in the cervical carcinoma and cervical intraepithelial neoplasia. Methods Using tissue microarray and immunohistochemical staining to detect the expression of hCTR1 in 52 cases of cervical carcinoma, 94 cases of CIN and 23 cases of normal cervical squamous epithelia. Results ①The manual tissue microarray was satisfied in IHC staining,and it satisfied the requirement of our studying multiple samples,the detetectable rate was 89.4%.②The positive retas of hCTR1 in SCC(69.2%) and in the level CIN(CIN 2-3, 73.1%) were significantly higher than those of normal cervical squamous epithelium(17.4%) and CIN 1(29.6%) (P<0.05). Other groups are no significant different(P>0.05). ③No significant relationships were found between the expression of hCTR1 and clinical pathological features. Conclusions Using manual tissue microarray made all the cases under the same experiment standard, could minimize the experimental error. hCTR1 expression rates became higher and higher from normal cervical squamous epithelia, CIN to cervical squamous cell carcinoma.hCTR1 regulates the cisplatin concentration in tumor cells so that it may be used as a potential biomarker of cervical carcinoma.

    Study on the disorder of coagulation in the patients with chronic heart failure

    Objective:Study on the function and condition of the vascular endothelial system and coagulation system in the patients with the chronic heart failure(CHF), assay the mechanisms of the dysfunction. Methods:Using the IL ACL-9000 blood coagulation instrument assay the von Willebrand Factor antigen(vWF:Ag). The TF:A was measured by chromogenic substrate method. The TAT was measured by ELISA. Results:Compared the control with the patients group, vWF:Ag, TF:A and TAT were increased significantly in the patients group(P<0.01). vWF:Ag and TF:A in the HF patients with NYHA Class Ⅱ were remarkably higher than the NYHA Class Ⅰ(P<0.01). vWF:Ag, TF:A and TAT in the HF patients with NYHA Class Ⅲ were remarkably higher than the NYHA Class Ⅱ(P<0.01). There was no relationship between the vWF:Ag and the TF:A, the TAT(r=0.2128, r=0.4003, P>0.05), and the strong positive relationship between the TF:A and the TAT(r=0.6946,P<0.01). Conclusion:There was severe vascular endothelial cells damage in the patients with HF, and the endothelial injury caused the coagulation system to activate, and brought the worse thrombosis risk. The level of the vWF:Ag, TAT and TF:Ag were closely connected with heart failure progress in CHF patients.

    The clinical research of umbilic cord blood stem cell transplantation in non-posthepatitic cirrhosis

    【Abstract】 Objective: To evaluate and assess the efficacy of umbilic cord blood stem cell transplantation through hepatic artery in the therapy of patients with non-posthepatitic cirrhosis. Methods: 10-15ml umbilic cord blood stem cells were transplanted into the liver through hepatic artery in 12 patients with non-posthepatitic cirrhosis. Alanine aminotransferase(ALT), albumin(ALB), total bilirubin(TB) and fibrinogen(FIB) were observed at pre-transplantation,1 week ,1 month and 3 months after transplantation were measured and analyzed. Results: All patients were transplanted successfully. The symptoms such as anorexia, fatigue,and distension/ascites begun improved at 1 week post-transplantation and showed significant improvement at 3 months post-transplantation (P<0.05).ALT increased at 1 week post-transplantation compared with pre-transplantation (P<0.01),while ALB decreased slightly at 1 week after transplantation(P<0.05), TB showed insignificant increase after transplantation(P>0.05), while FIB decreased insignificantly at the same time(P>0.05).All variable resumed to pre-transplantation level at 1month post-transplantation(P>0.05).There was significant improvement 3 months after transplantation(P<0.05),but the level of ALB and FIB were still under the lower limit. No severe effect and serious complication occurred. No occupying lesions was detected in liver by CT or B-ulreasound at 3 month post-transplantation. Conclusion: The transplantation of umbilic cord blood stem cell through hepatic artery in therapy of patients with non-posthepatitic cirrhosis was a safe and recommendable method with creditable short-term efficacy,but still need further research.

    Abstract Objective: To introduce an innovative operation style of performing microscope-assisted fasciculated ligation of the spermatic vein at the annulus inguinalis externalis. Methods: 133 cases of microscope-assisted fasciculated ligation of the spermatic vein at the annulus inguinalis externalis were retrospectively analyzed. The current operation style and the limitations were discussed. Results: Microscope-assisted fasciculated ligation of the spermatic vein at the annulus inguinalis externalis had relatively less surgical injury and complication, better postoperative recovery in patients with higher improvement rate of postoperative semen. Conclusions: Compared to the standard microscope-assisted ligation of the spermatic vein at the annulus inguinalis externalis, microscope-assisted fasciculated ligation of the spermatic vein could significantly improve the operative efficiency, shorten the operative time. It would be suitable for promotion in the broad masses of grass-roots hospitals.

    Abstract:Objective To use the noninvasive check means by Multi-slice Spiral CT Angiography to evaluate the value and clinical meaning in adult’s coronary artery anatomic abnomalies.Methods: MSCTA and clinical data of 760 patients were trospectively analysed.Results: Congenital Anatomic Variation of Coronary Artery 202 patients (26.58%, 202/760) were found by MSCTA among 760 patients. Take CAG test result as the "gold standard", the sensitivity of coronary artery anatomic abnomalies in MSCTA examination = 96.47%, specificity = 97.44%. Positive predictive value = 96.47%, negative predictive value = 97.44%.Conclusion: MSCT in the evaluation of anomalous coronary artery anatomy with high specificity, is a noninvasive and reliable evaluation of anomalous coronary anatomy in the first choice of imaging examination method.

    【Abstract】Purpose: Research how does hypothyroidism affect the metabolic indicators of pregnant women with gestational diabetes and the pregnancy outcome. Method: Choose 98 cases of pregnant women with gestational diabetes who made an examination and delivered between Jan. 2012 to Apr. 2012 in Tianjin Central Obstetrics and Gynecology Hospital. After the pregnant women of the third trimester came to hospital waiting for the parturition, fasting determine triglyceride(TC), cholesterol (TG), high density lipoprotein (HDL - C), low density lipoprotein (LDL - C), thyroid stimulating hormone (TSH), creatinine, urea nitrogen, uric acid, cystatin C, neonatal blood glucose testing blood glucose after childbirth. Grouped compare these cases according to TSH 0.3-3.0 MIU/ L. In one case, TSH<0.3 MIU/ L, and this is the hyperthyroidism group. In 62 cases, TSH 0.3-3.0 MIU/ L, and this is the normal contrast group. In 35 cases, TSH>3.0 MIU/ L, and this is the hypothyroidism group. Because there is only one case in the hyperthyroidism group, no statistical significance shows here. Just compare the clinical metabolic indicators and the pregnancy outcomes, including the incidence of gestational hypertension, placental abruption, premature birth, neonatal asphyxia, neonatal hypoglycemia, and neonatal birth weight, between the hypothyroidism group and the normal contrast group. Result: The averages of cholesterol, LDL, creatinine in the hypothyroidism group (TSH>3) are higher than which are in the normal contrast group (TSH 0.3-3), and the distinction has statistical significance. All the other metabolic indicators do not have statistical significance. Comparing the pregnancy outcomes of two groups, the incidences of gestational hypertension and premature birth in the hypothyroidism group are higher than which are in the normal contrast group, and the distinction has statistical significance. And comparing the incidences of premature rupture of membranes, placental abruption, postpartum hemorrhage, neonatal hypoglycemia, neonatal asphyxia between these two groups, the distinction do not have statistical significance, while the incidences of premature rupture of membranes, and postpartum hemorrhage are obviously higher in the hypothyroidism group. Conclusion: Hypothyroidism may aggravate the incidences of the metabolic disorders and adverse pregnancy outcomes of the pregnant women with GDM. It is recommended that pregnant women with GDM should screen the thyroid function, and make timely treatment to thyroid function abnormality.

Method 76 cases were randomly divided into application viscera abortion which only used mifepristone and misoprostol get medicine flow (control group) and prepared with premarin medicine flow (the observation group), two groups of patients medicine flow success rate, blood loss, etc are compared. Result The observation group operation flow rate, traditional medicine with reduced blood loss was better than control. Conclusion the compatibility with mifepristone influences misoprostol treatment can speed up the natural emptying viscera miscarriage embryo organization and shorten the time abortion completely after miscarriage bleeding time.
